OBJECTIVES To evaluate association of three single-nucleotide polymorphisms (SNPs) of growth hormone receptor (GHR) gene with mandibular prognathism (MP) and relationships between mandibular morphology and GHR gene SNPs in the Korean population. MATERIALS AND METHODS A total of 325 subjects were divided into two groups based on sagittal maxillomandibular relationship by the lateral cephalography: the MP and control groups. From the SNPs in the GHR gene, three SNPs (rs6180, rs6182, and rs6184) were selected. SNP genotyping was performed using direct sequencing. The craniofacial measurements of lateral cephalography were analyzed. RESULTS We found lack of association between GHR and MP. However, in analysis according to the values of cephalometric measurements, rs6180 was significantly associated with ANB, SNB, effective mandibular length, and SNMP in females. Additionally, rs6182 and rs6184 were significantly associated with ramal height in males. CONCLUSION GHR SNPs may affect not only the sagittal development of mandible but also the vertical development of ramal height, and GHR SNPs may gender-differently influence mandibular morphology. This finding supports that the GHR might be susceptible on mandibular morphogenesis in the Korean population.

OBJECTIVE To investigate the accuracy of automated identification of cephalometric landmarks using the cascade convolutional neural networks (CNN) on lateral cephalograms acquired from nationwide multi-centres. SETTINGS AND SAMPLE POPULATION A total of 3150 lateral cephalograms were acquired from 10 university hospitals in South Korea for training. MATERIALS AND METHODS We evaluated the accuracy of the developed model with independent 100 lateral cephalograms as an external validation. Two orthodontists independently identified the anatomic landmarks of the test data set using the V-ceph software (version 8.0, Osstem, Seoul, Korea). The mean positions of the landmarks identified by two orthodontists were regarded as the gold standard. The performance of the CNN model was evaluated by calculating the mean absolute distance between the gold standard and the automatically detected positions. Factors associated with the detection accuracy for landmarks were analysed using the linear regression models. RESULTS The mean inter-examiner difference was 1.31 ± 1.13 mm. The overall automated detection error was 1.36 ± 0.98 mm. The mean detection error for each landmark ranged between 0.46 ± 0.37 mm (maxillary incisor crown tip) and 2.09 ± 1.91 mm (distal root tip of the mandibular first molar). Objective To investigate the effect of local injection of mineralized hybrid nanoparticles loading dentin matrix protein-1 (DMP-1) and matrix metalloproteinase-13 (MMP-13) complex (P-NPs) on the bone remodeling on atrophic alveolar ridges (AAR) ahead of orthodontic tooth movement (OTM). Settings and sample population Four beagles were randomly allocated into Group C (OTM only) and Group NP (OTM with P-NPs injection). Experimental model of AAR was prepared in 8 mandibular quadrants after extraction of the third premolars (n=4 per Group). Materials and methods Reciprocal traction of the second and fourth premolars was performed toward AAR for 8 weeks. P-NPs were prepared by loading recombinant DMP-1 and MMP-13 complex into calcium carbonate (CaCO3 )-mineralized hybrid nanoparticles, and injected at 0, 3, 6 weeks. The rate of OTM and the bone remodeling characteristics were compared between Groups using fluorescent microscopic analysis and micro-structural histomorphometric analysis. Results Group NP revealed higher bone volume fraction and higher trabecular ratio with lower bone mineral density than Group C on AAR area. Meanwhile, the root movement towards AAR was facilitated in Group NP representing more bodily movement than Group C. Conclusion Non-invasive intervention of P-NPs injection suggested a clinical potential to facilitate translational movement into the AAR with sustaining woven bone-like microstructural environment.

Signal transducer and activator of transcription 3 (STAT3) has been considered as a potential target for development of anticancer therapeutics. Here, we report a novel mechanism by which the cyclopentenone prostaglandin, 15-deoxy-Δ12,14 -prostaglandin J2 (15d-PGJ2 ) functions as an allosteric inhibitor of STAT3. 15d-PGJ2 inhibits phosphorylation, dimerization, nuclear translocation, and transcriptional activity of STAT3 in H-Ras-transformed human mammary epithelial cells (MCF10A-Ras) through the Michael addition reaction at cysteine 259 of STAT3. Comparative studies with 15d-PGJ2 analogues reveal that both C12-C13 and C9-C10 double bonds conjugated to the carbonyl group in the cyclopentenone ring of 15d-PGJ2 are essential for STAT3 binding. Antiproliferative and pro-apoptotic activities of 15d-PGJ2 in MCF10A-Ras cells are attributable to covalent modification of STAT3 on Cys259, and mimic the effects induced by mutation of this amino acid.

