1. miR‐92a‐3p promotes breast cancer proliferation by regulating the KLF2/BIRC5 axis

Abstract

Abstract Background Breast cancer remains the most common malignancy in females around the world. Recently, a growing number of studies have focused on gene dysregulation. In our previous study, Krüppel‐like factors (KLFs) were found to play essential roles in breast cancer development, among which KLF2 could function as a tumor suppressor. Nevertheless, the underlying molecular mechanism remains unclear. Methods miR‐92a‐3p was identified as the upstream regulator of KLF2 by starBase v.3.0. The regulation of KLF2 by miR‐92a‐3p was verified by a series of in vitro and in vivo assays. Further exploration revealed that Baculoviral IAP Repeat Containing 5 (BIRC5) was the target of KLF2. ChIP assay, dual‐luciferase reporter analysis, quantitative real‐time PCR, and western blot were performed for verification. Results miR‐92a‐3p functioned as a tumor promoter by inhibiting KLF2 by binding to its 3'‐untranslated region (3'‐UTR). In addition, KLF2 could transcriptionally suppress the expression of BIRC5. Conclusion Collectively, our results uncovered the miR‐92a‐3p/KLF2/BIRC5 axis in breast cancer and provided a potential mechanism for breast cancer development, which may serve as promising strategies for breast cancer therapy.

Abstract

Abstract InP quantum dots (QDs) are a promising and environment‐friendly alternative to Cd‐based QDs for in vitro diagnostics and bioimaging applications. However, their poor fluorescence and stability severely limit their biological applications. Herein, we synthesize bright (∼100%) and stable InP‐based core/shell QDs by using cost‐effective and low‐toxic phosphorus source, and then aqueous InP QDs are prepared with quantum yield over 80% by shell engineering. The immunoassay of alpha‐fetoprotein can be detected in the widest analytical range of 1–1000 ng ml−1 and the limit of detection of 0.58 ng ml−1 by using those InP QDs‐based fluorescent probes, making it the best‐performing heavy metal‐free detection reported so far, comparable to state‐of‐the‐art Cd‐QDs‐based probes. Furthermore, the high‐quality aqueous InP QDs exhibit excellent performance in specific labeling of liver cancer cells and in vivo tumor‐targeted imaging of live mice. Overall, the present work demonstrates the great potential of novel high‐quality Cd‐free InP QDs in cancer diagnosis and image‐guided surgery.

Abstract

Owing to early diagnosis and rapid development of treatments for cancers, the five-year survival rate of all cancer types has markedly improved worldwide. Over time, however, there has been an increase in the number of cancer patients who develop coronary artery disease (CAD) due to different causes. First, many risk factors are shared between cancer and CAD. Second, inflammation and oxidative stress are common underlying pathogeneses in both disorders. Lastly, cancer therapy can result in endothelial injury, coronary artery spasm, and coagulation, thereby increasing the risk of CAD. As more cancer patients are being diagnosed with CAD, specialized cardiac care should be established to minimize the cardiovascular mortality of cancer survivors. Abstract

Yam is a common ‘medicine food homology’ vegetable in Asia, and its peel is often considered a food residue during processing or cooking. In this work, the effects of hydrogen peroxide modification on the dietary fibres (DFs) from Chinese yam peel (CYP) were investigated. The structural characteristics of soluble dietary fibre (SDF), insoluble dietary fibre (IDF), modified soluble dietary fibre (MSDF) and modified insoluble dietary fibre (MIDF) were analysed using Fourier‐transform infrared spectroscopy, X‐ray diffraction, differential scanning calorimetry, granularity analysis, scanning electron microscopy and GC‐MS (monosaccharide composition). As results, after modification with hydrogen peroxide, the sizes of the DFs were reduced and MIDF exposed more cellulose. Experiments on the physicochemical and functional properties of DFs showed that MSDF and MIDF obtained a better water holding capacity, oil absorption capacity, swelling capacity and absorption abilities with altered structures, which is of great importance in food processing and development.

Abstract

Light harvesting (LH) and carrier transport abilities of a photoactive layer, which are both crucial for optoelectronic devices such as solar cells and photodetectors (PDs), are typically hard to be synergistically improved. Taking perovskite as an example, a freeze-drying recrystallization method is used to construct porous films with improvements of both LH and carrier transport ability. During the freeze-drying casting process, the rapid solvent evaporation produces massive pores, the sizes of which can be adjusted to exploit the Mie scattering for enhancement of the LH ability. Meanwhile, owing to the strong iconicity, the interface between perovskite nanocrystals fused during recrystallization, which favors carrier transport. Subsequently, PDs based on these Mie porous and interface-fused films show a high on/off ratio of more than 104 and an external quantum efficiency value of 658 % under 9 V bias and 520 nm light irradiation.

Abstract

All-inorganic perovskites have high carrier mobility, long carrier diffusion length, excellent visible light absorption, and well overlapping with localized surface plasmon resonance (LSPR) of noble metal nanocrystals (NCs). The high-performance photodetectors can be constructed by means of the intrinsic outstanding photoelectric properties, especially plasma coupling. Here, for the first time, inorganic perovskite photodetectors are demonstrated with synergetic effect of preferred-orientation film and plasmonic with both high performance and solution process virtues, evidenced by 238% plasmonic enhancement factor and 106 on/off ratio. The CsPbBr3 and Au NC inks are assembled into high-quality films by centrifugal-casting and spin-coating, respectively, which lead to the low cost and solution-processed photodetectors. The remarkable near-field enhancement effect induced by the coupling between Au LSPR and CsPbBr3 photogenerated carriers is revealed by finite-difference time-domain simulations. The photodetector exhibits a light on/off ratio of more than 106 under 532 nm laser illumination of 4.65 mW cm−2. The photocurrent increases from 0.67 to 2.77 μA with centrifugal-casting. Moreover, the photocurrent rises from 245.6 to 831.1 μA with Au NCs plasma enhancement, leading to an enhancement factor of 238%, which is the most optimal report among the LSPR-enhanced photodetectors, to the best of our knowledge. The results of this study suggest that all-inorganic perovskites are promising semiconductors for high-performance solution-processed photodetectors, which can be further enhanced by Au plasmonic effect, and hence have huge potentials in optical communication, safety monitoring, and biological sensing.
