Increasing demand for river water now conflicts with an increasing desire to maintain riparian ecosystems. Efficiently managing river flows for riparian vegetation requires an understanding of the time scale of flow effects, but this information is limited by the absence of long-term studies of vegetation change in response to flow variation. To investigate the influence of short- and long-term flow variability and dam operation on riparian vegetation, we determined the occurrence of 107 plant species in 133 permanent plots of known inundating discharge along the Gunnison River in Colorado on five different occasions between 1990 and 2013. Individual species moved up and down the gradient of inundating discharge coincident with increases and decreases in mean annual flow, and the correlations between flow and species occurrence were strongest when flows were weighted by time before vegetation sampling with a median half-life of 1.5 years. Some tall, rhizomatous, perennial species, however, responded to flows on a longer time scale. Logistic regression of species occurrence showed a significant relation with inundation duration for 70 out of 107 species. Plot species richness and total vegetative cover decreased in association with desiccation at low inundation durations and with fluvial disturbance at high inundation durations. Within-plot similarity in species occurrence between years decreased strongly with increasing inundation duration. Moderate inundation durations were dominated by tall, rhizomatous, perennial herbs, including invasive Phalaris arundinacea (reed canary grass). Over the 23-year study period, species richness declined, and the proportion of rhizomatous perennials increased, consistent with the hypothesis that decreases in flow peaks and increases in low flows caused by flow regulation have decreased establishment opportunities for disturbance-dependent species. In summary, annual-scale changes in vegetation were strongly influenced by flow variation, and decadal-scale changes were influenced by decreases in fluvial disturbance from upstream flow regulation beginning decades prior to the onset of this study.

Management of riparian plant invasions across the landscape requires understanding the combined influence of climate, hydrology, geologic constraints and patterns of introduction. We measured abundance of nine riparian woody taxa at 456 stream gages across the western USA. We constructed conditional inference recursive binary partitioning models to discriminate the influence of eleven environmental variables on plant occurrence and abundance, focusing on the two most abundant non-native taxa, Tamarix spp. and Elaeagnus angustifolia, and their native competitor Populus deltoides. River reaches in this study were distributed along a composite gradient from cooler, wetter higher-elevation reaches with higher stream power and earlier snowmelt flood peaks to warmer, drier lower-elevation reaches with lower power and later peaks. Plant distributions were strongly related to climate, hydrologic and geomorphic factors, and introduction history. The strongest associations were with temperature and then precipitation. Among hydrologic and geomorphic variables, stream power, peak flow timing and 10-yr flood magnitude had stronger associations than did peak flow predictability, low-flow magnitude, mean annual flow and channel confinement. Nearby intentional planting of Elaeagnus was the best predictor of its occurrence, but planting of Tamarix was rare. Higher temperatures were associated with greater abundance of Tamarix relative to P. deltoides, and greater abundance of P. deltoides relative to Elaeagnus. Populus deltoides abundance was more strongly related to peak flow timing than was that of Elaeagnus or Tamarix. Higher stream power and larger 10-yr floods were associated with greater abundance of P. deltoides and Tamarix relative to Elaeagnus. Therefore, increases in temperature could increase abundance of Tamarix and decrease that of Elaeagnus relative to P. deltoides, changes in peak flow timing caused by climate change or dam operations could increase abundance of both invasive taxa, and dam-induced reductions in flood peaks could increase abundance of Elaeagnus relative to Tamarix and P. deltoides.

SUMMARY 1. Riparian vegetation composition, structure and abundance are governed to a large degree by river flow regime and flow-mediated fluvial processes. Streamflow regime exerts selective pressures on riparian vegetation, resulting in adaptations (trait syndromes) to specific flow attributes. Widespread modification of flow regimes by humans has resulted in extensive alteration of riparian vegetation communities. Some of the negative effects of altered flow regimes on vegetation may be reversed by restoring components of the natural flow regime. 2. Models have been developed that quantitatively relate components of the flow regime to attributes of riparian vegetation at the individual, population and community levels. Predictive models range from simple statistical relationships, to more complex stochastic matrix population models and dynamic simulation models. Of the dozens of predictive models reviewed here, most treat one or a few species, have many simplifying assumptions such as stable channel form, and do not specify the time-scale of response. In many cases, these models are very effective in developing alternative streamflow management plans for specific river reaches or segments but are not directly transferable to other rivers or other regions. 3. A primary goal in riparian ecology is to develop general frameworks for prediction of vegetation response to changing environmental conditions. The development of riparian vegetation-flow response guilds offers a framework for transferring information from rivers where flow standards have been developed to maintain desirable vegetation attributes, to rivers with little or no existing information. 4. We propose to organise riparian plants into non-phylogenetic groupings of species with shared traits that are related to components of hydrologic regime: life history, reproductive strategy, morphology, adaptations to fluvial disturbance and adaptations to water availability. Plants from any river or region may be grouped into these guilds and related to hydrologic attributes of a specific class of river using probabilistic response curves. 5. The transcriptional response to damaging agents is of fundamental significance for understanding mechanisms responsible for cell survival and genome maintenance. However, how damage signals are transmitted to the transcriptional apparatus is poorly understood. Here we identify two new regulators of the UV response transcriptome: Snf1, a nutrient-sensing kinase, and Rad23, a nucleotide excision repair factor with no previously known function in transcriptional control. Over half of all UV-responsive genes are dependent on Snf1 or Rad23 for proper regulation. After irradiation, Snf1 targets the Mig3 repressor, a new effector of the UV response. Snf1 and Rad23 are both required for the displacement of Mig3 from the UV-activated HUG1 promoter, and Rad23's activity is functionally linked to the proteasome 19S regulatory particle. Our data reveal overlapping functions for Snf1 and Rad23 in UV-responsive transcriptional regulation and provide mechanistic insight into the action of these factors at a UV-activated promoter. These results also highlight how diverse environmental stimuli are processed by a limited repertoire of signalling molecules to result in tailored patterns of gene expression.

