The EU Forestry Strategy 2030 includes an ambitious reforestation/afforestation plan aimed at planting 3 billion trees that should lead the European Union toward carbon neutrality by 2050. We argue that important ecological aspects were not sufficiently considered in the Strategy. The consequences of large-scale afforestation plans on biodiversity-rich open areas were poorly considered. “Where to plant” is a key aspect, as one of the targets for planting trees may be biodiversity-rich open areas, that are already declining due to natural shrub and tree encroachment. We suggested that urban areas and areas near linear infrastructures should be themain targets of afforestation. Concerning “What to plant” non-native species should be avoided in all cases. Among native species, certain geno-ecotypes may not equally perform under the effects of climate change. We therefore suggest a throughout revision of the EU Forestry Strategy 2030 with the inclusion of ecologically driven principles.

Inherited or somatic mutations in the MRE11, RAD50 and NBN genes increase the incidence of tumours, including medulloblastoma (MB). On the other hand, MRE11, RAD50 and NBS1 protein components of the MRN complex are often overexpressed and sometimes essential in cancer. In order to solve the apparent conundrum about the oncosuppressive or oncopromoting role of the MRN complex, we explored the functions of NBS1 in an MB-prone animal model.We generated and analysed the monoallelic or biallelic deletion of the Nbn gene in the context of the SmoA1 transgenic mouse, a Sonic Hedgehog (SHH)-dependent MB-prone animal model. We used normal and tumour tissues from these animal models, primary granule cell progenitors (GCPs) from genetically modified animals and NBS1-depleted primary MB cells, to uncover the effects of NBS1 depletion by RNA-Seq, by biochemical characterisation of the SHH pathway and the DNA damage response (DDR) as well as on the growth and clonogenic properties of GCPs.We found that monoallelic Nbn deletion increases SmoA1-dependent MB incidence. The southern branch of the Elizabeth River near Portsmouth, Virginia, USA, is one of the most creosote-polluted subestuaries in North America and the former location of the Atlantic Wood US Environmental Protection Agency Superfund site. We previously demonstrated that adult Atlantic Wood killifish collected in situ had severe hepatic lesions, including hepatoblastoma and hepatocellular carcinoma, as well as suppressed circulating antibody responses compared to a historical reference site. Moreover, several innate immune functions were higher in Atlantic Wood fish, including elevated expression of hepatic cyclooxygenase-2 (COX-2), suggesting a proinflammatory environment. To further examine the potential of Atlantic Wood contaminants to modulate innate immune function(s), the present study used RAW264.7 mouse macrophages as an in vitro model to develop new approach methodologies for rapid screening. Lipopolysaccharide (LPS)-stimulated nitric oxide secretion by macrophages is a rapid, sensitive, and predictive in vitro system for screening potentially immunotoxic contaminants as single compounds or as complex mixtures. Compared to the reference site, filter-sterilized Atlantic Wood sediment extracts (water accommodated fractions) induced nitric oxide and IL-6 secretion as well as inducible nitric oxide synthase and COX-2 proteins at levels comparable to or higher than those induced by LPS treatments alone. Extracts also increased phagocytic activity by macrophages. Using a limulus lysate assay, we show that bacterial endotoxin levels in Atlantic Wood extracts are higher than in reference extracts and that polymyxin-B chelation ameliorates proinflammatory effects. These findings illuminate the reality of sediment constituents other than toxic compounds previously associated with developmental abnormalities and carcinogenesis in killifish from the Atlantic Wood site. Perhaps these data also suggest the presence of contaminant-adapted consortia of sediment microbes at many heavily polluted sites worldwide compared to less contaminated sites. Key points It is unclear whether the visual input that accompanies a perturbation of a standing person can affect whether a recovery step is taken. Visual motion speeds were manipulated during unexpected forward and backward shoulder pulls. Visual motion that appeared slower than actual body motion reduced the initial in-place resistance to the perturbation. As a result of the modulation of the in-place response, less pull force was needed to trigger a step when visual velocity appeared slower than normal. The visuomotor postural response occurred earlier and was larger when the full-field visual input was paired with a mechanical perturbation. Abstract The present study aimed to determine how visual motion evoked by an upper body perturbation during standing affects compensatory postural responses. This was investigated by rotating the visual field forwards or backwards about the ankle, time-locked to a forwards or backwards shoulder pull. Kinematic, kinetic and electromyographic responses were recorded to a range of pull forces over 160 trials in 12 healthy adults (mean ± SD = 31 ± 5.8 years). Stepping threshold forces and in-place postural responses were compared between conditions. When the visual field moved in the same direction as the pull, so that the apparent velocity of the body was reduced (SLOW condition), the pull-force required to induce a step was less than when the visual field either rotated in the opposite direction (FAST) or was unaltered (NATURAL). For in-place responses, the body was displaced further in the direction of the pull in the SLOW condition. This was the result of a reduction in the resistive force from lower leg muscles 130 ms after the visual motion onset. In trials with no pull, the visual motion induced postural responses that were later (290 ms) and had smaller amplitudes compared to when visual motion is paired with an unexpected perturbation of the body. The results suggest that the apparent speed of the visual environment during a perturbation does influence whether a compensatory step is taken, not via a direct effect on the decision to step but by modulating the initial in-place response.

