Anthracnose disease of sorghum is caused by Colletotrichum sublineola, a filamentous fungus. The genetic basis of resistance to anthracnose in sorghum is largely unclear, especially in Senegalese sorghum germplasm. In this study, 163 Senegalese sorghum accessions were evaluated for response to C. sublineola, and a genome‐wide association study (GWAS) was performed to identify genetic variation associated with response to C. sublineola using 193,727 single nucleotide polymorphisms (SNPs) throughout the genome. Germplasm diversity analysis showed low genetic diversity and slow linkage disequilibrium (LD) decay among the Senegalese accessions. Phenotypic analysis resulted in relatively low differences to C. sublineola among the tested population. Genome‐wide association study did not identify any significant association based on a strict threshold for the number of SNPs available. However, individual analysis of the top eight SNPs associated with relative susceptibility and resistance identified candidate genes that have been shown to play important roles in plant stress tolerance in previous studies. This study identifies sorghum genes whose annotated properties have known roles in host defense and thus identify them as candidates for use in breeding for resistance to anthracnose.

PURPOSE Use standardized methods to determine how assessment of protective value of radiation-protective garments changes under conditions employing standard beam qualities, scatter-mimicking primary beams, and a modified Hp (10) measurement. METHODS The shielding properties of radiation-protective garments depend on the spectrum of beam energies striking the garment and the attenuation properties of materials used to construct the garment, including x-ray fluorescence produced by these materials. In this study the primary beam spectra employed during clinical interventional radiology and cardiology procedures (clinical primary beams, CPB) were identified using radiation dose structured reports (RDSR) and fluoroscope log data. Monte Carlo simulation was used to determine the scattered radiation spectra produced by these CPB during typical clinical application. For these scattered spectra, scatter-mimicking primary beams (SMPB) were determined using numerical optimization-based spectral reconstruction that adjusted kV and filtration to produce the SMPB that optimally matched the scattered spectrum for each CPB. The penetration of a subset of SMPB through four radiation-protective garments of varying compositions and nominal thicknesses was measured using a geometry specified by the International Electrotechnical Commission (IEC). The diagnostic radiological index of protection (DRIP), which increases with increasing penetration through a garment, was calculated using these measurements. Penetration through the same garments was measured for standard beams specified by the American Society of Testing and Materials (ASTM). Finally, 10 mm of PMMA was affixed to the inside of each garment and the DRIP remeasured in this configuration to simulate Hp (10). RESULTS The SMPB based on actual CPB were in general characterized by lower kV (range 60-76) and higher half-value layer (HVL, range 3.44-4.89 mm Al) than standard beam qualities specified by ASTM (kV range 70-85; HVL range 3.4-4.0 mm Al). A lead garment of nominal thickness 0.5 mm (D) had a DRIP of 0.8%, two lead-free garments of 0.5 mm nominal thickness had DRIPs of 1.2% (A) and 2.2% (B), and a lead-free bilayer (C) had a DRIP of 1.4%. When standard beam qualities specified by the ASTM were used, the DRIP for D was 2.2%, 175% higher than the DRIP measured using SMPB, and for A, B, and C was 2.8%, 3.2%, and 2.9%, respectively. This was 133%, 45%, and 107% higher than the DRIP measured using SMPB. Differences between the DRIP of lead-alternative garments and the lead garment were reduced when measured with 10 mm of PMMA. Using this method, the measured DRIPs were 2.2% (A), 3.1% (B), 2.5% (C), and 2.3% (D). CONCLUSIONS Penetration of radiation through radiation-protective garments depended strongly on the methods and X-ray spectra used for evaluation. The DRIP was higher (i.e., protective value was lower) for lead-alternative garments than for lead garments in this evaluation. The DRIP was lower for all garments when SMPB based on actual clinical beam quality data were used to measure penetration compared to ASTM standard beams. Differences in penetration between lead-alternative and lead garments were less when the DRIP was measured with 10 mm of PMMA between the garment and the chamber.

