OBJECTIVE Methotrexate (MTX) can be used as an alternative for patients with Crohn's disease (CD) who are intolerant of thiopurine. This retrospective study aimed to provide some clues about MTX treatment in Chinese patients with CD. METHODS Medical records of 27 adult patients with CD who were treated with MTX between 2012 and 2017 at Renji Hospital were reviewed. MTX was administered at 15 mg or 20 mg intramuscularly once per week. The remission and response rates and adverse reactions of MTX were recorded and analyzed. RESULTS Thirteen (48.1%) of the patients achieved remission for more than 12 months, whereas four (14.8%) responded clinically. Eight (29.6%) patients discontinued MTX due to adverse events. The mean age of those who maintained remission was significantly younger than that of those who did not ([35.62 ± 10.99] years vs. [45.43 ± 11.93] years, P < 0.05). The pretreatment C-reactive protein (CRP) level was higher in the group who maintained remission than that in those who did not ([17.20 ± 17.26] mg/L vs. [6.98 ± 5.66] mg/L, P

BTBR mice exhibit several autistic-like behaviors and are currently used as a model for understanding mechanisms that may be responsible for the pathogenesis of autism. Ras/Raf/ERK1/2 signaling has been suggested to play an important role in neural development, learning, memory, and cognition. Two studies reported that a deletion of a locus on chromosome 16 containing the mitogen-activated protein kinase 3 (MAPK3) gene, which encodes ERK1, is associated with autism. In the present study, Ras/Raf/ERK1/2 signaling was found to be up-regulated in BTBR mice relative to matched control B6 mice, to further suggest involvement in the pathogenesis of autism. To further characterize the developmental pattern of Ras/Raf/ERK1/2 signaling, varying stages during development were sampled to reveal an up-regulation in newborn and 2-week old BTBR mice relative to age-matched B6 mice. By the age of 3-week, Ras/Raf/ERK1/2 signaling in the brain of BTBR mice was unaltered relative to B6 mice, with this trend maintained in 6-week samples. These results suggest that the alteration of Ras/Raf/ERK signaling in the early developmental stages in mice could contribute to the noted autistic phenotype. Furthermore, these findings support the value of BTBR mice to serve as a human analog for autistic etiological research and aid in a better understanding of the developmental mechanisms of autism.

The effects of aluminum (Al) and cadmium (Cd) single and combinational treatments on two barley varieties differing in Al tolerance were examined in terms of chemical forms of Al and Cd in the rhizosphere soil and their plant availability. Our results showed that acidic-soluble inorganic Al, humic acidic Al, and residue Cd were the predominant form(s) of Al and Cd in the rhizosphere soil of both barley varieties. The contents of different Al and Cd forms, especially for the exchangeable form, as well as Al and Cd contents in plants significantly increased under stress treatments. Meanwhile, stress treatments significantly reduced grain yield. In comparison with the single metal treatments, combinational stress increased the contents of exchangeable Al and Fe–Mn-oxide-bound Cd, but showed a decrease or no effect on the other forms. As a result, combinational stress led to a higher Al but a lower Cd accumulation in plants, and more reduction in grain yield. Increase in the contents of bioavailable Al and Cd forms in the stressed rhizosphere soil and in the concentrations of Al or Cd in plant parts was higher in the Al sensitive variety.

Memory performance, brain excitatory amino acid and acetylcholinesterase activity of chronically aluminum (Al) exposed mice in response to soy isoflavones (SI) treatment was investigated in the study. Forty eight mice were allotted randomly into a control group, an Al exposed group (100 mg/kg Al) and an Al exposed group treated with SI (100 mg/kg Al + 60 mg/kg SI) for 60 days. Chronic Al exposure significantly impaired long memory performance in mice as assessed using a passive avoidance task test (χ(2) analysis, p < 0.05). Interestingly, SI treatment markedly improved the memory performance score in the Al exposed mice. This improvement was associated with a total reversal of Al-induced increases in acetylcholinesterase activity in the cerebral cortex and hippocampus of mice. The Al exposure also led to significant decreases in brain levels of aspartic and glutamic acids, two excitatory amino acid neurotransmitters; whereas SI treatment partially reversed the decreased aspartic and glutamic acid contents in the hippocampus. The results suggest that SI can improve long memory performance in the Al exposed mice, possibly by modulating the metabolism of brain acetylcholine and amino acid neurotransmitters.

This experimental study is part of the research program related to the aerodynamic characteristics of impingement in a confined channel. Experimental research on better understanding flow structure in confined channel with impingement cooling is rarely found in open literature. The main purposes of this project are (1) gaining greater insight into the structure of the impingement jet flow and flow in the outlet hole; (2) understanding the effect of impingement hole arrangement build up on the flow structure and aerodynamic parameters within holes and channels with different aspect ratios. The test models are two confined channels with three staggered circular jet holes, and a single large size outlet hole placed downstream of the jet plate. The structure parameters of these orifices are different. In this paper, detailed flow field in the outlet hole was measured with a straight five-hole probe, and the discharge coefficient of outlet holes with different aspect ratios was also studied in each channel. Experimental data shows that the jet arrangement has little influence on the flow behavior of the outlet hole when the aspect ratio of the passage was 1, but it played an important role on the discharge coefficient. Distinctively different flow patterns were found in two models with the aspect ratios of 3 and 5, while the variation of the discharge coefficient showed a slight influence on impingement hole arrangement. © 2007 Wiley Periodicals, Inc. Breast cancer specimens from 600 women were assayed for estrogen receptors (ER) using an immunocytochemical assay (ICA) employing the monoclonal antiestrophilin antibody H222 Sp gamma. Results showed significant correlation with biochemical ER determinations as well as with tumor grade and menopausal status. In 449 cases, results of progesterone receptor assay by ICA using the monoclonal anti-PgR antibody KD 68, also correlated significantly with biochemical PgR measurements. The ERICA/PgRICA positivity was significantly more frequent in postmenopausal white women. Colloid carcinomas were most likely to be ERICA positive and PgRICA positive whereas medullary carcinomas were most often negative. In 47 patients with advanced mammary carcinoma, results of ERICA and PgRICA were more closely related to endocrine response than those of ER and PgR by dextran-coated charcoal assay (DCC). In 339 women with Stage I or Stage II breast cancer, ERICA was significantly associated with disease-free survival. Analysis by Cox's proportional hazard model, however, showed PgRICA to be the best predictor of survival and disease-free survival in 197 women at the same stages of disease. These data indicate that ICA is more predictive of prognosis than biochemical ER and PgR. The ease of ICA performance coupled with these results indicate that the method is an acceptable substitute for DCC in analyzing breast cancers for ER/PgR.
