Background: Only 10% of dietary iron is absorbed in the duodenum which implies that 90% (approximately 9 mg day(-1)) reaches the lower small intestine and colon. Therefore the purpose of this study was to assess the iron transport capacity of the rat proximal colon and to determine whether iron absorption is regulated by changes in dietary iron content., Materials and Methods: Rats were fed for 14 days on either iron adequate (44 mg Fe kg(-1) diet) or iron-deficient (< 0.5 mg Fe kg(-1) diet) diets. The 59Fe transport across the colonic epithelium and its subsequent appearance in the blood were measured in vivo. In separate studies the colon was excised and used to measure divalent metal transporter expression., Results: Divalent metal transporter (DMT1) was expressed at the apical membrane of the surface epithelium in rat proximal colon. In animals fed an iron-deficient diet, DMT1 mRNA and protein expression were increased. This was accompanied by a significant increase in tissue 59Fe uptake., Conclusions: The proximal colon can absorb non-haem iron from the intestinal lumen. The purpose of this mechanism remains to be elucidated.

Background: The intestinal handling of dextran, an alpha-1,6-linked glucose polymer, is poor compared with starch, and some ingested dextran might therefore reach the lower small intestine. As luminal sugar up-regulates SGLT1 (sodium-dependent glucose transporter) locally, we report the effects of a dextran-enriched diet on jejunal and ileal brush border membrane (BBM) glucose uptake., Methods: Rats were maintained on a diet containing 65% maltodextrin or 32.5% maltodextrin + 32.5% dextran (10 kD or 40 kD) for 8-10 days, and the kinetics of phlorizin-sensitive [3H]-glucose uptake by purified BBM vesicles was determined., Results: Ingestion of 40-kD but not 10-kD dextran increased Vmax for jejunal and ileal glucose uptake (+64.3% and +61.8% respectively, both P < 0.02). The transport response to 40-kD dextran was in keeping with lower levels of expired H2 at the end of the feeding period. High-performance liquid chromatography (HPLC) analysis of luminal contents indicated extensive hydrolysis of ingested dextran. Finally, 3-h jejunal exposure to 40-kD dextran in vivo increased the Vmax for glucose uptake by jejunal BBM., Conclusion: It is likely that increased SGLT1-mediated glucose uptake after short or longer term mucosal exposure to dextran results from luminal dextran per se or a hydrolysis product. The clinical implications of this up-regulation are discussed.

Haemolytic anaemia induced by phenylhydrazine (PZ) promotes iron absorption across rat small intestine. This present study investigates the role of the brush border potential difference (Vm) and mucosal reducing activity in the response. In addition, quantitative autoradiography was used to assess PZ-induced changes in the villus localization of brush border iron uptake. Iron transfer from duodenum to blood was increased significantly 5 days after treatment with PZ. Autoradiography showed that most brush border iron uptake occurred at the upper villus region and the maximal rate was increased fourfold by PZ. Duodenal villus length was increased in PZ-treated rats. PZ treatment did not influence mucosal reducing activity but Vm, measured using duodenal sheets, increased from -50 to -57 mV (P < 0.001) and this was due to a reduced brush border sodium permeability. Thus, an expanded absorptive surface and an enhanced electrical driving force for iron uptake across the duodenal brush border are important adaptations for increased iron absorption in PZ-induced haemolytic anaemia.

Acute hypoglycaemia enhances intestinal sugar uptake but the mechanisms involved are unknown. Results from the present study show increased galactose movement across the brush border and basolateral membranes of isolated upper, but not mid-villus, jejunal enterocytes 45 min after intravenous administration of insulin to rats at a level which reduced by half plasma glucose concentration. Incubation of upper villus cells from uninjected animals with insulin (100 mU ml-1) for 40 min was without effect on brush border or basolateral sugar transport. Insulin treatment of rats did not affect glucose uptake by brush border vesicles prepared from upper villus cells when the process was driven by an inwardly directed 100 mM sodium thiocyanate gradient. In contrast, glucose uptake using a 100 mM inwardly directed sodium chloride gradient was reduced by 49% following hypoglycaemia. It is concluded that the enhanced sugar uptake following insulin hypoglycaemia involves both brush border and basolateral membranes of only the most mature villus cells at the villus tip. Upregulation of Na(+)-sugar cotransport at the brush border is best explained by an increased electrochemical driving force for Na(+)-sugar cotransport rather than increased numbers of transporters. The transport response is not due to a direct effect of insulin on the enterocyte and the possible systemic factors involved are discussed.

Two strains of rat, Sprague-Dawley and inbred Piebald Virol Glaxo pigmented (PVG) strain, were dosed orally with hypertonic sodium chloride at a dose of 0.25, 0.5, 1.0 or 1.33 g/kg. Gastric epithelial cell proliferation was compared 16 hours after a single dose. The rats were given intraperitoneal bromodeoxyuridine (20 mg/kg) 1 hour before sacrifice, and cells undergoing DNA synthesis (S-phase) were assessed using a monoclonal antibody to bromodeoxyuridine. The number of labelled cells per gastric gland was counted using video image analysis, assessing ten low-power fields (160-180 gastric glands) per rat. Tissue injury was graded for submucosal oedema, inflammation and necrosis; it was minimal after dosing with 0.25 and 0.5 g/kg of sodium chloride. The PVG rats were more susceptible to tissue injury after dosing with 1.0 or 1.33 g/kg of sodium chloride: submucosal oedema, 80% in the PVG and 10% in the Sprague-Dawley; inflammation, 70% compared with 10%; necrosis, 70% compared with 20%. The number of labelled cells per fundic gland increased with increasing dose concentration of sodium chloride and the response was similar for both strains of rat. Plasma gastrin concentration at the time of sacrifice was significantly higher in the PVG rat for the 0.5, 1.0 and 1.33 g/kg doses. These strain differences may be useful in the further evaluation of the mechanisms of sodium chloride-induced tissue damage and repair.

The mechanisms involved in the increased Na(+)-dependent nutrient uptake across intestine of diabetic animals are poorly understood. Here we have studied the effect of acute (7d) and chronic (30-40d) diabetes on the autoradiographic localization of 3H-L-valine accumulation by rat jejunal villi and on enterocyte migration rate. In control rats, Na(+)-dependent valine uptake was confined to enterocytes on the upper 20-23% of the villus. In intestine from diabetic rats, however, this area was extended to occupy the upper 42-45% of an enlarged villus surface. Hyperphagia was not responsible for the expanded functional surface and systemic factors are therefore implicated in the adaptive response. Enterocyte migration rate was found to be unaffected by diabetes but an increased villus height in this condition resulted in an additional 13.5 h in enterocyte lifespan. These data are compatible with the hypothesis that during diabetes the earlier maturation of enterocyte absorptive function produces an epithelial surface containing a higher proportion of mature enterocytes.

The effects of neomycin sulphate on galactose absorption have been studied using in vivo and in vitro preparations of rat small intestine. Neomycin (10(-3)M) produced an increase in the maximum transport capacity (Jmax) for the active component of absorption in vivo. The apparent Kt for absorption was unaffected. The antibiotic caused a dose-dependent increase in the potential difference across the mucosal membrane (Vm) measured in vitro, a maximal effect being seen at a concentration of 10(-4)M. Furthermore, the magnitude of the depolarization induced by the addition of galactose (4 mM) to the mucosal fluid was enhanced by neomycin (10(-4)M). Phlorhizin (10(-4)M) abolished the galactose-induced depolarization in both the absence and presence of the antibiotic. It is concluded that neomycin increases the electrical driving force for Na+ during Na+-coupled galactose entry into the enterocyte.
