Abstract

International audience; Natural products from marine animals show high potential for the development of new medicines, but drug development based on these compounds is commonly hampered by their low natural abundance. Since many of these metabolites are suspected or known to be produced by uncultivated bacterial symbionts, the rapidly growing diversity of sequenced prokaryotic genomes offers the opportunity to identify alternative, culturable sources of natural products computationally. In this work, we investigated the potential of using this sequenced resource to facilitate the production of meroterpenoid-like compounds related to those from marine sources. This genome-mining strategy revealed a biosynthetic gene cluster for highly modified cytotoxic meroterpenoids related to pelorol and other compounds isolated from sponges. Functional characterization of the terpene cyclase MstE showed that it generates an ent-sterol-like skeleton fused to an aryl moiety from an open-chain precursor and is therefore a promising tool for the chemoenzymatic preparation of synthetically challenging chemical scaffolds.

Abstract

The sea star Oreaster reticulatus contains, along with the known ophidiacerebrosides C–E, nine new glycosphingolipids named oreacerebrosides A–I. Their structures were elucidated by a combination of spectroscopic methods and chemical degradation. All compounds contain a 4,8,10-triunsaturated sphingoid base. Oreacerebrosides A–C have a β-glucopyranoside as the sugar residue, as found in ophidiacerebrosides C–E and in all known compounds of this type; in contrast, oreacerebrosides D–I are the first examples of β-galactosylceramides containing this unusual sphingoid base. Four representative glycosphingolipids were tested for cytotoxic activity on rat glioma C6 cells and were shown to be mildly cytotoxic; the glucosylceramides were more active than the galactosylceramides. In addition, oreacerebroside I, but not ophidiacerebroside E, was shown to exert proangiogenic activity and was able to increase VEGF-induced human endothelial cell proliferation.(© Wiley-VCH Verlag GmbH & Co. Abstract

Electrospray ionization (ESI) and liquid secondary ionization (LSI) mass spectrometry were applied for characterization of glycosphingolipids (GSLs) isolated in their peracetylated form from four Agelas marine sponge species. Since peracetylated glycosphingolipids are not soluble in solvents traditionally used for ESI, lithium chloride was added to the samples in order to obtain lithium cationized molecules. Although the preferred fragmentation seems to be the sequential loss of acetic acid molecules, it was found that tandem mass spectra obtained from peracetylated diglycosyl ceramides might provide direct information about the structure of the long-chain base (formation of W''/Z0 fragments). The utility of ESI and LSI in the analysis of these compounds has also been compared. It was found that the tandem mass spectra obtained by LSI-MS/MS experiments could provide information about the chain-length (carbon atom number) variations within a certain ceramide mass. Thus, from one of our samples, 25 different ceramide compositions have been identified from 8 precursor (Z0) ions. Comparison of the two ionization modes (LSI and ESI) highlights the fact that molecular mass distributions obtained by LSI-MS, especially the presence of unsaturated species, have to be interpreted carefully. For the first time a direct high-performance liquid chromatography (HPLC)/ESI-MS method was used for characterization of complex mixtures of peracetylated GSLs. The results demonstrate that HPLC/ESI-MS is able to analyze mono- and diglycosylated GSLs, and other kinds of glycolipids that are actually present in the sample. Abstract

Three new glycosphingolipids (2a−4a) were isolated as their peracetates (2b−4b) from the marine ascidian Microcosmus sulcatus, and their structures were elucidated by spectroscopic and chemical analysis. Abstract

In order to obtain N-benzyl-3,5-dicarbethoxy-2,6-dimethyl-4-phenyl-1,4-dihydropyridine 1 as a lead compound of pharmacological interest, the classical Hantzsch synthetic method and the modified Collie procedure were used. However, only a very low yield of 1 was obtained in a mixture with larger amounts of some by-products (2, 3, 4). Compound 1 was then synthesized in satisfactory yield via a different method, together with another by-product (9). The structures of all by-products were elucidated and possible mechanisms leading to the formation of such compounds are described.

Abstract

A specimen of the marine sponge Agelas longissima was analyzed for glycosphingolipids. Three cerebrosides 1a-3a were isolated, each as a mixture of homologs. The structure of the new glycosphingolipid 1a was elucidated by extensive NMR studies of its peracetate derivative and chemical analysis. Compound 2a, previously isolated from another specimen of A. longissima, exhibits in the new specimen a remarkably different alkyl chain composition.

Abstract

Colorectal cancer (CRC) is one of the most common malignant tumours worldwide. Diarylheptanoids, secondary metabolites isolated from Zostera marina, are of interest in natural products research due to their biological activities. Zosterabisphenone B (ZBP B) has recently been shown to inhibit the viability of CRC cells. The aim of this study was to investigate the therapeutic potential of ZBP B for targeting human CRC cells. Cell viability was determined using the MTT assay. Flow cytometry and Western blot analyses were used to assess apoptosis and autophagy. A CRC xenograft model was used to evaluate the in vivo effect of ZBP B. No cytotoxic effect on HCEC cells was observed in the in vitro experiments. ZBP B caused morphological changes in HCT116 colon cancer cells due to an increase in early and late apoptotic cell populations. Mechanistically, ZBP B led to an increase in cleaved caspase-3, caspase-8, caspase-9, PARP and BID proteins and a decrease in Bcl-2 and c-Myc proteins. In the xenograft model of CRC, ZBP B led to a reduction in tumour growth. These results indicate that ZBP B exerts a selective cytotoxic effect on CRC cells by affecting apoptotic signalling pathways and reducing tumour growth in mice.
