Abstract The US Federal Government supports hundreds of watershed monitoring efforts from which solute fluxes can be calculated. Although instrumentation and methods vary between studies, the data collected and their motivating questions are remarkably similar. Nevertheless, little effort toward their compilation has previously been made. The MacroSheds project has developed a future‐friendly system for harmonizing daily time series of streamflow, precipitation, and solute chemistry from 169+ watersheds, and supplementing each with watershed attributes. Here, we describe the breadth of MacroSheds data, and detail the steps involved in rendering each data product. We provide recommendations for usage and discuss when other datasets might be more suitable. The MacroSheds dataset is an unprecedented resource for watershed science, and for hydrology, as a small‐watershed supplement to existing collections of streamflow predictors, like CAMELS and GAGES‐II. The MacroSheds platform includes a web dashboard for visualization and an R package for data access and analysis.

Cardiac catheterization remains the gold standard for the diagnosis and management of pediatric pulmonary hypertension (PH). There is lack of consensus regarding optimal anesthetic and airway regimen. This retrospective study describes the anesthetic/airway experience of our single center cohort of pediatric PH patients undergoing catheterization, in which obtaining hemodynamic data during spontaneous breathing is preferential. A total of 448 catheterizations were performed in 232 patients. Of the 379 cases that began with a natural airway, 274 (72%) completed the procedure without an invasive airway, 90 (24%) received a planned invasive airway, and 15 (4%) required an unplanned invasive airway. Median age was 3.4 years (interquartile range [IQR] 0.7-9.7); the majority were either Nice Classification Group 1 (48%) or Group 3 (42%). Vasoactive medications and cardiopulmonary resuscitation were required in 14 (3.7%) and eight (2.1%) cases, respectively; there was one death. Characteristics associated with use of an invasive airway included age <1 year, Group 3, congenital heart disease, trisomy 21, prematurity, bronchopulmonary dysplasia, WHO functional class III/IV, no PH therapy at time of case, preoperative respiratory support, and having had an intervention ( p  < 0.05). A composite predictor of age <1 year, Group 3, prematurity, and any preoperative respiratory support was significantly associated with unplanned airway escalation (26.7% vs. 6.9%, odds ratio: 4.9, confidence interval: 1.4-17.0). This approach appears safe, with serious adverse event rates similar to previous reports despite the predominant use of natural airways. Purpose: Body composition MRI captures the distribution of fat and lean tissues throughout the body, and provides valuable biomarkers of obesity, metabolic disease, and muscle disorders, as well as risk assessment. Highly reproducible protocols have been developed for 1.5T and 3T MRI. The purpose of this work was to demonstrate the feasibility and test-retest repeatability of MRI body composition profiling on a 0.55T whole-body system., Methods: Healthy adult volunteers were scanned on a whole-body 0.55T MRI system using the integrated body RF coil. Experiments were performed to refine parameter settings such as TEs, resolution, flip angle, bandwidth, acceleration, and oversampling factors. The final protocol was evaluated using a test-retest study with subject removal and replacement in 10 adult volunteers (5 M/5F, age 25-60, body mass index 20-30)., Results: Compared to 1.5T and 3T, the optimal flip angle at 0.55T was higher (15°), due to the shorter T1 times, and the optimal echo spacing was larger, due to smaller chemical shift between water and fat. Overall image quality was comparable to conventional field strengths, with no significant issues with fat/water swapping or inadequate SNR. Repeatability coefficient of visceral fat, subcutaneous fat, total thigh muscle volume, muscle fat infiltration, and liver fat were 11.8 cL (2.2%), 46.9 cL (1.9%), 14.6 cL (0.5%), 0.1 pp (2%), and 0.2 pp (5%), respectively (coefficient of variation in parenthesis)., Conclusions: We demonstrate that 0.55T body composition MRI is feasible and present optimized scan parameters. Background: Blenderized formula (BF) can be used in place of standard enteral formula and is composed of blenderized whole foods and purees. Benefits of using BF include reduced vomiting and retching, in part related to a higher viscosity. Commercially prepared BFs vary significantly in viscosity when comparing available products. Viscosity can be measured using a viscometer, although this requires expensive and sophisticated equipment. We proposed an alternative method to describe viscosity of BFs using the International Dysphagia Diet Standardisation Initiative (IDDSI) framework. This flow test provides a simple and objective method to describe liquid thickness and requires only a 10-mL luer tip syringe and stopwatch., Methods: Full-strength BFs were measured using the IDDSI flow test and compared with a standard enteral formula. BFs were then diluted until a slightly thick viscosity was achieved, which correlated to the viscosity of the reference formula., Results: IDDSI flow-test results indicated that undiluted BFs ranged in viscosity, from slightly thick to extremely thick, when measured at room temperature. The volume of water required to achieve a slightly thick viscosity ranged from 0 to 270 mL per serving, resulting in a wide variation in dilution of formula., Conclusion: The IDDSI flow test was simple to perform and can serve as a tool to determine the viscosity of BFs., (© 2020 American Society for Parenteral and Enteral Nutrition.)

