Abstract The development of cathode materials with controllable physicochemical structures and explicit catalytic sites is important in rechargeable Zn–air batteries (ZABs). Covalent organic frameworks (COFs) have garnered increasing attention owing to their facile synthesis methods, ordered pore structure, and selectivity of functional groups. However, the sluggish kinetics of oxygen evolution reaction (OER) or oxygen reduction reaction (ORR) inhibit their practical applications in ZABs. Herein, nucleophilic substitution is adopted to synthesize pyridine bi‐triazine covalent organic framework (denoted as O‐COF), and meanwhile, ionothermal conversion synthesis is employed to load MOx (M=Fe, Co) onto carbon nanosheet (named as FeCo@NC) to modulate the electronic structure. The Fe, Co‐N codoped carbon material possesses a large portion of pyridinic N and M‐N, high graphitization, and a larger BET surface area. An outstanding bifunctional activity has been exhibited in FeCo@NC, which provides a small voltage at 10 mA cm−2 for OER (E10 = 1.67 V) and a remarkable half‐wave voltage for ORR (E1/2 = 0.86 V). More impressively, when assembling ZABs, it displays notable rate performance, significant specific capacity (783.9 mAh gZn−1), and satisfactory long‐term endurance. This method of regulating covalent organic framework and ionothermal synthesis can be extended to design diverse catalysts.

Abstract Variants in potassium channel‐related genes are one of the most important mechanisms underlying abnormal neuronal excitation and disturbances in the cellular resting membrane potential. These variants can cause different forms of epilepsy, which can seriously affect the physical and mental health of patients, especially those with refractory epilepsy or status epilepticus, which are common among pediatric patients and are potentially life‐threatening. Variants in potassium ion channel‐related genes have been reported in few studies; however, to our knowledge, no systematic review has been published. This study aimed to summarize the epilepsy phenotypes, functional studies, and pharmacological advances associated with different potassium channel gene variants to assist clinical practitioners and drug development teams to develop evidence‐based medicine and guide research strategies. PubMed and Google Scholar were searched for relevant literature on potassium channel‐related epilepsy reported in the past 5–10 years. Various common potassium ion channel gene variants can lead to heterogeneous epilepsy phenotypes, and functional effects can result from gene deletions and compound effects. Administration of select anti‐seizure medications is the primary treatment for this type of epilepsy. Most patients are refractory to anti‐seizure medications, and some novel anti‐seizure medications have been found to improve seizures. Use of targeted drugs to correct aberrant channel function based on the type of potassium channel gene variant can be used as an evidence‐based pathway to achieve precise and individualized treatment for children with epilepsy. Plain Language Summary In this article, the pathogenesis and clinical characteristics of epilepsy caused by different types of potassium channel gene variants are reviewed in the light of the latest research literature at home and abroad, with the expectation of providing a certain theoretical basis for the diagnosis and treatment of children with this type of disease.

Abstract Acrylamide (AA) is a neurotoxic and potentially carcinogenic contaminant, usually generated during food processing. MicroRNAs are a class of noncoding small RNAs, which serve essential pathogenic roles in linking exposure to toxic contaminants with their pathological outcomes. Our previous research identified miR‐27a‐5p as a key regulator in AA toxicity, as it induces mitochondria‐dependent apoptosis by targeting Btf3 in rats. However, the mechanisms by which miR‐27a‐5p is regulated to exert its toxic effects remain unclear. Here, the transcription factors that bind to the promoter of miR‐27a‐5p and their potential regulatory functions are investigated in cell lines and rats. The results showed that nuclear factor kappa‐B 1 (NF‐κB1) is a promising transcription factor from bioinformatic analysis. The luciferase reporter assay demonstrated that NF‐κB1 enhances the transcriptional activity of the miR‐27a‐5p promoter, whereas the cleavage under targets and tagmentation assay successfully verified the specific binding sites at −305/−298 bp. Furthermore, the inhibition of NF‐κB1 resulted in the suppression of miR‐27a‐5p expression and its associated target pathways, with additional validation provided by in vivo experimentation. Consequently, the upregulation of miR‐27a‐5p induced by AA and its subsequent intrinsic apoptosis can be attributed to the binding of NF‐κB1 to the miR‐27a‐5p promoter. This binding event may serve as the initial step in the molecular network regulated by miR‐27a‐5p that underlies AA toxicity.

Abstract Leveraging Gravity Recovery and Climate Experiment mascon products spanning from April 2002 to September 2023, we, for the first time, ascertain the substantial influence of cryospheric mass variations on Earth's Chandler wobble (CW). Further, in contrast to traditional analysis conducted in the excitation domain, this study focuses on the polar motion domain and incorporates the wavelet analysis technique. Our findings reveal some intriguing phenomena: Between 2006 and 2020, the cryosphere contributed an average amplitude of approximately 4.85 mas to CW, equivalent to 5.05%, with its impact escalating to about 11 mas from 2018 to 2022, representing a fourfold rise in its contribution ratio to approximately 20%. This marked surge can be attributed to the more erratic glacier mass balance results from ongoing climate change. Moreover, there is a pronounced decrease in the CW signal post‐2018, which starkly contrasts with cryospheric contribution, suggesting a potential linkage to climate change yet warrants further investigation.

Abstract The browning of white fat has become a hot topic as it could help to lose weight and improve metabolic health. Therefore, this study aims to explore the in vivo browning or thermogenesis effect of 6‐gingerol (6G), a natural bioactive compound of ginger, and to clarify the underlying mechanism. The results showed that 6G limited weight gain, improved cold tolerance, and increased energy expenditure in high‐fat diet‐induced obese mice. Moreover, histological and immunohistochemical analysis revealed that 6G activated brown adipose tissue (BAT) and induced more beige cells in epididymal adipose tissue (eWAT). Meanwhile, eWAT browning was marked by appearance of multilocular lipid droplets, a higher level of uncoupling protein 1 (UCP1), and increased mitochondrial content. The potential molecular mechanism of eWAT browning was further investigated by RNA‐seq, qPCR, and western blot assays. Results indicated that 6G significantly induced the expression of beige and thermogenesis‐specific markers in eWAT and triggers thermogenesis mainly through a phosphoinositide 3‐kinase (PI3K) / protein kinase B (AKT)‐ribosomal protein S6 (RPS6) axis. Consistently, in vitro, data showed that pretreatment of primary white adipocytes with PI3K/AKT pathway inhibitor significantly counteracted the 6G‐induced effects. This study reveals, for the first time, that 6G induces browning in adipose tissue by modulating the PI3K/AKT‐RPS6 and thermogenesis pathways, thereby alleviating obesity.

