In this study, the antibacterial and antibiofilm properties of pomegranate (Punica granatum L.) extracts (PPLs) prepared by 10g of pomegranate peels (PPL10) and 100 mL of different solvents (ethanol, methanol, and their acid combinations, and water) were investigated as sources of bioactive compounds against food related bacteria. The antimicrobial activities of the extracts were measured by using agar well diffusion assay. The acid-treated extracts exhibited the highest antimicrobial activities (31-34 mm). The biofilm formations were assessed by using a microplate reader (570 nm) after crystal violet staining. The prevention and removal of bacterial biofilms were also evaluated by using extracts prepared by 5g of initial pomegranate peel (PPL5) with the different solvents or 200 ppm chlorine solution. All PPL5 and PPL10 extracts diluted at different ratios inhibited and removed biofilms and the highest antibiofilm effects were up to 80% by acid-treated extracts. In addition, PPL5s and PPL10s were as effective as or more effective than chlorine for the prevention of biofilms. Therefore, from an economical perspective the PPL5s could represent promising candidates as natural antibiofilm agents for food industry.

Gadolinium-based contrast agents (GBCAs), because of their paramagnetic characteristics, are the ones mostly used in MRI to increase signal intensity. However, the use of contrast media has raised concerns on cellular toxic risks of these agents. Studies showed the accumulation of gadolinium after injection to humans with or without renal impairment. Also, there are findings obtained under in vitro and/or in vivo conditions that revealed conflicting results for their cytotoxic and genotoxic effects. Some of them declared damage in cells and genetic material; some others did not. Abnormal cell growth and genetic aberration are critical because they may lead to carcinogenesis in somatic cells or may be transferred to the next generations through germ cells. Therefore, understanding the effect of GBCAs on cells is important for their safer usage in clinical administrations to generate high-quality contrast-enhanced magnetic resonance images. Because of all these reasons, cellular toxicities—mainly genotoxic and cytotoxic effects—of GBCAs were reviewed in this paper.

BACKGROUND We aimed to evaluate the effectiveness of preoperative fine-needle aspiration biopsies (FNAB) by the postthyroidectomy pathology results. METHOD Seven hundred and ninety-five patients with FNAB and following thyroid operations which have been performed between April 2008 and December 2019 were included in this study. By comparing the results of the FNAB and final pathologies, the specificity, sensitivity, FNR, false positivity ratio (FPR), accuracy and also the effect of nodule diameter on these have been evaluated. In Bethesda III subgroup according to FNAB, we investigated the malignancy rates and in whom this risk has been increased more. RESULTS In our study, the sensitivity of FNAB is 73.40%, the specificity is 95.33%, the accuracy is 91.81%, FNR is 26.60% and FPR is 4.67%. In the patients with nodules ≥4 cm and

Permanent hypoparathyroidism is a severe clinical condition accompanied by low parathyroid hormone level. Conventional treatment requires lifelong medication, and daily drug usage has some side effects. To avoid this circumstance, transplantation is an alternative and curative option. Microencapsulation may be used as a transplantation approach particularly to evade immune response. In order to define treatment of permanent hypoparathyroidism, a 37-year-old female recipient who has permanent hypoparathyroidism was evaluated for 3 years. Routine tests, viral markers, and T and B lymphocyte cross-match tests were analyzed. In addition intradermal skin test was performed for ultrapure alginate. Microencapsulation of cultured parathyroid cells was performed with ultrapure alginate. Cell suspension was prepared and spheroids were generated with calcium chloride. Afterward, transplantation was performed with a laparoscopic approach in the omental tissue. The recipient was discharged from the hospital without complications. Serum calcium, parathyroid hormone (PTH), and phosphorus levels were observed throughout 1 year. During the follow-up period, no complications were observed. Serum calcium levels were increased significantly on day 10 and PTH levels were increased on day 25 as well. According to our knowledge, this is the first study where ultrapure alginate-based microencapsulated parathyroid cells were transplanted in the omental tissue. A significant increment of PTH levels was detected. Microencapsulated parathyroid cells showed the functionality of this technique for more than 1 year. This study showed that using ultrapure alginate-based microencapsulation without immunosuppression appears to be a promising technique. © 2019 International Center for Artificial Organs and Transplantation and Wiley Periodicals, Inc.

