Emerging and endemic mosquito-borne viruses can be difficult to detect and monitor because they often cause asymptomatic infections in human or vertebrate animals or cause nonspecific febrile illness with a short recovery waiting period. Some of these pathogens circulate into complex cryptic cycles involving several animal species as reservoir or amplifying hosts. Detection of cases in vertebrate hosts can be complemented by entomological surveillance, but this method is not adapted to low infection rates in mosquito populations that typically occur in low or nonendemic areas. We identified West Nile virus circulation in Camargue, a wetland area in South of France, using a cost-effective xenomonitoring method based on the molecular detection of virus in excreta from trapped mosquitoes. We also succeeded at identifying the mosquito species community on several sampling sites, together with the vertebrate hosts on which they fed prior to being captured using amplicon-based metabarcoding on mosquito excreta without processing any mosquitoes. Mosquito excreta-based virus surveillance can complement standard surveillance methods because it is cost-effective and does not require personnel with a strong background in entomology. This strategy can also be used to noninvasively explore the ecological network underlying arbovirus circulation.

To implement and evaluate a simultaneous multi-slice balanced SSFP (SMS-bSSFP) perfusion sequence and compressed sensing reconstruction for cardiac MR perfusion imaging with full left ventricular (LV) coverage (nine slices/heartbeat) and high spatial resolution (1.4 × 1.4 mmA preliminary study was performed to evaluate the performance of blipped controlled aliasing in parallel imaging (CAIPI) and RF-CAIPI with gradient-controlled local Larmor adjustment (GC-LOLA) in the presence of fat. A nine-slice SMS-bSSFP sequence using RF-CAIPI with GC-LOLA with high spatial resolution (1.4 × 1.4 mmFat signal leakage was significantly higher for blipped CAIPI than for RF-CAIPI with GC-LOLA (7.9% vs. 1.2%, p = 0.010). All 10 SMS-bSSFP perfusion datasets resulted in 16/16 diagnostic myocardial segments. There were no significant differences between the SMS and conventional acquisitions in terms of image quality (2.6 ± 0.6 vs. 2.7 ± 0.2, p = 0.8) or perceived SNR (2.8 ± 0.3 vs. 2.7 ± 0.3, p = 0.3). Inter-reader variability was good for both image quality (ICC = 0.84) and perceived SNR (ICC = 0.70). Myocardial sharpness was improved using the SMS sequence compared to the conventional sequence (0.37 ± 0.08 vs 0.32 ± 0.05, p 0.001). There was no significant difference between measurements of upslope index for the SMS and conventional sequences (0.11 ± 0.04 vs. 0.11 ± 0.03, p = 0.84).SMS-bSSFP with multiband factor 3 and compressed sensing reconstruction enables cardiac MR perfusion imaging with three-fold increased spatial coverage and improved myocardial sharpness compared to a conventional sequence, without compromising perceived SNR, image quality, upslope index or number of diagnostic segments.

The fossil history of turtle and whale barnacles (Coronuloidea: Chelonibiidae, Platylepadidae, Coronulidae and †Emersoniidae) is fragmentary and has only been investigated in part. Morphological inferences and molecular phylogenetic analyses on extant specimens suggest that the roots of whale barnacles (Coronulidae) are to be found among the chelonibiid turtle barnacles, but the hard-part modifications that enabled early coronuloids to attach to the cetacean skin are still largely to be perceived. Here, we reappraise a fossil chelonibiid specimen from the Miocene of insular Tanzania that was previously referred to the living species Chelonibia caretta. This largely forgotten specimen is here described as the holotype of the new species †Chelonibia zanzibarensis. While similar to C. caretta, †C. zanzibarensis exhibits obvious external longitudinal parietal canals occurring in-between external longitudinal parietal septa that abut outwards to form T-shaped flanges, a character so far regarded as proper of the seemingly more derived Coronulidae and Platylepadidae. Along with these features, the presence of a substrate imprint on the shell exterior indicates that †C. zanzibarensis grasped its host's integument in much the same way as coronulids and platylepadids, albeit without the development of macroscopic parietal buttresses and bolsters. Thin section analyses of the inner parietal architecture of some extant and extinct coronuloids conclusively demonstrate that vestiges of comparable external parietal microstructures are present in some living members of Chelonibiidae. This observation strengthens the unity of Coronuloidea while significantly contributing to our understanding of the evolution of the coronuloid shell structure in adapting to a diverse spectrum of hosts.

