Abstract The atmospheric CO2 growth rate is a fundamental measure of climate forcing. NOAA's growth rate estimates, derived from in situ observations at the marine boundary layer (MBL), serve as the benchmark in policy and science. However, NOAA's MBL‐based method encounters challenges in accurately estimating the whole‐atmosphere CO2 growth rate at sub‐annual scales. Here we introduce the Growth Rate from Satellite Observations (GRESO) method as a complementary approach to estimate the whole‐atmosphere CO2 growth rate utilizing satellite data. Satellite CO2 observations offer extensive atmospheric coverage that extends the capability of the current NOAA benchmark. We assess the sampling errors of the GRESO and NOAA methods using 10 atmospheric transport model simulations. The simulations generate synthetic OCO‐2 satellite and NOAA MBL data for calculating CO2 growth rates, which are compared against the global sum of carbon fluxes used as model inputs. We find good performance for the NOAA method (R = 0.93, RMSE = 0.12 ppm year−1 or 0.25 PgC year−1). GRESO demonstrates lower sampling errors (R = 1.00; RMSE = 0.04 ppm year−1 or 0.09 PgC year−1). Additionally, GRESO shows better performance at monthly scales than the NOAA method (R = 0.76 vs. 0.47, respectively). Due to CO2's atmospheric longevity, the NOAA method accurately captures growth rates over 5‐year intervals. GRESO's robustness across partial coverage configurations (ocean or land data) shows that satellites can be promising tools for low‐latency CO2 growth rate information, provided the systematic biases are minimized using in situ observations. Along with accurate and calibrated NOAA in situ data, satellite‐derived growth rates can provide information about the global carbon cycle at sub‐annual scales.

Abstract In this monocentric prospective study, the influence on long‐term outcomes of peripheral blood levels of monocytic‐myeloid‐derived suppressive cells (M‐MDSC) was investigated in 56 patients with acute leukemia (myeloid n = 47; lymphoid n = 9) before and after (Days+60/+90) allogeneic hematopoietic stem cell transplantation (Allo‐HSCT). A risk of relapse was found to be associated with a level of pregraft M‐MDSC above 1.4% by ROC curve analysis. In multivariate analysis, this threshold retained a strong statistical significance (HR: 5.94 [2.09–16.87], p = 0.001). Considering only the group of patients who were in complete remission prior to Allo‐HSCT (n = 44), a significant prediction of relapse was found to be associated, in multivariate analysis, with a level of pregraft M‐MDSC above 1.4% (HR: 55.01 [14.95–202.37], p

Abstract The Surface Ocean CO2 Atlas (SOCAT) of CO2 fugacity (fCO2) observations is a key resource supporting annual assessments of CO2 uptake by the ocean and its side effects on the marine ecosystem. SOCAT data are usually released with a lag of up to 1.5 years which hampers timely quantification of recent variations of carbon fluxes between the Earth System components, not only with the ocean. This study uses a statistical ensemble approach to analyze fCO2 with a latency of one month only based on the previous SOCAT release and a series of predictors. Results indicate a modest degradation in a retrospective prediction test for 2021–2022. The generated fCO2 and fluxes for January–August 2023 show a progressive reduction in the Equatorial Pacific source following the La Niña retreat. A breaking‐record decrease in the northeastern Atlantic CO2 sink has been diagnosed on account of the marine heatwave event in June 2023.

Abstract Congenital erythropoietic porphyria (CEP), or Gunther disease, is a rare genetic disease responsible for severe dermatologic, hepatic and/or haematological damages related to the deficient activity of the uroporphyrinogen III synthase. Allogeneic stem cell transplantation (Allo‐SCT) represents the only curative treatment and few allotransplanted cases have been reported in children but not in adults. Here we report for the first time the successful cure of a 46‐year old man with CEP with a 5‐year follow‐up after Allo‐SCT.

