Aim In the context of intensifying threats of climate change on marine communities, ecological models are widely applied for conservation strategies, though polar studies remain scarce given the limited number of datasets available. Correlative (e.g. species distribution models, SDM) and mechanistic (e.g. dynamic energy budget models, DEB) modelling approaches are usually used independently in studies. Using both approaches in integrative, hybrid models could help to better estimate the species potential ecological niche, as mechanistic and correlative models complement each other very well, giving more insights into species potential response to fast-changing environmental conditions. Location The study focusses on the Baie du Morbihan, a silled basin located in the east of the Kerguelen Islands (sub-Antarctic). Methods A hybrid, correlative-mechanistic model was implemented to predict the response of the endemic sea urchin Abatus cordatus (Verrill, 1876). We compared the performances of classic and integrated approaches to predict A. cordatus distribution according to two dates representing seasonal contrasts. Two integrated approaches were studied and performed by either (1) including the spatial projection of the DEB model as an input layer inside the SDM ('integrated SDM-DEB') or (2) using a Bayesian procedure to use DEB model outputs as priors of the SDM ('integrated Bayesian' approach). Results Results show higher performances of 'integrated Bayesian' approaches to evaluate A. cordatus potential ecological niche compared with 'classic' and 'integrated SDM-DEB' methods. The influence of environmental conditions on model predictions is further captured with these Bayesian procedures and better highlights the environmental influence on the species-predicted distribution. Model performance is good for the different simulations, and uncertainty in predictions is well-highlighted. Main conclusions The good performances of 'integrated Bayesian' approaches to estimate species potential ecological niche opens perspectives for future applications to a broad panel of natural examples, noteworthy for decision-making and conservation management purposes.

There is a large amount of co-product generated by the sea urchin fisheries around the world, as well as a growing interest in removing large quantities of undersize and low value sea urchins from barren areas in the northern Atlantic and Pacific coasts. The authors believe there is scope to develop a hydrolysate product from this and this study gives preliminary observations on the characteristics of hydrolysate from the sea urchin Strongylocentrotus droebachiensis. The biochemical composition for S. droebachiensis were; water 64.1%, protein 3.4%, oil 0.9% and ash 29.8%. Amino acid composition, molecular weight distribution, lipid-class and fatty acid composition are also presented. The authors suggest a sensory-panel mapping be undertaken on future sea urchin hydrolysates. Possible uses for the hydrolysate are unclear at this stage but the combination of amino acids and the relatively high levels of Glycine, Aspartic acid, and Glutamic acid should be further investigated.

BACKGROUND Sea urchins are model organisms for studying the spatial-temporal control of gene activity during development. The Southern California species, Lytechinus pictus, has a sequenced genome and can be raised in the laboratory from egg to egg in 4 to 5 months. RESULTS Here, we present new techniques for generating parthenogenetic larvae of this species and include a gallery of photomicrographs of morphologically abnormal larvae that could be used for transcriptomic analysis. CONCLUSIONS Comparison of gene expression in parthenogenotes to larvae produced by fertilization could provide novel insights into gene expression controls contributed by sperm in this important model organism. Knowledge gained from transcriptomics of sea urchin parthenogenotes could contribute to parthenogenetic studies of mammalian embryos.

Shifts in microbial communities represent a rapid response mechanism for host organisms to respond to changes in environmental conditions. Therefore, they are likely to be important in assisting the acclimatization of hosts to seasonal temperature changes as well as to variation in temperatures across a species' range. The Persian/Arabian Gulf is the world's warmest sea, with large seasonal fluctuations in temperature (20℃ - 37℃) and is connected to the Gulf of Oman which experiences more typical oceanic conditions (32℃ in the summer). This system is an informative model for understanding how symbiotic microbial assemblages respond to thermal variation across temporal and spatial scales. Here, we elucidate the role of temperature on the microbial gut community of the sea urchin Echinometra sp. EZ and identify microbial taxa that are tightly correlated with the thermal environment. We generated two independent datasets with a high degree of geographic and temporal resolution. The results show that microbial communities vary across thermally variable habitats, display temporal shifts that correlate with temperature, and can become more disperse as temperatures rise. The relative abundances of several ASVs significantly correlate with temperature in both independent datasets despite the300 km distance between the furthest sites and the extreme seasonal variations. Notably, over 50% of the temperature predictive ASVs identified from the two datasets belonged to the family Vibrionaceae. Together, our results identify temperature as a robust predictor of community-level variation and highlight specific microbial taxa putatively involved in the response to thermal environment.

