1. One in, one out: Generic circumscription within subtribe Manilkarinae (Sapotaceae)

Abstract

International audience; Previous phylogenetic studies have demonstrated that the Manilkarinae are a monophyletic subtribe if Northia is excluded. The subtribe consists of four genera: Faucherea, Labourdonnaisia, Labramia and Manilkara. However, the same phylogenetic studies also raised taxonomic issues concerning unclear generic delimitations and unresolved relationships. The current study's aims are: to resolve these taxonomic issues using a molecular phylogeny based on hundreds of nuclear markers sequenced from a representative sampling of taxa across the four genera; to find relevant morphological characters allowing the distinction of the clades retrieved with the phylogeny; and finally to understand the evolutionary history of the subtribe by conducting a divergence time estimation and ancestral state reconstructions. Our phylogeny shows a well-resolved backbone with four main lineages: the Labramia clade, the main clade of Manilkara, a clade in which all species of Labourdonnaisia and Faucherea are mixed, and a clade of three Pacific Manilkara species. The main clade of Manilkara is retrieved as sister to Labramia, and the Labourdonnaisia-Faucherea clade is clearly assessed as sister to the three Pacific Manilkara species. As a consequence, Faucherea is synonymized with Labourdonnaisia, and the three Pacific Manilkara are considered to be a separate genus, for which the name Abebaia is resurrected. We provide emended descriptions for Labourdonnaisia and Abebaia as well as the necessary new combinations. The ancestral state reconstruction of flower characters shows that ancestral Manilkarinae were characterized by a hexamerous corolla, well-developed dorsal appendages and staminodes, and a pubescent ovary. These character states have been retained in the main Manilkara clade, but surprisingly also in Abebaia, which appears as a cryptic genus. The lack of dorsal appendages and the reduction of staminodes observed in Labourdonnaisia appeared after the split from Abebaia. The increase in corolla merism observed mainly in the Mascarene Labourdonnaisia, which was used to separate it from Faucherea, appears to be a derived state, which evolved separately in a few species during the radiation of Labourdonnaisia on Madagascar and the Mascarenes. The glabrous ovary state observed in Labramia also constitutes a derived synapomorphic state in the genus.

Abstract

1. Urban environments are heterogeneous landscapes of social and environmental features, with important consequences for human–nature entanglements, such as that of mosquito-borne disease. 2. Investigations into this intra-urban heterogeneity in mosquito dynamics find conflicting results, likely due to the complex socio-ecological interactions and the importance of place-based context. Integrative research, which synthesizes multiple disciplines and epistemologies, can place ecological results into their social context to explore these place-based differences and reveal novel solutions for mosquito-borne disease management. 3. Here, we develop an integrative approach to understanding spatial patterns of mosquito burdens in urban systems by combining entomological surveys, semi-structured interviews and sketch maps. We highlight this approach using a mixed-method study conducted in Bengaluru, India, a rapidly urbanizing city with a high burden of mosquito-borne disease. 4. Although we found no evidence for a difference in mosquito abundance across an urban gradient, there were differences in individuals' everyday experiences with mosquitoes. These differences were mediated by how individuals moved through outdoor space and their vulnerability to hazards in these spaces. 5. This example of integrative research illustrates what can be gained from the inclusion of multiple epistemologies, particularly for research in urban systems.