Iron is an essential trace element especially in cell proliferation, and growth for various cellular events. An increasing amount of research has shown that iron metabolism is altered in tumour cells which usually have rapid growth rates. However, the number of studies on iron metabolism, and calcium regulation are limited in drug-resistant tumour cells. Previously, we have shown that modulation of iron metabolism through iron chelation regulated the intracellular calcium, and increased the doxorubicin sensitivity. In the present study, we investigated the effects of iron on mRNA expression profiles of fifteen key genes (IP3 R1/2/3, RYR1/2, SERCA1/2/3, NCX1/2/3, PMCA1/2/3, and PMCA4) related to calcium homeostasis in the parental cell line K562 and its subclone doxorubicin-resistant K562 cells. According to the ΔΔCt method with a two-fold expression difference (P < .05) as a cut-off level, although iron showed differential effects on most of the genes, IP3 R and PMCA genes were especially determined to have changed significantly. These results show that iron metabolism is an important metabolism due to changes in the expression of genes involved in calcium regulation and is a new perspective to overcome cancer/drug resistance.

We examine the relation between the investment horizons of insiders and the information content of their trading activity regarding future stock returns. We conjecture that an insider’s investment horizon establishes a benchmark for expected patterns of continued trading behavior, and thus helps to identify unexpected insider trades, which should be more informative in efficient markets. Consistent with this conjecture, the trades of short horizon insiders are both more unexpected and more informed than those of long horizon insiders, on average. Short horizon insiders and their firms also tend to display attributes that are associated with a greater focus on short-termism.

To investigate the efficiency of Superb Microvascular Imaging (SMI; Toshiba Medical Systems, Tustin, CA) in assessing inflammation of the synovium in the knee of patients with juvenile idiopathic arthritis (JIA) compared with power Doppler ultrasound (PDUS).Both knees of patients with a diagnosis of clinically active JIA were examined with grayscale ultrasound (US). The knees with positive US and physical examination findings were included in group A, whereas the knees with positive US findings despite negative physical examination findings were included in group B. The observers calculated the vascularity index (VI) by manually drawing a region of interest onto the thickest part of the synovium using PDUS and SMI.The median SMI VI (observer 1, 4.9% [interquartile range (IQR), 3.6%]; observer 2, 4.1% [IQR, 4.6%]) exceeded the median PDUS VI (observer 1, 1.5% [IQR, 1.8%]; observer 2, 1.5 [IQR, 1.9%]; P .0001). In group B, the median SMI VI (observer 1, 2.85% [IQR, 8%]; observer 2, 3.1% [IQR, 6.3%]) exceeded the median PDUS VI (observer 1, 0.5% [IQR, 2%]; observer 2, 0.55% [IQR, 2.3%]; P = .002 and .001 for observers 1 and 2, respectively). No significant differences were observed between groups concerning the PDUS VI and SMI VI (P .05).Superb Microvascular Imaging was superior to PDUS in depicting blood flow in the hypertrophied synovial tissue in the knee of patients with clinically active JIA. Superb Microvascular Imaging seems to a promising tool and a valuable adjunct to conventional US in assessing inflammation of the synovial tissue in patients with JIA.