Sea urchins are key grazers in coastal seas, where they can survive a variety of conditions and diets, enhancing their ecological impact on kelp forests and other ecosystems. Using 16S rRNA gene sequencing, we characterized bacterial communities associated with guts of the two dominant sea urchin species in southern California, the red urchin Mesocentrotus franciscanus, and the purple urchin Strongylocentrotus purpuratus. Our results show that the two urchin species have distinct gut microbiomes that vary with habitat. The taxonomic composition of their microbiomes suggests that they may facilitate digestion of food and be a source of nutrition themselves. These results highlight the role of microbiomes within macroorganisms as an extended ecological trait, and suggest that microbes may be crucial to resource use and partitioning in co‐occurring species.

Ocean acidification (OA) from seawater uptake of rising carbon dioxide emissions impairs development in marine invertebrates, particularly in calcifying species. Plasticity in gene expression is thought to mediate many of these physiological effects, but how these responses change across life history stages remains unclear. The abbreviated lecithotrophic development of the sea urchin Heliocidaris erythrogramma provides a valuable opportunity to analyse gene expression responses across a wide range of life history stages, including the benthic, post-metamorphic juvenile. We measured the transcriptional response to OA in H. erythrogramma at three stages of the life cycle (embryo, larva, and juvenile) in a controlled breeding design. The results reveal a broad range of strikingly stage-specific impacts of OA on transcription, including changes in the number and identity of affected genes; the magnitude, sign, and variance of their expression response; and the developmental trajectory of expression. The impact of OA on transcription was notably modest in relation to gene expression changes during unperturbed development and much smaller than genetic contributions from parentage. The latter result suggests that natural populations may provide an extensive genetic reservoir of resilience to OA. Taken together, these results highlight the complexity of the molecular response to OA, its substantial life history stage specificity, and the importance of contextualizing the transcriptional response to pH stress in light of normal development and standing genetic variation to better understand the capacity for marine invertebrates to adapt to OA.

Antarctic marine environments are at risk from petroleum fuel spills as shipping activities in the Southern Ocean increase. Knowledge of the sensitivity of Antarctic species to fuels under environmentally realistic exposure conditions is lacking. We determined the toxicity of 3 fuels, Special Antarctic Blend diesel (SAB), marine gas oil (MGO), and intermediate fuel oil (IFO 180) to a common Antarctic sea urchin, Sterechinus neumayeri. Sensitivity was estimated for early developmental stages from fertilization to the early 4-arm pluteus in toxicity tests of up to 24 d duration. The effects of the water accommodated fractions (WAFs) of fuels were investigated under different exposure scenarios to determine the relative sensitivity of stages and of different exposure regimes. Sensitivity to fuel WAFs increased through development. Both MGO and IFO 180 were more toxic than SAB, with median effect concentration values for the most sensitive pluteus stage of 3.5, 6.5, and 252 µg/L total hydrocarbon content, respectively. Exposure to a single pulse during fertilization and early embryonic development showed toxicity patterns similar to those observed from continuous exposure. The results show that exposure to fuel WAFs during critical early life stages affects the subsequent viability of larvae, with consequent implications for reproductive success. The sensitivity estimates for S. neumayeri that we generated can be utilized in risk assessments for the management of Antarctic marine ecosystems. Sea urchin Mesocentrotus nudus, Glyptocidaris crenularis, and Strongylocentrotus intermedius gonad protein isolates (mnGPIs, gcGPIs, and siGPIs) were extracted by isoelectric solubilization/precipitation (ISP) from the defatted gonads, and their functional properties were compared. Sodium dodecyl sulfate polyacrylamide gel electrophoresis results showed the similar protein pattern between each protein isolate and defatted gonad, indicating the high efficiency of ISP processing for protein recovery. Amino acid profileconfirmed that the mnGPIs and siGPIs could be potential sources of essential amino acid in nature. As regard to functional properties, mnGPIs showed higher water- and oil- holding capacities followed bysiGPIs and gcGPIs and all protein isolates presented great foaming property. As for emulsifying activity index (EAI), mnGPIs, gcGPIs, and siGPIs showed the minimum solubility and EAI at pH 5, 3, and 4, respectively, and behaved a pH-dependent manner. The gcGPIs revealed the highest EAI from pH 6 to 8 among the samples. In addition, circular dichroism showed increased content of β-sheet at the expense of α-helix and β-turn, suggesting the structure denaturation of the protein isolates. Indeed, no statistical difference was observed between secondary structure of mnGPIs and siGPIs. Moreover, ISP processing increased free sulfhydryl content of sea urchin protein isolates, but no difference was observed among the samples. Furthermore, siGPIs revealed the highest amount of total sulfhydryl and disulfide bonds, whereas both defatted gonads and protein isolates from G. crenularis presented the maximum surface hydrophobicity. These results suggest that gonad protein isolates from three species of sea urchin possess various functionalities and therefore can be potentially applied in food system. PRACTICAL APPLICATION: Sea urchin M. nudus, G. crenularis, and S. intermedius gonads are edible, whereas the functional properties of protein isolates from sea urchin gonad remain unknown. In this case, the extraction and comparison of three species of sea urchin gonad protein isolates will not only confirm functional properties but also screen food ingredients with suitable functions. In this study, functionalities of protein isolates derived from M. nudus, G. crenularis, and S. intermedius gonads would provide potential application in bakery food and meat products or as emulsifier candidates in food system.