Abstract Stakeholder engagement (SkE) in research is currently experiencing significant growth within the fields of environmental and sustainability sciences. Stakeholder engagement ensures the relevance of research questions to societal expectations and the uptake and salience of the co‐produced knowledge and results for their use in the decision‐making process. In a context of societal challenges regarding biodiversity conservation and the sustainability of marine and freshwater social‐ecological systems (SESs), participatory approaches constitute key methods in applied research involving actions and decision‐making. There are, however, many gaps in the practical, conceptual and ethical ways stakeholders have been involved in research. We propose here a systematic map of the literature on SkE in research on marine and aquatic SESs carried out on French European and overseas territories, in order to draw up the first comprehensive overview of how SkE has developed and changed since 1945. This systematic map will identify a representative list of scientific articles on SkE in French marine and freshwater social‐ecological research. The literature search will include both academic literature (e.g. peer‐reviewed articles, reviews, meta‐analyses) and grey literature (e.g. reports, working papers) using the most relevant search engines for the scientific literature published between 1945 and 2023. Retrieved publications will be reviewed for relevance according to a predefined set of eligibility/ineligibility criteria by a group of trained reviewers. The eligibility check will be done in two successive screening steps: (1) title and abstract and (2) full text, each independently performed by two reviewers. All retained literature will be subjected to coding and metadata extraction using the Sysrev platform. No validity assessment will be undertaken. A database of the metadata extracted will be provided, along with a narrative description of the evidence base, and a set of figures and tables summarizing the relevant characteristics of the studies. This systematic map will provide a reliable overview of SkE conducted in research projects on French aquatic social‐ecological systems to strengthen the science–society relationship and help future research projects implement efficient and sustainable SkE processes in France and elsewhere.

Abstract The magnitude and spatial pattern of anomalous net biome exchange (NBE) induced by the 2015/16 El Niño over Amazonian rainforests remain uncertain. We here investigated them using multi‐model posterior NBE products in the Orbiting Carbon Observatory‐2 (OCO‐2) version 10 modeling intercomparison project. Results suggest that relative to the annual NBE average in 2017/18, larger anomalous carbon release occurred over the eastern and northern Amazonian rainforests in 2015/16, with a total flux of approximately 0.4 PgC yr−1 after assimilating satellite‐observed column CO2 concentrations (XCO2) over land. We further find that this anomalous spatial pattern was predominantly determined by soil dryness, while the total positive NBE anomaly was dominated by higher temperature with its contribution of approximately 68～70%. We believe that atmospheric inversions assimilating more satellite‐observed XCO2 in future can provide us more comprehensive understanding how Amazonian rainforests cope with the abiotic stresses induced by El Niño events.

Abstract The impact of pre‐transplant anti‐severe acute respiratory syndrome coronavirus‐2 (SARS‐CoV‐2) vaccine in 20 recipients of allogeneic hematopoietic stem cell transplantation (Allo‐HSCT) and/or their donors is reported here, showing that the persistence of anti‐SARS‐CoV‐2 antibodies can be detected in almost all patients, whatever the type of vaccine used, and up to 9 months post transplant. Also, an anti‐SARS‐CoV‐2 spike glycoprotein CD3+ T‐cell response could be detected in six (35%) of 17 evaluable patients. This study provides a rationale to consider anti‐SARS‐CoV‐2 vaccination of both recipients and donors before Allo‐HSCT.

Abstract Background Post‐transplant cyclophosphamide (PTCY) alone as graft‐versus‐host disease (GVHD) prophylaxis may avoid/reduce short‐ and mid‐term toxicities of drugs commonly used for GVHD prophylaxis, accelerate immune reconstitution after the graft to decrease infections and facilitate the early integration of adjunct maintenance therapies to prevent relapse. Objective A prospective phase 2 study was designed in order to assess the feasibility and safety of PTCY as a sole GVHD prophylaxis in adult patients receiving a Baltimore‐based reduced‐intensity conditioning (RIC) peripheral blood (PB) allogeneic hematopoietic stem cell transplantation (Allo‐HSCT) with a matched donor. Study design Patients were planned to be included stepwise up to 59 evaluable PTCY recipients, in order to be able to stop the protocol in case of excessive corticosteroid resistant grade 3–4 severe acute GVHD (aGVHD). Because a high incidence of grade 2–4 aGVHD was observed after analysis of the first 27 patients, the protocol was amended to test the addition of 1 day of anti‐thymoglobulin to PTCY. In spite of this, the trial had to be stopped after 38 treated patients, because of an unacceptable rate of grade 3–4 aGVHD. Donors were matched related to 12 patients and unrelated to 26. Results With a median follow‐up of 29.6 months, 2‐year overall, disease‐free and GVHD‐free relapse‐free (GRFS) survivals were respectively 65.4%, 62.1% and 46.9%. Cumulative incidences of grade 2–4 and 3–4 aGVHD at day 100 were 52.6% and 21.1%, respectively, while that of moderate/severe chronic(c) GVHD was 15.7% at 2 years. Addition of ATG to PTCY did influence neither aGVHD, cGVHD nor GRFS. Conclusion Despite paradoxically good survivals, especially GRFS, this study failed to demonstrate that PTCY (± ATG) alone can be used for Baltimore‐based RIC PB Allo‐HSCT with matched donors. Other combinations should be tested to try and avoid long‐term use of immunosuppressive drugs following Allo‐HSCT in this setting.