Mot1 is a conserved Snf2/Swi2-related transcriptional regulator that uses ATP hydrolysis to displace TATA-binding protein (TBP) from DNA. Several models of the enzymatic mechanism have been proposed, including Mot1-catalyzed distortion of TBP structure, competition between Mot1 and DNA for the TBP DNA-binding surface, and ATP-driven translocation of Mot1 along DNA. Here, DNase I footprinting studies provide strong support for a ‘DNA-based' mechanism of Mot1, which we propose involves ATP-driven DNA translocation. Mot1 forms an asymmetric complex with the TBP core domain (TBPc)–DNA complex, contacting DNA both upstream and within the major groove of the TATA Box. Contact with upstream DNA is required for Mot1-mediated displacement of TBPc from DNA. Using the SsoRad54–DNA complex as a model, DNA-binding residues in Mot1 were identified that are critical for Mot1–TBPc–DNA complex formation and catalytic activity, thus placing Mot1 mechanistically within the helicase superfamily. We also report a novel ATP-independent TBPc displacement activity for Mot1 and describe conformational heterogeneity in the Mot1 ATPase, which is likely a general feature of other enzymes in this class.

Objectives: To derive a prediction rule using data available in the emergency department (ED) to identify a group of patients hospitalized for the treatment of heart failure who are at low risk of death and serious complications. Methods: The authors analyzed data for all 33,533 patients with a primary hospital discharge diagnosis of heart failure in 1999 who were admitted from EDs in Pennsylvania. Candidate predictors were demographic and medical history variables and the most abnormal examination or diagnostic test values measured in the ED (vital signs only) or on the first day of hospitalization. The authors constructed classification trees to identify a subgroup of patients with an observed rate of death or serious medical complications before discharge < 2%; the tree that identified the subgroup with the lowest rate of this outcome and an inpatient mortality rate < 1% was chosen. Results: Within the entire cohort, 4.5% of patients died and 6.8% survived to hospital discharge after experiencing a serious medical complication. The prediction rule used 21 prognostic factors to classify 17.2% of patients as low risk; 19 (0.3%) died and 59 (1.0%) survived to hospital discharge after experiencing a serious medical complication. Conclusions: This clinical prediction rule identified a group of patients hospitalized from the ED for the treatment of heart failure who were at low risk of adverse inpatient outcomes. Model performance needs to be examined in a cohort of patients with an ED diagnosis of heart failure and treated as outpatients or hospitalized.

Mot1 is an essential Snf2/Swi2-related ATPase and TATA-binding protein (TBP)-associated factor (TAF). In vitro, Mot1 utilizes ATP hydrolysis to disrupt TBP–DNA complexes, but the relationship of this activity to Mot1's in vivo function is unclear. Chromatin immunoprecipitation was used to determine how Mot1 affects the assembly of preinitiation complexes (PICs) at Mot1-controlled promoters in vivo. We find that the Mot1-repressed HSP26 and INO1 promoters are both regulated by TBP recruitment; inactivation of Mot1 leads to increased PIC formation coincident with derepression of transcription. For the Mot1-activated genes BNA1 and URA1, inactivation of Mot1 also leads, remarkably, to increased TBP binding to the promoters, despite the fact that transcription of these genes is obliterated in mot1 cells. In contrast, levels of Taf1, TFIIB, and RNA polymerase II are reduced at Mot1-activated promoters in mot1 cells. These results suggest that Mot1-mediated displacement of TBP underlies its mechanism of repression and activation at these genes. We suggest that at activated promoters, Mot1 disassembles transcriptionally inactive TBP, thereby facilitating the formation of a TBP complex that supports functional PIC assembly.

