The aim of this study was to clarify the role of blood in the early stage of development of endometriotic lesions by developing a syngeneic transplantation model using immunocompetent mice.Endometriotic lesions were induced in C57BL/6 mice by an intraperitoneal injection of endometrial fragments plus saline or endometrial fragments plus blood. Some endometrial fragments plus blood were injected with heparin, hirudin or tissue plasminogen activator (tPA). Endometriotic lesions on days 1, 3 and 5 were evaluated by gross and microscopic findings.The areas of endometriotic lesions in the blood group (6.4 ± 1.7 mmBlood accelerates the early stage of development of endometriotic lesions when endometrial fragments plus blood are injected. Blood property might be involved in early endometrial-peritoneal interactions.

DURABLE IMMUNE-MEDIATED PROTECTION FROM ACUTE LYMPHOBLASTIC LEUKEMIA (ALL) IS INDUCED BY CPG OLIGODEOXYNUCLEOTIDES (CPG ODN) IN A SYNGENEIC MOUSE MODEL Alix Seif;David Barrett;Valerie Brown;Stephan Grupp;Gregor Reid; Pediatric Blood & Cancer

We have studied the endocytosis of tritium-labelled and of non-radioactive agarose in mouse macrophages in vilro. The endocytosis was greatest and most rapid in syngeneic mouse serum and in human serum, reaching a plateau after 12 h of incubation. Ten per cent serum was the minimum concentration giving optimal ingestion. The endocytosis appeared to be regulated by mechanisms involving complement factors C3 and B. Different pretreatments of sera, inactivating or depleting C3 and B, resulted in 70–80% reduction of endocytosis, Preincubation of agarose in untreated serum increased the endocytosis of agarose in heat-inactivated serum three-fold, indicating that the essential factors were bound to agarose. Antibodies against C3 and B reduced endocytosis moderately hut significantly.

The requirement of thymus-derived cells for the immune response to trinitrophenylated syngeneic mouse red cells (TNP-MRC) was investigated. In three sets of experiments the following results were obtained: a) irradiated mice which were reconstituted with bone marrow cells alone showed a better anti-TNP response after injection with TNP-MRC than those reconstituted with both bone marrow cells and thymocytes. b) Anti-thymocyte serum augmented the mouse anti-TNP response to TNP-MRC. c) Nude thymusless mice showed a better anti-TNP response to TNP-MRC than their normal littermate controls. These results indicate that the anti-TNP response of mice to TNP-MRC does not require thymus-derived helper cells. Moreover, thymus-derived cells have a suppressive effect on the anti-TNP response.

Using a syngeneic mouse sarcoma of recent origin and demonstrable immunogenicity, the effects of removal or retention of regional lymph nodes upon development of local recurrences and distant metastases were studied. Irrespective of extent of operation, early tumor removal prevented development of distant metastases while delayed tumor excision was followed by distant metastases in about half the animals so treated. Significant and comparable incidences of local recurrence were noted after early operations with deliberate postoperative wound seeding with tumor and after late operations without wound seeding, whereas early operation without wound seeding resulted in a negligible incidence of local recurrence.

The purpose of the study was to determine the effect of the inoculation of syngeneic mouse mammary tumor cells on the incidence of spontaneous tumors and mortality in a group of RIII female mice which normally have a high incidence of primary breast tumors. Animals were inoculated with either untreated, heat-treated or irradiated tumor cells or a combination of cells. One group received no injections and served as a control. The incidence of tumor development and mortality was recorded for each animal used in the study. Significant delays in tumor onset and subsequent death of the animals were noted in those groups that had been injected with heat-treated cells as compared to those animals that had been inoculated with non-heated cells (either irradiated or untreated) or the untreated controls. No detectable differences were found between the groups receiving non-heated cells and the untreated control group. It was found that the protection derived from inoculation of heat-treated tumor cells was not due to decreased tumor growth rate, but was more probably related to the destruction of a portion of the tumor cell population. Protection Contre L'adenocarcinome Mammaire Spontane de la Souris par Inoculation de Cellules de Tumeurs Mammaires Sygeneiques Traitees par la Chaleur La presente etude avait pour but de determiner l'effet de l'inoculation de cellules de tumeurs mammaires syngeneiques sur l'incidence de tumeurs spontanees et la mortalite dans un groupe de souris femelles RIII,chez lesquelles on constate normalement une forte incidence des tumeurs mammaires primitives. Nous avons administre aux animaux des cellules tumorales non traitees, traitees par la chaleur ou irradiees, ou encore un melange de cetlules. Un groupe n'a recu aucune injection et a servi de temoin. Nous avons enregistre l'incidence du developpement tumoral et la mortalite pour chaque animal utilise au cours de l'etude. Nous avons note des retards importants dans l'apparition des tumeurs et la mort des animaux dans les groupes inocules avec des cellules traitees a la chaleur, par comparaison avec les animaux ayant recu des cellules irradiees ou non traitees et avec les animaux temoins. Aucune difference decelable n'apparit entre les groupes inocules avec des cellules non chauffees et le groupe temoin. Nous avons constate que la protection due a l'inoculation de cellules tumorales chauffees ne provient pas d'une diminution de la vitesse de croissance des tumeurs, mais bien plutot de la destruction d'une partie de la population de cellules tumorales.