Abstract The ability of global transport models to go up in resolution becomes discriminating for greenhouse gas atmospheric inversions. This paper describes the porting on Graphics Processing Units of the global transport model currently used in the European operational Copernicus Atmosphere Monitoring Service (CAMS) for CO2 and N2O inversions. It represents an important milestone to achieve sub‐degree resolution. The code includes not only the direct model but also its tangent‐linear and its adjoint versions which are needed in variational inversions. Tests were carried out for CO2 at a resolution of 2.50° in longitude, 1.27° in latitude and 79 layers in the vertical, corresponding to 1,626,768 3D cells, 4.5 times more than the current standard resolution of the model used in the CAMS reanalyzes. A month's worth of computation of the tangent‐linear and of the adjoint versions now takes 2.5 min, including 50 s for reading meteorological data.

Graft versus host disease (GVHD) prophylaxis with posttransplantation cyclophosphamide (PTCY) has been established to reduce severe GVHD, and thereby potentially reducing nonrelapse mortality (NRM) after allogeneic stem cell transplantation (alloSCT). We evaluated the predictive capacity of established NRM-risk scores in patients receiving PTCY-based GVHD prophylaxis, and subsequently developed and validated a novel PTCY-specific NRM-risk model. Adult patients (n = 1861) with AML or ALL in first complete remission who received alloSCT with PTCY-based GVHD prophylaxis were included. The PTCY-risk score was developed using multivariable Fine and Gray regression, selecting parameters from the hematopoietic cell transplantation-comorbidity index (HCT-CI) and European Group for Blood and Marrow Transplantation (EBMT) score with a subdistribution hazard ratio (SHR) of ≥1.2 for 2-year NRM in the training set (70% split), which was validated in the test set (30%). The performance of the EBMT score, HCT-CI, and integrated EBMT score was relatively poor for discriminating 2-year NRM (c-statistic 51.7%, 56.6%, and 59.2%, respectively). The PTCY-risk score included 10 variables which were collapsed in 3 risk groups estimating 2-year NRM of 11% ± 2%, 19% ± 2%, and 36% ± 3% (training set, c-statistic 64%), and 11% ± 2%, 18% ± 3%, and 31% ± 5% (test set, c-statistic 63%), which also translated into different overall survival. Collectively, we developed an NRM-risk score for acute leukemia patients receiving PTCY that better predicted 2-year NRM compared with existing models, which might be applicable to the specific toxicities of high-dose cyclophosphamide.

Abstract The poor prognosis of acute T‐cell lymphoblastic leukemia (T‐ALL) and T‐cell lymphoblastic lymphoma (T‐LBL) in older adults and patients with relapsed/refractory illness is an unmet clinical need, as there is no defined standard of care and there are few treatment options. Abnormally elevated CD38 expression in T‐ALL and T‐LBL is associated with tumor expansion and disease development, making CD38 a potential target for anti‐T‐ALL and T‐LBL treatment. Isatuximab is a monoclonal antibody that binds to a specific epitope on CD38. The purpose of the study was to assess the efficacy and safety of isatuximab monotherapy in a phase 2, multicenter, one‐arm, open‐label study in patients with relapsed or refractory T‐ALL or T‐LBL (Clinical Trials.gov identifier NCT02999633). The primary endpoint was to assess the efficacy of isatuximab by overall response rate (ORR). An interim analysis based on the efficacy and safety of isatuximab in the first 19 patients enrolled was scheduled, however only 14 patients were enrolled in the study. No patient achieved complete response (CR) or CR with incomplete peripheral recovery. Most patients (11 [78.6%]) developed progressive disease and had progressive disease as their best response. A total of 10 (71.4%) patients had treatment emergent adverse events considered treatment‐related, with infusion reactions as the most frequent drug‐related TEAE, occurring in 8 (57.1%) patients. Despite the low efficacy of isatuximab in the current study, it is likely that the use of immunotherapy medication in T‐ALL will be expanded through logically targeted approaches, together with advances in the design of T‐cell therapy and clinical experience and will provide restorative options beyond chemotherapy and targeted treatments.