Abstract

Due to the shortage of multi-organ donors, human pancreatic islet transplantation has now been extended to islets originating from obese subjects. In this study, our aim is to compare the respective sensitivity of human islets from lean vs obese donors to chronic high glucose or high palmitate.Human islets were isolated from pancreases harvested from brain-dead multi-organ donors. Islets were cultured during 72 hours in the presence of moderate (16.7 mmol/L) or high (28 mmoL/L) glucose concentrations, or glucose (5.6 mmoL/L) and palmitate (0.4 mmoL/L), before measurement of their response to glucose.We first observed a greater insulin response in islets from obese donors under both basal and high-glucose conditions, confirming their hyperresponsiveness to glucose. When islets from obese donors were cultured in the presence of moderate or high glucose concentrations, insulin response to glucose remained unchanged or was slightly reduced, as opposed to that observed in lean subjects. Moreover, culturing islets from obese donors with high palmitate also induced less reduction in insulin response to glucose than in lean subjects. This partial protection of obese islets is associated with less induction of inducible nitric oxide synthase in islets, together with a greater expression of the transcription factor forkhead box O1 (FOXO1).Our data suggest that in addition to an increased sensitivity to glucose, islets from obese subjects can be considered as more resistant to glucose and fatty acid excursions and are thus valuable candidates for transplantation.背景: 由于缺乏多器官供体, 人胰岛移植目前已衍生到肥胖者来源的胰岛移植。在这项研究中, 我们旨在比较瘦人和肥胖者的胰岛对慢性高糖或棕榈酸盐的敏感性。 方法: 从脑死亡的多器官捐赠者的胰腺中分离胰岛。胰岛在中等浓度(16.7 mmol/L)或高浓度(28 mmol/L)葡萄糖培养基中, 或是葡萄糖(5.6 mmol/L)和棕榈酸盐(0.4 mmol/L)存在下培养72h, 然后测定其对葡萄糖的反应。 结果: 我们首先观察到肥胖捐赠者的胰岛在基础和高糖条件下都有更大的胰岛素反应, 证实了其对葡萄糖的高反应性。但当来自肥胖捐赠者的胰岛在中高浓度葡萄糖存在的情况下继续培养时, 胰岛对葡萄糖的反应则保持不变或略有降低, 这与在瘦的捐赠者中观察到的相反。此外, 用高棕榈酸培养后的肥胖捐赠者胰岛也比瘦捐献者的对于葡萄糖的胰岛素反应减少得少。这种对肥胖者胰岛的部分保护作用, 与较少诱导一氧化氮合成酶有关, 以及转录因子FOXO1的较高表达相关。 结论: 我们的结果表明, 除了对葡萄糖的敏感性增加外, 肥胖者的胰岛可以被认为对葡萄糖和脂肪酸剧增更有抵抗力, 因此是有价值的移植候选者。.

Abstract

International audience; The European Academy of Allergy and Clinical Immunology (EAACI) supports three journals: Allergy, Pediatric Allergy and Immunology, and Clinical and Translational Allergy. EAACI's major goals include supporting the promotion of health, in which the prevention of allergy and asthma plays a critical role, and disseminating the knowledge of allergic disease to all stakeholders. In 2018, the remarkable progress in the identification of basic mechanisms of allergic and respiratory diseases as well as the translation of these findings into clinical practice were observed. Last year's highlights include publication of EAACI guidelines for allergen immunotherapy, many EAACI Position Papers covering important aspects for the specialty, better understanding of molecular and cellular mechanisms, identification of biomarkers for disease prediction and progress monitoring, novel prevention and intervention studies, elucidation of mechanisms of multimorbidities, introduction of new drugs to the clinics, recently completed phase three clinical studies, and publication of a large number of allergen immunotherapy studies and meta-analyses.

Abstract

International audience; Cancer is currently one of the deadliest diseases worldwide. Based on the high incidence of this disease, the side-effects associated with current chemotherapies and the appearance of drug resistance, considerable efforts have been directed towards the development of new anticancer drugs with new modes of action during. Metal-based compounds were shown to be particularly attractive candidates due to their metabolic mechanisms which differ substantially to organic drugs. Of special interest in this context are organometallic ruthenium(II) complexes of the type [Ru(η6-arene)(pta)Cl2] (arene = p-cymene, toluene, benzene, etc.; pta = 1,3,5-triaza-7-phosphaadamantane), abbreviated RAPTA. Complementary to chemotherapy, photoactivated chemotherapy (PACT) is a technique that has received increasing attention towards the development of treatment of numerous kinds of cancer. With this in mind, we have designed a photoactive RAPTA-type complex bearing azide ligands. The diazide complex, [Ru(η6-p-cymene)pta(N3)2], is inert in water but slowly releases the azide ligand on exposure to light. Consequently, the in vitro cytotoxicity of the complex in the dark as well as upon light exposure at 450 nm in human cervical carcinoma (HeLa) and non-cancerous retinal pigment epithelium (RPE-1) cells was investigated. While the cytotoxicity of the complex was found to be modest in the dark, an increase in toxicity upon light exposure was observed.

