The objective of this study was to characterise the nutritional potential of leaves and identify a diversity centre with low cyanide and high nutrient content among 178 Latin American cassava genotypes. This field‐based collection, represents the seven diversity centres, held at The International Center for Tropical Agriculture (CIAT Palmira, Colombia) by the Cassava Program. The cyanide, all‐trans‐β‐carotene and lutein concentrations in cassava leaves ranged from 346 ‐ 7484 ppm dry basis (db), from 174‐547 μg.g‐1 db and 15 ‐ 181 μg · g‐1 db respectively. Cassava leaves also showed significant levels of essential amino acids leucine, lysine, phenylalanine, valine and threonine, and average total protein content of 26.24 g · 100g‐1 db. Among seven diversity centres, South American rainforest group showed low cyanide and high carotene content in leaves. In addition, VEN77 and PAN51 genotypes stood out for having low cyanide in leaves and roots and high carotene in leaves. This genetic diversity can be used to select high potential progenitors for breeding purposes.

Le nouvel acide amine soufre isole du champignon Xerocomus Subtomentosus est caracterise par la formule brute C8H10N2O4S. Des etudes analytiques et des reactions de degradations ont montre qu'il s'agissait d'un acide α-amine bibasique derivant du thiazole. II s'agit de l'acide α-amino γ-(carboxy-4 thiazolyl-2) butyrique. La synthese de cette, substance corrobore la structure chimique proposee du compose naturel. The new Sulfur-containing amino acid which has been isolated from the mushroom Xerocomus Subtomentosus is characterized by the general formula C8H10N2O4S. Analytical studies and degrading reactions have shown that it was an α-amino dibasic acid derived from thiazol: the α-amino γ-[2-(4-carboxy)thiazolyl]butyric acid. The synthesis of that compound supports the chemical structure proposed for the natural compound.

Zusammenfassung In den Versuchen, uber die hier berichtet wird, wurde die Durchlassigkeit der Schleimhaut aus dem kranioventralen Bereich des Pansens fur Aminosauren in vitro gepruft. Sie schwankte stark von Tier zu Tier. Der unidirektionale Flux von der Lumen- zur Blutseite war etwa dem Konzentrationsunterschied proportional (Abb. 3). Die Permeabilitatskonstanten lagen zwischen 0,15 und 0,50 · 10−5 cm · sec−1. Das Besondere dieser Befunde bestand darin, das sich aus anfangs gleichen Konzentrationen von 14C-markierten Aminosauren zu beiden Seiten der Schleimhaut (0,1–0,5 μMol/ml je Aminosaure) im Laufe von 4 Stunden ein Konzentrations- und Aktivitatsunterschied entwickelte mit einer hoheren Aminosaurenkonzentration auf der Lumenseite (Abb. 4). Es zeigte sich, das besonders bei Alanin und Serin ein Konzentrationsgefalle zwischen der Blut- und der Lumenseite der Schleimhaut auftritt (Abb. 5). Summary In vitro studies on the secretion of amino acids in the rumen Rumen mucosa is readily permeable to a number of electrolytes, water, and physiologically metabolites of rumen fermentation. Its permeability for amino acids has not yet been fully understood. Surviving mucosal preparations from 2 years old bulls were incubated in saline (pH 7,2 300 mOsmol/1, 37 °C, saturated with 95% O2 and 5% CO2) for four hours. Sixteen 14C-labelled amino acids at concentrations of 0,1 to 0,5 μMol/ml were added to both sides of the mucosa in the presence of ATP and Vit. B6. A concentration- and radioactivity gradient developed during incubation with time. Higher concentrations were found at the lumen side of the mucosa (Fig. 4). The regression equations of total activity versus time at different sides of the mucosa were significantly different (p < 0,001, Fig. 4). Amino acid analysis of the bathing fluids revealed an uneven amino acids distribution between the two sides with higher concentrations in the fluids taken from the lumen side of the mucosa. It remains open whether this was due to an active transport of amino acids from the blood to the lumen side (Fig. 5). Highest concentration differences at the end of four hours incubation were observed for alanine and serine with about 17–18% higher concentrations at the lumen side (Fig. 5). Permeability constants were calculated from unidirectional fluxes from lumen- to blood side. They ranged from 0,15–0,5 · 10−5 cm · sec−1 but showed great variations from animal to animal (Fig. 3). Dans ces experiences, on teste la permeabilite de la muqueuse du