Samples from bovine viral diarrhoea virus (BVDV)-positive cattle were gathered by Scottish diagnostic laboratories and used to produce a Biobank of samples with associated location and identification data in support of the Scottish BVDV eradication scheme. The samples were subject to direct amplification and sequencing of the 5'-untranslated region (5'-UTR) to define the viral types and subtypes present. From 2693 samples collected prior to 2016, approximately 2300 sequences were obtained, representing 8 BVDV type 1 subtypes. No BVDV type 2 samples were detected. The samples came from all regions of the UK but 66 per cent were from Scotland. Analysis of the sequences showed great diversity in the 5'-UTR, with 1206 different sequences. Many samples carried virus with identical 5'-UTR sequences; often from single locations, but there were also examples of the same sequence being obtained from samples at several different locations. This work provides a resource that can be used to analyse the movement of BVDV strains both within Scotland and between Scotland and other nations, particularly in the latter stages of the Scottish eradication programme, and so inform the advice available to both livestock keepers and policymakers., (British Veterinary Association.)

The current study is a replication and extension of previous research that has found support for a relationship between posttraumatic numbing and hyperarousal. This study examined this association while controlling for depression in 345 motor vehicle accident survivors. Additionally, the relationships among specific hyperarousal symptoms and numbing were explored. Results provided further evidence for an association between hyperarousal and numbing, even while controlling for the influence of depression, and revealed that all hyperarousal symptoms (except hypervigilance) contribute to this association.

The hemocompatibility of a TiN/TiC/diamond-like carbon (DLC) multilayer structure, deposited on titanium substrates for use as coatings for a heart valve prosthesis, has been studied through the adsorption of blood proteins and the adhesion and attachment of blood platelets. All of the surfaces were characterized by stylus profilometry and water contact angles. The adsorption of albumin and fibrinogen to the surfaces was assessed using the Amido Black assay, whereas platelet attachment was studied by scanning electron microscopy and quantified using stereological techniques. The degree of platelet spreading on the surfaces was seen to correlate with differences in surface energy, indicated from contact angle measurements. The greatest spreading was seen on the more hydrophilic surfaces. When studying protein adsorption to the surfaces, no correlation could be determined between contact angle results and levels of adsorption, although the most hydrophilic surfaces did appear to promote greater amounts of fibrinogen adsorption. Thrombus formation was observed to some degree on all of the surfaces, with the exception of the DLC coating. This coating also promoted less spreading of platelets than the other surfaces. The good hemocompatibility of the DLC coating is attributed to its hydrophobicity and smooth surface, resulting in a higher ratio of albumin to fibrinogen than any of the other surfaces., (Copyright 2000 John Wiley & Sons, Inc.)

The physical, chemical, and biological properties of pulsed laser deposited (PLD) and plasma sputtered (PS) hydroxyapatite (HA) coatings were compared. Human osteoblast-like cell responses to these coatings in vitro were assayed for proliferation and phenotypic expression. PS coatings formed smooth and continuous thin films that followed the contours of the substrate surface. PLD coatings consisted of numerous spheroidal micro- and macroparticles. The crystallinity of all coatings was quantified by comparison with the HA target used for both the PS and PLD processes. The XRD and FTIR results indicated that unannealed PLD coatings deposited at room temperature had X-ray spectra consistent with an amorphous structure and were found to dissolve after only a few hours in saline solution. Annealing at 400 degrees C increased the crystallinity (87-98%), which resulted in improved stability and cell activity. The PS coatings showed greater chemical stability than the unannealed PLD coatings and contained an approximate 15% crystalline phase, increasing to 65% postannealing. Cell proliferation and alkaline phosphatase production were significantly higher on unannealed PS specimens than the other coating treatments. There may be benefits in engineering the presence of a minor percentage of a microcrystalline phase in an amorphous or nanometer scale polycrystalline HA structure., (Copyright 2000 John Wiley & Sons, Inc.)

Plasma-sputtered hydroxyapatite (HA) thin coatings ( approximately 1 microm) were deposited onto uncoated and (TiAlV)N-coated Ti-6Al-4V-alloy substrates at low temperatures. The (TiAlV)N coating interlayer was deposited by reactive sputtering. Depositions were achieved by utilizing unbalanced and balanced magnetrons in a capacitively coupled RF plasma. Characterization of the thermostability, bioerosion resistance, and chemical composition of the coating layer was examined by scanning electron microscopy (SEM), energy-dispersive X-ray analysis (EDX), and Fourier transform infrared spectroscopy (FTIR). The results show that for deposition temperatures as low as 67 degrees C, the crystalline phase of the HA coating still is clearly detectable and that the underlying (TiAlV)N coating can increase the crystallinity and thermostability of the HA coating before and after heat treatment. The thin ( approximately 1 microm) sputtered HA coating shows strong HA characteristic peaks in the FTIR spectra even after a 30-day dissolution test. The experimental results show that a multilayer structure comprised of a bioinert (TiAlV)N and bioactive HA coating has the potential to improve the biocompatibility of implant materials. The bioinert (TiAlV)N coating also may provide a long-term stable interface between bone tissue and an alloy implant after the bioactive HA coating is remodeled by the surrounding tissue., (Copyright 1999 John Wiley & Sons, Inc.)

The liver plays an important rôle in protecting the organism from potentially toxic chemical insults through its capacity to convert lipophiles into more water-soluble metabolites which can be efficiently eliminated from the body via the urine. This protective ability of the liver stems from the expression of a wide variety of xenobiotic biotransforming enzymes whose common underlying feature is their ability to catalyse the oxidation, reduction and hydrolysis (Phase I) and/or conjugation (Phase II) of functional groups on drug and chemical molecules. The broad substrate specificity, isoenzyme multiplicity and inducibility of many of these enzyme systems make them particularly well adapted to handling the vast array of different chemical structures in the environment to which we are exposed daily. However, some chemicals may also be converted to more toxic metabolites by certain of these enzymes, implying that variations in the latter may be important predisposing factors for toxicity. Pharmacogenetic defects of xenobiotic biotransformation enzymes, a subclass of inborn errors of metabolism which are manifested only upon drug challenge, introduce marked variation into human populations for the pharmacokinetics and pharmacodynamics of therapeutic and toxic agents, and thus may have important clinical consequences for drug efficacy and toxicity.

Urinary metabolites excreted after oral caffeine were quantified in a healthy sample (n = 68) from the Toronto population by HPLC analyses. The profile of metabolites, assessed by examining particular metabolite ratios, was found to differ widely among subjects. Ratios denoting cytochrome P-450-dependent activities were shown to be interethnically variable between oriental and Caucasian groups, whereas those indicative of xanthine oxidase activity exhibited neither significant interindividual variation nor an ethnic difference. It was also shown that a ratio providing an index of polymorphic N-acetyltransferase activity holds promise as a simple marker for acetylator status in man.

In the course of investigations into variability in the metabolism of caffeine in human populations, urinary levels of 5-acetylamino-6-formylamino-3-methyluracil (AFMU), a newly discovered ring-opened metabolite of caffeine, were found to be both bimodally distributed and interethnically variable in samples (Caucasian: n = 42; Oriental: n = 26) from the Toronto population. To test the premise that the polymorphic N-acetyltransferase enzyme (E.C.2.3.1.5) could be responsible for the production of AFMU, 20 of the subjects were phenotyped for acetylator status using sulfamethazine (SMZ). Concordance for all subjects between AFMU production and SMZ acetylation strongly suggests that the acetylation polymorphism is involved in the formation of AFMU in man.
