The emergence and sustained transmission of novel pathogens are exerting an increasing demand on the diagnostics sector worldwide, as seen with the ongoing severe acute respiratory coronavirus 2 (SARS-CoV-2) pandemic and the more recent public health concern of monkeypox virus (MPXV) since May 2022. Appropriate and reliable viral inactivation measures are needed to ensure the safety of personnel handling these infectious samples. In the present study, seven commercialized diagnosis buffers, heat (56°C and 60°C), and sodium dodecyl sulfate detergent (2.0%, 1.0%, and 0.5% final concentrations) were tested against infectious SARS-CoV-2 and MPXV culture isolates on Vero cell culture. In this paper, real-time results for a novel continuous-time adaptive tracking controller algorithm for nonlinear multiple input multiple output systems are presented. The control algorithm includes the combination of a recurrent high order neural network with block control transformation using a high order sliding modes technique as control law. A neural network is used to identify the dynamic plant behavior where a filtered error algorithm is used to train the neural identifier. A decentralized high order sliding mode, named the twisting algorithm, is used to design chattering-reduced independent controllers to solve the trajectory tracking problem for a robot arm with three degrees of freedom. Stability analyses are given via a Lyapunov approach.

Chronic kidney disease (CKD) patients with established nephrology care have a high rate of tunneled dialysis catheters (TDC) as first vascular access when transitioning to hemodialysis (HD). We sought to identify factors associated with this problem. Patients who started HD and had prior CKD care within our renal clinic were categorized according to access type at incident HD. Clinical factors, all estimated glomerular filtration rates (eGFR), renal clinic attendance records, hospital admissions in the 6 months preceding HD start, and patient participation in predialysis education course were analyzed. Three hundred thirty-eight patients initiated HD, 107 received pre-HD CKD care within our clinics. Seventy patients started with a TDC. All groups started HD at similar eGFR values. The trajectory of eGFR decline in the 6 months prior to HD start was significantly more rapid in the TDC group. Patients in the TDC group had more acute health events in the prior 6 months. Multivariate modeling showed that failure to attend a predialysis education course and having a more rapid rate of eGFR decline in the 6 months prior to dialysis initiation were both associated with TDC use. Patients with CKD nephrology care who initiated HD with a TDC as first vascular access had a more rapid rate of decline in eGFR in the months preceding dialysis start and were less likely to have attended our predialysis education course. This appears to correspond with the observed increased number of emergency and hospital visits in the 6 months prior to end-stage renal disease.

The use of ferroelectric materials as adsorbents or catalysts could be a promising way to control surface chemical reactions by tuning the ferroelectric polarization. In this context, the oxidation of carbon monoxide over well-defined SrTiO3(100) and BaTiO3/SrTiO3(100) perovskite surfaces was investigated under stoichiometric gas-phase conditions at a total pressure of 3 Torr, in the 100–300 °C temperature range. To this aim, an ultrahigh vacuum-compatible reactor equipped with external laser heating, and specially designed to detect small yields of gaseous products by mass spectrometry, was used. Prior to the catalytic tests, with the help of Auger electron spectroscopy, a sample cleaning procedure under low oxygen pressure was established in order to remove carbon impurities. The as-prepared surfaces appear poorly active in CO oxidation, but their activity increases if a high-temperature pre-annealing treatment under ultrahigh vacuum is applied in order to produce oxygen vacancies, which act as adsorption and/or reaction sites. The activity of the bare SrTiO3 substrates, whether doped with Nb or not, is similar to that of the supported BaTiO3 films, but the latter lose less activity upon evacuation/reaction cycles. Isotopic labeling experiments using 18O2 show that the reaction involves CO (and O2) dissociation, and that lattice oxygen participates to the reaction via the so-called Mars–van Krevelen mechanism. Anomalous small angle X-ray scattering is used to determine the distribution of divalent ions in a neutralized polyelectrolyte gel of sodium polyacrylate in the vicinity of the volume transition. At the five different energies of the incident beam used to vary the contrast, the scattering curves have similar shapes, and are separated only by constant multiplying factors. This result, in conjunction with SANS results from the same sample, indicates that the divalent ion (strontium) is confined on the polymer backbone.

Elastomers are soft materials that can be reinforced by dispersing into them nanosized solid particles. Common examples of the latter are silica or carbon black aggregates. However, the mechanism of reinforcement is still not yet fully understood. Our work consists in investigating by small-angle X-ray scattering (SAXS) the structure of the aggregate network spreading throughout the matrix in the initial sample and its modification during and after straining (elongation). The goal is to relate the macroscopic mechanical behaviour with the structure of the aggregate network. The present paper is a qualitative overview of recent results obtained on well defined composites.

HSV infections are treated efficiently and prevented by acyclovir, although resistant strains have been reported. Resistance to acyclovir involves mainly mutations in the viral gene encoding thymidine kinase; mutations may lead to an altered or, more frequently, deficient TK. These acyclovir-resistant TK deficient strains are not able to reactivate from a latent infection in an experimental model, compared to TK positive strains. A case is reported of a bone marrow transplant child who developed HSV infection at 11 days post-transplantation. Acyclovir-resistant HSV 1 was isolated on day 19 post-transplantation. The patient was cured of his infection. A resistant virus was detected 20 months later that harboured the same TK gene mutation as the first resistant virus. This mutation is an insertion of one guanine in a homopolymer repeat of seven guanines located at codon 146 of TK. It has previously been reported and associated with the expression of a deficient TK activity and the ability to reactivate in mice. These results corroborate the clinical relevance of this mutation, which is associated with acyclovir-resistant recurrent infections in humans.

