As a new and green type of nanomaterials, lignin nanoparticles (LNPs) have been considered as high-value renewable materials for application in many fields. However, the industrialization of LNPs faces many challenges, such as high manufacturing costs and small-scale production. Here, a simple but rapid and reversible approach for the fabrication of LNPs was provided via switching pH environments. The LNPs were regularly shaped in the acetonitrile/water system, and their size appeared to be very homogeneous. The alternation of forming and dissolving of LNPs could be repeated many times simply by alternately adding acid and alkaline solutions. Lignin, as the most abundant aromatic biopolymer in nature, has attracted great attention due to the complexity and richness of its functional groups for value-added applications. The yield of production of lignin and the reactivity of prepared lignin are very important to guarantee the study and development of lignin-based chemicals and materials. Various fractionation techniques have been developed to obtain high yield and relatively high-purity lignin as well as carbohydrates (hemicelluloses and celluloses) and to reduce the condensed and degraded nature of conventional biorefinery lignin. Herein, novel and efficient biomass fractionation and lignin fractionation towards lignin valorization are summarized and discussed.

Lignin, as the most abundant aromatic renewable biopolymer in nature, has long been regarded as waste and simply discarded from the pulp and paper industry. In recent years, with many breakthroughs in lignin chemistry, pretreatment, and processing techniques, a lot of the inherent bioactivities of lignin, including antioxidant activities, antimicrobial activities, biocompatibilities, optical properties, and metal-ion chelating and redox activities, have been discovered and this has opened a new field not only for lignin-based materials but also for biomaterials. In this Review, the biological activities of lignin and drug/gene delivery and bioimaging applications of various types of lignin-based material are summarized. In addition, the challenges and limitations of lignin-based materials encountered during the development of biomedical applications are also discussed.

The traditional method to isolate cellulose nanocrystals (CNCs) is to subject cellulosic materials to strong acid hydrolysis by mineral acids, which usually causes problems such as corrosion of equipment, the need for large amounts of water, the difficulty of acid recovery, and over-degradation of cellulose. Thus, a green and sustainable approach for the preparation of CNCs was developed where mild acid hydrolysis with diluted oxalic acid was used. The reaction kinetics of different preparation parameters, such as reaction temperature, oxalic acid dose, addition of HCl, and reaction time were thoroughly investigated. A high yield of up to 85 % was achieved by mild oxalic acid hydrolysis in comparison to the yield of 35 % using the most common approach with sulfuric acid hydrolysis. The CNCs from the above approach have a high thermal stability, that is, a maximum thermal degradation temperature of 350 °C in comparison to 200 °C when sulfuric acid hydrolysis was used. Importantly, oxalic acid solutions were readily recovered, and exhibited consistently high performance in several continuous runs of reaction. The hydrolysates contained mostly monomeric sugars, which could be further utilized for chemical or biofuel production.

There is growing interest in utilizing galactoglucomannan, the main hemicellulose in softwoods, for various applications such as cosmetics, pharmaceuticals, textiles, alimentary, and health products, as well as for the production of fuels. For fuel production and for using the rare sugars as platform chemicals, the hemicelluloses need to be hydrolyzed to sugar monomers, and for this purpose, low-molecular-mass extracts are favorable. However, for the other applications high molecular masses are required, which presents an even greater challenge for extraction. The ability to optimize the extraction process according to the needs of further processing, by using solely water as the solvent, is a key issue in the environmentally friendly utilization of this versatile raw material. The goal of this work is to study how the average molar mass of hemicelluloses extracted from spruce sapwood can be influenced by altering the experimental conditions. The main parameters influencing the extraction and hydrolysis of the hemicelluloses, namely, extraction time, temperature, pH, and chip size, were studied. The results show that it is feasible to develop an extraction process for harvesting spruce hemicelluloses, also of large molar masses, for industrial applications by using pressurized hot water extraction.

A green chemoenzymatic pathway for the synthesis of conducting polyaniline (PANI) composites is presented. Laccase-catalyzed polymerization in combination with anionic polysaccharides is used to produce polysaccharide/PANI composites, which can be processed into flexible films or coated onto cellulose surfaces. Different polysaccharide templates are assessed, including κ-carrageenan, native spruce O-acetyl galactoglucomannan (GGM), and TEMPO-oxidized cellulose and GGM. The resulted conducting biocomposites derived from natural materials provide a broad range of potential applications, such as in biosensors, electronic devices, and tissue engineering.

New types of hydrogels derived from O-acetyl galactoglucomannan (AcGGM) hemicellulose have been synthesized and characterized. The objective of this work was to analyze the sorption capacity (S) of three types of hydrogels containing AcGGM derivatives incorporated into the carboxylic groups of the polymer chain in the AA hydrogel, sulfonic groups in the APA hydrogel, and amide groups in the acrylamide (Aam) hydrogel. These hydrogels are capable of interacting and removing ions such as cadmium [Cd(II)], copper [Cu(II)], lead [Pb(II)], nickel [Ni(II)], and zinc [Zn(II)]. The results show that AA and Aam hydrogels had a lower sorption capacity of ions compared to the APA hydrogel, which had a high sorption capacity. The maximal sorption capacity was determined by the successive enrichment method, obtaining Pb(II) amount of 48.3 mg/g of AA hydrogel, 65.8 mg/g of APA hydrogel, and 40.8 mg/g of Aam hydrogel. Hence, Pb(II) ions are greatly retained by the three hydrogels. The chemist's guide to the galactosyl unit: A chemo-enzymatic process is developed for the multivalent functionalization of cellulose surfaces via regioselective oxidation of heteropolysaccharides with galactose 6-oxidase. Reductive amination, surface sorption, and click chemistry enable the assembly of (bio)chemically active cellulose surfaces for applications ranging from functional biocomposites to in vitro diagnostics.

Li C, Li Y, Wang X, Wang Z, Cai J, Wang L, Zhao Y, Song H, Meng X, Ning X, Xu C, Lin M, Li L & Geng J (2012) Histopathology 60, 953–963 Elevated expression of astrocyte elevated gene-1 (AEG-1) is correlated with cisplatin-based chemoresistance and shortened outcome in patients with stages III–IV serous ovarian carcinoma Aims: To correlate astrocyte elevated gene-1 (AEG-1) expression with the clinicopathological features and outcome of patients with stages III–IV ovarian serous carcinoma, and to clinically assess the involvement of AEG-1 in acquired cisplatin resistance. Methods and results: The frequency and intensity of immunohistochemical AEG-1 expression increased in a step-wise fashion from normal to chemosensitive to chemoresistant tissues. These observations were confirmed by Western blot analysis. AEG-1 expression level was correlated with lymph nodal metastasis, histological differentiation, residual tumour size and response to primary chemotherapy. Five-year progression-free survival (PFS) and overall survival (OS) rates were lower in the high-expression group than that in the low-expression group. AEG-1 overexpression was an independent but poor prognostic factor in the OS and PFS of these patients, as determined by multivariate Cox regression analysis. Multivariate logistic regression analysis revealed that the presence of cisplatin-based chemoresistance was significantly associated with expression level of AEG-1 and the degree of residual disease (P = 0.0001 and P = 0.0027, respectively). Conclusion: Our findings indicate that tumour AEG-1 overexpression is associated with poor prognosis and cisplatin resistance in advanced serous ovarian cancer.
