2. Microtensile Bond Strength of Lithium Disilicate to Zirconia with the CAD-on Technique

Abstract

Purpose Recently, a novel technique was introduced to combine lithium disilicate and zirconia into one restoration. The purpose of this study was to compare the microtensile bond strength of veneering ceramic to a zirconia core in two techniques: the e.max® CAD-on technique and the Press-on technique. Materials and Methods Group A was prepared by veneering sintered zirconia blocks (e.max® ZirCAD) with lithium disilicate blocks (e.max® CAD) using the CAD-on technique according to manufacturer's instructions. Group B was prepared by taking sintered e.max® ZirCAD blocks and veneering them with fluorapatite glass-ceramic (e.max® ZirPress) using the Press-on technique according to manufacturer's instructions. Each block was loaded in a dynamic cyclic loading machine. The blocks were then sectioned into 1 × 1 mm2 beams (n = 43) using a precision saw, thermocycled, and loaded in tension until failure on a universal testing machine. A mean and standard deviation were determined per group. Data were analyzed using an unpaired t-test (α = 0.05). Results The mean microtensile bond strengths were 44.0 ± 13.8 MPa for the CAD-on technique and 14.9 ± 8.8 MPa for the Press-on technique. Significant differences were found between the two groups (p = 2.7E−19). Conclusions The CAD-on technique (lithium disilicate/zirconia) resulted in greater microtensile bond strength than the Press-on technique (fluorapatite glass-ceramic/zirconia).

Abstract

Deferoxamine is a hydroxylamine which binds ferric ions to form a highly stable complex. Since iron is thought to be required at a critical stage for cell proliferation, we investigated the effect of deferoxamine on the proliferative activity of human leukaemic cell populations in vitro by means of 3 permanent cell lines, HL60, U937 and 8402. We found deferoxamine to be a potent inhibitor of DNA synthesis and proliferation of leukaemic cells, acting by accumulating treated cells at the early S phase of the cell cycle. Suppression of leukaemic proliferation was obtained at deferoxamine concentrations in the range usually achieved in the treatment of patients for iron overload. Deferoxamine might therefore warrant further investigation as a potentially useful agent for leukaemia chemotherapy.

Abstract

In the light of the experience gained studying various sets of muscarinic ligands related to a rigid model, flexible structures such as alkyltrimethylammonium salts, ethers and esters of choline have been prepared and their affinities (pD2) have been studied. The quantitative structure-activity relationships (QSAR) show that the pD2 value is strongly dependent on hydrophobic and steric parameters of the ligand substituent. Moreover, two indicator variables are considered to take ito account extra-activities connected with ligand special structural features. The overall QSAR confirm that the electrostatic attraction, the hydrophobic bonding and the polar interaction holding the ligand on the receptor are individually weak; however, these three forces collectively permit a more complete utilization of the receptor binding site.

Abstract

Two sets of substantial picolyl and furfuryltrimethylammonium salts have been synthesized and their affinities (expressed as pD2) have been studied with the aim of characterizing muscarinic receptor spaces to which the ligand structural features bind. The new data were combined with those relative to a set of substituted benzyltrimethylammonium salts previously studied to formulate an overall QSAR which showed that the pD2 value is strongly dependent on hydrophobic and steric parameters of the ring substituent. Moreover, two indicator variables were considered to take into account the extra-activity associated with special structural features of the heterocyclic system and the further increase in activity associated with a "cooperative interaction" between the lipophilic binding of the substituent and a polar interaction of the ring heteroatom. The results confirm our earlier predictions and offer a model for ligand binding which gives a consistent view of the topology of the ligand attachment points on the receptor.