Purpose: To evaluate the sensitivity of the diagnostic radiological index of protection (DRIP), used to quantify the protective value of radioprotective garments, to procedural factors in fluoroscopy in an effort to determine an appropriate set of scatter-mimicking primary beams to be used in measuring the DRIP. Methods: Monte Carlo simulations were performed to determine the shape of the scattered x-ray spectra incident on the operator in different clinical fluoroscopy scenarios, including interventional radiology and interventional cardiology (IC). Two clinical simulations studied the sensitivity of the scattered spectrum to gantry angle and patient size, while technical factors were varied according to measured automatic dose rate control (ADRC) data. Factorial simulations studied the sensitivity of the scattered spectrum to gantry angle, field of view, patient size, and beam quality for constant technical factors. Average energy (Eavg) was the figure of merit used to condense fluence in each energy bin to a single numerical index. Results: Beam quality had the strongest influence on the scattered spectrum in fluoroscopy. Many procedural factors affect the scattered spectrum indirectly through their effect on primary beam quality through ADRC, e.g., gantry angle and patient size. Lateral C-arm rotation, common in IC, increased the energy of the scattered spectrum, regardless of the direction of rotation. The effect of patient size on scattered radiation depended on ADRC characteristics, patient size, and procedure type. Conclusions: The scattered spectrum striking the operator in fluoroscopy is most strongly influenced by primary beam quality, particularly kV. Use cases for protective garments should be classified by typical procedural primary beam qualities, which are governed by the ADRC according to the impacts of patient size, anatomical location, and gantry angle.

The productivity and profitability of sorghum [ (L.) Moench] is reduced by susceptibility to fungal diseases, such as anthracnose ( P. Henn.). A limited number of resistant accessions are present in the temperate-adapted germplasm; other exotic sources of resistance are not currently available for breeding programs. Among 335 accessions available to breeders from a previously genotyped sorghum association panel (SAP), we found that 75 were resistant to anthracnose. A phylogenetic analysis of these accessions showed high genetic diversity and multiple resistance sources. Genome-wide association scans (GWAS) were conducted using 268,289 single-nucleotide polymorphisms to identify loci associated with anthracnose resistance. Using logistic regressions for binary measures of resistance responses, we identified three loci within a region on chromosome 5 that have been previously associated with three sources of anthracnose resistance. A GWAS limited to Caudatum germplasm identified an association with a region on chromosome 1 and with the same previous region on chromosome 5. Candidate genes within these loci were related to R-gene families, signaling cascades, and transcriptional reprogramming, suggesting that the resistance response is controlled by multiple defense mechanisms. The strategic integration of exotic resistant germplasm into the SAP is needed to identify additional rare resistance alleles via GWAS.