Summary Background: Bispectral index (BIS) is a signal processing device that potentially is a pharmacodynamic measure of the effects of anaesthesia on the central nervous system. Methods: In this prospective blinded study, we investigated the pharmacodynamic relationship between BIS, haemodynamic changes during anaesthesia and endtidal nonsteady state concentrations of sevoflurane in 30 children, mean age 3.3 ± 1.1 years, who were undergoing tonsillectomy and adenoidectomy. A standardized anaesthetic technique was used and included induction and maintenance with sevoflurane, nitrous oxide and oxygen. BIS, heart rate, mean arterial pressure (MAP) and endtidal sevoflurane (ETsevo) concentrations were continuously recorded and specifically noted at the time of intubation, placement of Dingman gag, incision of adenoid, adenoid out, incision of tonsil, tonsil one out, tonsil two out, last agent off, first spontaneous movement, first eye opening and extubation. The anaesthetist was blinded to BIS throughout the procedure. Objectives: Existing clinical practice guidelines for ordering prothrombin time (PT) and partial thromboplastin time (PTT) tests in the emergency department (ED) include physician expectation of an invasive procedure as a criterion. This study sought to determine whether this criterion accurately identifies patients who undergo an invasive procedure and whether an amalgam of these guidelines identifies patients at low risk of an adverse medical outcome. Methods: A prospective observational cohort study of adults treated in a university medical center ED between July 1997 and March 1998. Physicians were surveyed at order placement to determine the presence of guideline criteria. Adverse clinical outcomes defined as an International Normalized Ratio [INR] > 1.3 or PTT > 39.9 seconds combined with consequent directed medical therapy were abstracted from ED and the first 24 hours of inpatient medical records. Results: The sensitivity of ED physician expectation of an invasive procedure was 60.0%; therefore, it was excluded from both the PT and PTT guidelines. There were 553 patients with a PT test order; test order indications were absent in 190 (34.4%), of which one (0.5%, 95% CI = 0.1% to 2.9%) had adverse outcomes. There were 547 patients with a PTT test; test order indications were absent in 226 (41.3%), of which three (1.3%, 95% CI = 0.4 to 3.8%) had adverse outcomes. All three were taking warfarin when they presented to the ED. Conclusions: Emergency department physician expectation of an invasive procedure for patients with PT or PTT test orders is an insensitive predictor of patients who undergo such procedures. Clinical practice guidelines that exclude this criterion identify ED patients at the time of ED presentation at low risk for adverse medical outcomes in the ED or shortly after admission.

Study objectives To examine the psychometric and cardiac effects of pseudoephedrine at 1 and 3 atmospheres (atm) of pressure (0 and 66 feet of sea water, respectively), and to make recommendations about the agent's safety in the diving environment. Design Double-blind, placebo-controlled, crossover study. Setting Monoplace hyperbaric chamber of a university hospital. Subjects Thirty active divers (mean age 38 yrs). Intervention A bank of seven tests was used to assess cognitive function during four different simulated dive combinations: placebo-1 atm, placebo3 atm, pseudoephedrine-1 atm, and pseudoephedrine-3 atm. Measurements and main results Heart rate and cardiac rhythm were recorded during all dives. Repeated-measures analysis of variance was used to analyze the effects of pseudoephedrine, depth, and drug-depth interaction. No significant, independent effects of pseudoephedrine were seen on any of the seven psychometric test scores (p>0.05), although the drug tended to increase anxiety scores (p=0.092). Depth resulted in a significant increase in anxiety scores (p=0.021) and a significant decrease in verbal fluency test scores (p=0.041); it had no significant effects on the other five psychometric tests (p>0.05). Pseudoephedrine caused a significant increase (p=0.036) in mean heart rate, and depth caused a significant decrease (p=0.013). Neither pseudoephedrine nor depth affected cardiac rhythm. Conclusion Pseudoephedrine does not cause significant alterations in psychometric performance at 3 atm of pressure that might increase the risk of diving. Depth causes significant adverse effects on anxiety levels and semantic memory at 3 atm. Pseudoephedrine and depth have significant but opposite effects on heart rate; although, these effects are unlikely to be clinically significant during diving. It is unlikely that pseudoephedrine adds significant risk to the diver.

