We have previously demonstrated that the rate of fluid shear stress (ΔSS) can manipulate the fate of mesenchymal stem cells (MSCs) to osteogenic or chondrogenic cells. However, whether ΔSS is comparable to other two means of induction medium and substrate stiffness that have been proven to be potent in differentiation control is unknown. In this study, we subjected MSCs to 1-7 days of osteogenic or chondrogenic chemical induction, or 1-4 days of 37 or 86 kPa of substrate stiffness induction, followed by 20 min of Fast ΔSS (0-0') or Slow ΔSS (0-2'), which is a laminar FSS that linearly increased from 0 to 10 dyn/cm 2 in 0 (Fast) or 2 min (Slow) and maintained at 10 dyn/cm 2 for a total of 20 min. We found that 20 min of ΔSS could compete with 5 days' chemical and 2 days' substrate stiffness inductions. Our study confirmed that ΔSS is a powerful tool to control the differentiation of MSCs, which stressed the possible application in MSCs linage specification.

In this study, effect of swirling addition on the liquid mixing behavior of multi-orifice-impinging transverse jet mixer has been investigated by planar laser induced fluorescence as well as large eddy simulation (LES). In the case of swirling addition into the jet flow, there exists an optimized swirling jet angle or optimized jet-to-cross velocity ratio for the fixed mixer configuration. A larger swirling jet angle will make the flow dominated by the swirling, resulting in a slower mixing process. Interaction of swirling crossflow with no-swirling injected streams, or with swirling injected streams in the opposite direction is beneficial for the mixing. LES predictions show that many small vortices are produced homogenously due to intensified impingement in the case of opposite swirling directions, leading to a relative fast mixing process in several milliseconds. Whereas the mixing is restrained when the swirling directions of two flows are the same.

Caryolanes, known as β-caryophyllene alcohols, are widely occurring sesquiterpenes in plants. From the cultures of marine Streptomyces sp. AH25, two new caryolane sesquiterpenes, micaryolanes A and B (1 and 2), together with caryolan-1,9β-diol (3) were isolated. Their structures were elucidated by extensive analyses of HR-MS and NMR spectroscopic data. The absolute configurations were assigned via the CD data of the in situ formed [Rh2 (OCOCF3 )4 ] complex and supported by comparison of experimental and calculated specific rotation values. Compounds 1-3 exhibited no activities against Hep3B or MG-63 cell lines or against Gram-positive and Gram-negative bacteria. The results not only enriched the caryolane family, but also proved bacteria as a productive source of terpene metabolites.

HSPC159 is a novel human galectin‐related protein that has been shown to be involved in carcinogenesis. Little is known about HSPC159 expression and function in breast cancer. Herein we showed that HSPC159 was aberrantly expressed in both breast cancer cell lines and tumor tissues and that its expression was associated with poor prognosis of breast cancer patients. Using gain‐ and loss‐of‐function methods we found that HSPC159 enhanced breast cancer cell proliferation and metastasis in vitro and in vivo. Mechanistically, HSPC159 was found to induce epithelial‐mesenchymal transition (EMT) and the F‐actin polymerization process of breast cancer cells. Moreover, HSPC159 promoted proliferation, migration and invasion through activating the PI3K/Akt signaling pathway in breast cancer. In conclusion, our findings showed that HSPC159 contributed to breast cancer progression through the PI3K/Akt pathway and might serve as a potential therapeutic target for the treatment of breast cancer.