Rationale Perfluorocarbons such as perfluoromethylcyclohexane (c-C6F11-CF3) are important man-made chemicals that have many uses including plasma processing, blood substitutes and atmospheric tracers. It is important to know the kinetics and thermodynamics of the reactions of thermal electrons with these molecules since they are potentially harmful greenhouse gases that can accumulate in the atmosphere. Methods The least-squares fits of the temperature dependence of electron-capture detection and atmospheric pressure negative ion mass spectrometry to a kinetic model are used to determine the electron affinities of c-C6F11-CF3, activation energies for the formation of c-C6F10-CF3, and c-C6F11 anions and single bond dissociation energies. These are supported by semi-empirical quantum mechanical calculations. These techniques were previously used to characterize superoxide, NO and SF6 anions. Results The literature electron affinities: (eV) c-C6F11-CF3, 1.06, c-C6F10-CF3, 3.9, c-C6F11, 3.5 and D(R1-CF3), 3.8; D(R-F), 4.3 are supported. Additional electron affinities for c-C6F11-CF3, from 0.5 to 1.5 eV are assigned to excited states. The ground state electron affinity is 3.0(2) eV from the photodetachment threshold. Pseudo one-dimensional anionic Morse potentials illustrating the mechanism for the reaction of thermal electrons with c-C6F11-CF3 are presented. The major anion peaks in perfluorokerosene-L are identified. An experimental setup for studying thermal electron capture reactions at variable temperatures, pressures and concentrations proposed by Herder in 2004 is presented. Conclusions There are multiple anions of c-C6F11-CF3 more stable than the neutral. Electron-capture detection and atmospheric pressure negative ion mass spectrometry are effective general methods for determining multiple electron affinities similar to the photodetachment, flowing afterglow, magnetron, negative surface ionization, swarm and beam procedures. Semi-empirical theoretical calculations support experimental results. The USDA-ARS National Plant Germplasm System maintains a Zimbabwe sorghum collection of 1235 accessions from different provinces. This germplasm has not been extensively employed in US breeding programmes due to the lack of phenotypic and genetic characterization. Therefore, 68 accessions from Zimbabwe were phenotyped, and evaluated for their anthracnose response for two consecutive years, and genetically characterized with 21 simple sequence repeat markers. Phenotypic analysis showed significant differences among accessions with plant height and panicle length being the most variable traits. Likewise, 25 accessions were anthracnose resistant, nine showed variable responses and 34 were susceptible. Genetic analysis identified 174 alleles with an average of 8.3 alleles and 11.8 genotypes per locus and a polymorphic information content of 0.60. These results reflect a moderate genetically diverse germplasm. Neighbour-joining clustering analysis revealed that the majority of anthracnose-resistant accessions showed high genetic relatedness; therefore, this germplasm might represent one to six new sources of resistances. Results presented herein show that the Zimbabwe collection contains valuable germplasm for breeding programmes and is an important source of anthracnose resistance.

Leaves and stalks of many sorghum genotypes accumulate dark red or purple pigments upon wounding while some plants, called ‘tan,’ do not. Grains with unpigmented ‘white’ pericarps grown on tan plants are more desirable for food. The hypothesis tested was that pigments in plants protected grain against the panicle diseases grain mould and head smut. Near-isogenic tan or purple plant colour genotypes with white grain were planted at Lincoln and Ithaca, NE and Corpus Christi, TX. The field grown grain was plated onto semi-selective media to detect the presence of grain colonisation by mould genera Alternaria, Fusarium and Curvularia .M oreFusarium and Curvularia spp. were recovered from grain grown at Corpus Christi than the Nebraska locations; however, there was no indication that the grain from purple plants was more resistant to the three fungal genera. Most fungi were identified morphologically as Alternaria alternata. Molecular identification of Fusarium species, using translation elongation factor 1-α gene sequences, showed that Fusarium thapsinum and Fusarium proliferatum infected grain at all three locations. Head smut disease of panicles, caused by the fungus Sporisorium reilianum, was assessed at Corpus Christi; surprisingly, purple plants had significantly greater disease incidence than tan plants. We propose that the tan plant colour lines with white grain are promising for development of foodgrade sorghums not more susceptible than pigmented lines to grain mould and head smut.

Summary objectives To test the hypothesis that Artesunate–mefloquine paediatric (AS+MEF) is as effective as Artemether–lumefantrine (AL) in treating acute uncomplicated malaria in children. methods In an open label, randomized controlled clinical trial, children aged 6–59 months were randomized to receive AS+MEF or AL. Both drug regimens were given for 3 days, and follow-up was for 28 days. The primary endpoint was the 28-day cure rate and was defined as proportion of patients with PCR-corrected cure rate after 28 days of follow-up. results One hundred and fifty-six patients with confirmed uncomplicated P. falciparum malaria were randomly assigned to receive AS+MEF (n = 77) or AL (n = 79). PCR-corrected day 28 cure rates for per protocol (PP) populations were 99% for AS+MEF and 97% (P = 1) for AL. For the intention to treat (ITT) population, cure rates were 96% for AS+MEF and 92% (P = 0.49) for AL. Both regimens were well tolerated. conclusion AS+MEF is as effective as AL, and both combinations were efficacious and safe.