Abstract Autophagy‐lysosomal pathway (ALP), a lysosome‐mediated self‐renewal process, is crucial for cell survival and death. Acrylamide (AA) is a neurotoxic compound produced during food thermal processing, and the mechanism underlying AA‐induced neurotoxicity remains elusive. In this study, we explored whether dysregulated ALP was involved in AA‐induced neurotoxicity and the underlying mechanism. We first evaluated the toxic effects of AA on the activation of apoptosis and NLRP3 pathway in human glioma U251 cells. We found that AA‐induced autophagy activation with the accumulation of an autophagy substrate P62, which implies the occurrence of autophagy‐lysosomal disorders. By using autophagy agonist PP242 and siRNA interfering ATG5, we demonstrated that ALP dysregulation contributed to AA‐induced apoptosis and NLRP3 inflammasome pathway activation. In addition, AA triggered ALP dysfunction by decreasing the expression of transcription factor EB (TFEB), and TFEB overexpression restored the lysosomal‐associated proteins and protected against AA‐induced apoptosis and inflammasome activation. Moreover, the autophagy agonist rapamycin restored AA‐induced ALP dysfunction by upregulating TFEB and prevented neurotoxicity. Overall, our study provides novel insights into the role of disrupted ALP in AA‐induced neurotoxicity and highlights that TFEB can be developed as a promising intervention target against AA‐induced neurotoxicity.

Abstract Browning is a severe problem in the juice industry, which is tremendously affected by quinone (Q) reactivity. Here, the formation rate, electrophilicity, and oxidizing ability of different quinones were investigated. LC/MS was applied to monitor the loss of polyphenols and quinones in model systems during storage. Mathematical modeling of collected LC/MS data was conducted to derive the rate constants of various reactions. Polyphenols containing catechol or pyrogallol groups are more susceptible to oxidation. Periodic acid and quinone mediated oxidation were faster than the autoxidation of polyphenols. Chlorogenic acid (CQA) and 4‐methylcatechol (4MC) only containing B ring can hardly contribute to browning. They must combine with A ring to form yellow polymers. Furthermore, the electrophilicity and oxidizing ability order of quinone is: CQA‐Q ≈ 4MC‐Q > catechin quinone (CAT‐Q) ≈ epicatechin quinone (EC‐Q) > gallic acid quinone (GA‐Q). It was possible that CAT‐Q and EC‐Q would undergo nucleophilic addition with their A rings and GA‐Q's extra hydroxyl group might contribute to the electrons, resulting in lower reactivity. This work innovatively evaluated the reactivity of diverse polyphenols and quinones, attempting to illustrate their contributions to nonenzymatic browning. These findings provide a new perspective to restrain the browning in the juice processing industry.

Abstract Astringency is a major sensory property and plays an important role in the overall quality of juice and wines. A major contributor to astringency is condensed tannins, which can precipitate with salivary glycoproteins. And thus, it is vital to develop methods to measure the tannin accurately. Recently, the interest in the application of condensed tannin assays has been increasing in various fields. Condensed tannin assays broadly include the determination of concentration, activity, mean degree of polymerization and its derivative flavan‐3‐ol sulfonates content. In this guideline, the mechanisms and the practice of the most common condensed tannin assays applied in research labs are discussed in detail.

Abstract Background To compare the response and safety of Ixazomib/Lenalidomide/Dexamethasone (IRd) and Bortezomib/Lenalidomide/Dexamethasone (VRd) treatment in newly diagnosed multiple myeloma (MM). Methods This was a single‐center retrospective analysis in Xiangtan Central Hospital. A total of 52 newly diagnosed MM patients from June 2019 to June 2021 were enrolled and divided into the IRd (n = 21) and VRd (n = 31) groups. After 4 cycles of chemotherapy, the best response and adverse events were recorded. Moreover, the progression‐free survival (PFS) and overall survival (OS) were calculated. Results Patients in IRd group and VRd group showed similar PFS (Log‐rank p = 0.70), OS (Log‐rank p = 0.61) and overall response rate (83.87% vs 90.48%, p = 0.803). In addition, patients in VRd group showed lower Eastern Cooperative Oncology Group scores (p = 0.047), and higher incidence of peripheral sensory neuropathy (0.00% vs 19.35%, p = 0.032) than that of patients in IRd group. Conclusion Compared to VRd regimen, IRd had the similar efficacy, better safety, and may be more convenient for patients with poor basic condition for newly diagnosed MM. This study provides an insight for physicians to use IRd as first‐line treatment in MM.

Abstract Cognitive dysfunction is a common postoperative neurological complication in patients undergoing valve replacement surgery. This study aimed to compare the effects of sevoflurane versus propofol‐based total intravenous anesthesia on the incidence of cognitive dysfunction following valve replacement surgery. This multicenter, randomized, controlled double‐blinded study was conducted in three teaching hospitals in China. Patients receiving on‐pump valve replacement surgery were enrolled. Stratified block randomization was used to randomly assign patients 1:1 to receive sevoflurane (1.0–1.5 MAC) or propofol (2.0–3.0 mg/kg/h) for anesthesia maintenance. The primary outcome was the incidence of cognitive dysfunction assessed by four cognitive tests before, as well as 7–14 days after surgery. Patients were randomly assigned to receive sevoflurane anesthesia (n = 144) or propofol‐based total intravenous anesthesia (n = 145). The incidence of postoperative cognitive dysfunction in the sevoflurane anesthesia group (31.9%) was significantly lower than that in the total intravenous anesthesia group (43.4%; relative risk 0.61, 95% confidence interval [CI]: 0.38–0.97, p = 0.044). There was no difference in the incidence of delirium between patients receiving sevoflurane and total intravenous anesthesia (27.8% [35/144] vs. 25.9% [35/145], 1.10, 95% CI: 0.64 to 1.90, p = 0.736). There was a significant difference in the Katz Index on day 3 after surgery (3 [0.9) vs. 3 (1.0], 0.095, 95% CI: 0.05 to 0.43, p = 0.012). No difference was observed in other outcomes between the two groups. For patients undergoing on‐pump valve replacement surgery, sevoflurane anesthesia had a smaller effect on cognitive function and independence in daily life activities compared with propofol anesthesia.

Abstract Aim Early mobilization (EM) improves critical illness survivors' clinical outcomes. This study examines ICU quasi‐specialty nurses' EM‐related knowledge, attitudes and practices (KAP). Design This cross‐sectional study was conducted at a provincial ICU specialty nurse training site from 2019 to 2021. KAP data were collected using a self‐report questionnaire. Methods Participants were registered nurses enrolled in the training programme; 485 nurses from 188 hospitals completed the questionnaire. Results Of the participants, 37.7% had EM training, and 30.7% reported their wards performed EM. Median (IQR) scores for knowledge, attitudes and practices were 14.0 (4.0), 47.0 (9.0) and 37.0 (8.0), belonging to medium, high and medium levels. Scores were lowest for EM scope, implementation indicators and out‐of‐bed mobilization. There were significant deviations in KAP scores amongst nurses at different levels of hospitals with EM training and whose departments performed EM. Significant positive correlations between knowledge and practice, and attitude and practice were identified.