OBJECTIVES In order to evaluate the influence of the genetic load of 49 genetic variants known to be associated with PD on the age at onset as well as on clinical outcome parameters. BACKGROUND PD patients show a large variability in phenotype and progression reflecting interindividual heterogeneity. This might be influenced by a diverse genetic architecture. METHODS Six hundred seventeen PD patients were included in this study and stratified by their "genetic load," which is based on the weighted odds ratios of 49 genetic variants known to be associated with PD from genome-wide association studies. Clinical parameters (H & Y, UPDRS-III, MMSE, and Beck's Depression Inventory) were evaluated cross-sectionally and in a subgroup longitudinally over 8 years. RESULTS PD patients with the highest genetic load were younger at disease onset, whereas severity of clinical parameters were similar compared to patients with the lowest genetic load. These findings could be confirmed regarding progression to clinical endpoints in the longitudinal analysis. CONCLUSION A high genetic load is associated with a younger age at onset, which, in turn, might possibly promote more effective compensatory mechanisms resulting in a similar rate of disease progression. © 2018 International Parkinson and Movement Disorder Society.

OBJECTIVES To evaluate the ability of shear wave elastography (SWE) to detect renal parenchymal scar formation in patients with vesicoureteral reflux. METHODS We prospectively evaluated 49 patients with unilateral grade 2 or higher-degree VUR. All patients underwent dimercaptosuccinic acid (DMSA) scintigraphy for evaluation of the renal parenchymal scar. After the DMSA scan, 2 radiologists, who were blinded to clinical data and each other's measurements, evaluated the kidneys of the patients using SWE. The kidneys were divided into 3 parts: upper pole, middle region, and lower pole, and 3 regions of interest were placed to each part. Shear wave velocity (SWV) values were calculated using meters per second as a unit and recorded for each region. Afterward, SWV values were compared to DMSA results. RESULTS There was no significant difference between the observers' mean SWV values of kidneys with VUR without scar formation (mean ± SD, 2.11 ± 0.06 and 2.09 ± 0.05 m/s) and the contralateral normal kidney SVW values (2.11 ± 0.06 and 2.10 ± 0.05 m/s; P = .936 and .724, respectively). Background To describe the role and benefits of intraoperative sonographic (US) guidance in intracavitary brachytherapy of cervical cancer. Methods The data of 142 patients who received tandem-based intracavitary brachytherapy for cervical cancer between January 2010 and June 2015 were retrospectively reviewed. US guidance was carried out for tandem selection and appropriate application. The complications and applicator conformity were assessed with planning CT. Results Intracavitary brachytherapy was performed under US guidance for 412 insertions in 113 consecutive patients with cervical cancer. Before we started to use US guidance, applications were done in 29 patients: uterine perforation occurred in two patients (6.9%), the tandem length was short in two patients (6.9%), the tandem length was long in four patients (13.8%), and tandem was in myometrium in three patients (10.3%). We then decided to use US guidance routinely. With US guidance, only 1 of 113 patients had uterine perforation (0.9%), tandem length was short in only one patient (0.9%), and tandem was in myometrium in one patient (0.9%). Conclusions Real-time US provided safe and effective guidance for intracavitary brachytherapy of cervical cancer resulting in decreased rates of perforations and misplacement of applicators. © 2017 Wiley Periodicals, Inc. An amyloid goiter is the presence of amyloid protein in the thyroid in sufficient amounts to produce enlargement of the gland, accompanied by fat deposition of varying extents. It can be seen in long-standing inflammatory disorders such as familial Mediterranean fever. Imaging findings depend on the amount of fat and amyloid deposition; however, the main imaging finding is diffuse fatty infiltration of the thyroid. Herein, the multimodality imaging features in 3 cases of amyloid goiters secondary to familial Mediterranean fever are presented.

Several months before information becomes public, the level of short interest contains value-relevant information about publicly traded corporations. Short interest predicts future bad news, negative earnings surprises, and downward revisions in analyst earnings forecasts. This informational content is stronger for stocks that are harder to short. We also find that nearly half of the well-known cross-sectional relation between short interest and future stock returns is related to future changes in firms’ value-relevant information. Our results suggest that short interest predicts future returns, in part, due to short sellers’ ability to uncover unfavorable information about firms.