Larvae of many echinoids are known to be phenotypically plastic and capable of changing the growth rate of their post-oral arms depending on the microalgae concentration in their habitat. As literature data show, developing larvae use chemosensation to detect algae in the environment and "adjust" the rate of growth of their post-oral arms through dopamine signaling. According to our results, dopamine has a significant effect on the post-oral arm growth in early larvae of two sea urchin species, Mesocentrotus nudus and Strongylocentrotus intermedius. The dopamine effect depends on concentration: the higher the dopamine concentration in the water, the shorter the post-oral arms. We suggest that the pattern of response to variation in dopamine concentration, manifested by early larvae of both species, is similar to that observed at different concentrations of microalgae.

The proliferation of sea urchins can decimate macroalgal forests in coastal ecosystems, leading to persistent barren seascapes. While kelp forests are among the most productive ecosystems on the planet, productivity in these urchin barrens is dramatically reduced. Moreover, urchins inhabiting these food-depauperate barrens face starvation and many survive in these barrens for years or decades. Urchins in barrens can persist by eating food subsidies from drift algae, pelagic salps, tubeworms, as well as encrusting and filamentous algae, microbial mats, and slow-growing species resistant to herbivory. Despite both food from endogenous production and exogenous subsidies, many urchins in barrens likely experience prolonged food deprivation. This resource limitation may create a trade-off between reproduction and survival; for example, fecundity of purple sea urchins (Strongylocentrotus purpuratus) is 99.9% lower in barrens. Despite food constraints, red sea urchins (Mesocentrotus franciscanus), the dominant urchin species at our study sites, can live in excess of 100 years and barrens in Haida Gwaii, British Columbia (BC), Canada, have persisted for at least 143 years. While these phenomena are widespread and well documented, the bioenergetic adaptations that allow urchins to persist in these food-depauperate barrens remain poorly understood. To quantify habitat-specific differences in metabolic rates and energy reserves (as measured by gonadal mass), we conducted respirometry on and measured gonadal mass in M. franciscanus at three locations in BC inside and outside of adjacent kelp forest and barrens habitat. Here we demonstrate that M. franciscanus in barrens versus kelp forests have substantially lower energy reserves and, importantly, also exhibit dramatic reductions in size-specific resting metabolic rates (RMR), even after standardizing by metabolically active body mass. On average, gonadal mass was 44.6% lower and RMR scaled to metabolically active body mass was 40% lower in barrens urchins than in kelp forest urchins. Such a shift in metabolic rate may provide a mechanism that facilitates barren state stability over long time scales as M. franciscanus can lower energetic demands while they wait for small pulses of food, scrape by on low-productivity resources, and suppress recruitment of macroalgae for months, years, or decades.

Sea urchins are used as a model organism for research on developmental biology and gene regulatory networks during early development. Gene knockdown by microinjection of morpholino antisense oligonucleotide (MASO) has been used to analyze gene function in early sea urchin embryos. However, as the effect of MASO is not long lasting, it is impossible to perturb genes expressed during late development by MASO. Recent advances in genome editing technologies have enabled gene modification in various organisms. We previously reported genome editing in the sea urchin Hemicentrotus pulcherrimus using zinc-finger nuclease (ZFN) and transcription activator-like effector nuclease (TALEN); however, the efficiencies of these technologies were not satisfactory. Here, we applied clustered regularly interspaced short palindromic repeat (CRISPR)-CRISPR-associated nuclease 9 (Cas9) technology to knock out the Pks1 gene in H. pulcherrimus. When sgRNAs targeting Pks1, which is required for the biosynthesis of larval pigment, were microinjected into fertilized eggs with SpCas9 mRNA, high-efficiency mutagenesis was achieved within 24 hr post fertilization and SpCas9/sgRNA-injected pluteus larvae had an albino phenotype. One of the sgRNAs yielded 100% mutagenesis efficiency, and no off-target effect was detected. In addition, the albino phenotype was maintained in juvenile sea urchins after metamorphosis, and the knockout sea urchins survived for at least one year and grew to albino adult sea urchins. These findings suggest that knockout adult sea urchins were successfully established and the CRISPR-Cas9 system is a feasible method for analyzing gene functions from late developmental to adult stage.