Abstract Organic near‐infrared (NIR) photodetectors with essential applications in medical diagnostics, night vision, remote sensing, and optical communications have attracted intensive research interest. Compared with most conventional inorganic counterparts, organic semiconductors usually have higher absorption coefficients, and their thin active layer could be sufficient to absorb most incident light for effective photogeneration. However, due to the relatively poor charge mobility of organic materials, it remains challenging to inhibit the photogenerated exciton recombination and effectively extract carriers to their respective electrodes. Herein, this challenge was addressed by increasing matrix conductivities of a ternary active layer (D–A–D structure NIR absorber [2TT‐oC6B]:poly(N,N′‐bis‐4‐butylphenyl‐N,N′‐bisphenyl)benzidin [PolyTPD]:[6,6]‐phenyl‐C61‐butyric acid methyl ester [PCBM] = 1:1:1) upon in situ incident light illumination, significantly accelerating charge transport through percolated interpenetrating paths. The greatly enhanced photoconductivity under illumination is intrinsically related to the unique donor–acceptor molecular structures of PolyTPD and 2TT‐oC6B, whereas stable intermolecular interaction has been verified by systematic molecular dynamics simulation. In addition, an ultrafast charge transfer time of 0.56 ps from the NIR aggregation‐induced luminogens of 2TT‐oC6B absorber to PolyTPD and PCBM measured by femtosecond transient absorption spectroscopy is beneficial for effective exciton dissociation. The solution‐processed organic NIR photodetector exhibits a fast response time of 83 μs and a linear dynamic range value of 111 dB under illumination of 830 nm. Therefore, our work has opened up a pioneering window to enhance photoconductivity through in situ photoirradiation and benefit NIR photodetectors as well as other optoelectronic devices.

Background Despite optimized risk factor control, people with prior cardiovascular disease remain at high cardiovascular disease risk. We assess the immediate‐ and longer‐term impacts of new vascular and nonvascular events on quality of life (QoL) and hospital costs among participants in the REVEAL (Randomized Evaluation of the Effects of Anacetrapib Through Lipid Modification) trial in secondary prevention. Methods and Results Data on demographic and clinical characteristics, health‐related quality of life (QoL: EuroQoL 5‐Dimension‐5‐Level), adverse events, and hospital admissions during the 4‐year follow‐up of the 21 820 participants recruited in Europe and North America informed assessments of the impacts of new adverse events on QoL and hospital costs from the UK and US health systems' perspectives using generalized linear regression models. Reductions in QoL were estimated in the years of event occurrence for nonhemorrhagic stroke (−0.067 [United Kingdom], −0.069 [US]), heart failure admission (−0.072 [United Kingdom], −0.103 [US]), incident cancer (−0.064 [United Kingdom], −0.068 [US]), and noncoronary revascularization (−0.071 [United Kingdom], −0.061 [US]), as well as in subsequent years following these events. Myocardial infarction and coronary revascularization (CRV) procedures were not found to affect QoL. All adverse events were associated with additional hospital costs in the years of events and in subsequent years, with the highest additional costs in the years of noncoronary revascularization (£5830 [United Kingdom], $14 133 [US Medicare]), of myocardial infarction with urgent CRV procedure (£5614, $24722), and of urgent/nonurgent CRV procedure without myocardial infarction (£4674/£4651 and $15 251/$17 539). Conclusions Stroke, heart failure, and noncoronary revascularization procedures substantially reduce QoL, and all cardiovascular disease events increase hospital costs. These estimates are useful in informing cost‐effectiveness of interventions to reduce cardiovascular disease risk in secondary prevention. Abstract An engineered bladder construct that mimics the structural and functional characteristics of native bladder is a promising therapeutic option for bladder substitution. We previously showed that pedicled vascularized smooth muscle tissue fabricated by grafting smooth muscle cell (SMC) sheets onto an axial capsule vascular bed had the potential for reliable bladder reconstruction. In this study, we investigated the feasibility of buccal mucosa graft (BMG) integration with the pedicled vascularized smooth muscle tissue to generate a full‐layer pedicled vascularized bladder construct. BMG transplanted onto vascularized smooth muscle tissue showed good survival and developed into a pedicled vascularized bladder construct with full‐layer structures, appropriate thickness, abundant vascularization, and effective barrier function. Then the full‐thickness bladder defects were, respectively, reconstructed by pedicled capsule tissue (pedicled capsule group), nonpedicled vascularized bladder construct (nonpedicled construct group), and pedicled vascularized bladder construct (pedicled construct group). The picrosirius red (PSR) staining and immunohistochemistry results showed minimal fibrosis, maximal smooth muscle proportion, and high vascular density in the pedicled construct group. A continuous mucosal layer was observed only in the pedicled construct group. Moreover, morphological and functional studies revealed better bladder compliance and good ductility in the pedicled construct group. Overall, these results suggested that the BMG could be well integrated with vascularized smooth muscle tissue and generated a pedicled, fully vascularized, and structurally organized bladder construct, which facilitated structural remodeling and functional recovery and could become an alternative to bladder reconstruction.

Abstract Purpose To investigate the efficacy and safety of pyrotinib in treating patients with human epidermal growth factor receptor type 2 (HER2)‐positive breast cancers with brain metastasis. Patients and Methods This is a multicenter retrospective study, and the HER2‐positive breast cancer patients with brain metastasis were studied. The enrolled patients were given pyrotinib 400 mg orally once per day for 21 days as one cycle, and evaluated every two cycles. All relevant data were detected for final assessments including medical history, clinical examination, histopathology, immunohistochemistry, radiographic imaging, treatment outcome, and adverse events. Results Forty‐two female patients in total were enrolled in this study. The objective response rate (ORR) and disease control rate (DCR) of central nervous system (CNS), were found in 20 of 42 (47.6%) and in 39 of 42 (92.8%), respectively, while for extra‐CNS, the respective ORR and DCR were in 9 of 38 (23.6%) and in 36 of 38 (94.7%), respectively. The compounded ORR and DCR were seen in 17 of 42 (40.4%) and in 39 of 42 (92.8%), respectively. The improvement rate of craniocerebral symptoms after treatment was (19/19) 100% and the median duration was 15 months. The median effective time of brain metastases and other metastases was 43 and 50 days. The median follow‐up time was 22 months (interquartile range, 16.0–24.3 months). The median time for progression in brain metastasis was 16.6 months. The median time to progress for our group patients was 11.1 months. Sixteen patients (36%) with adverse reactions were recorded in the study. Conclusion Pyrotinib combined with chemotherapy/radiotherapy or alone showed significantly greater local control rates and progression free survival (PFS), with manageable toxicity for patients with HER2‐positive breast cancer with brain metastases, and further follow‐up will provide an overall survival (OS) data.

In this paper, a model to predict the residual shear capacity of RAC beams with corroded stirrups was established, and the simulation analysis was conducted on recycled concrete beams with different corrosion level using ANSYS. The reliability of the finite element analysis model was verified by comparing the finite element results with experimental results.