OBJECTIVES To evaluate three-dimensional (3D) craniofacial changes induced by a non-surgical tooth-bone-borne rapid palatal expander (TBB-RPE) according to the symmetrical pattern of expansion, to investigate the 3D changes between the sides in patients with asymmetric expansion, and to identify the related factors of asymmetric expansion. SETTING AND SAMPLE POPULATION Sixty-six patients (mean age: 19.3 ± 5.7 years) treated with TBB-RPE were divided into a symmetric expansion group (Group S, n = 46) or asymmetric expansion group (Group A, n = 20). Group S was subdivided into Group Ss (n = 27), with bilateral frontomaxillary suture (FMS) split, and Group Sn (n = 19), with no FMS split. MATERIALS AND METHODS Pre- and post-expansion cone-beam computed tomography images were superimposed, and the common coordinated system was set. All landmarks were designated as coordinate pairs, and treatment changes were automatically calculated. Analysis of variance was conducted for intergroup comparison of craniofacial changes, and logistic regression analysis was performed to identify the related factors of asymmetric expansion. RESULTS The frequency of asymmetric expansion was 30.3%. Group A with unilateral FMS split showed less craniofacial changes than Group Ss and more changes than Group Sn. Group A exhibited different nasomaxillary displacement between the two halves, showing greater changes in the FMS-split side. Among the tested six variables (age, gender, Angle's classification, unilateral crossbite, maxillary cant and chin deviation), chin deviation was uniquely associated with asymmetric expansion. CONCLUSIONS Tooth-bone-borne rapid palatal expander had a risk of asymmetric expansion, especially in facial asymmetric patients with chin deviation, producing different craniofacial changes from symmetric expansion.

Mechanism-based chemical transformation of 15-deoxy-Δ12, 14 -PGJ2 (15d-PGJ2 ) resulted in a series of new NF-E2-related factor-2 (Nrf2) activators and detailed elucidation of the function of each electrophilic binding site. In addition, HO-1 expression resulting from Nrf2 activation through enhanced dissociation of the Keap1-Nrf2 complex by the new activators was proved.

This study was performed to investigate the compliance, safety, dosage modifications (dose reduction and/or schedule change [including permanent S‐1 withdrawal]), and clinical parameters that predict S‐1 dosage modification in gastric cancer patients receiving adjuvant S‐1 chemotherapy. One hundred and forty‐nine patients who underwent curative D2 surgery and received adjuvant S‐1 chemotherapy were enrolled. S‐1 was administered orally (40 mg/m(2) twice daily on days 1–28 every 6 weeks) for 1 year. For patients unable to tolerate S‐1, the dosage was reduced or the schedule was changed to a 3‐weekly schedule of 2 weeks on treatment followed by 1 week off treatment. The planned 1‐year treatment was completed in 73.8% of patients; 69 patients required dosage modification because of toxicity. The most frequent cause of dosage modification was enterocolitis (37 patients; defined as ≥ grade 2 abdominal pain and/or ≥ grade 2 diarrhea). Most dosage modification occurred during the early cycles of treatment (within the first 3 months). Severe toxicities (≥ grade 3) included neutropenia (13.4%), abdominal pain (8.1%) and diarrhea (8.1%). In multivariate analyses, decreased relative dose intensity was related to poor disease‐free survival independent of stage, and only low creatinine clearance predicted S‐1 dosage modification. In conclusion, although adjuvant S‐1 therapy has a high compliance rate, meticulous monitoring of adverse events is required in the early period of treatment. Background and Objective Both Corticision and low-level laser therapy (LLLT) are known to affect the rate of tooth movement. Our objective was to investigate the combined effects of Corticision and LLLT on the tooth movement rate and paradental remodeling in beagles. Study Design/Materials and Methods The maxillary second premolars (n = 24) of 12 beagles were randomly divided into four groups (n = 6 per group) based on the treatment modality: group A, only orthodontic force (control); group B, orthodontic force plus Corticision; group C, orthodontic force plus LLLT; group D, orthodontic force plus Corticision and LLLT. Results Ratios of second premolar-to-canine movement were greater by 2.23-fold in group B and 2.08-fold in group C, but 0.52-fold lesser in group D than in group A. The peak velocity was observed at an earlier stage of tooth movement in group B but at a later stage in group C during the 8-week treatment period. Summary Retinoblastoma protein (Rb) plays a key role in cell cycle control, cell differentiation, and apoptosis in animals. In this study, we used virus-induced gene silencing (VIGS) to investigate the cellular functions of Rb in higher plants. VIGS of NbRBR1, which encodes the Nicotiana benthamiana Rb homolog, resulted in growth retardation and abnormal organ development. At the cellular level, Rb suppression caused prolonged cell proliferation in tissues that are normally differentiated, which indicates that Rb is a negative regulator of plant cell division. Furthermore, differentiation of the epidermal pavement cells and trichomes was partially retarded, and stomatal clusters formed in the epidermis, likely due to uncontrolled cell division of stomata precursor cells. Rb suppression also caused extra DNA replication in endoreduplicating leaf cells, suggesting a role of Rb in the endocycle. These Rb phenotypes were accompanied by stimulated transcription of E2F and E2F-regulated S-phase genes. Thus, disruption of Rb function in plants leads to ectopic cell division in major organs that correlates with a delay in cell differentiation as well as increased endoreduplication, which indicates that Rb coordinates these processes in plant organ development.