A dramatic life history switch that has evolved numerous times in marine invertebrates is the transition from planktotrophic (feeding) to lecithotrophic (nonfeeding) larval development—an evolutionary tradeoff with many important developmental and ecological consequences. To attain a more comprehensive understanding of the molecular basis for this switch, we performed untargeted lipidomic and proteomic liquid chromatography-tandem mass spectrometry on eggs and larvae from three sea urchin species: the lecithotroph Heliocidaris erythrogramma, the closely related planktotroph Heliocidaris tuberculata, and the distantly related planktotroph Lytechinus variegatus. We identify numerous molecular-level changes possibly associated with the evolution of lecithotrophy in H. erythrogramma. We find the massive lipid stores of H. erythrogramma eggs are largely composed of low-density, diacylglycerol ether lipids that, contrary to expectations, appear to support postmetamorphic development and survivorship. Rapid premetamorphic development in this species may instead be powered by upregulated carbohydrate metabolism or triacylglycerol metabolism. We also find proteins involved in oxidative stress regulation are upregulated in H. erythrogramma eggs, and apoB-like lipid transfer proteins may be important for echinoid oogenic nutrient provisioning. These results demonstrate how mass spectrometry can enrich our understanding of life history evolution and organismal diversity by identifying specific molecules associated with distinct life history strategies and prompt new hypotheses about how and why these adaptations evolve.

To investigate the effects of family, stocking density, and their interaction on mortality, growth rate, and size dispersal in the sea urchin Strongylocentrotus intermedius, 1,440 juveniles from six full‐sib families were exposed to three stocking densities, that is, low stocking density (LD), middle stocking density (MD), and high stocking density (HD), and reared for 10 months in sea‐based suspended cages. The results demonstrated significant family effects on cumulative mortality rate (CMR); specific growth rate for test diameter (SGR for TD); specific growth rate for body weight (SGR for BW); coefficient of variation for body weight at the 10th month (CV for BW₁₀); and significant stocking density effects on SGR for TD, SGR for BW, and CV for BW₁₀. Statistically significant family by stocking density interaction was only found in SGR for TD. A certain degree of family‐ranking inversions occurred in SGR for TD. The present study provides evidence for the existence of family by stocking density interaction on the growth rate of test diameter in the family selection for S. intermedius. More attention should be paid to this interaction effect to select correct parents in S. intermedius.

Multiple anthropogenic stressors are causing a global decline in foundation species, including macrophytes, often resulting in the expansion of functionally different, more stressor‐tolerant macrophytes. Previously subdominant species may experience further positive demographic feedback if they are exposed to weaker plant–herbivore interactions, possibly via decreased palatability or being structurally different from the species they are replacing. However, the consequences of the spread of opportunistic macrophytes for the local distribution and life history of herbivores are unknown.The green alga, Caulerpa filiformis, previously a subdominant macrophyte on low intertidal‐shallow subtidal rock shores, is becoming locally more abundant and has spread into warmer waters across the coast of New South Wales, Australia.In this study, we measured (a) the distribution and abundance of a key consumer, the sea urchin Heliocidaris erythrogramma, across a seascape at sites where C. filiformis has become dominant, (b) performed behavioral field experiments to test the role of habitat selection in determining the local distribution of H. erythrogramma, and (c) consumer experiments to test differential palatability between previously dominant higher quality species like Ecklonia radiata and Sargassum sp. and C. filiformis and the physiological consequences of consuming it.At all sites, urchin densities were positively correlated with distance away from C. filiformis beds, and they actively moved away from beds. Feeding experiments showed that, while urchins consumed C. filiformis, sometimes in equal amounts to higher quality algae, there were strong sublethal consequences associated with C. filiformis consumption, mainly on reproductive potential (gonad size). Specifically, the gonad size of urchins that fed on C. filiformis was equivalent to that in starved urchins. There was also a tendency for urchin mortality to be greater when fed C. filiformis.Overall, strong negative effects on herbivore life‐history traits and potentially their survivorship may establish further positive feedback on C. filiformis abundance that contributes to its spread and may mediate shifts from top‐down to bottom‐up control at locations where C. filiformis has become dominant., The spread of the previously subdominant Caulerpa filiformis along the coast of NSW, Australia has strong lethal and sublethal effects on a key herbivore population. We performed sea urchin surveys, behavioral experiments, and consumer experiments and found that C aulerpa has strong negative effects on their populations. These results may establish reinforcing feedbacks on the spreading algae and mediate shifts from top‐down to bottom‐up control.

