Abstract

Accumulating evidence indicates that oxymatrine may exert protective effects on the cardiovascular system. This study was designed to evaluate the antiarrhythmic effects as well as the electrophysiological properties of oxymatrine. The antiarrhythmic activity of oxymatrine was observed in a rat model of arrhythmia induced by coronary ligation. Action potential duration (APD), L-type calcium current (I(Ca-L) ), transient outward potassium current (I(to) ) and inward rectifier potassium current (I(K1)) in rat ventricular myocytes were recorded by utilizing the whole cell patch-clamp technique. The results showed that administration of oxymatrine significantly delayed the onset of ventricular arrhythmia, decreased the duration of ventricular arrhythmia and reduced the arrhythmia score of arrhythmic rats. The beneficial effects of oxymatrine may be related to the shortening of APD through reduction of I(Ca-L) , enhancement of I(to) and inhibition of I(K1).

Abstract

Counting of Escherichia coli DH5α-cell suspensions in PBS is performed using a microflow cytometer based on a photonic-microfluidic integrated device. Side-scattered light signals are used to count the E. coli cells. A detection efficiency of 92% is achieved when compared with the expected count from a hemocytometer. The detection efficiency is correlated to the ratio of sample to sheath flow rates. It is demonstrated that E. coli can be easily distinguished from beads of similar sizes (2-4 μm) as their scattering intensities are different.

Abstract

Interferon-gamma (IFN-gamma) is a pleiotropic cytokine that plays an important role in regulating cellular immune responses. Regulation of IFN-gamma expression is considered to be strictly controlled at the transcriptional level. Two single-nucleotide polymorphisms (SNPs) within the human IFN-gamma promoter (at positions -183 and -155) are considered to influence the promoter activity by altering the acting transcription factor-1 (AP-1) binding. We sought to assess the association between the SNPs of the IFN-gamma promoter and the host susceptibility to hepatitis B virus (HBV) infection, as well as its interaction with age and gender. No polymorphism at position-155 was detected in any of the participants, but a significant difference was found in the polymorphism at position -183 between the cases and controls (G/T and T/T vs. GG; P < 0.01, odds ratio (OR) = 4.50 (95% confidence interval (CI) = 2.23-9.09). A susceptibility analysis revealed a gradually increased trend of the OR value from the young to the old group (OR = 3.03, 4.17, and 5.56). Similarly, the association of the -183 polymorphism was markedly different in females (OR = 5.71). Our data suggest that the polymorphism at position -183 of the IFN-gamma gene promoter may be associated with susceptibility to HBV infection, and age and gender factors are coordinative risk factors., (Copyright 2005 Wiley-Liss, Inc.)