rumen (region cranio-ventrale) aux acides amines in vitro. Elle varie fortement d'un animal a l'autre. Le flux a sens unique depuis la lumiere jusqu'au sang est approximativement proportionnel a la difference de concentration (fig. 3). Les constantes de permeabilite se trouvent entre 0,15 et 0,50 cm · sec−1. Il est curieux d'observer qu'a partir de concentrations identiques d'acides amines marques au C14, des deux cotes de la muqueuse (0,1–0,5 μMol/ml pour chaque acide amine), se developpe en 4 heures une difference de concentration et d'activite, avec une concentration plus elevee en acides amines du cote de la lumiere (fig. 4). L'alanine et la serine en particulier entrainent une chute de concentration entre le sang et la lumiere de la muqueuse (fig. 5). Con estos ensayos se examino in vitro la permeabilidad para los aminoacidos de la mucosa de la region craneoventral ruminal. La misma oscilaba mucho de animal a animal. El flux unidireccional de la cara de luz a la sanguinea era proporcional aproximadamente a la diferencia de concentraciones (fig. 3). Las constantes de permeabilidad se hallaban entre 0,15 y 0,50 cm. · seg.−1. Lo mas sorprendente de estos hallazgos consistia en que de las concentraciones iguales al principio de aminoacidos marcados en 14C a ambos lados de la mucosa (0,1–0,5 μmol/ml por cada aminoacido) se desarrollaba en el transcurso de 4 horas una diferencia de concentracion y actividad con aumento de aminoacidos en la cara de luz (fig. 4). Apreciose que sobre todo en la alanina y serina aparece una caida de concentracion entre al cara sanguinea y la de luz de la mucosa (fig. 5).

Zusammenfassung Bei Schafen wurde die Frage der Aminosaurenresorption und -sekretion durch die Pansenwand gepruft. In den gewaschenen Pansen wurde eine kunstliche Pansenflossigkeit mit 18 Aminosauren, von denen 15 mit M14C markiert waren, in einer Konzentration von 15 nMol/ml je Aminosaure und einer spezifischen Radioaktivitat von 0,021 μ;Ci/μMol gegeben. Wahrend des sechsstundigen Versuches wurde die kunstliche Pansenflussigkeit kontinuierlich mit CO2 begast. Der Zuflus von Speichel in den Pansen wurde mit Hilfe eines Speichelfangers unterbunden. Zum Ausgleich fur den durch den Speichelentzug entstandenen Elektrolyt- und Flussigkeitsverlust wurde eine dem Salzgehalt des Speichels entsprechende Losung kontinuierlich in die V. jugularis infundiert. Aus der Veranderung der Gesamtradioaktivitat, der spezifischen Radioaktivitat und der Konzentration der Aminosauren in der kunstlichen Pansenflussigkeit wurde festgestellt, das mit Ausnahme von Prolin alle in der Test-flussigkeit vorhandenen Aminosauren sowohl resorbiert als auch sezerniert werden. Auf Grund der Ergebnisse dieser Untersuchungen kann die Frage, ob Aminosauren aus dem Pansen resorbiert werden oder in die Pansenflussigkeit ubertreten, dahingehend beantwortet werden, das die einzelnen Aminosauren in unterschiedlichem Ausmas ein- und ausstromen. Prolin und Arginin sind in der kunstlichen Pansenflussigkeit nach 60 bzw. 270 Minuten nicht mehr nachweisbar. Histidin und Tryptophan nehmen im Laufe des Versuches ab. Der Ausstrom dieser Aminosauren ubersteigt den Einstrom. Phenylalanin, Tyrosin und Threonin, Serin, Methionin, Asparaginsaure und Cystin stromen mit etwa gleichgroser Rate ein und aus. Ihre Konzentration bleibt wahrend des Versuches gleich und ihre spezifische Markierung nimmt ab. Valin, Leucin und Isoleucin stromen in groserem Mase in den Pansen ein als aus. Ihr Nettoeinstrom ist allerdings gering. Bei Glutaminsaure, Alanin und Glycin dagegen ist ein nennenswerter Nettoeinstrom in den Pansen zu verzeichnen. Die Konzentration dieser Aminosauren steigt im Laufe von sechs Stunden von 15 nMol/ml auf 37, bzw. 45 und 90 nMol/ml. Der Nettoeinstrom ist bei den Aminosauren am grosten, die verstarkt in beiden Richtungen durch die Pansenwand diffundieren. Die physiologische Bedeutung des Nettoeinstroms einiger nicht essentieller Aminosauren wird im Hinblick auf die Zufuhr von Stickstoff und umsetzbarer Energie in den Pansen diskutiert. Summary Model studies on the passage of amino-acids through the rumen wall in vivo 2. Permeability of the rumen wall for individual amino-acids The bidirectional fluxes of amino acids through the rumen wall have been studied using two one year old fistulated wethers. The animals were fed twice daily with 300 g. of a concentrate diet and hay ad lib. three months before