The detection of light intensity and subsequent phototaxis are essential for the fitness of sea urchins in intertidal and shallow subtidal waters, where light intensity varies in accordance with the depth and other hydrographic conditions. The molecular basis of photoresponse, however, remains largely unknown. We compared the expression of SiOpsin4, SiPax6, and SiOpsin5 among tube feet, coelomocytes, gonads, and gut of the sea urchin Strongylocentrotus intermedius. All three genes showed the significantly highest expression in tube feet. To test whether r‐opsin genes (for example, Opsin4) and Pax6 in tube feet, rather than chaopsin genes (for example, Opsin5), are involved in the molecular responses to light intensity, we investigated the expression of Opsin4, Pax6, and Opsin5 among four light intensities. Both Opsin4 and Pax6 expressions were significantly higher in the sea urchins at 1,500 lx than at 100, 300, and 700 lx, indicating a co‐upregulation of Opsin4 and Pax6 expressions in sea urchins exposed to the high light intensity (for example, 1,500 lx). However, there was no significant difference of SiOpsin5 expression among the four light intensity groups. Consistent with our hypothesis, there was no significant difference in either Opsin4 or Pax6 expression between the two light intensity environments (low light intensity environment: 50–100 lx; high light intensity environment: 330–1400 lx). This indicates that positive phototaxis and negative phototaxis probably regulate the expression of Opsin4 and Pax6 to a similar level when sea urchins are exposed to the high and low light intensities, respectively. The present study provides new insights into the molecular basis of light intensity detection and subsequent phototaxis of eyeless species.

Coelomic fluid aspiration has been utilized in echinoderms in research and clinical settings. Detailed procedural descriptions for coelomic fluid sampling in sea urchins (class Echinoidea) are lacking, and samples are prone to contamination. The objectives of this study were to (1) standardize a technique for coelomic fluid collection in long-spined sea urchin Diadema antillarum that optimizes the diagnostic quality of the sample utilizing diagnostic imaging, (2) identify coelomic fluid bacterial isolates (using Biolog GEN III MicroLog and 16s rDNA sequencing), and (3) compare positive cultures to animal weight, holding time prior to sampling, water temperature, and gross fluid appearance. Seventy Diadema antillarum from the Florida Keys collected in two groups (March and September 2015) were utilized. Positive cultures for bacterial contamination were identified in 5% and 44%, respectively, of animals in the sampling groups. Vibrio spp. was the predominant genus identified. Positive cultures were more frequent in the group with smaller-sized animals, increased holding times, and elevated water temperatures. Deviation from clear-pink gross coelomic fluid appearance did not reliably predict bacterial contamination. A standardized technique for coelomocentesis was defined. The use of the proposed coelomocentesis methodology may facilitate improved health evaluations of sea urchins and may be applicable to research, conservation efforts, and disease investigations.

BACKGROUND AND PURPOSE Sperm from many species share the sperm-specific Ca2+ channel CatSper that controls the intracellular Ca2+ concentration and, thereby, the swimming behaviour. A growing body of evidence suggests that the mechanisms controlling the activity of CatSper and its role during fertilization differ among species. A lack of suitable pharmacological tools has hampered the elucidation of the function of CatSper. Known inhibitors of CatSper exhibit considerable side effects and also inhibit Slo3, the principal K+ channel of mammalian sperm. The compound RU1968 was reported to suppress Ca2+ signaling in human sperm by an unknown mechanism. Here, we examined the action of RU1968 on CatSper in sperm from humans, mice, and sea urchins. EXPERIMENTAL APPROACH We resynthesized RU1968 and studied its action on sperm from humans, mice, and the sea urchin Arbacia punctulata by Ca2+ fluorimetry, single-cell Ca2+ imaging, electrophysiology, opto-chemistry, and motility analysis. KEY RESULTS RU1968 inhibited CatSper in sperm from invertebrates and mammals. The compound lacked toxic side effects in human sperm, did not affect mouse Slo3, and inhibited human Slo3 with about 15-fold lower potency than CatSper. Moreover, in human sperm, RU1968 mimicked CatSper dysfunction and suppressed motility responses evoked by progesterone, an oviductal steroid known to activate CatSper. Finally, RU1968 abolished CatSper-mediated chemotactic navigation in sea urchin sperm. CONCLUSION AND IMPLICATIONS We propose RU1968 as a novel tool to elucidate the function of CatSper channels in sperm across species.