Terrestrial compound protein (Cpro) can be potentially used to replace fishmeal (FM) in the marine carnivorous teleost, golden pompano (Trachinotus ovatus). Four isonitrogenous (45%) and isolipidic (12%) diets named FM30, AP80, PP80, and CP80 were formulated. FM30 (control) contained 30% FM and 25% basic protein, while AP80, PP80, and CP80 only contained 6% FM, where 80% FM and 25% basic protein of control diet were completely replaced by animal protein, plant protein, and Cpro, respectively. After golden pompano juveniles (initial weight: 10.32 ± 0.09 g) were, respectively, fed the four diets in floating sea cages for 10 weeks, the growth performance, intestinal digestive enzyme activity, and immune responses, protein metabolism indices of the CP80 group were similar to or better than those of the FM30 group (P>0.05), and significantly better than those of the AP80 and PP80 groups. Specifically, the weight gain (WG), feed conversion ratio (FCR), activity of alanine transaminase (ALT), growth hormone (GH), and insulin-like growth factor-1 (IGF-1) contents of serum, mRNA level of interleukin-10 (il-10), zonula occludens-2 (zo-2), claudin-3, claudin-12, and eukaryotic translation initiation factor 4G (eif4g) were significantly higher, and the activity of α-amylase (AMS), lipase (LPS) in the foregut and midgut, interleukin-8 (il-8) expression in the intestine was significantly lower than that in the CP80 group, compared with those in AP80 and PP80 groups (P0.05), but was significantly lower than those of AP80 and PP80 groups (P

Our recent study demonstrated that diet with blend oil (named BO1) as lipid, which is designed on the base of essential fatty acid requirement of Trachinotus ovatus, achieved good performance. Here, to confirm its effect and investigate the mechanism, three isonitrogenous (45%) and isolipidic (13%) diets (D1-D3) only differing in dietary lipids, which were, respectively, fish oil (FO), BO1, and blend oil 2 (BO2) consisting of FO and soybean oil at 2 : 3, were formulated and used to feed the T. ovatus juveniles (average initial weight: 7.65 g) for 9 weeks. The results showed that the weight gain rate of fish fed D2 was higher than that of fish fed D3 (P0.05). Correspondingly, compared with the D3 group, fish of the D2 group exhibited better oxidative stress parameters such as lower serum malondialdehyde content and inflammatory indexes in the liver such as the lower expression level of genes encoding four interleukin proteins and tumor necrosis factor α, as well as higher hepatic immune-related metabolites such as valine, gamma-aminobutyric acid, pyrrole-2-carboxylic acid, tyramine, l-targinine, p-synephrine, and butyric acid (P

Abstract Along with the increasing of infections of COVID‐19, nurses are needed more in caring patients with COVID‐19. The aim of this study is to explore the real intention and influencing factors of the nurses' willingness to participate in public health emergency in facing the COVID‐19. A total of 10 nurses who volunteered to care patients with COVID‐19 were selected. Data were collected by semi‐structured interviews and analyzed by content analysis method based on the theory of planned behavior. Three main categories were attitude, subjective norms and perceived behavior control. Under the first category, two subcategories were included: nurses' personal and professional value expectation, patriotism. Two subcategories were yielded for the second category: support from family and friends, role model impact of important people. In addition, three subcategories were identified for the third category: physical condition, professional knowledge and skills, national measures and incentive policies. Nurses with high willingness have seven features: high value expectation, patriotism, generous family support, good physical condition, experienced professional knowledge and skills, awareness of national measures and incentive policies. Combined with our previous quantitative study, developing tailored training programs to improve nurses’ professional value expectation, knowledge and skills in coping with unwilling attitude and weak perceived behavior control are effective ways.

Abstract Indoor photovoltaics (IPVs) are expected to power the Internet of Things ecosystem, which is attracting ever‐increasing attention as part of the rapidly developing distributed communications and electronics technology. The power conversion efficiency of IPVs strongly depends on the match between typical indoor light spectra and the band gap of the light absorbing layer. Therefore, band‐gap tunable materials, such as metal‐halide perovskites, are specifically promising candidates for approaching the indoor illumination efficiency limit of ∼56%. However, perovskite materials with ideal band gap for indoor application generally contain high bromine (Br) contents, causing inferior open‐circuit voltage (VOC). By fabricating a series of wide‐bandgap perovskites (Cs0.17FA0.83PbI3−xBrx, 0.6 ≤ x ≤ 1.6) with varying Br contents and related band gaps, it is found that, the high Br vacancy (VBr) defect density is a significant reason that leading to large VOC deficits apart from the well‐accepted halide segregation. The introduction of I‐rich alkali metal small‐molecule compounds is demonstrated to suppress the VBr and increase the VOC of perovskite IPVs up to 1.05 V under 1000 lux light‐emitting diode illumination, one of the highest VOC values reported so far. More importantly, the modules are sent for independent certification and have gained a record efficiency of 36.36%.

Abstract Background Lymphopenia can decrease immune function of the host and is a known risk factor for poor prognosis in malignant tumors. Radiation induced lymphopenia was common in patients with breast cancer and was also reported to have a negative effect on long‐term outcome. Aims Lymphopenia may be associated with baseline immune status before radiotherapy (RT). This study aimed to explore the rate and risk factors of lymphopenia before start of the adjuvant RT in patients with breast cancer. Methods Patients with invasive breast cancer treated from March 2015 to February 2020 and with peripheral lymphocyte counts (PLC) available within 7 days from the beginning of RT were eligible for this study. Data were presented as mean and 95% confidence interval unless otherwise specified. The risk factors of low PLC before RT were identified using univariate and multivariable linear regressions. Results A total of 1012 consecutive patients met the study criteria. The mean PLC before RT commencement was 1.58*109/L (95%CI: 1.55–1.62*109/L) with 15.2% (95%CI: 13.1%–17.6%) CTCAE defined lymphopenia, rendering 12.3%, 2.6%, 0.3%, and 0% for grade 1, 2, 3 and 4 respectively. Univariate and multivariable linear regression showed prior chemotherapy was the most significant risk factor (p .05). A total of 912 (90.1%, 95%CI: 88.1%–91.9%) patients had chemotherapy before adjuvant RT in this study. In patients with HR+/HER2‐ breast cancer, 69.0% (95%CI: 63.0%–74.5%) N0 and 98.1% (95%CI: 95.1%–99.5%) N1 had also received chemotherapy. Conclusions Patients with breast cancer might have lymphopenia from prior chemotherapy at the start of adjuvant RT which could have negative effect on long‐term outcome. It is also noted that most of the patients with HR+/HER2‐, early‐stage breast cancer were treated with aggressive chemotherapy without knowing the risk of chemotherapy induced lymphopenia. Future study on predictive or prognostic multigene assays is warranted to avoid unnecessary chemotherapy and subsequent lymphopenia in patients with low risk breast cancer.

Abstract Background Cell‐free messenger RNA (cf‐mRNA) and long non‐coding RNA (cf‐lncRNA) are becoming increasingly important in liquid biopsy by providing biomarkers for disease prediction, diagnosis and prognosis, but the simultaneous characterization of coding and non‐coding RNAs in human biofluids remains challenging. Methods Here, we developed polyadenylation ligation‐mediated sequencing (PALM‐Seq), an RNA sequencing strategy employing treatment of RNA with T4 polynucleotide kinase to generate cell‐free RNA (cfRNA) fragments with 5′ phosphate and 3′ hydroxyl and RNase H to deplete abundant RNAs, achieving simultaneous quantification and characterization of cfRNAs. Results Using PALM‐Seq, we successfully identified well‐known differentially abundant mRNA, lncRNA and microRNA in the blood plasma of pregnant women. We further characterized cfRNAs in blood plasma, saliva, urine, seminal plasma and amniotic fluid and found that the detected numbers of different RNA biotypes varied with body fluids. Abstract Gallbladder cancer (GBC) is a rare but the most malignant type of biliary tract tumor. It is usually diagnosed at an advanced stage and conventional treatments are unsatisfactory. As a proteasome inhibitor, bortezomib (BTZ) exhibits excellent antitumor ability in GBC. However, the long‐term treatment efficacy is limited by its resistance, poor stability, and high toxicity. Herein, BTZ‐encapsulated pH‐responsive copolymeric nanoparticles with estrone (ES‐NP(BTZ; Ce6)) for GBC‐specific targeted therapy is reported. Due to the high estrogen receptor expression in GBC, ES‐NP(BTZ; Ce6) can rapidly enter the cells and accumulate near the nucleus via ES‐mediated endocytosis. Under acidic tumor microenvironment (TME) and 808 nm laser irradiation, BTZ is released and ROS is generated by Ce6 to destroy the “bounce‐back” response pathway proteins, such as DDI2 and p97, which can effectively inhibit proteasomes and increase apoptosis. Compared to the traditional treatment using BTZ monotherapy, ES‐NP(BTZ; Ce6) can significantly impede disease progression at lower BTZ concentrations and improve its resistance. Moreover, ES‐NP(BTZ; Ce6) demonstrates similar antitumor abilities in patient‐derived xenograft animal models and five other types of solid tumor cells, revealing its potential as a broad‐spectrum antitumor formulation.