We prove long-time stability of linearly extrapolated BDF2 (BDF2LE) timestepping methods, together with finite element spatial discretizations, for incompressible Navier-Stokes equations (NSE) and related multiphysics problems. For the NSE, Boussinesq, and magnetohydrodynamics schemes, we prove unconditional long time L2 stability, provided external forces (and sources) are uniformly bounded in time. We also provide numerical experiments to compare stability of BDF2LE to linearly extrapolated Crank-Nicolson scheme for NSE, and find that BDF2LE has better stability properties, particularly for smaller viscosity values. © 2016 Wiley Periodicals, Inc. The prognostic importance of platelet–lymphocyte ratio (PLR) is already known for various artery diseases. In this study, the relationship between PLR and severity of erectile dysfunction (ED) is examined in patients with impotence. The data from patients suffering from erection problems was screened retrospectively. Detailed medical history, age, International Index of Erectile Function-5 (IIEF-5) scores, fasting blood glucose, lipid, whole blood count, and hormone profile values were examined. Patients with no ED were selected as the control group. All men answered the IIEF-5 questions and were then classified according to their scores. Patients were determined to have severe ED (scores 5–7), moderate ED (scores 8–16), or mild ED (scores 17–21). An IIEF-5 score greater than 21 was accepted for the control group. Rationale Urinary liver fatty acid binding protein (L-FABP) has been evaluated as a promising early biomarker of renal ischemia in human kidney transplant patients. The use of L-FABP in clinical practice requires that this biomarker be associated with an analytical method that combines specificity, accuracy and robustness. This study aimed to evaluate an optimized multiple reaction monitoring (MRM) method using ultrafast liquid chromatography coupled with tandem mass spectrometry to measure urinary L-FABP levels in renal transplant recipients. Methods Purified recombinant human L-FABP tryptic standard was analyzed by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS/MS) and liquid chromatography (LC)/MS/MS to select for peptides that provided specificity and adequate response in developing an MRM method for urinary L-FABP quantification. Human urine samples collected from kidney transplant recipients were isolated, concentrated, precipitated and trypsin digested before mass spectrometric analysis of L-FABP. L-FABP levels were also measured in urine samples by enzyme immunoassay. Results The tryptic peptide ion MH(+) of (50) FTITAGSK(57) (m/z 824) provided an adequate signal and was used for quantification of L-FABP under conditions employed for LC/MS/MS analysis. MALDI-TOF-MS/MS spectra obtained by collision-induced dissociation of the parent MH(+) ion (50) FTITAGSK(57) resulted in a y3 product ion that was used for quantitative analysis by the MRM method. Urinary L-FABP content measured by both ELISA and LC/MS/MS after transplantation was significantly higher compared to before transplantation levels. The Spearman correlation coefficient between the two methods was statistically significant. Intra-day and inter-day coefficients of variation provided good repeatability and reproducibility for validation of LC/MS/MS analysis. Conclusions LC/MS/MS quantification of L-FABP may provide a new reference method to determine changes in this potential biomarker in human kidney transplant patients.

I provide evidence that stocks experiencing unusually low trading volume over the week prior to earnings announcements have more unfavorable earnings surprises. This effect is more pronounced among stocks with higher short-selling constraints. These findings support the view that unusually low trading volume signals negative information, since, under short-selling constraints, informed agents with bad news stay by the sidelines. Changes in visibility or risk-based explanations are insufficient to explain the results. This evidence provides insights into why unusually low trading volume predicts price declines.