Abstract

International audience; Introduction Emicizumab (Hemlibra (R)) recently became available and requires an adaptation for managing bleeding, suspected bleeding and emergency or scheduled invasive procedures in haemophilia A patients with inhibitor. This implicates a multidisciplinary approach and redaction of recommendations for care that must be regularly adapted to the available data. Aim The following text aims to provide a guide for the management of people with haemophilia A with inhibitor treated with emicizumab in case of bleeding or invasives procedures. Methods The French network on inherited bleeding disorders (MHEMO), the French Reference Centre on Haemophilia (CRH), in collaboration with the French Working Group on Perioperative Haemostasis (GIHP) have been working together to make proposals for the management of these situations. Results Haemostatic treatment and other medications should be given stepwise, according to the severity and location of the bleeding or the risk of bleeding of the procedure as well as the haemostatic response obtained at each step in order to ensure an optimal benefit/risk ratio. Conclusion The lack of data means that it is only possible to issue proposals rather than recommendations.

Abstract

International audience; AIM: The worldwide increase in obesity and type 2 diabetes (T2D) represents a major health challenge. Chronically altered lipids induced by obesity further promote the development of T2D, and the accumulation of toxic lipid metabolites in serum and peripheral organs may contribute to the diabetic phenotype. METHODS: To better understand the complex metabolic pattern of lean and obese T2D and non-T2D individuals, we analysed the lipid profile of human serum, skeletal muscle and visceral adipose tissue of two cohorts by systematic mass spectrometry-based lipid analysis. RESULTS: Lipid homeostasis was strongly altered in a disease- and tissue-specific manner, allowing us to define T2D signatures associated with obesity from those that were obesity independent. Lipid changes encompassed lyso-, diacyl- and ether-phospholipids. Moreover, strong changes in sphingolipids included cytotoxic 1-deoxyceramide accumulation in a disease-specific manner in serum and visceral adipose tissue. The high amounts of non-canonical 1-deoxyceramide present in human adipose tissue most likely come from cell-autonomous synthesis because 1-deoxyceramide production increased upon differentiation to adipocytes in mouse cell culture experiments. CONCLUSION: Taken together, the observed lipidome changes in obesity and T2D will facilitate the identification of T2D patient subgroups and represent an important step towards personalized medicine in diabetes.

Abstract

International audience; Benthic foraminifera are single-celled eukaryotes that make a protective organic, agglutinated or calcareous test. Some agglutinated, single-chambered taxa, including Psammophaga Arnold, 1982, retain mineral particles in their cytoplasm, but the selective mechanism of accumulation is not clear. Here, we report the ability of a foraminiferal species to select and accumulate zircons and other heavy minerals in their cytoplasm. In particular, the use of Scanning Electron Microscope coupled with an Energy Dispersive X-ray microanalysis system (SEM–EDS) enabled a representative overview of the mineral diversity and showed that the analysed Psammophaga zirconia sp. nov. individuals contained dominantly crystals of zircon (51%), titanium oxides (27%), and ilmenite (11%) along with minor magnetite and other minerals. The studied specimens occur in the shallow central Adriatic Sea where the sediment has a content of zircon below 1% and of other heavy minerals below 4%. For that reason we hypothesize that: (i) P. zirconia may be able to chemically select minerals, specifically zircon and rutile; (ii) the chemical mechanism allowing the selection is based on electrostatic interaction, and it could work also for agglutinated foraminifera (whether for ingestion, like Xenophyophores, or incorporation in the test as in many other described taxa). In particular, this aptitude for high preferential uptake and differential ingestion or retention of zircon is reported here for the first time, together with the selection of other heavy minerals already described in members of the genus Psammophaga. They are generally counted among early foraminifera, constructing a morphologically simple test with a single chamber. Our molecular phylogenetic study confirms that P. zirconia is a new species, genetically distinctive from other Psammophaga, and occurs in the Adriatic as well as in the Black Sea.

Abstract

International audience; Patients with psoriatic arthritis (PsA) may not be optimally treated. The impact of the disease extends beyond skin and joint symptoms, impairing quality of life. This indicates that the adoption of a patient-focused approach to PsA management is necessary. An expert multidisciplinary working group was convened, with the objective of developing an informed perspective on current best practice and needs for the future management of PsA. Topics of discussion included the barriers to current best practice and calls to action for the improvement of three areas in PsA management: early and accurate diagnosis of PsA, management of disease progression and management of the impact of the condition on the patient. The working group agreed that, to make best use of the available of diagnostic tools, clinical care recommendations and effective treatments, there is a clear need for healthcare professionals from different disciplines to collaborate in the management of PsA. By facilitating appropriate and rapid referral, providing high quality information about PsA and its treatment to patients, and actively involving patients when choosing management plans and setting treatment goals, management of PsA can be improved. The perspective of the working group is presented here, with recommendations for the adoption of a multidisciplinary, patient-focused approach to the management of PsA.