The formation of the cerebellum is highly coordinated to obtain its characteristic morphology and all cerebellar cell types. During mouse postnatal development, cerebellar progenitors with astroglial‐like characteristics generate mainly astrocytes and oligodendrocytes. However, a subset of astroglial‐like progenitors found in the prospective white matter (PWM) produces astroglia and interneurons. Characterizing these cerebellar astroglia‐like progenitors and distinguishing their developmental fates is still elusive. Here, we reveal that astrocyte cell surface antigen‐2 (ACSA‐2), lately identified as ATPase, Na+/K+ transporting, beta 2 polypeptide, is expressed by glial precursors throughout postnatal cerebellar development. In contrast to common astrocyte markers, ACSA‐2 appears on PWM cells but is absent on Bergmann glia (BG) precursors. In the adult cerebellum, ACSA‐2 is broadly expressed extending to velate astrocytes in the granular layer, white matter astrocytes, and to a lesser extent to BG. Cell transplantation and transcriptomic analysis revealed that marker staining discriminates two postnatal progenitor pools. One subset is defined by the co‐expression of ACSA‐2 and GLAST and the expression of markers typical of parenchymal astrocytes. These are PWM precursors that are exclusively gliogenic. They produce predominantly white matter and granular layer astrocytes. Another subset is constituted by GLAST positive/ACSA‐2 negative precursors that express neurogenic and BG‐like progenitor genes. This population displays multipotency and gives rise to interneurons besides all glial types, including BG. In conclusion, this work reports about ACSA‐2, a marker that in combination with GLAST enables for the discrimination and isolation of multipotent and glia‐committed progenitors, which generate different types of cerebellar astrocytes., Glial cells of the cerebellum are still insufficiently classified. In this study we describe the subclassification of GLAST+ precursors in the prospective white matter using immunophenotyping, transcriptomics, and transplantation assays. A subclass of GLAST+ cells, which co‐express astrocyte cell surface antigen‐2 (ACSA‐2), generates only parenchymal astrocytes—as revealed by protein expression, their genetic signature and transplantation. The absence of the astrocyte marker (ACSA‐2) reveals GLAST+ cells to maintain a multipotent potential, giving rise to interneurons, glia, and Bergmann glia.

Lignocellulosic biomass is an abundant byproduct from cereal crops that can potentially be valorized as a feedstock to produce biomaterials. Zea mays CINNAMYL ALCOHOL DEHYDROGENASE 2 (ZmCAD2) is involved in lignification, and is a promising target to improve the cellulose‐to‐glucose conversion of maize stover. Here, we analyzed a field‐grown zmcad2 Mutator transposon insertional mutant. Zmcad2 mutant plants had an 18% lower Klason lignin content, whereas their cellulose content was similar to that of control lines. The lignin in zmcad2 mutants contained increased levels of hydroxycinnamaldehydes, i.e. the substrates of ZmCAD2, ferulic acid and tricin. Ferulates decorating hemicelluloses were not altered. Phenolic profiling further revealed that hydroxycinnamaldehydes are partly converted into (dihydro)ferulic acid and sinapic acid and their derivatives in zmcad2 mutants. Syringyl lactic acid hexoside, a metabolic sink in CAD‐deficient dicot trees, appeared not to be a sink in zmcad2 maize. The enzymatic cellulose‐to‐glucose conversion efficiency was determined after 10 different thermochemical pre‐treatments. Zmcad2 yielded significantly higher conversions compared with controls for almost every pre‐treatment. However, the relative increase in glucose yields after alkaline pre‐treatment was not higher than the relative increase when no pre‐treatment was applied, suggesting that the positive effect of the incorporation of hydroxycinnamaldehydes was leveled off by the negative effect of reduced p‐coumarate levels in the cell wall. Taken together, our results reveal how phenolic metabolism is affected in CAD‐deficient maize, and further support mutating CAD genes in cereal crops as a promising strategy to improve lignocellulosic biomass for sugar‐platform biorefineries.