The partitioning of different grain‐size classes in gravity flow deposits is one of the key characteristics used to infer depositional processes. Turbidites have relatively clean sandstones with most of their clay deposited as part of a mudstone cap or as a distal mudstone layer, whereas sand‐bearing debrites commonly comprise mixtures of sand grains and interstitial clay; hybrid event beds develop alternations of clean and dirty (clay‐rich) sandstones in varying proportions. Analysis of co‐genetic mudstone caps in terms of thickness and composition is a novel approach that can provide new insight into gravity flow depositional processes. Bed thickness data from the ponded Castagnola system show that turbidites contain more clay overall than do hybrid event beds. The Castagnola system is characterized by deposits of two very different petrographic types. Thanks to this duality, analyses of sandstone and mudstones composition allow inference of which proportion of the clay in each of the deposit types was acquired en route. In combination with standard sedimentological observations the new data allow insight into the likely characteristics of their parent flows. Clean turbidites were deposited by lower concentration, long duration, erosive, muddy turbidity currents which were more efficient at fractionating clay particles away from their basal layer. Hybrid event beds were deposited by shorter duration, higher‐concentration, less‐erosive sandier flows which were less efficient at clay fractionation. The results are consistent with data from other turbidite systems (for example, Marnoso‐arenacea). The approach represents a new method to infer the controls on the degree of clay partitioning in gravity flow deposits.

In the history of diagnostics, breath analysis was one of the first method used until the breakthrough of biochemical testing technology. Today, breath analysis has made a comeback with the development of gas analyzers and e‐noses, demonstrating its power in its applicability for diagnosing a wide range of diseases. The physical basis of multiple chemical sensitivity (MCS), an emerging environmental disease, is difficult to understand because it is based on the scenario of chronic hypoxia, with a complex of chemical compounds that trigger the syndrome and result in multiple symptoms. The aim of this study was to investigate MCS by analyzing exhaled volatile organic compounds (VOCs). The volatile, metabolic picture could be a putative gold standard for understanding and diagnosing the disease. The study was based on recording in resting condition using the noninvasive passive e‐nose contactless breath test, the Olfactory Real‐Time Volatile Organic Compounds (ORT‐VOC) test in MCS, and control samples. The VOCs profile distinguished between disease and health. It also distinguished the gender‐related volatile profile with significant robustness. The results trace a putative compensatory physiological pathway elicited by increased lactate, leading to acidosis, and hyperventilation, resulting in the production of specific VOCs. We conclude that breath testing is a valuable tool to investigate the hypoxia‐related VOC profile, facilitating MCS diagnosis.

Chemically skinned fibres allow the study of human muscle contractile function in vitro. A particularly important parameter is specific force (SF), that is, maximal isometric force divided by cross-sectional area, representing contractile quality. Although SF varies substantially between studies, the magnitude and cause of this variability remains puzzling. Here, we aimed to summarize and explore the cause of variability in SF between studies. A systematic search was conducted in Medline, Embase and Web of Science databases in June 2020, yielding 137 data sets from 61 publications which studied healthy, young adults. Five-fold differences in mean SF data were observed. Adjustments to the reported data for key methodological differences allowed between-study comparisons to be made. However, adjustment for fibre shape, swelling and sarcomere length failed to significantly reduce SF variance (I2 = 96%). Interestingly, grouping papers based on shared authorship did reveal consistency within research groups. In addition, lower SF was found to be associated with higher phosphocreatine concentrations in the fibre activating solution and with Triton X-100 being used as a skinning agent. Although the analysis showed variance across the literature, the ratio of SF in single fibres containing myosin heavy chain isoforms IIA or I was found to be consistent across research groups. In conclusion, whilst the skinned fibre technique is reliable for studying in vitro force generation of single fibres, the composition of the solution used to activate fibres, which differs between research groups, is likely to heavily influence SF values.