Studies of echinoid microscopic anatomy over the last two centuries have created a number of inaccuracies and mistakes that have accumulated in the descriptions of the intricate organization of the coelomic system of Echinoidea. To clarify the situation, we reconstructed the axial complex and radial complex of the echinoid Strongylocentrotus pallidus. The water ring is located between the perivisceral coelom and the perioral coelom. The oral haemal ring lies between the coelothelia of the water-vascular, perivisceral, and perioral rings. The axial part of the axial organ communicates with the oral haemal ring in interradius CD, but the axial coelom does not form the axocoelomic perihaemal ring. The ventral intestinal haemal vessel originates from the oral haemal ring in radius A, and then branches into a network of capillaries, from which the dorsal intestinal vessel starts. The pericardial coelom envelopes the pericardial part of the axial organ, the lacunae of which communicate with the haemocoel of the body wall and with the axial part of the axial organ. The genital haemal ring and the dorsal intestinal vessel communicate with the axial organ. The genital coelom passes in the CD interradius on the side opposite to the hindgut. There is a somatocoelomic perihaemal ring, which sends a pair of coelomic outgrowths into each radius, accompanied by a radial haemal vessel in the oral part. The mistakes and inaccuracies of early descriptions of the echinoid axial complex are listed. The axial complex and associated structures of sea urchins are compared with other eleutherozoans.

During gastrulation, one of the most important morphogenetic events in sea urchin embryogenesis, the gut bends toward the ventral side to form an open mouth. Although the involvement of transforming growth factor-β (TGF-β) signals in the cell-fate specification of the ectoderm and endoderm along the dorsal-ventral axis has been well reported, it remains unclear what controls the morphogenetic behavior of gut bending. Here, using two sea urchin species, Hemicentrotus pulcherrimus and Temnopleurus reevesii, we show that TGF-β signals are required for gut bending toward the ventral side. To search for the common morphogenetic cue in these two species, we initially confirmed the expression patterns of the dorsal-ventral regulatory TGF-β members, nodal, lefty, bmp2/4, and chordin, in T. reevesii because these factors are appropriate candidates to investigate the cue that starts gut bending, although genetic information about the body axes is entirely lacking in this species. Based on their expression patterns and a functional analysis of Nodal, the dorsal-ventral axis formation of T. reevesii is likely regulated by these TGF-β members, as in other sea urchins. When the Alk4/5/7 signal was inhibited by its specific inhibitor, SB431542, before the late gastrula stage of T. reevesii, the gut was extended straight toward the anterior tip region, although the ectodermal dorsal-ventral polarity was normal. By contrast, H. pulcherrimus gut bending was sensitive to SB431542 until the prism stage. These data clearly indicate that gut bending is commonly dependent on a TGF-β signal in sea urchins, but the timing of the response varies in different species.

Background: Photoreception-associated genes of the Pax-Six-Eya-Dach network (PSEDN) are deployed for many roles in addition to photoreception development. In this first study of PSEDN genes during development of the pentameral body in sea urchins, we investigated their spatial expression in Heliocidaris erythrogramma. Results: Expression of PSEDN genes in the hydrocoele of early (Dach, Eya, Six1/2) and/or late (Pax6, Six3/6) larvae, and the five hydrocoele lobes, the first morphological expression of pentamery, supports a role in body plan development. Pax6, Six1/2 and Six3/6 were localized to the primary and/or secondary podia and putative sensory/neuronal cells. Six1/2 and Six3/6 were expressed in the neuropil region in the terminal disc of the podia. Dach was localized to spines. Sequential up regulation of gene expression as new podia and spines formed was evident. Rhabdomeric opsin and pax6 protein were localized to cells in the primary podia and spines. Conclusion: Our results support roles for PSEDN genes in development of the pentameral body plan, contributing to our understanding of how the most unusual body plan in the Bilateria may have evolved. Development of sensory cells within the Pax-Six expression field is consistent with the role of these genes in sensory cell development in diverse species. This article is protected by copyright. Acrylic ester resin (AER) is a promising oil-absorbing material for oils and organic solvents absorption because it exhibits high oil retention ability, good swelling properties, and reusability while also being inexpensive, large-scale production, and environmentally benign. However, the AER is not fully utilized owing to its low oil absorption capacity and slow oil absorption rate. In this work, we combine porous structures of MnO2 and swelling properties of AER and design a novel route to fabricate sea urchin-like MnO2/AER hybrids. In this route, the sea urchin-like MnO2 was prepared by a facile hydrothermal method in acidic media. The sea urchin-like MnO2/AER hybrids with different wt% were synthesized by a microwave polymerization route. The resin hybrids exhibited very rough surfaces, which are beneficial for organic molecules absorption and diffusion into the interiors of materials. The resin hybrids can remove oils and organic solvents from water with high selectivity and absorption capacity, and can absorb not only floating oil but also heavy organic solvents underwater. The maximum absorption capacity of resin hybrids was found at the MnO2 content of 3%, and maximum absorption capacity up to 32.5 times of their own weight, higher than that of the pure AER. The performances were mainly attributed to the unique porous structure of MnO2 and swelling properties of AER, indicating the potential application of such oil-absorbing resins to introduce the porous inorganic materials for developing new oils and organic solvents absorbent for removal of organic pollutants from the wastewater. The globiferous pedicellariae of the venomous sea urchin Toxopneustes pileolus contains several biologically active proteins. We have cloned the cDNA of one of the toxin components, SUL-I, which is a rhamnose-binding lectin (RBL) that acts as a mitogen through binding to carbohydrate chains on target cells. Recombinant SUL-I (rSUL-I) was produced in Escherichia coli cells, and its carbohydrate-binding specificity was examined with the glycoconjugate microarray analysis, which suggested that potential target carbohydrate structures are galactose-terminated N-glycans. rSUL-I exhibited mitogenic activity for murine splenocyte cells and toxicity against Vero cells. The three-dimensional structure of the rSUL-I/l-rhamnose complex was determined by X-ray crystallographic analysis at a 1.8 A resolution. The overall structure of rSUL-I is composed of three distinctive domains with a folding structure similar to those of CSL3, a RBL from chum salmon (Oncorhynchus keta) eggs. The bound l-rhamnose molecules are mainly recognized by rSUL-I through hydrogen bonds between its 2-, 3-, and 4-hydroxy groups and Asp, Asn, and Glu residues in the binding sites, while Tyr and Ser residues participate in the recognition mechanism. It was also inferred that SUL-I may form a dimer in solution based on the molecular size estimated via dynamic light scattering as well as possible contact regions in its crystal structure.