The apolipoprotein E (apoE)-derived peptide (141-155)2 has a neurotoxic effect, implying that apoE itself could be a source of toxicity in Alzheimer's disease brain. We characterized the toxicity of this peptide on superior cervical ganglion (SCG) neurons and compared the death with the apoptotic death that occurs after nerve growth factor (NGF) deprivation in these cells. A dose of 10 microM apoE (141-155)2 resulted in the death of approximately 50% of the neurons within 24 h. Nuclear condensation and DNA fragmentation preceded the death. However, most inhibitors of NGF deprivation-induced death, including the caspase inhibitor Boc-aspartyl(O-methyl)fluoromethyl ketone and genetic deletion of bax-/-, had no effect on the toxicity. Inclusion of depolarizing levels of potassium did block the toxicity. Receptor-associated peptide (RAP), an antagonist for apoE receptors, did not protect cells in either SCG or hippocampal cultures. In addition, RAP had no effect on internalization of the apoE peptide. These data support the observation that apoE (141-155)2 is neurotoxic but suggest that the neurotoxicity is distinct from classical apoptosis or necrosis. Furthermore, these results indicate that the toxic effect may occur independently of members of the low-density lipoprotein receptor gene family.

Ships’ protests have been used for centuries as legal documents to record and detail damages and indemnify Captains from fault. We use them in this article, along with data extracted through forensic synoptic analysis (McNally, 1994, 2004) to identify a tropical or subtropical system in the North Atlantic Ocean in 1785. They are shown to be viable sources of meteorological information. By comparing a damaging storm in New England in 1996, which included an offshore tropical system, with one reconstructed in 1785, we demonstrate that the tropical system identified in a ship’s protest played a significant role in the 1785 storm. With both forensic reconstruction and anecdotal evidence, we are able to assess that these storms are remarkably identical. The recurrence rate calculated in previous studies of the 1996 storm is 400–500 years. We suggest that reconstruction of additional years in the 1700s would provide the basis for a reanalysis of recurrence rates, with implications for future insurance and reinsurance rates. The application of the methodology to this new data source can also be used for extension of the hurricane database in the North Atlantic basin, and elsewhere, much further back into history than is currently available.

Radiation-induced skin burns can be produced by high doses of fluoroscopic X rays. Though uncommon, such injuries can cause considerable distress to the patient and they can lead to deep ulcers requiring skin grafts. Factors that increase the chance of a burn can be readily identified and in nearly all instances they can be avoided or minimized. We discuss these issues and use case illustrations to point out how burns can be avoided.

Purpose: Ionization chambers remain the standard for calibration of air-kerma rate measuring devices. Despite their strong energy-dependent response, solid state radiation detectors are increasingly used, primarily due to their efficiency in making standardized measurements. To test the reliability of these devices in measuring air-kerma rates, we compared ion chambers measurements with solid-state measurements for various mobile fluoroscopes operated at different beam qualities and air-kerma rates. Methods: Six mobile fluoroscopes (GE OEC models 9800 and 9900) were used to generate test beams. Using various field sizes and dose rate controls, copper attenuators and a lead attenuator were placed at the image receptor in varying combinations to generate a range of air-kerma rates. Air-kerma rates at 30 centimeters from the image receptors were measured using two 6-cm3 ion chambers with electrometers (Radcal, models 1015 and 9015) and two with solid state detectors (Unfors Xi and Raysafe X2). No error messages occurred during measurements. However, about two months later, one solid-state device stopped working and was replaced by the manufacturer. Two out of six mobile fluoroscopic units were retested with the replacement unit. Results: Generally, solid state and ionization chambers agreed favorably well, with two exceptions. Before replacement of the detector, the Xi meter when set in the “RF High” mode deviated from ion chamber readings by factors of 2 and 10 with no message indicating error in measurement. When set in the “RF Low” mode, readings were within −4% to +3%. The replacement Xi detector displayed messages alerting the user when settings were not compatible with air-kerma rates. Conclusion: Air-kerma rates can be measured favorably well using solid-state devices, but users must be aware of the possibility that readings can be grossly in error with no discernible indication for the deviation.