Summary Historical structures and preserving their cultural values are crucial issues for humanity, not only because of their link to important certain periods in the past, but also their unique architectural features. Although many historical structures continue to offer services while keeping their historical values, some of them are at high risk in seismic prone regions. Therefore, the understanding of their structural performance under strong ground motions has been of significant importance to the civil societies as well as engineering community. The structural systems of historical buildings, generally, consist of masonry walls or piers. The behavior of such walls is controlled by either deformation or force. Seismic performance evaluation of historical structures can be carried out within the framework of performance-based evaluation principles. This paper mainly discusses the basic principles to be considered in performance-based seismic evaluation of historical structures. Proposed seismic hazard levels, evaluation of existing seismic hazard, selection of earthquake ground motions as well as site geology, geological and tectonic settings of the area, seismic activity of the region and local soil conditions are needed for a thorough performance evaluation. Summary In this study, the antibiofilm activity of citric acid treatment on Staphylococcus aureus strains isolated from raw milk samples was evaluated. For this purpose, the prevention and removal of biofilm formation of S. aureus strains by citric acid treatments (2% and 10%) for 20 min were investigated for comparison with peracetic acid treatment (0.3%) on both microtitration plate and stainless steel coupons. The results indicated that the prevention and removal of biofilm formation and the numbers of prevented or removed S. aureus strains using citric acid treatments were observed to be higher than those using peracetic acid treatment on both surfaces. The prevention and removal of biofilm formation were substantially higher when the concentration of citric acid treatment increased from 2% to 10% and the stainless coupons were used. The results show that citric acid can be used as an alternative disinfectant in controlling biofilm formation in the dairy industry.

Fournier's gangrene is a rare but highly mortal infectious disease characterised by fulminant necrotising fasciitis involving the genital and perineal regions. The objective of this study is to analyse the demographics, clinical feature and treatment approaches as well as outcomes of Fournier's gangrene. Data were collected retrospectively from medical records and operative notes. Patient data were analysed by demographics, aetiological factors, clinical features, treatment approaches and outcomes. Twelve patients (five female and seven male) were enrolled in this study. The most common aetiology was perianal abscess (41·6%). Wound cultures showed a mixture of microorganisms in six (50%) patients. For faecal diversion, while colostomy was performed in six cases (50%), Flexi-Seal was used in two cases (16·6%). In four patients (33·4%), no faecal diversion was performed. Negative pressure wound therapy (NPWT) system was effective in the last four patients (33·4%). The mean hospitalisation period in patients who used NPWT was 18 days, while it was 20 days in the others. NPWT in Fournier's gangrene is a safe dressing method. It promotes granulation formation. Flexi-Seal faecal management is an alternative method to colostomy and provides protection from its associated complications. The combination of two devices (Flexi-Seal and NPWT) is an effective and comfortable method in the management of Fournier's gangrene in appropriate patients.

Introduction The purpose of this study was to assess the incidence, distribution and MRI characteristics of pelvic bone complications after radiation therapy. Methods The medical charts of 345 patients who received pelvic radiation therapy were retrospectively reviewed. A total of 122 patients, 99 women and 23 men, with a mean age of 57 (range 32–87 years) were included in this study. The MR images were retrospectively reviewed by two radiologists. Results Fatty replacement of bone marrow was seen in all patients. Pelvic bone complications and focal red marrow changes were identified in a total of 38 patients (31.1%). Pelvic insufficiency fracture was diagnosed in 17 patients (13.9%, with a total of 64 lesions) and radiation osteitis was diagnosed in 5 patients (4.1%, with a total of 13 lesions). Avascular necrosis of the femoral head was detected in one patient (0.8%). Focal red bone marrow changes were seen in 15 patients (12.3%). The median time from the end of radiotherapy to the diagnosis of pelvic bone complications or changes was 25 months (range 2–45 months). The 1-, 2-, and 3-year cumulative incidences were 22%, 41% and 49%, respectively. The distribution of insufficiency fractures was as follows: sacral ala, sacral body, ilium, acetabulum, pubis and lumbar spinal vertebra. The distribution of radiation osteitis was as follows: sacral ala, ilium and pubis. Conclusion Radiation-induced pelvic bone complications are not uncommon, and knowledge of characteristic imaging patterns is essential in order to rule out bone metastases and to avoid inaccurate or excessive treatment.