the experiments started. The emptied and washed rumen was filled with 2.45 1. of physiological saline pH 6.6 ± 0.1 and 325 mOsm., polyethyleneglycole (PEG) as a marker and antibiotics (c. f. Table 2). Furthermore the artificial ruminal fluid contained 18 amino acids at a concentration of 15 nMol/ml. each, 15 of them uniformly labelled with 14C. The saliva was collected during the six hours of the experiments. The artificial ruminal fluid was gassed continuously with CO2. A sodium—containing saliva solution (c. f. Table 1) was infused intravenously at a constant rate in order to account for the loss of sodium carbonate with saliva. During the six hours of incubation, pH and osmolarity including the concentrations of Na+, K+, Cl− and that of PEG remained approximately constant, indicating, that there was no nettflux of the three ions and of water (c. f. Table 3). Total nitrogen in the artificial ruminal fluid increased during three hours of incubation from 25 to 150 μg/ml. and reached a fairly constant level at that time (c. f. Fig. 1). The time courses of concentrations of 18 individual amino acids are shown in Fig. 3. Fig. 4 represents the initial and final concentrations of the amino acids in the artificial ruminal fluid. During the experimental period of six hours the concentrations of glycine, alanine and glutamic acid increased in the rumen from 15 nMoles/ml. to 90, 45 and 37 nMoles/ml. resp. This increase occurred mainly in the second half of the incubation period. From the linear decrease of total 14C-radioactivity on a semilogarithmic plot during the first three hours of the experiment (c. f. Fig. 5) allowing an estimate of the mean outflow of the amino acids through the ruminal wall. An outflow of 112.3 μMoles/hour was calculated equivalent to a rate of 17% per hour. From the decrease in specific radioactivity of the individual amino acid it could be deduced, that the nettinflux of the amino acid was positively correlated with its permeability. The physiological significance of the findings is discussed. Resume Recherche d'un modele pour l'etude du passage des acides amines a travers la paroi de la panse in vivo Communication 2: La permeabilite de la paroi de la panse pour les acides amines pris individuellement On a examine le probleme de la resorption et de la secretion des acides amines au travers de la paroi de la panse. On a introduit dans la panse lavee un liquide stomacal artificiel contenant 18 acides amines, dont 15 etaient marques au 14C, a une concentration de 15 n Mol/ml pour chaque acide et une radioactivite specifique de 0,021 μCi/μMol. On a ajoute continuellement, pendant l'essai d'une duree de six heures, du CO2, sous forme de gaz, dans le liquide de la panse. On a empeche l'arrivee de salive dans la panse a l'aide de capteur de salive. Pour compenser la perte en electrolyte et liquide resultant du retrait de la salive, on a infuse continuellement dans la veine jugulaire une solution dont la teneur en sel correspondait a la salive. On a etabli a partir du changement de la radioactivite totale et specifique et a partir de la concentration en acides amines dans le liquide artificiel de la panse, qu'a l'exception de la proline, tous les acides amines presents dans la solution de base etaient aussi bien resorbes que secretes. D'apres les resultats de ces recherches, a la question de savoir si les acides amines sont resorbes hors de la panse ou passent dans le liquide de la panse, on peut repondre que chaque acide amine penetre ou sort en quantite differente. On ne peut plus mettre en evidence la proline et l'arginine dans le liquide artificiel de la panse apres 60 et respectivement 270 minutes. L'histidine et le tryptophane diminuent au cours de l'experience. La sortie de ces acides amines surpasse l'entree. La phenylalanine, la tyrosine, la threonine, la serine, la methionine, l'acide aspartique et la cystine entrent et sortent dans les memes proportions; leur concentration reste egale pendant l'experience et leur marquage specifique diminue. La valine, la leucine et l'isoleucine penetrent en plus grande quantite dans la panse qu'elles n'en sortent. La quantite qui penetre est toutefois minime. On notera que la quantite d'acide glutamique, d'alanine et de glycine qui entre dans la panse est considerable. La concentration de ces acides amines passe pendant six heures de 15 n Mol/ml a 37, respectivement 45 et 90 n Mol/ml. Al