Recently, preinduction of the heat shock response has been shown to protect CNS neurons undergoing various stressful insults, e.g., heat, ischemia, or exposure to excitotoxins. However, it is not known which of the proteins induced by the heat shock response mediate the protective effects. Previous correlative evidence points to a role for the highly stress-induced 72-kDa heat shock protein (hsp72). However, it is not known whether hsp72 expression alone can protect against a range of acute neuronal insults. We constructed a herpes simplex virus-1 vector carrying the rat brain stress-inducible hsp72 gene and the Escherichia coli lacZ (marker) gene. Infection with the vector caused hippocampal neurons to coexpress hsp72 and beta-galactosidase. Infection with a control vector led to marker gene expression only. Overexpression of hsp72 protected cultured hippocampal neurons against a heat shock but not against the metabolic toxin 3-nitropropionic acid or the excitotoxin glutamate. This is the first published report of protection following heat shock protein transfection in CNS neurons.

Doubly alkylated ketones can be prepared from methyl ketones and two different alcohols by using a sequential one-pot iridium- and rhodium-catalyzed process [reaction of (VI)].

Purpose: To develop a new algorithm for multiple channel radiochromic film calibration and to evaluate the accuracy of the algorithm with virtual simulated film images. Methods: The proposed calibration method was based on least-square optimization of discrepancies between measured and estimated optical density of the three colour channels, with the inclusion of the weighted dose gradient as penalty to this minimization problem. The regularization parameter λ was selected using the L-curve method to reduce the L2-norm of the dose gradient while preserving the data consistency. The situation of λ = 0 represented no regularization and the least-square solution would be obtained. The algorithm was tested with 4 virtual film images which were produced from dose planes exported from the planning system. Zero-mean Gaussian noise were added to simulate the real situation. The resultant dose maps calculated by both least-square method and regularization method were then compared with the original dose planes. Results: Comprehensive study of the 4 simulated cases with the L-curve method yielded an optimal regularization parameters of 2×10−8. The dose planes generated by least-square method showed significant noise retained when compared with that from the regularization method. In performing Gamma Index analysis with tolerance of 1% and 1 mm, it was observed that in the least-square solution, failed points exist everywhere on the dose plane, while only a small amount in the sharp dose gradient region of the regularized solution. It was also noted that the number of failed point in the least-square solution can be 20 to 150 times more than that in the regularized solution. Conclusion: The calibration algorithm proposed in this study successfully recovered the original dose plane from the noise contaminated film image with the regularization method. Regularization should be a better option to the least-square one in solving the radiochromic film calibration problem.

Approximately 4000 women per year in the United States require radiotherapy during pregnancy. This report presents data and techniques that allow the medical physicist to estimate the radiation dose the fetus will receive and to reduce this dose with appropriate shielding. Out-of-beam data are presented for a variety of photon beams, including cobalt-60 gamma rays and x rays from 4 to 18 MV. Designs for simple and inexpensive to more complex and expensive types of shielding equipment are described. Clinical examples show that proper shielding can reduce the radiation dose to the fetus by 50%. In addition, a review of the biological aspects of irradiation enables estimates of the risks of lethality, growth retardation, mental retardation, malformation, sterility, cancer induction, and genetic defects to the fetus.

The olefin metathesis reaction has underpinned spectacular achievements in organic synthesis in recent years. Arguably, metathesis has now become the foremost choice for a carbon–carbon double bond disconnection. Despite this general utility, de novo routes to heteroaromatic compounds using the cross-metathesis (CM) reaction have only recently emerged as an efficient strategy. This approach allows a convergent union of simple, functionalised, three- to four-carbon olefinic core building blocks, to generate furans, pyrroles and pyridines with a high degree of control of substitution pattern in the product.

Heck reactions are performed on α,β-unsaturated -δ-sulfonamido intermediates (III), derived from cross-metathesis, to allow the installment of substituents at the β-position.