Studies on federalism, regionalism and devolution often tackle the processes of institutional change in a country's polity merely as transitions from an institutional pattern to another, thus mainly focusing on the role of legal frameworks and Constitutional setting in the transition from an equilibrium to another. Such an approach overlooks the role of incremental dynamics: what happens after a new constitutional equilibrium has been reached? Does it stand still, crystallized by formal institutions, or does it evolve in time? This article illustrates the relevance of incremental institutional changes that occur after a major Constitutional reform and shows how an alteration in the distribution of resources among actors may heavily affect the territorial arrangement and the intergovernmental relations (IGRs) of a country. It focuses on the Italian case and the change in its IGRs between 2001 and 2016. Through a secondary analysis, a scrutiny of the trilateral game among the State, the Regions and the Local Authorities - played by reshaping and implementing the initial arrangement through gradual, piecemeal and layered modifications - will be conducted. Evidence is displayed through Dente (1997) resources (legal, political, financial and cognitive) framework. The main finding is that the change in the pattern turns out to be less visible, sometimes ambiguous and sticky, and more uncertain than a change of the pattern, but equally deep and effective. In the conclusion, some interpretative hypotheses for further research on the relevance of change in the pattern and its generalization for multi-level system countries are proposed.

The architecture of foreland basins and the resulting distribution of clastic sediments are related to the constant interplay between tectonics and sedimentation. Specifically, basin floor modifications strongly influence dimensions, continuity and connections of sand‐size and fine‐grained deposits. Given the increasing need to identify deep potential reservoir deposits, the large‐scale definition of clastic porous targets and their seals is a matter of interest for oil and gas industry. Here, we present the reconstruction of the Po Plain and Northern Adriatic Foreland Basin (with an extent of ca. 40,000 km2) and its Pliocene–Pleistocene evolution, as an example of a sedimentary clastic system controlled by strongly non‐cylindrical foreland geometry. The study is based on the basin‐scale mapping of six unconformity‐bounded sequences, performed by interpreting a dense network of seismic lines and correlating well‐log data. This provides a three‐dimensional model of the step‐by‐step evolution of the basin and a description of the sediment dispersal pattern. We found that the basin records the change from a continuous (cylindrical) to highly fragmented (non‐cylindrical) foredeep geometry during Late Pliocene. In the Northern Apennines case, the main factors driving the development of a non‐cylindrical geometry are mainly related to inherited inhomogeneity in the downgoing block linked to its Mesozoic extensional faulting, and the relative orientation of these lineaments with respect to the direction of orogen migration. During the late Pliocene–Pleistocene the two directions progressively became close to parallel, and the Northern Apennines system reacted changing from a cylindrical to a non‐cylindrical state.

The ageing process is characterized by a decline in several physiological functions resulting in a reduced capability to maintain homeostasis. The lowered homeostatic capacity seems to involve the carotid body (CB), whose role is to modulate ventilation and tissue oxygen supply. It thus plays a prime role in all ageing processes. Ageing causes marked changes in CB morphology. In older animals, it is enlarged and shows a concomitant decrease in the percentage of chemoreceptor tissue, as well as a proliferation of type II cells. The carotid glomitis is present with aggregates of lymphocytes and fibrosis of the lobules. Type I cells are dehydrated, with a profound vacuolization, a shrinking nucleus, and lipofuscin accumulation. With increased age, human CB shows a reduction in the number and volume of mitochondria, fewer synaptic junctions between glomi, along with a reduction in CB content of neurotransmitters, leading to a sort of 'physiological denervation'. Ageing could be interpreted as a cumulative result of oxidative damage to cells, which derives from aerobic metabolism. Moreover, metabolic rate is tightly correlated with life duration; thus a loss in mitochondrial function is one of the prime factors affecting CB ageing processes. The age-related reduction in synaptic junctions might be a self-protective mechanism through which cells buffer themselves against an accumulation of reactive oxygen species. The correlation between hypoxia and the life duration of CB cells remains an open question until how and why cells sense oxygen is understood.

Curcumin, a phenolic compound extracted from Curcuma longa, exerts multiple pharmacological effects, including an antimicrobial action. Mycobacterium abscessus, an environmental, nontuberculous, rapidly growing mycobacterium, is an emerging human pathogen causing serious lung infections and one of the most difficult to treat, due to its multidrug resistance and biofilm-forming ability. We wanted to evaluate the antimicrobial and antivirulence activity of curcumin and its ability to synergize with antibiotics against a clinical M. abscessus strain (29904), isolated from the bronchoaspirate of a 66-year-old woman admitted to hospital for suspected tuberculosis. Curcumin [minimum inhibitory concentrations (MIC) = 128 mg/L] was synergic (fractional inhibitory concentration index ≤0.5) with amikacin, clarithromycin, ciprofloxacin, and linezolid, to which strain 29904 showed resistance/intermediate susceptibility. Curcumin at 1/8 × MIC significantly reduced motility, whereas at 4 × MIC, it completely inhibited 4- and 8-day mature biofilms. Synergistic combinations of curcumin and amikacin induced a general reduction in microbial aggregates and substantial loss in cell viability. Disruption of 4- and 8-day biofilms was the main effect detected when curcumin was the predominant compound. The present findings support previous evidence that curcumin is a potential antibiotic resistance breaker. Curcumin, either alone or combined with antibiotics, could provide a novel strategy to combat antibiotic resistance and virulence of M. abscessus.

