11. Potential drivers and consequences of regional phosphate depletion in the western subtropical North Pacific

Abstract

In regions of the nitrogen limited low latitude ocean, phosphate can also be depleted to levels initiating stress responses in marine microbes. Here, we associate a broad region of phosphate depletion in the subtropical North Pacific with different levels of phosphorus stress. Nutrient and aerosol addition experiments demonstrated primary nitrogen limitation of the bulk phytoplankton community, with supply of aerosols relieving this limitation. At northern sites with depleted phosphate, alkaline phosphatase activities were enhanced, indicating elevated phosphorus stress. Analysis of satellite- and model-derived aerosol loading showed that aerosol deposition was elevated in these regions. Surface rate measurements suggested that the regional enhancement in phosphate depletion was predominantly driven by elevated nitrogen fixation, likely stimulated by the coincident supply of aerosol iron. Such observations are important for predicting future biogeochemical responses in the subtropical North Pacific to changing aerosol supply.

12. Enhanced Phosphate Consumption Stimulated by Nitrogen Fixation Within a Cyclonic Eddy in the Northwest Pacific

Abstract

Mesoscale eddies are common in the subtropical Northwest Pacific, however, relatively little is known about their spatial variability and temporal evolution, and how these impact upper ocean biogeochemistry. Here we investigate these using observations of a cyclonic eddy carried out along four sequential transects. Consistent with previous observations of cyclonic eddies, the eddy core had doming isopycnals, bringing elevated nutrient waters nearer to the surface. However, we also found that the upper layer of the eddy above the nutricline had significantly lower phosphate concentrations within its core relative to its edge. We attributed this to elevated N 2 fixation within the eddy core, which was likely driven by enhanced subsurface iron supply, ultimately resulting in increased phosphate consumption. Eddy‐enhanced N 2 fixation was additionally supported by the elevation of nitrate + nitrite to phosphate ratios below the euphotic zone. Moreover, we observed that while the upward displacement of isopycnals within the eddy core led to an increase in phytoplankton biomass in the lower euphotic zone, there was no significant increase in total phytoplankton biomass across the entire euphotic zone. Cyclonic eddies in the subtropical North Pacific are projected to be becoming more frequent, implying that such dynamics could become increasingly important for regulating nutrient biogeochemistry and ultimately productivity of the region. 40. Characterisation of the spoilage bacterial microbiota in oyster gills during storage at different temperatures

Abstract

Background The spoilage bacterial community in oyster gill was investigated during storage at 4, 10 and 20°C. Aerobic plate counts and pH values were determined. Total bacterial DNA was extracted from oyster gill and bulk cells of plate count media. The major bacterial species during fresh or different temperatures storage were determined by polymerase chain reaction–denaturing gradient gel electrophoresis (PCR-DGGE). Results The initial aerobic plate count in oyster gill reached 6.70 log CFU g−1. PCR-DGGE fingerprinting analysis of the 16S rRNA gene V3 region revealed that most of the strains in fresh oyster gill belonged to the genera Lactococcus and Enterobacter. The major spoilage bacteria at a storage temperature of 20°C were Leuconostoc pseudomesenteroides, an uncultured bacterium, Cytophaga fermentans, Lactococcus lactis, Pseudoalteromonas sp., Enterococcus mundtii, Clostridium difficile and an uncultured Fusobacteria; those at 10°C were Lactococcus spp., Lactobacillus curvatus, Weissella confusa and C. difficile; those at 4°C were Lactococcus, Weissella, Enterobacter and Aeromonas. The other minor species were L. curvatus, Pseudomonas sp. and E. mundtii. Lactococcus spp. was the most common main spoilage bacteria in oyster gill during chilled storage. Conclusion PCR-DGGE revealed the complexity of the bacterial microbiota and the major bacteria species in oyster gill for fresh and storage. © 2013 Society of Chemical Industry

49. Integrity of cell-free DNA in maternal plasma extracellular vesicles as a potential biomarker for non-invasive prenatal testing.

Abstract

Objective: Large proportions of cell-free DNA (cfDNA) in plasma are localized in extracellular vesicles (EVs), which are secreted from placental cells. This study was conducted to reveal the integrity pattern of cfDNA in maternal plasma EVs (evcfDI) across gestation, and explore if evcfDI could be a potential biomarker in screening for aneuploid fetus in non-invasive prenatal testing (NIPT)., Methods: A total of 180 maternal plasma samples were collected during NIPT. Both evcfDNA and fetal evcfDNA (evcffDNA) were measured by quantitative PCR of LINE1 and SRY gene amplicons with different sizes. The evcfDI was calculated as the ratio of long to short fragments., Results: evcfDI is not affected by gestational age; whereas evcffDI has a mild decreasing trend with increasing gestational age (P = 0.048). evcfDI is significantly and negatively correlated with maternal body mass index (BMI; calculated as weight in kilograms divided by the square of height in meters: ≤18.5, 18.5-25, and ≥25) (P < 0.01) and age (<35 and ≥35 years) (P < 0.01). Mean evcfDI decreases from 2.113 in euploid controls to 0.681 in those with an aneuploid fetus in NIPT (P = 0.003)., Conclusion: Maternal clinical characteristics such as BMI and age could be innovative biomarkers to calibrate evcfDI, which was shown to be a potential indicator of an aneuploid fetus. Analysis of evcfDI based on quantitative PCR could serve as a novel, rapid, and low-cost NIPT strategy, which might facilitate testing at earlier gestations., (© 2021 International Federation of Gynecology and Obstetrics.)