Interferon inducible transmembrane proteins (IFITMs) are broad-spectrum antiviral factors. In cell culture the entry of many enveloped viruses, including orthomyxo-, flavi-, and filoviruses, is inhibited by IFITMs, though the mechanism(s) involved remain unclear and may vary between viruses. We demonstrate that Sindbis and Semliki Forest virus (SFV), which both use endocytosis and acid-induced membrane fusion in early endosomes to infect cells, are restricted by the early endosomal IFITM3. The late endosomal IFITM2 is less restrictive and the plasma membrane IFITM1 does not inhibit normal infection by either virus. IFITM3 inhibits release of the SFV capsid into the cytosol, without inhibiting binding, internalization, trafficking to endosomes or low pH-induced conformational changes in the envelope glycoprotein. Infection by SFV fusion at the cell surface was inhibited by IFITM1, but was equally inhibited by IFITM3. Furthermore, an IFITM3 mutant (Y20A) that is localized to the plasma membrane inhibited infection by cell surface fusion more potently than IFITM1. Together, these results indicate that IFITMs, in particular IFITM3, can restrict alphavirus infection by inhibiting viral fusion with cellular membranes. That IFITM3 can restrict SFV infection by fusion at the cell surface equivalently to IFITM1 suggests that IFITM3 has greater antiviral potency against SFV., (© 2016 The Authors. Background and Purpose: The phytocannabinoid Delta(9)-tetrahydrocannabivarin (Delta(9)-THCV) has been reported to exhibit a diverse pharmacology; here, we investigate functional effects of Delta(9)-THCV, extracted from Cannabis sativa, using electrophysiological techniques to define its mechanism of action in the CNS., Experimental Approach: Effects of Delta(9)-THCV and synthetic cannabinoid agents on inhibitory neurotransmission at interneurone-Purkinje cell (IN-PC) synapses were correlated with effects on spontaneous PC output using single-cell and multi-electrode array (MEA) electrophysiological recordings respectively, in mouse cerebellar brain slices in vitro., Key Results: The cannabinoid receptor agonist WIN 55,212-2 (WIN55) decreased miniature inhibitory postsynaptic current (mIPSC) frequency at IN-PC synapses. WIN55-induced inhibition was reversed by Delta(9)-THCV, and also by the CB(1) receptor antagonist AM251; Delta(9)-THCV or AM251 acted to increase mIPSC frequency beyond basal values. When applied alone, Delta(9)-THCV, AM251 or rimonabant increased mIPSC frequency. Pre-incubation with Delta(9)-THCV blocked WIN55-induced inhibition. In MEA recordings, WIN55 increased PC spike firing rate; Delta(9)-THCV and AM251 acted in the opposite direction to decrease spike firing. The effects of Delta(9)-THCV and WIN55 were attenuated by the GABA(A) receptor antagonist bicuculline methiodide., Conclusions and Implications: We show for the first time that Delta(9)-THCV acts as a functional CB(1) receptor antagonist in the CNS to modulate inhibitory neurotransmission at IN-PC synapses and spontaneous PC output. Delta(9)-THCV- and AM251-induced increases in mIPSC frequency beyond basal levels were consistent with basal CB(1) receptor activity. WIN55-induced increases in PC spike firing rate were consistent with synaptic disinhibition; whilst Delta(9)-THCV- and AM251-induced decreases in spike firing suggest a mechanism of PC inhibition.

Previous studies have shown that certain sulfated polysaccharides, such as dextran sulfate (DxS), can induce a rapid and sustained increase in the free intracellular calcium ion concentration ([Ca2+]i) of thymocytes, whereas they cannot induce a change in the [Ca2+]i of peripheral lymphocytes. This study examined this phenomenon in more detail and clearly demonstrated that only immature thymocytes can respond to DxS. Cortisone-resistant thymocytes failed to exhibit an increase in [Ca2+]i when exposed to DxS. Furthermore, analysis of [Ca2+]i changes at the single cell level using a flow cytometer demonstrated that only a subpopulation of immature thymocytes responded to DxS. Additional flow cytometry studies revealed that DxS and Con-A induce a [Ca2+]i response in different subpopulations of thymocytes, approximately 30% of thymocytes responding to Con-A and 15-20% to DxS. Thus, responsiveness to Con-A and DxS, in terms of increases in [Ca2+]i, may represent a novel marker for two different thymocyte subpopulations. Such metabolic markers could be used to complement the conventional serological methods currently employed to define stages in thymocyte differentiation.