High-level, acute exposures to individual polycyclic aromatic hydrocarbons (PAHs) and complex PAH mixtures result in cardiac abnormalities in developing fish embryos. Whereas acute PAH exposures can be developmentally lethal, little is known about the later life consequences of early life, lower level PAH exposures in survivors. A population of PAH-adapted Fundulus heteroclitus from the PAH-contaminated Superfund site, Atlantic Wood Industries, Elizabeth River, Portsmouth, Virginia, United States, is highly resistant to acute PAH cardiac teratogenicity. We sought to determine and characterize long-term swimming performance and cardiac histological alterations of a subteratogenic PAH mixture exposure in both reference killifish and PAH-adapted Atlantic Wood killifish embryos. Killifish from a relatively uncontaminated reference site, King's Creek, Virginia, United States, and Atlantic Wood killifish were treated with dilutions of Elizabeth River sediment extract at 24 h post fertilization (hpf). Two proven subteratogenic dilutions, 0.1 and 1.0% Elizabeth River sediment extract (total PAH 5.04 and 50.4 µg/L, respectively), were used for embryo exposures. Then, at 5-mo post hatching, killifish were subjected to a swim performance test. A separate subset of these individuals was processed for cardiac histological analysis. Unexposed King's Creek killifish significantly outperformed the unexposed Atlantic Wood killifish in swimming performance as measured by Ucrit (i.e., critical swimming speed). However, King's Creek killifish exposed to Elizabeth River sediment extract (both 0.1 and 1.0%) showed significant declines in Ucrit. Histological analysis revealed the presence of blood in the pericardium of King's Creek killifish. Although Atlantic Wood killifish showed baseline performance deficits relative to King's Creek killifish, their pericardial cavities were nearly free of blood and atrial and ventricular alterations. Implant sealing capability is a crucial issue in assessment of implant success and peri-implant marginal bone loss. Clinical studies demonstrated presence of viable bacteria in the internal part of functioning implants during tissue healing. For this study, a volatile organic compounds (VOCs) emission test was developed to evaluate the existing "seal" between implant and healing screw. Two kinds of implant-screw connection were compared: 1) internal hexagon; and 2) cone Morse.Fifteen patients were enrolled in the study, three males and 12 females, who required fixed prosthetic rehabilitation. A total of 37 implants was placed, 23 with a cone Morse taper internal connection and 14 with a screw-retained internal hexagon abutment. VOCs real-time measures were performed in the implant site immediately after removing the healing screw.Statistical analysis was carried out. Results showed VOCs maximum peak amplitude in cone Morse versus internal hexagon showed significant difference (P0.001), whereas VOCs time to peak showed no significant difference (P = 0.7).Use of the new methodology for the VOCs emission test may lead to important new data for understanding how the "failed" attachment of implant components, in two-part assemblies, may contribute to implant losses. In particular, study results support the hypothesis that the microgap of the implant-screw healing junction could cause differences in bacterial penetration. VOCs emission test evaluation represents a new diagnostic tool with an effective approach to quickly analyze, in real time, sealing capability of dental implants with healing screw interfaces.

The source of visual motion is inherently ambiguous such that movement of objects in the environment can evoke self-motion illusions and postural adjustments. Theoretically, the brain can mitigate this problem by combining visual signals with other types of information. A Bayesian model that achieves this was previously proposed and predicts a decreasing gain of postural response with increasing visual motion speed. Here we test this prediction for discrete, unidirectional, full-field visual rotations in the frontal plane of standing subjects. The speed (0.75–48 deg s–1) and direction of visual rotation was pseudo-randomly varied and mediolateral responses were measured from displacements of the trunk and horizontal ground reaction forces. The behaviour evoked by this visual rotation was more complex than has hitherto been reported, consisting broadly of two consecutive components with respective latencies of ∼190 ms and >0.7 s. Both components were sensitive to visual rotation speed, but with diametrically opposite relationships. Thus, the early component decreased with faster visual rotation, while the later component increased. Furthermore, the decrease in size of the early component was not achieved by a simple attenuation of gain, but by a change in its temporal structure. We conclude that the two components represent expressions of different motor functions, both pertinent to the control of bipedal stance. We propose that the early response stems from the balance control system attempting to minimise unintended body motion, while the later response arises from the postural control system attempting to align the body with gravity.