Epithelial-mesenchymal transition (EMT) is an evolutionarily conserved cellular program, which is a prerequisite for the metastatic cascade in carcinoma progression. Here, we evaluate the EMT process using the sea urchin Paracentrotus lividus embryo. In sea urchin embryos, the earliest EMT event is related to the acquisition of a mesenchymal phenotype by the spiculogenetic primary mesenchyme cells (PMCs) and their migration into the blastocoel. We investigated the effect of inhibiting the epidermal growth factor (EGF) signaling pathway on this process, and we observed that mesenchyme cell differentiation was blocked. In order to extend and validate our studies, we investigated the migratory capability and the level of potential epidermal growth factor receptor (EGFr) targets in a breast cancer cell line after EGF modulation. Altogether, our data highlight the sensitivity of the sea urchin embryo to anti-EMT drugs and pinpoint the sea urchin embryo as a valuable in vivo model system for studying EMT and the screening of anti-EMT candidates.

Increasing awareness of spatial and temporal variation in ocean pH suggests some marine populations may be adapted to local pH regimes and will therefore respond differently to present-day pH variation and to long-term ocean acidification. In the Northeast Pacific Ocean, differences in the strength of coastal upwelling cause latitudinal variation in prevailing pH regimes that are hypothesized to promote local adaptation and unequal pH tolerance among resident populations. In this study, responses to experimental seawater acidification were compared among six populations of purple sea urchins (Strongylocentrotus purpuratus) inhabiting areas that differ in their frequency of low pH exposure and that prior research suggests are locally adapted to seawater pH. Transcriptomic analyses demonstrate urchin populations most frequently exposed to low pH seawater responded to experimental acidification by expressing genes within major ATP producing pathways at greater levels than populations encountering low pH less often. Multiple genes within the tricarboxylic acid cycle, electron transport chain, and fatty acid beta oxidation pathways were up-regulated in urchin populations experiencing low pH conditions most frequently. These same metabolic pathways were significantly over-represented among genes both expressed in a population-specific manner and putatively under selection to enhance low pH tolerance. Collectively, these data suggest natural selection is acting on metabolic gene networks in order to redirect ATP toward acid-base regulatory processes and enhance tolerance of seawater acidification. As a trade-off, marine populations more tolerant of low pH may have less energy to put toward other aspects of fitness and to respond to additional ocean change. Predator–prey interactions may be strongly influenced by temperature variations in marine ecosystems. Consequently, climate change may alter the importance of predators with repercussions for ecosystem functioning and structure. In North-eastern Pacific kelp forests, the starfish Pycnopodia helianthoides is known to be an important predator of the purple sea urchin Strongylocentrotus purpuratus. Here we investigated the influence of water temperature on this predator–prey interaction by: (i) assessing the spatial distribution and temporal dynamics of both species across a temperature gradient in the northern Channel Islands, California, and (ii) investigating how the feeding rate of P.helianthoides on S.purpuratus is affected by temperature in laboratory tests. On average, at sites where mean annual temperatures were 16°C (equivalent to summer temperatures at sites where P.helianthoides were rare) reduced predation rates regardless of predator and prey sizes, although larger sea urchins were consumed only by large starfishes. These results clearly demonstrate that the effect of P.helianthoides on S.purpuratus is strongly mediated by temperature, and that the local abundance and predation rate of P.helianthoides on sea urchins will likely decrease with future warming. A reduction in top-down control on sea urchins, combined with other expected impacts of climate change on kelp, poses significant risks for the persistence of kelp forests in the future.