Abstract Currently, numerous pieces of evidence have demonstrated that the health gut microbiota contributed to physiological homeostasis for the host. Accumulating methods appear to detect microbial population, including denaturing gradient gel electrophoresis (DGGE), high‐throughput sequencing, and quantitative real‐time polymerase chain reaction (qPCR). With characteristics of flexibility and high‐sensitivity, qPCR technique plays a critical role in determining the dynamic alteration of bacterial abundances, which entirely exhibits the microbial activity in various external situations. In these guidelines, taking Akkermansia muciniphila for example, we provide experimental details in specific primer design and validation, standard plasmid construction, qPCR assay, and data analysis for microbial detection, which also can be used for bacterial determination in other environmental systems. In addition, we offer the solution preparation and operation procedures of in vitro gastrointestinal digestion system. Furthermore, these guidelines can contribute to verify the high‐throughput sequencing results and reveal the underlying mechanism of the microbial shift in response to the dietary intervention or the development of diseases.

The channel slope in the red-bed soft rock area is prone to instability and collapse due to the influence of the channel flow movement, rainfall, weathering, and other factors. Under long-term operation conditions, the sediment stripped from the channel side wall is liable to silt along the process of water flow transportation in the channel, which seriously leads to elevation of the channel bottom, increase of channel width, and reduction of the horseway, which affect the normal water conveyance and operation safety of channels. In view of the collapse and instability of Zhetang diversion irrigation channel project in Xinjiang hydraulic project, through on-site sampling and indoor rock mechanics test, the strength mechanical parameters of slope rock samples are obtained, and the indoor rheological test is carried out, and the triaxial rheological law is obtained. Under different levels of stress, the axial creep strain accounts for more than 50% of the total axial strain, indicating that the rheological effect of sample rock is obvious. Based on the strength parameters of rock samples, a numerical model is established. The stability of red-bed soft rock channel bank slope is studied by using the strength reduction method of finite element method. The seepage field of channel slope under conventional working conditions is analyzed, and the slope stability under different operating conditions is compared and calculated. The results show that the slope stability safety factor of Zhetang diversion irrigation channel slope is 1.60 under conventional working conditions and 1.33 under check working conditions, which are greater than the recommended value in the specification.

The re-alkalization technology of carbonated concrete can effectively restore the alkalinity of concrete, but the effect of re-alkalization is affected by many factors, such as the voltage, electrolyte concentration, and current conduction time. In this paper, the re-alkalization effect of carbonated concrete under different parameter changes is studied, and the feasibility of re-alkalization technology and the selection range of parameters in its application are determined. Results show that the voltage should be controlled below 12 V or above 16 V and can be adjusted properly; the electrolyte concentration should be controlled at 0.5 molL−1; and the current conduction time is determined according to the control model. Finally, a new re-alkalization control model is obtained by regression of test data to control the re-alkalization process.

This study evaluates the profitability and marketability efficiencies of digital firms ranked in Forbes’ list of top companies by using a two-stage network data envelopment analysis (DEA) model with multiplicative efficiency aggregation under the second-order cone programming (SOCP) and examines the respective impacts of the 1995–2001 dot-com bubble and the 2007–2009 global financial crisis on the companies’ efficiencies by applying impulse response function (IRF) analysis. The data of our 49 sampled companies are derived from the Compustat database. The covered period is 1999–2018. These results present the stable and increasing improvement of profitability and marketability efficiencies; in addition, two crisis events have no significant impact on the performance of digital firms. This research is supposed to offer a reasonable and objective evaluation model to measure the performance of digital firms, providing the managers and investors a reference for making their decision.

Blasting vibration is a widely studied harmful effect of rock blasting excavations. Many factors affect its dominant frequency, which makes analyzing, evaluating, and predicting it difficult. This study explored the factors influencing the dominant frequency of blasting vibrations in the case of a spherical charge. Based on symmetry, a theoretical analysis in terms of a spherical explosion source is generally sufficient to describe a cavity excited by a spherical explosion charge. The elastic cavity radius and the dominant frequency of vibration induced by the spherical blasting source are closely related. However, there is a lack of relevant research on cylindrical charges. Therefore, a calculation model for a single-hole cylindrical charge was established. There is a relationship between the corresponding dominant frequency and the range of the plastic zone. The results indicate that the dominant frequency of the blasting vibration for both cylindrical and spherical charges is closely related to the range of the plastic zone formed by the rock blasting. As the elastic cavity radius increases, both the zero-crossing and the dominant Fourier frequencies decrease, and the amplitude spectrum shifts to lower frequencies. However, increasing the cylindrical charge diameter causes more changes in the plastic zone in the direction perpendicular to the cylindrical explosive axis. Moreover, increasing the charge length causes more changes in the plastic zone along this axis. It is therefore difficult to identify a unique dimensional parameter that characterizes the range of the plastic zone formed by the blasting. Because the plastic zone around a cylindrical charge is less regular than around a spherical charge, the charge weight Q is a more favorable parameter than the elastic cavity radius a when used as the main influencing factor and in an attenuation analysis for the dominant frequency of cylindrical charge blasting.

Background. Accurate interpretation of lung function tests requires appropriate spirometry reference values derived from large-scale population-specific epidemiological surveys. The aim of this cross-sectional study was to establish normal spirometric values for the population of healthy, nonsmoking Han Chinese adults residing in Zhejiang province, China. Methods. We measured lung function parameters such as forced expiratory volume in 1 s, forced vital capacity, peak expiratory flow, maximal midexpiratory flow, and diffusion capacity for carbon monoxide and considered age, height, and weight as independent factors that may modify these parameters. The clinical data were divided into the study arm and validation group. The study arms were used to construct predictive equations using stepwise multiple linear regression, and data from the validation group were used to assess the robustness of the equations. Results. The 3866 participants were randomized into a study arm (n = 1,949) and a validation arm (n = 1,917). Lung function parameters had a negative association with age and a positive association with height. Data from the two groups were similar. Predictive equations were constructed from the study arm, and the validation group was used to test the feasibility of the reference equations. Conclusions. The reference values we derived can be used to evaluate lung function in this cohort in both epidemiological studies and clinical practice.