In this review paper, a critical assessment of the main degradation processes in three key components of solid oxide fuel cells and electrolysers (negative and positive electrodes and the interconnect) is undertaken, attempting prioritization of respective degradation effects and recommendation of the best approaches in their experimental ascertainment and numerical modeling. Besides different approaches to quantifying the degradation rate of an operating solid oxide cell (SOC), the latest advancements in microstructural representation (3D imaging and reconstruction) of SOC electrodes are reviewed, applied to the quantification of triple-phase boundary (TPB) lengths and morphology evolution over time. The intrinsic degradation processes in the negative (fuel) electrode and the positive (oxygen) electrode are discussed, covering first the composition and governing mechanisms of the respective electrodes, followed by a comprehensive evaluation of the most important factors of degradation during operation. By this systematic identification of dominant degradation processes, measurement techniques, and modeling approaches, the foundations are laid for the definition of meaningful accelerated stress testing of SOC cells and stacks, which will help the technology achieve the constantly more demanding durability targets in market applications.

In 2014, the Japanese beetle Popillia japonica (Coleoptera: Scarabaeidae) was first spotted in northern Italy in the Nature Park of the Ticino Valley, its first detection in continental Europe. This polyphagous invasive species has the potential to cause serious losses to horticulture and agriculture. Particularly for its management in a Nature Park, environmentally friendly strategies are necessary. To develop baseline data for a biological control approach to the Italian outbreak of P. japonica, we conducted laboratory and field experiments testing several indigenous and commercial strains of the entomopathogenic nematode (EPN) species Heterorhabditis bacteriophora and Steinernema carpocapsae against P. japonica larvae. In the laboratory, strains of H. bacteriophora caused greater mortality (ranging from 57% to 100%) than those of S. carpocapsae (3% to 77%). In micro-plot field tests carried out at three different times, the most virulent indigenous EPN strain, H. bacteriophora ItH-LU1, showed again the best results ranging from 44% to 93% against young larvae. Finally, in a large-plot field trial, the commercial H. bacteriophora product provided 46% larval mortality. This study shows that H. bacteriophora strains have good potential as biological control agents of larvae of the invasive P. japonica in northern Italy.

Astrocytes are the most abundant cell type of the central nervous system and cover a broad range of functionalities. We report here the generation of a novel monoclonal antibody, anti-astrocyte cell surface antigen-2 (Anti-ACSA-2). Flow cytometry, immunohistochemistry and immunocytochemistry revealed that Anti-ACSA-2 reacted specifically with a not yet identified glycosylated surface molecule of murine astrocytes at all developmental stages. It did not show any labeling of non-astroglial cells such as neurons, oligodendrocytes, NG2+ cells, microglia, endothelial cells, leukocytes, or erythrocytes. Co-labeling studies of GLAST and ACSA-2 showed largely overlapping expression. However, there were also notable differences in protein expression levels and frequencies of single-positive subpopulations of cells in some regions of the CNS such as cerebellum, most prominently at early postnatal stages. In the neurogenic niches, the dentate gyrus of the hippocampus and the subventricular zone (SVZ), again a general overlap with slight differences in expression levels were observed. ACSA-2 was unlike GLAST not sensitive to papain-based tissue dissociation and allowed for a highly effective, acute, specific, and prospective purification of viable astrocytes based on a new rapid sorting procedure using Anti-ACSA-2 directly coupled to superparamagnetic MicroBeads. In conclusion, ACSA-2 appears to be a new surface marker for astrocytes, radial glia, neural stem cells and bipotent glial progenitor cells which opens up the possibility of further dissecting the characteristics of astroglial subpopulations and lineages.

Mucosa-associated lymphatic tissue (MALT) lymphoma of the breast is an extremely rare disease; its pathogenesis is not clear because of the rarity of disease, and the best diagnostic method has yet to be established. The metabolic behavior of this lymphoma is not still clear because only a few case reports are present in literature describing the possible role of fluorine-18-fluorodeoxyglucose positron emission tomography/computed tomography (18F-FDG PET/CT) in this field. This report presents 4 cases of women with histologically proven breast MALT lymphoma who underwent 7 18F-FDG PET/CT throughout the course of disease. All patients underwent staging PET/CT showing in all cases an FDG avid lesion corresponding to breast lymphoma; 3 patients underwent 18F-FDG PET/CT also after chemotherapy. Our results suggest that breast MALT lymphomas are 18F-FDG-avid lymphomas. Positron emission tomography/computed tomography showed heterogeneous but high FDG uptake (mean maximum standardized uptake value 7.9), suggesting that it could be part of diagnostic workup and restaging process.