International audience; The high throughput characterization of protein N-termini is becoming an emerging challenge in the proteomics and proteogenomics fields. The present study describes the free N-terminome analysis of human mitochondria-enriched samples using trimethoxyphenyl phosphonium (TMPP) labelling approaches. Owing to the extent of protein import and cleavage for mitochondrial proteins, determining the new N-termini generated after translocation/processing events for mitochondrial proteins is crucial to understand the transformation of precursors to mature proteins. The doublet N-terminal oriented proteomics (dN-TOP) strategy based on a double light/heavy TMPP labelling has been optimized in order to improve and automate the workflow for efficient, fast and reliable high throughput N-terminome analysis. A total of 2714 proteins were identified and 897 N-terminal peptides were characterized (424 N-α-acetylated and 473 TMPP-labelled peptides). These results allowed the precise identification of the N-terminus of 693 unique proteins corresponding to 26% of all identified proteins. Overall, 120 already annotated processing cleavage sites were confirmed while 302 new cleavage sites were characterized. The accumulation of experimental evidence of mature N-termini should allow increasing the knowledge of processing mechanisms and consequently also enhance cleavage sites prediction algorithms. Abstract

International audience; Socio-cognitive conflict has been defined as a situation of confrontation with a disagreeing other. Previous research suggests that individuals can regulate conflict in a relational way, namely by focusing on social comparison between relative levels of competences. Relational conflict regulation has been described as yielding particularly negative effects on social interactions and learning, but has been understudied. The present research addresses the question of the origin of relational conflict regulation by introducing a fundamental distinction between two types of regulation, one based on the affirmation of one's own point of view and the invalidation of the other's (i.e., " competitive " regulation), the other corresponding to the protection of self-competence via compliance (i.e., " protective " regulation). Three studies show that these modes of relational conflict regulation result from the endorsement of distinct performance goals, respectively performance-approach goals (trying to outperform others) and performance-avoidance goals (avoiding performing more poorly than others). Theoretical implications for the literature on both conflict regulation and achievement goals are discussed.

International audience; In this paper we study a combinatorial optimization problem issued from on-board networks in satellites. In this kind of networks the entering signals (inputs) should be routed to amplifiers (outputs). The connections are made via expensive switches with four links available. The paths connecting inputs to outputs should be link-disjoint. More formally, we call it $\plk-$network an undirected graph with $p+\lambda$ inputs, $p+k$ outputs and internal vertices of degree four. A $\plk-$network is valid if it is tolerant to a restricted number of faults in the network, i.e. if for any choice of at most $k$ faulty inputs and $\lambda$ faulty outputs, there exist $p$ edge-disjoint paths from the remaining inputs to the remaining outputs. In the special case $\lambda=0$, a $\plk-$network is already known as a selector. Our optimization problem consists of determining $N\plk$, the minimum number of nodes in a valid $\plk-$network. For this, we present validity certificates and a gluing lemma from which derive lower bounds for $N\plk$. We also provide constructions, and hence upper bounds, based on expanders. The problem is very sensitive to the order of $\lambda$ and $k$. For instance, when $\lambda$ and $k$ are small compared to $p$, the question reduces to avoid certain forbidden local configurations. For larger values of $\lambda$ and $k$, the problem is to find graphs with a good expansion property for small sets. This leads us to introduce a new parameter called $\alpha$-robustness. We use $\alpha$-robustness to generalize our constructions to higher order values of $k$ and $\lambda$. In many cases, we provide asymptotically tight bounds for $N\plk$.

Abstract

The XXL survey currently covers two 25 sq. deg. patches with XMM observations of ~10ks. We summarise the scientific results associated with the first release of the XXL data set, that occurred mid 2016. We review several arguments for increasing the survey depth to 40 ks during the next decade of XMM operations. X-ray (z1 cluster density.