Abstract Background Tumor mutational burden (TMB) is a promising biomarker for stratifying patient subpopulation who would benefit from immune checkpoint blockade (ICB) therapies. Although great efforts have been made for standardizing TMB measurement, mutation calling and TMB quantification can be challenging in samples with low tumor content including liquid biopsies. The effect of varying tumor content on TMB estimation by different assay methods has never been systematically investigated. Method We established a series of reference standard DNA samples derived from 11 pairs of tumor–normal matched human cell lines across different cancer types. Each tumor cell line was mixed with its matched normal at 0% (control), 1%, 2%, 5%, and 10% mass‐to‐mass ratio to mimic the clinical samples with low tumor content. TMB of these reference standards was evaluated by both ∼1000× whole‐exome sequencing (wesTMB) and targeted panel sequencing (psTMB) at four different vendors. Both regression and classification analyses of TMB were performed for theoretical investigation and clinical practice purposes. Results Linear regression model was established that demonstrated in silico psTMB determined by regions of interest (ROI) as a great representative of wesTMB based on TCGA dataset. It was also true in our reference standard samples as the predicted psTMB interval based on the observed wesTMB captured the intended 90% of the in silico psTMB values. Although ∼1000× deep WES was applied, reference standard samples with less than 5% of tumor proportions are below the assay limit of detection (LoD) of wesTMB quantification. However, predicted wesTMB based on observed psTMB accurately classify (>0.97 AUC) for TMB high and low patient stratification even in samples with 2% of tumor content, which is more clinically relevant, as TMB determination should be a qualitative assay for TMB high and low patient classification. One targeted panel sequencing vendor using an optimized blood psTMB pipeline can further classify TMB status accurately (>0.82 AUC) in samples with only 1% of tumor content. Conclusions We developed a linear model to establish the quantitative correlation between wesTMB and psTMB. A set of DNA reference standards was produced in aid to standardize TMB measurements in samples with low tumor content across different targeted sequencing panels. This study is a significant contribution aiming to harmonize TMB estimation and extend its future application in clinical samples with low tumor content including liquid biopsy.

Abstract This study aimed to investigate the relationship between inflammation‐related T‐helper cell polarization and the receptor activator for nuclear factor‐κB ligand (RANKL)/osteoprotegerin (OPG) ratio, which is associated with bone resorption or remodeling of chronic periodontitis patients. Gingival crevicular fluid (GCF) and gingival tissues were obtained from periodontally healthy individuals (PH group) and chronic periodontitis patients (CP group). The GCF levels of IFN‐γ, IL‐4, IL‐17, and IL‐10 linked to T‐helper cell polarization toward the Th1, Th2, Th17, and Treg phenotypes, respectively, were determined by ELISA. The expression levels of these cytokines and the polarized T‐helper cells in gingival tissues were assessed through immunohistochemical and immunofluorescence assays. In addition, the RANKL and OPG expression levels in gingival tissues were detected by immunohistochemical assays, and linear regression analysis was used to identify the potential relationship between T‐helper cell polarization and the RANKL/OPG ratio. In total, 22 individuals and 35 patients were enrolled in the present study. In both GCF and gingival tissues, increased levels of IL‐17 and the decreased levels of IL‐4 and IL‐10 were observed in the CP group. When polarized T‐helper cells were identified in gingival tissues, more Th1 and Th17 cells were found in the CP group, whereas more Th2 and Treg cells were found in the PH group. Although there was no significant difference in OPG expression between the two groups, the RANKL/OPG ratio in the CP group was higher than that in the PH group. The linear regression analysis showed that the presence of more Th1 and Th17 cells correlated with a higher RANKL/OPG ratio, whereas the presence of more Th2 cells correlated with a lower RANKL/OPG ratio. Th1 and Th17 cells are positively correlated and Th2 cells are negatively correlated with the RANKL/OPG ratio. Our data suggest that T‐helper cell polarization is closely linked to the RANKL/OPG ratio in gingival tissues from chronic periodontitis patients.

As the most basic structure, the concrete-filled steel tubular (CFST) frame has been widely used in various structures and systems. Compared with conventional reinforced concrete structures and steel structures, CFST structures in strong earthquake showcase more complicated strength and deformation behavior because there are many factors underlying the failure mode. Furthermore, according to the specifications at home and abroad, the corresponding design method to achieve reasonable failure modes for CFST structures has not been clarified. Based on a destructive test on steel beam-CFST plane frames under constant axial load and lateral load, the fiber mode method and solid element model method are adopted to simulate the failure process of the test frames. Based on finite element model simulations and tests, the fiber model method is proposed to carry out the pushover analysis on the CFST frame structures. The factors behind the reasonable failure mode of steel beam-concrete-filled circular steel tubular (CFCST) frame structures are analyzed. Furthermore, the law and influencing factors behind the ratio of flexural capacity of column to beam, the ratio of line stiffness of beam to column, and the ratio of axial compression on the deformation, bearing capacity, and failure modes of the structure are discussed. Some suggestions on the design of reasonable failure mode of steel beam-concrete-filled circular steel tubular (CFCST) frame structures are proposed.

Purpose. To explore the diagnostic value of fractional exhaled nitric oxide (FeNO), small airway function, and a combined of both in differentiating cough-variant asthma (CVA) from typical asthma (TA). Methods. A total of 206 asthma subjects, including 104 CVA and 102 TA, were tested for pulmonary function, bronchial provocation test and FeNO. The correlation between FeNO, small airway function and other pulmonary indicators was analyzed by single correlation and multiple regression analysis. The receiver operating characteristic (ROC) curve was established to evaluate the diagnostic efficiency of FeNO, small airway function, and their combination and to predict the optimal cut-off point. Results. All the respiratory function parameters and small airway function indicators in TA group were significantly different from those in CVA group, and FeNO value was significantly higher than that in CVA group. In addition, the area under the ROC curve (AUC) was estimated to be 0.660 for FeNO, 0.895 for MMEF75%/25%, 0.873 for FEF50%, 0.898 for FEF25%, 0.695 for Fres, 0.650 for R5-R20, and 0.645 for X5. The optimal cut-off points of FeNO, MMEF75%/25%, FEF50%, FEF25%, Fres, R5-R20 and X5, were 48.50 ppb, 60.02%, 63.46%, 45.26%, 16.63 Hz, 0.38 kPa·L−1·s−1, and −1.32, respectively. And the AUC of FeNO combined with small airway function indexes FEF25%, Fres, R5-R20, and X5 were prior than single indicators. Conclusion. FeNO and small airway function indexes might have great diagnostic value for differentiating CVA from TA. The combination of FeNO and FEF25%, Fres, R5-R20, and X5 provided a significantly better prediction than either alone.