rumen lavado se le aporto un liquido ruminal artificial con 18 aminoacidos, de los cuales 15 estaban marcados con 14C, en una concentracion de 15 nMol/ml. por aminoacido y una radiactividad de 0,021 μCi/μMol. Durante las seis horas que duro el ensayo, se gaseifico continuamente con CO2 el liquido artificial de la panza. El aflujo de saliva a la panza se impidio con ayuda de un colector salival. Para compensar la perdida de electrolitos y liquido creada por la sustraccion de saliva, se infundio de continuo en la v. yugular una solucion correspondiente al contenido salino salival. De la modificacion de la radiactividad total, de la radiactividad especifica y de la concentracion de aminoacidos en el liquido artificial de la panza, se pudo derivar que con excepcion de la prolina son tanto resorbidos como segregados todos los aminoacidos presentes en el liquido problema. A la vista de los resultados de estas experiencias, se puede contestar la cuestion de si los aminoacidos son resorbidos de la panza o si pasan al liquido ruminal en el sentido de que los distintos aminoacidos afluyen y efluyen en cantidades distintas. La prolina y arginina ya no son identificables en el liquido artificial de la panza tras 60 resp. 270 minutos. La histidina y el triptofano disminuyen a lo largo del ensayo. El reflujo de estos aminoacidos sobrepasa el aflujo. La fenilalanina, tirosina y treonina, serina, metionina, acido aspartico y cistina fluyen adentro y afuera una tasa casi igual de grande. Su concentracion permanece igual durante el ensayo y disminuye su marcacion especifica. La valina, leucina e isoleucina afluyen en cantidad mayor a la panza que la que sale. Sin embargo, su aflujo neto es bien escaso. Ahora bien, en acido glutamico, alanina y glicina se registra un aflujo neto a la panza digno de mencion. La concentracion de estos aminoacidos aumenta en el curso de seis horas de 15 nMol/ml. a 37 resp. 45 y 90 nMol/ml. El aflujo neto es maximo en los aminoacidos que difunden de manera reforzada en ambas direcciones a traves de la pared ruminal. Se discute la importancia fisiologica del aflujo neto de algunos aminoacidos no esenciales con respecto al aporte de nitrogeno y de energia convertible a la panza.

An experiment was conducted to determine true and apparent ileal amino-acid digestibility of a native cultivar of wheat (Mahdavi), autoclaved wheat (120°C for 30 min) and spaghetti by-products available in Iran. One hundred 21-day-old broiler chickens were fed a standard corn–soybean meal starter diet from day 0 to 28 post hatch. At 28 days 80 chicks were distributed according to possessing very nearly the same average bodyweight to 20 wire cages. The experimental units were allocated at random to five dietary treatments with four replicates per treatment. The basal diet contained corn and soybean meal as the major ingredients. Three test diets were formulated containing wheat, autoclaved wheat and spaghetti by-products as the sole source of dietary protein and each test diet was combined with a basal diet 50:50 on a weight basis to form three assay diets. The apparent and true digestibility of amino acid in the test ingredients were estimated from those in the assay diets basal/test diet mixtures, using the difference method. Zusammenfassung Veranderungen der Aminosaurekonzentrationen in Plasma und Leber cobaltdepletierter Rinder Die vorliegende Studie sollte die Wechselbeziehung zwischen dem Stoffwechsel des Cobalamins und der Aminosauren in cobaltdepletierten Rindern untersuchen. Um einen Cobaltmangel zu induzieren, erhielten zwei Gruppen uber 43 Wochen eine Grundration auf Maissilagebasis, die entweder cobaltarm (83 μg Co/kg) war, oder mit Cobalt bis zu einem Gesamtgehalt von 200 μg Co/kg supplementiert wurde. Der Cobaltmangcl lies sich aufgrund eines Appetitmangels, einem verminderten Wachstum und stark erniedrigter Vitamin B12-Konzentrationen in Serum und Leber diagnostizieren. Cobaltmangel bei Rindern, der in erster Linie eine Kombination aus verminderter Futteraufnahme und einem Vitamin B12-Mangel war, lies weder einen Einflus auf die Konzentrationen des Gesamtproteins und Albumins im Serum, noch eine quantitative Veranderung des Serumcreatinins und Leberammoniums erkennen. Allerdings kam es im Cobaltmangel zu einer starken Anreicherung des Harnstoffs im Serum, wahrend die Aktivitaten der Transaminasen abfielen. Cobaltmangel rief zahlreiche Veranderungen