Recognition of the TATA box by the TATA-binding protein (TBP) is a highly regulated step in RNA polymerase II-dependent transcription. Several proteins have been proposed to regulate TBP activity, yet the TBP domains responsive to all these regulators have not been defined. Here we describe a new class of TBP mutants that increase transcription from core promoters in vivo. The majority of these mutations alter amino acids that cluster tightly on the TBP surface, defining a new TBP regulatory domain. The mutant TBP proteins are defective for binding the transcriptional regulator yNC2, are resistant to inhibition by yNC2 in vitro and exhibit allele-specific genetic interactions with yNC2. These results provide strong biochemical and genetic evidence that TBP is directly repressed in vivo, and define a new TBP domain important for transcriptional regulation.

To explore how high flows limit the streamward extent of riparian vegetation we quantified the effects of sediment mobilization and extended inundation on box elder (Acer negundo) saplings along the cobble-bed Gunnison River in Black Canyon of the Gunnison National Monument, Colorado, USA. We counted and aged box elders in 144 plots of 37.2 m2, and combined a hydraulic model with the hydrologic record to determine the maximum shear stress and number of growing-season days inundated for each plot in each year of the record. We quantified the effects of the two mortality factors by calculating the extreme values survived during the lifetime of trees sampled in 1994 and by recounting box elders in the plots following a high flow in 1995. Both mortality factors can be modeled as threshold functions; box elders are killed either by inundation for more than 85 days during the growing season or by shear stress that exceeds the critical value for mobilization of the underlying sediment particles. Construction of upstream reservoirs in the 1960s and 1970s reduced the proportion of the canyon bottom annually cleared of box elders by high flows. Furthermore, because the dams decreased the magnitude of high flows more than their duration, flow regulation has decreased the importance of sediment mobilization relative to extended inundation. We use the threshold functions and cross-section data to develop a response surface predicting the proportion of the canyon bottom cleared at any combination of flow magnitude and duration. This response surface allows vegetation removal to be incorporated into quantitative multi-objective water management decisions. Copyright © 1999 John Wiley & Sons, Ltd.

Flow variability plays a central role in structuring the physical environment of riverine ecosystems. However, natural variability in flows along many rivers has been modified by water management activities. We quantified the relationship between flow and establishment of the dominant tree (plains cottonwood, Populus deltoides subsp. monilifera) along one of the least hydrologically altered alluvial reaches of the Missouri River: Coal Banks Landing to Landusky, Montana. Our purpose was to refine our understanding of how local fluvial geomorphic processes condition the relationship between flow regime and cottonwood recruitment. We determined date and elevation of tree establishment and related this information to historical peak stage and discharge over a 112-yr hydrologic record. Of the excavated trees, 72% were established in the year of a flow >1400 m3/s (recurrence interval of 9.3 yr) or in the following 2 yr. Flows of this magnitude or greater create the necessary bare, moist establishment sites at an elevation high enough to allow cottonwoods to survive subsequent floods and ice jams. Almost all cottonwoods that have survived the most recent flood (1978) were established >1.2 m above the lower limit of perennial vegetation (active channel shelf). Most younger individuals were established between 0 and 1.2 m, and are unlikely to survive over the long term. Protection of riparian cottonwood forest along this National Wild and Scenic section of the Missouri River depends upon maintaining the historical magnitude, frequency, and duration of floods >1400 m3/s. Here, a relatively narrow valley constrains lateral channel movement that could otherwise facil- itate cottonwood recruitment at lower flows. Effective management of flows to promote or maintain cottonwood recruitment requires an understanding of locally dominant fluvial geomorphic processes.

The intense demand for river water in arid regions is resulting in widespread changes in riparian vegetation. We present a direct gradient method to predict the vegetation change resulting from a proposed upstream dam or diversion. Our method begins with the definition of vegetative cover types, based on a census of the existing vegetation in a set of 1 ° 2 m plots. A hydraulic model determines the discharge necessary to inundate each plot. We use the hydrologic record, as defined by a flow duration curve, to determine the inundation duration for each plot. This allows us to position cover types along a gradient of inundation duration. A change in river management results in a new flow duration curve, which is used to redistribute the cover types among the plots. Changes in vegetation are expressed in terms of the area occupied by each cover type. We applied this approach to riparian vegetation of the Black Canyon of the Gunnison National Monument along the Gunnison River in Colorado. We used TWINSPAN to cluster plots according to species occurrence. This analysis defined three vegetative cover types that were distinct in terms of inundation duration. Quantitative changes in the extent of cover types were estimated for three hypothetical flow regimes: two diversion alternatives with different minimum flows and a moving average modification of historical flows. Our results suggest that (1) it is possible to cause substantial changes in riparian vegetation without changing mean annual flow, and (2) riparian vegetation is especially sensitive to changes in minimum and maximum flows. Principal advantages of this method are simplicity and reliance on relatively standard elements of plant community ecology and hydrologic engineering. Limitations include use of a single environmental gradient, restrictive assumptions about changes in channel geometry, representation of vegetation as quasi—equilibrium cover types, and the need for model validation.