Abstract Background Age‐related cataracts (ARC) is the most common blinding eye disease worldwide, and its incidence tend to become younger. However, the relationship between genetic factors and mechanisms is not fully understood. The aim of the study was to further clarify the relationship between ARC and genetic mechanisms in East Asian populations and to elucidate the pathogenesis. Methods The study collected 191 sporadic cataracts and 208 healthy people from the eastern provinces of China, with an average age of about 60 years. All participants were subjected to a comprehensive ophthalmic clinical examination and peripheral blood samples were collected and their genomic DNA was extracted. Mutations were screened among 792 candidate genes to enhance understanding of the disease through targeted capture and high‐throughput sequencing. Results We identified novel candidate susceptibility gene, which may serve as a potential susceptibility factor leading to an increase in the incidence of age‐related cataracts. Three novel loci are associated with age‐related cataracts significant significance: rs129882 in DBH (p = 5.27E‐07, odds ratio = 3.9), rs1800280 in DMD (p = 2.85E‐06, odds ratio = 1.4) and rs2871776 in ATP13A2 (p = 4.18E‐05, odds ratio = 0.04). Gene–gene interaction analysis revealed that the most significant interactions between genes include the interaction between DBH and TUB (rs17847537 in TUB, rs129882 in DBH, p‐value = 2.12E‐14), and the interaction between DBH and DMD (rs1800280 in DMD, rs129882 in DBH, p‐value = 2.12E‐14). Pathway analysis shows that the most significant processes are concentrated in response to light stimulation (adjusted p‐Value = 5.56E‐03), response to radiation (adjusted P‐Value = 5.56E‐03), abiotic stimulus (adjusted p‐Value = 5.56E‐03). eQTL analysis shows that DBH rs129882 could regulate the expression of DBH mRNA in various tissues including retina. Conclusion Our study indicates rs129882 and rs1800280 loci are associated with age‐related cataracts, which enlarge the gene map of age‐related cataracts.

Abstract Background Noninvasive prenatal testing (NIPT) is one of the most commonly employed clinical measures for screening of fetal aneuploidy. Fetal Fraction (ff) has been demonstrated to be one of the key factors affecting the performance of NIPT. Accurate quantification of ff plays vital role in NIPT. Methods In this study, we present a new approach, the accurate Quantification of Fetal Fraction with Shallow‐Coverage sequencing of maternal plasma DNA (FF‐QuantSC), for the estimation of ff in NIPT. The method employs neural network model and utilizes differential genomic patterns between fetal and maternal genomes to quantify ff. Results Our results show that the predicted ff by FF‐QuantSC exhibit high correlation with the Y chromosome–based method on male pregnancies, and achieves the highest accuracy compared with other ff estimation approaches. We also demonstrate that the model generates statistically similar results on both male and female pregnancies. Conclusion FF‐QuantSC achieves high accuracy in ff quantification. The method is suitable for application in both male and female pregnancies. Since the method does not require additional information upon NIPT routines, it can be easily incorporated into current NIPT settings without causing extra costs. We believe that FF‐QuantSC shall provide valuable additions to NIPT.

Abstract Background Panel‐based targeted exome sequencing was used to analyze the genetic and clinical findings of targeted genes in a cohort of northeast Chinese with retinitis pigmentosa. Methods A total of 87 subjects, comprising 23 probands and their family members (total patients: 32) with confirmed retinitis pigmentosa were recruited in the study. Panel‐based targeted exome sequencing was used to sequence the patients and family members, all subjects with retinitis pigmentosa underwent a complete ophthalmologic examination. Results Of the 23 probands, the clinical manifestations include night blindness, narrowing of vision, secondary cataracts, choroidal atrophy, color blindness, and high myopia, the average age of onset of night blindness is 12.9 ± 14 (range, 0–65; median, 8). Posterior subcapsular opacities is the most common forms of secondary cataracts (nine cases, 39.1%), and peripheral choroidal atrophy is the most common form of secondary choroidal atrophy (12 cases, 52.2%). Of these probands with complication peripheral choroidal atrophy, there were eight probands (66.7%, 8/12) caused by the pathogenic variation in USH2A gene. A total of 17 genes and 45 variants were detected in 23 probands. Among these genes, the commonest genes were USH2A (40%; 18/45), RP1 (15.6%; 7/45), and EYS (8.9%; 4/45), and the top three genes account for 56.5% (13/23) of diagnostic probands. Among these variants, comprising 22 (48.9%) pathogenic variants, 14 (31%) likely pathogenic variants, and nine (20%) uncertain clinical significance variants, and 22 variants was discovered first time. Most of the mutations associated with RP were missense (53.3%, 24/45), and the remaining mutation types include frameshift (35.6%, 16/45), nonsense (6.7%, 3/45), and spliceSite (4.4%, 2/45). Among the probands with mutations detected, compound heterozygous forms was detected in 13 (56.5%, 13/23) probands, and digenic inheritance (DI) forms was detected in five (21.7%, 5/23) probands. Conclusion Panel‐based targeted exome sequencing revealed 23 novel mutations, recognized different combinations forms of variants, and extended the mutational spectrum of retinitis pigmentosa and depicted common variants in northeast China.

Abstract Background Panel‐based targeted exome sequencing was applied to identify the pathogenic variants and genetic characteristics of retinitis pigmentosa (RP) in two Chinese families, and to gain a deeper understanding of the relationship between clinical manifestations and genotypes. Methods A total of 17 subjects, comprising two probands (total patients: four subjects) and their family member, were recruited in this study. All subjects underwent comprehensive ophthalmic examinations and clinical evaluations, and the complete history and medical records were collected according to the standard procedures. All participants were screened using the multigene panel test (Target_Eye_792_V2 chip), and Sanger sequencing was used to confirm the candidate variants. Results Among these two families, a total of three novel mutations in the EYS gene were identified in patients, including a homozygous frameshift mutation c.9252_9253insT detected in two patients in one family, and the compound heterozygous splicesite mutation c.5644+2T>C and frameshift mutation c.1920_1923delTGAG detected in two patients in the another family. All patients in both families had early onset of night blindness and poor visual acuity, and with typical posterior capsule opacification. The mutation co‐segregated within all recruited individuals. In addition, one patient with compound heterozygous mutations was found to have typical blue‐blindness symptoms and detected a previously reported disease‐causing mutation c.235G>A in OPN1SW gene, which caused blue blindness manifestations and was first discovered in patient combined with RP causative genes. Conclusions Panel‐based targeted exome sequencing was used to identify three novel variants of RP causative gene, and we also detected a known pathogenic variants of blue‐blindness causative genes in two patients. Our finding will provide a powerful basis for genetic counseling and enhance our current understanding of the genetics factors for RP families.

Purpose. To investigate the efficacy of management of high myopic foveoschisis (MF) with a modified surgical technique of arc-shaped foldback fovea-sparing internal limiting membrane (ILM) peeling. Methods. A 23-gauge vitrectomy was performed in five patients with high MF. A long strip of ILM was peeled at the temporal side of the central fovea. Next, an ILM forceps was used to grasp the outer side of the ILM flap, and it was moved forward slowly from the outside to the paracentral fovea, followed by folding ILM back in an arc-shaped manner and then removing it. The above operations were repeated, and all ILM flaps were removed from the outside to paracentral fovea until a narrow strip of ILM remained. Finally, the narrow strip of ILM was excised using a vitreous cutter. Results. At the patients’ last visits, the foveoschisis almost disappeared completely and the fovea reattached. The central macular thickness statistically decreased from 399.0 ± 96.33 μm preoperatively to 164.60 ± 34.20 μm postoperatively (t = 4.289; P=0.013). The preoperative mean logarithm of the minimum angle of resolution best-corrected visual acuity (1.64 ± 0.65) significantly improved to 0.72 ± 0.18 postoperatively (t = 3.265, P=0.031). The average follow-up time was 11.80 ± 3.35 months (range; 8–16 months). Conclusion. The arc-shaped foldback fovea-sparing ILM peeling technique for high MF is safe and effective.