An HIV-negative infant presented with VZV primary infection during the maintenance therapy for mega-karyoblastic leukaemia. The lesions were initially vesicular and necrotic but became verrucous and hyperkerato tic. A clinical resistance to acyclovir was suspected and confirmed by histologic and virologic studies. The patient was successfully treated by foscarnet. Conclusion: resistance of VZV to acyclovir may occur after a short treatment in a non-AIDS patient. ± Acute leukaemia, acyclovir, foscarnet, Varicella zoster virus

Presently, several works question the effects of dehydroepiandrosterone (DHEA) reported in vivo and designate its 7-hydroxylated metabolites as native antiglucocorticoids and potent mediators in the triggering of immune response. Among mouse tissues and organs, and second to liver, the largest production of 7alpha-and 7beta-hydroxylated derivatives of DHEA takes place in brain microsomes. To contribute to identification of cytochromes P450 (CYPs) responsible for 7alpha- and 7beta-hydroxy-DHEA production, effects of CYP inhibitors and of several steroid hormones on DHEA 7-hydroxylation were examined. Using mouse brain microsomes as a source of enzyme, we report now that strong and smaller inhibitions of DHEA 7alpha-hydroxylation were obtained with ketoconazole and alpha-naphthoflavone, respectively, and that neither changed DHEA 7beta-hydroxylation. Metyrapone and antipyrine also inhibited 7alpha-hydroxylation, but by contrast, significantly increased 7beta-hydroxylation of DHEA. This indicated that at least, two different CYPs were responsible for 7alpha- and 7beta-hydroxylation of DHEA. Steroids sharing a 3beta-hydroxylated structure with DHEA, namely pregnenolone, 5-androstene-3beta,17beta-diol and 3beta-hydroxy-5alpha-androstan-17-one, were strong inhibitors of DHEA 7alpha-hydroxylation (non-competitive inhibition with pregnenolone, Ki=2.0 +/- 0.3 microM). In contrast, 7beta-hydroxylation yields were not decreased by the 3beta-hydroxysteroids tested. Moderate inhibition of 7alpha- and 7beta-hydroxylation was obtained with 3-oxosteroids, namely testosterone, progesterone, corticosterone and 4-androsten-3,17-dione. Taken together, these data indicate specific inhibition patterns of DHEA 7alpha- and 7beta-hydroxylation by CYP inhibitors and steroid hormones in mouse brain microsomes and may be used as criteria necessary for identification of the responsible CYP species.

In many tissues from different species, pregnenolone and dehydroepiandrosterone (DHEA) are hydroxylated mainly at the 7 alpha position by a cytochrome P450 (P450)-containing microsomal enzyme complex. In addition, 7-hydroxysteroids have been shown to activate immune processes in mice. The reported production of 7 beta-hydroxypregnenolone and 7 beta-hydroxy-DHEA was not supported by formal identification, and the P450 responsible for 7 alpha-hydroxylation and 7 beta-hydroxylation of pregnenolone and DHEA have not been identified. Based on results of analyses by crystallization to constant specific activity and gas chromatography/mass spectrometry, we report that mouse-liver and mouse-brain microsomes carried out 7 beta-hydroxylation of pregnenolone and DHEA, and that yeast-expressed mouse cytochrome P450-1A1 (P450 1A1) transformed pregnenolone into 7 beta-hydroxypregnenolone (Km = 25.1 +/- 0.4 microM, turnover number = 979 +/- 30 pmol.min-1.nmol-1 mouse P450 1A1). Neither 7-hydroxy derivatives of DHEA nor 7 alpha-hydroxypregnenolone was produced by P450 1A1. The presence of P450 1A1 in liver and brain microsomes was shown by Western blot analysis, and induction of mouse P450 1A1 by beta-naphthoflavone resulted in increased 7 beta-hydroxylation of pregnenolone in liver microsomes. Studies of the brain-microsome 7 beta-hydroxylating enzyme with pregnenolone or DHEA gave Km of 5.0 microM and 4.9 microM, respectively, and Vmax of 4.5 pmol.min-1.mg-1 and 6.1 pmol.min-1.mg-1, respectively, and showed the absence of cross-inhibitions between the two steroids. These findings indicate that, in addition to unidentified P450, P450 1A1 is involved in 7 beta-hydroxylation of pregnenolone and may contribute in part to the production of the 7-hydroxylated steroids necessary for activation of immune defenses.

Results Median age 20 months (±12 months), 15 female (50%) 15 male(50%). IgA2 deficiency 16 (53%), IgG4 and IgA2 deficiencies 13 (43%), IgG4 deficiency 27 (90%), 29 patients had distal renal tubular acidosis (96%) 1 patient with proximal renal tubular acidosis. About food allergies, cow’s milk 28(93%), egg 18 (60%), wheat 0, corn 8 (26.6%), soy 2 (6.6%), non IgE mediated to cow’s milk 6 (20%), egg 6 (20%), soy 7 (23%), wheat 4 (13%), corn 1 (3.3%). The formula used to cow’s milk allergy was aminoacid based formula 19 (63.3%), extensively hydrolized formula 11 (36.6%). Gastroesophageal reflux was associated in 83.3%, wheezing 75%, diarrhea 33.3%, atopic dermatitis 25%, rhinitis 83.3%, urticaria 19.4%, constipacion 2.8%, blood stools 2.8%.