bei den Konzentrationen der Plasmaaminosauren hervor. Cobaltdepletierte Rinder zeigten eine um 53% geringere Methioninkonzentration im Blutplasma als die Kontrolltiere, aber auch die Analyse anderer essentieller Aminosauren lies signifikant verminderte Valin-, Leucin-, Isoleucin- und Threoninspiegel erkennen. Auch nicht-essentielle Aminosauren zeigten sich durch den Cobaltmangel signifikant beeinflust, wobei verminderte Arginin- und Thyrosinspiegel, aber auch geringere Taurinkonzentrationen deutlich wurden. Die starksten Veranderungen traten jedoch beim Plasmaserin und — homoeystein auf. Rinder mit Cobaltmangel zeigten 2,7 fach hohere Serinkonzentrationen und 4,8 fach hohere Homocysteinspiegel im Blutplasma als die Kontrolltiere. Im Gegensatz dazu blieben die Konzentrationen der Aminosauren in der Leber weitestgehend unbeeinflust. Die einzigen Veranderungen waren leichte erhohte Lysin-, Glutamin- und Serinkonzentrationen. Aus den Ergebnissen last sich ableiten, das die veranderten Konzentrationen der Plasmaaminosauren in engem Zusammenhang mit dem erniedrigten Vitamin B12-Status stehen, allerdings konnten auch die reduzierte Futteraufnahme und die mogliche verminderte bakterielle Aktivitat im Pansen zu den Veranderungen beitragen. Die stark veranderten Konzentrationen des Homocysteins, Methionins, Taurins und Serins im Blutplasma lassen sich jedoch ursachlich auf verminderte Vitamin B12 Speicher zuruckfuhren und spiegeln somit den Cobaltmangelzustand am besten wider.

Zur Identifizierung organischer stickstoffhaltiger niedermolekularer Stoffe in Klaranlagenablaufen wurden verschiedene Ablaufproben einer Belebtschlamm-Klaranlage mit Nitrifikationsstufe, in der kommunales Abwasser behandelt wird (10000 EWG), herangezogen. Nach Entwicklung von Anreicherungsmethoden an Reversed-Phasen wurden die angereicherten Stoffgemische ohne Derivatisierung unter verschiedenen Bedingungen gaschromatographisch (GC) aufgetrennt und massenspektrometrisch (MS) ausgewertet. Die unter Anwendung des PBM-Algorithmus ausgewerteten GC/MS-Spektren der angereicherten Proben (Anreicherungsfaktor 1000...20000) ergaben 133 Stickstoffverbindungen, die mit einer Wahrscheinlichkeit von mindestens 30% ausgegeben wurden. Davon wurden siebzehn Verbindungen mit einer Ubereinstimmungszahl > 90% und zehn mit einer Ubereinstimmungszahl zwischen 80 und 90% identifiziert. Darunter auch 9-Methylacridin, das in neun von zehn verschiedenen Proben gefunden wurde. Der erfaste niedermolekulare Anteil am DON (geloster organisch gebundener Stickstoff) wurde zu 20% abgeschatzt, wobei der hohermolekulare Anteil am DON maximal 70% betragt. Identification of Nitrogen-containing Compounds of Low Molecular Weight in Effluents of Biologically Treated Municipal Wastewater Enriching methods based on reversed phases were developed for the investigation of low-molecular compounds containing nitrogen in municipal wastewater effluent. The enriched mixtures were treated gas chromatographically without derivatization under different conditions. By application of the PBM-algorithm to the GC/MS spectra of the enriched samples (concentration factor 1000...20000), 133 nitrogen-containing compounds could be identified with a probability of occurence of at least 30% put out. Seventeen of these compounds were identified with > 90% probability and another ten with 80...90% probability. 9-Methylacridine was found in nine of ten processed samples investigated. The nitrogen content of the 133 identified compounds was estimated at about 20% of the low-molecular fraction of DON (dissolved organic nitrogen).

The effect of boiling and soaking in water on the mimosine and protein contents of young leaves, pods and seeds of the tropical leguminous tree Leucaena leucocephala (leucaena) was investigated. It was observed that the soluble protein content in leucaena tissue was overestimated because of the presence of mimosine. A more accurate method of determining the soluble protein content in leucaena tissue containing mimosine was developed and used in this study. It was found that boiling removed mimosine from young leaves, pods and seeds of leucaena but it also reduced the amount of soluble protein significantly. Soaking in water for 24 h eliminated 97% of mimosine from young leaves, pods and split seeds of leucaena, and up to 20% of mimosine from the whole young seeds without significantly reducing the soluble protein content.