Objective. Renal ischemia/reperfusion injury (RI/RI) is the main cause of acute kidney injury. Total glucosides of paeony (TGP) are a traditional Chinese medicine. This study was aimed at exploring the role of TGP in RI/RI and its underlying mechanism of action. Methods. Rat RI/RI models were constructed by surgical operation. Serum creatinine (Scr) and blood urea nitrogen (BUN) were used to evaluate renal function. The levels of proinflammatory cytokines were detected by ELISA. RI/RI was simulated by hypoxia/reoxygenation (H/R) treatment in renal cells in vitro. The lncRNA XIST (XIST) expression was analyzed by qRT-PCR. Then, the viability and apoptosis of renal cells were detected by MTT and flow cytometry assay. Additionally, dual-luciferase reporter assay was used to determine the interactions among XIST, microRNA-124-3p (miR-124-3p), and ITGB1. Results. TGP improved renal function and inhibited inflammatory responses after RI/RI. XIST expression was highly expressed in rat RI/RI models and H/R-treated renal cells, whereas treatment with TGP downregulated the XIST expression. Additionally, TGP increased viability and attenuated apoptosis and inflammation of H/R-treated renal cells via inhibiting XIST. Moreover, XIST was competitively bound to miR-124-3p, and ITGB1 was a target of miR-124-3p. miR-124-3p overexpression or ITGB1 inhibition rescued the reduction effect on viability and mitigated the promoting effects on cell apoptosis and inflammation caused by XIST overexpression in H/R-treated renal cells. Conclusions. In vivo, TGP attenuated renal dysfunction and inflammation in RI/RI rats. In vitro, TGP inhibited XIST expression to modulate the miR-124-3p/ITGB1 axis, alleviating the apoptosis and inflammation of H/R-treated renal cells.

Locality preserving projection (LPP) is one of the most representative linear manifold learning methods and well exploits intrinsic structure of data. However, the performance of LPP remarkably degenerate in the presence of outliers. To alleviate this problem, the authors propose a robust LPP, namely LPP‐L21. LPP‐L21 employs L2‐norm as the distance metric in spatial dimension of data and L1‐norm as the distance metric over different data points. Moreover, the authors employ L1‐norm to construct similarity graph, this helps to improve robustness of algorithm. Accordingly, the authors present an efficient iterative algorithm to solve LPP‐L21. The authors’ proposed method not only well suppresses outliers but also retains LPP's some nice properties. Experimental results on several image data sets show its advantages.

The possibility of axillary lymph node metastasis differs in different breast cancer patients and is the strongest prognostic indicator in breast cancer. The existing studies mainly explored the relationship of axillary ultrasound imaging and axillary lymph node metastasis, without exploring whether ultrasound imaging of breast tumour can affect and perform axillary lymph node prediction. Therefore, this Letter proposes a novel particle space-time distribution model to find the correlation between contrast-enhanced ultrasonography of breast tumour and axillary lymphatic metastasis. Starting from the imaging principle of dynamic contrast-enhanced ultrasonography, the particle space-time distribution model not only comprises space-time features of contrast-enhanced ultrasonography with an encoder–decoder network, but also the flow field information of microbubble particles is integrated into the space-time features that better serves the metastasis prediction by enhancing the particle distribution information. Extensive experiments on real patients have demonstrated that dynamic contrast-enhanced ultrasonography of breast tumour can be used to predict the probability of lymphatic metastasis. This conclusion can be interpretable from the clinical and pathological perspectives.

Abstract Background Synaptic plasticity is the basis of memory formation. The pathological manifestations of abnormal glucose metabolism in the nervous system of sporadic Alzheimer's disease (SAD) may affect synaptic plasticity, thus causing memory damage. As a traditional Chinese medicine compound preparation, the mechanism by which Shenzhiling (SZL) oral liquid can alleviate the cognitive impairment of SAD by improving synaptic plasticity remains unclear. Objective: This article mainly discusses whether SZL can exert a protective synaptic effect as mediated by glutamate receptors and glycogen synthesis kinase 3β (GSK3β); further, it discusses whether SZL can improve cognitive function in SAD. Methods C57BL/6 mice were used as a SAD model after injection with streptozotocin (STZ) into the bilateral lateral ventricles; mice of the same background were injected with artificial cerebrospinal fluid into bilateral ventricles and were used as a control group. After 3 months of exposure to the intervention, the step‐down test was carried out. Western blot was used to detect the levels of NMDAR2B, p‐NMDAR2B, mGlu5, GSK3β, and p‐GSK3β in the hippocampus of mice. Immunohistochemical analysis was used to observe the number of GSK3β‐positive cells in the CA1 region of mouse hippocampus. Results The memory retention ability of mice was significantly improved after 3 months of SZL treatment, and the expression levels of NMDAR2B, mGlu5, and GSK3β were significantly changed. Conclusion Shenzhiling provides a potential for the treatment for SAD with traditional Chinese medicine.

The AC-DC hybrid microgrid is a credible evolution path for the microgrid. State estimation in complex distribution network is a significant foundation for the safe operation. In AC-DC hybrid microgrid, centralised state estimation algorithms have quite a few deficiencies such as large communication capacity and privacy protection. In response to these obstacles, this paper proposed a novel distributed state estimation algorithm. First, a three-stage state estimation model was established for AC-DC hybrid microgrid, in which the second stage is a nonlinear transform for the linearisation of the nonlinear state estimation progress so that the first and third stage can solve the linear state estimation problem. Furthermore, the Lagrangian relaxation method was ultilised for the distributed problem solution. In the proposed algorithm, AC subsystems and DC subsystems respectively execute state estimation through local information, and the overall consistency of the system state estimation can be eventually achieved according to the transfer iteration of the boundary information. The computational efficiency and convergence of the proposed method was ensured owing to the combination of bilinear theory and Lagrangian relaxation. Simulation results indicate that the proposed method can achieve its goal and be superior to existing method in efficiency.

Applying disaster system theory and with reference to the mechanisms that underlie agricultural drought risk, in this study, crop yield loss levels were determined on the basis of hazards and environmental and hazard-affected entities (crops). Thus, by applying agricultural drought risk assessment methodologies, the spatiotemporal distribution of maize drought risk was assessed at the national scale. The results of this analysis revealed that the overall maize drought risk decreases gradually along a northwest-to-southeast transect within maize planting areas, a function of the climatic change from arid to humid, and that the highest yield loss levels are located at values between 0.35 and 0.45. This translates to drought risks of once in every 10 and 20 years within 47.17% and 43.31% of the total maize-producing areas of China, respectively. Irrespective of the risk level, however, the highest maize yield loss rates are seen in northwestern China. The outcomes of this study provide the scientific basis for the future prevention and mitigation of agricultural droughts as well as the rationalization of related insurance.