Purpose: Lightweight lead-free or lead-composite protective garments exploit k-edge interactions to attenuate scattered X-rays. Manufacturers specify the protective value of garments in terms of lead equivalence at a single kVp. This is inadequate, as the protection provided by such garments varies with radiation quality in different use conditions. We present a method for matching scattered X-ray spectra to primary X-ray spectra. The resulting primary spectra can be used to measure penetration through protective garments, and such measurements can be weighted and summed to determine a Diagnostic Radiation Index for Protection (DRIP). Methods: Scattered X-ray spectra from fluoroscopic procedures were modeled using Monte Carlo techniques in MCNP-X 2.7. Data on imaging geometry, operator position, patient size, and primary beam spectra were gathered from clinical fluoroscopy procedures. These data were used to generate scattered X-ray spectra resulting from procedural conditions. Technical factors, including kV and added filtration, that yielded primary X-ray spectra that optimally matched the generated scattered X-ray spectra were identified through numerical optimization using a sequential quadratic programming (SQP) algorithm. Results: The primary spectra generated with shape functions matched the relative flux in each bin of the scattered spectra within 5%, and half and quarter-value layers matched within 0.1%. Themore » DRIP for protective garments can be determined by measuring the penetration through protective garments using the matched primary spectra, then calculating a weighted average according to the expected clinical use of the garment. The matched primary spectra are specified in terms of first and second half-value layers in aluminum and acrylic. Conclusion: Lead equivalence is inadequate for completely specifying the protective value of garments. Measuring penetration through a garment using full scatter conditions is very difficult. The primary spectra determined in this work allow for practical primary penetration measurements to be made with equipment readily available to clinical medical physicists.« less

Background The prognosis of children with high-grade astrocytomas of the central nervous system is grim and has not been substantially improved by conventional chemoradiotherapy. We performed a multi-institutional phase I study to determine the toxicities and tolerance of concurrent external beam radiation of the brain and a unique dose-schedule of paclitaxel as a radiation sensitizer. Procedure Paclitaxel was delivered intravenously as a continuous 24 h/day, 7 days/week infusion during the entire 6-week course of fixed schedule standard radiation therapy. The dose of paclitaxel was escalated in patient cohorts in standard phase I design. Results Eleven patients (eight brain stem gliomas, one glioblastoma multiforme, and two gliomatosis cerebri) were treated. Dose-limiting toxicity was encountered in the two patients treated at 6 mg/(m2/24 h), both of whom developed severe obstipation requiring prolonged hospitalization. Conclusions We have shown in this first study of its kind that paclitaxel can be administered safely to children as a 6-week continuous infusion concurrent with cranial irradiation. The maximally tolerated and recommended phase II dose is 4 mg/(m2/day). The benefits of taxanes as clinical radiation sensitizers for children with high-grade gliomas, if any, remain to be determined. Med Pediatr Oncol 2001;37:390–392. © 2001 Wiley-Liss, Inc.

Task Group 6 of the Diagnostic X-Ray Imaging Committee of the American Association of Physicists in Medicine (AAPM) was appointed to develop performance standards for diagnostic x-ray exposure meters. The recommendations as approved by the Diagnostic X-Ray Imaging Committee and the Science Council of the AAPM are delineated in this report and provide specifications on meter precision, calibration accuracy, calibration reference points, linearity, energy dependence, exposure rate dependence, leakage, amplification gain settings, directional dependence, the stem effect, constancy checks, and calibration intervals. The report summarizes recommendations for meters used in mammography, general purpose radiography including special procedures, computed tomography, and radiation safety surveys for x-ray radiography.

Sources of variations in the measurement of half‐value layer (HVL) in film–screen mammography are evaluated. Two principal sources are the energy responses of ionization chambers and the elemental compositions of the 1100 aluminum‐alloy attenuation materials. Methods in measuring the thickness of attenuation materials are also an important potential source of error. These factors as well as the influence of geometry of measurement, position of measurement in the field, and precision of radiation output are examined. Recommendations for consistency in measurement are provided.