SEA (sea urchin sperm protein, enterokinase, agrin) domains, many of which possess autoproteolysis activity, have been found in a number of cell surface and secreted proteins. Despite high sequence divergence, SEA domains were also proposed to be present in dystroglycan based on a conserved autoproteolysis motif and receptor-type protein phosphatase IA-2 based on structural similarity. The presence of a SEA domain adjacent to the transmembrane segment appears to be a recurring scheme in quite a number of type I transmembrane proteins on the cell surface, such as MUC1, dystroglycan, IA-2, and Notch receptors. By comparative sequence and structural analyses, we identified dystroglycan-like proteins with SEA domains in Capsaspora owczarzaki of the Filasterea group, one of the closest single-cell relatives of metazoans. We also detected novel and divergent SEA domains in a variety of cell surface proteins such as EpCAM, α/e-sarcoglycan, PTPRR, collectrin/Tmem27, amnionless, CD34, KIAA0319, fibrocystin-like protein, and a number of cadherins. While these proteins are mostly from metazoans or their single cell relatives such as choanoflagellates and Filasterea, fibrocystin-like proteins with SEA domains were found in several other eukaryotic lineages including green algae, Alveolata, Euglenozoa, and Haptophyta, suggesting an ancient evolutionary origin. In addition, the intracellular protein Nucleoporin 54 (Nup54) acquired a divergent SEA domain in choanoflagellates and metazoans. This article is protected by copyright. All rights reserved.

Using immune cells of sea urchin Strongylocentrotus droebachiensis in early development as a model, the cellular protective mechanisms against ionic and poly(allylamine)-coated silver nanoparticle (AgNPs; 14 ± 6 nm) treatments at 100 μg L-1 were investigated. Oxidative stress, heat shock protein expression, and pigment production by spherulocytes were determined as well as AgNP translocation pathways and their multiple effects on circulating coelomocytes. Sea urchins showed an increasing resilience to Ag over time because ionic Ag is accumulated in a steady way, although nanoAg levels dropped between 48 h and 96 h. A clotting reaction emerged on tissues injured by dissolved Ag (present as chloro-complexes in seawater) between 12 h and 48 h. Silver contamination and nutritional state influenced the production of reactive oxygen species. After passing through coelomic sinuses and gut, AgNPs were found in coelomocytes. Inside blood vessels, apoptosis-like processes appeared in coelomocytes highly contaminated by poly(allylamine)-coated AgNPs. Increasing levels of Ag accumulated by urchins once exposed to AgNPs pointed to a Trojan-horse mechanism operating over 12-d exposure. However, under short-term treatments, physical interactions of poly(allylamine)-coated AgNPs with cell structures might be, at some point, predominant and responsible for the highest levels of stress-related proteins detected. The present study is the first report detailing nano-translocation in a marine organism and multiple mechanisms by which sea urchin cells can deal with toxic AgNPs. International audience; In the coastal ocean, temporal fluctuations in pH vary dramatically across biogeo-graphic ranges. How such spatial differences in pH variability regimes might shape ocean acidification resistance in marine species remains unknown. We assessed the pH sensitivity of the sea urchin Strongylocentrotus purpuratus in the context of ocean pH variability. Using unique male–female pairs, originating from three sites with similar mean pH but different variability and frequency of low pH (pH T ≤ 7.8) exposures, fertilization was tested across a range of pH (pH T 7.61–8.03) and sperm concentrations. High fertilization success was maintained at low pH via a slight right shift in the fertilization function across sperm concentration. This pH effect differed by site. Urchins from the site with the narrowest pH variability regime exhibited the greatest pH sensitivity. At this site, mechanistic fertilization dynamics models support a decrease in sperm–egg interaction rate with decreasing pH. The site differences in pH sensitivity build upon recent evidence of local pH adaptation in S. purpuratus and highlight the need to incorporate environmental variability in the study of global change biology.