Abstract Introduction Various surgical centers tend to postpone a kidney transplantation (KT) to the following morning than to operate at night‐time. The objective of our study was to assess whether there was any difference between daytime and night‐time renal transplantation in our institution. Method This study is a retrospective monocentric study including all the KTs that were performed between 2012 and 2013 by transplant expert surgeons in our institution. Clavien‐Dindo (CD) complications were classified according to 7 variables going from 1 to 5. Time before postgraft diuresis and delayed graft function (DGF) were also analyzed. Two groups of patients were formed according to threshold value of incision time (6.30 p.m.). Data comparison were performed using the Kruskal–Wallis nonparametric test. Outcomes of allogeneic hematopoietic cell transplantation (allo-HCT) for adult acute lymphoblastic leukemia (ALL) have improved over time. Studies have shown that total body irradiation (TBI) is the preferable type of myeloablative conditioning (MAC). However, outcomes based on central nervous system (CNS) involvement, namely CNS-positive versus CNS-negative, have not been compared. Here, we evaluated outcomes of 547 patients (CNS-positive = 96, CNS-negative = 451) who were allografted in the first complete remission (CR1) between 2009 and 2019. Primary endpoint was leukemia-free survival (LFS). Median follow-up was not different between the CNS-positive and CNS-negative groups (79 versus 67.2 months, P = 0.58). The CNS-positive group were younger (median age 31.3 versus 39.7 years, P = 0.004) and were allografted more recently (median year 2012 versus 2010, P = 0.003). In both groups, MAC was the preferred approach (82.3% versus 85.6%, P = 0.41). On multivariate analysis, the CNS-positive group had higher incidence of relapse (RI) (hazard ratio [HR] = 1.58 [95% confidence interval (CI) = 1.06-2.35], P = 0.025), but no adverse effect on LFS (HR = 1.38 [95% CI = 0.99-1.92], P = 0.057) or overall survival (OS) (HR = 1.28 [95% CI = 0.89-1.85], P = 0.18). A subgroup multivariate analysis limited to CNS-positive patients showed that a TBI-based MAC regimen resulted in better LFS (HR = 0.43 [95% CI = 0.22-0.83], P = 0.01) and OS (HR = 0.44 [95% CI = 0.21-0.92], P = 0.03) and lower RI (HR = 0.35 [95% CI = 0.15-0.79], P = 0.01). Another subgroup analysis in CNS-negative patients showed that MAC-TBI preparative regimens also showed a lower RI without a benefit in LFS or OS. While a MAC-TBI allo-HCT regimen may not be suitable to all, particularly for older patients with comorbidities, this approach should be considered for patients who are deemed fit and able to tolerate.

Abstract Background Three different scoring systems have been developed to assess pre‐transplant comorbidity in allogeneic hematopoietic stem cell transplantation (Allo‐HSCT): the Hematopoietic Cell Transplantation‐Specific Comorbidity Index, the Comorbidity/Age index, and the Augmented Comorbidity/Age index. All were devised to predict overall survival (OS) and disease‐free survival (DFS) survivals and non‐relapse mortality (NRM) in patients receiving HLA‐matched Allo‐HSCT, but their performance has scarcely been studied in the haploidentical Allo‐HSCT setting with post‐transplant cyclophosphamide, a procedure in constant expansion worldwide. Methods To address this issue, their impact on survivals and NRM was examined in a cohort of 223 patients treated with haploidentical Allo‐HSCT in four different centers. Results With a median follow‐up of 35.6 months, 3‐year OS, DFS, and NRM were 48.1% ± 4%, 46.3% ± 4%, and 30.0% ± 3%, respectively. Abstract The aim of this study was to assess the potential impact of the kinetics of serum levels of seven cytokines during induction in acute myeloid leukemia (AML) patients. Indeed, the role of cytokines, in the pathophysiology and response to therapy of AML patients, remains under investigation. Here, we report on the impact of peripheral levels of two cytokines, the Fms‐like tyrosine kinase 3 ligand (FL) and interleukin‐6 (IL‐6), evaluated during first‐line intensive induction. A new risk stratification can be proposed, which supersedes the ELN 2017 classification to predict survivals in AML patients by examining the kinetic profile of these cytokines during the induction phase. It segregates three groups of, respectively, high‐risk, characterized by a stagnation of low FL levels, intermediate risk, with dynamic increasing FL levels and high IL‐6 at day 22, and favorable risk with increasing FL levels but low IL‐6 at day 22.

Abstract We have retrospectively compared survivals between acute myeloid leukemia (AML)/myelodysplastic syndrome (MDS) patients who received either a clofarabine/busulfan (CloB2A2) or a fludarabine/busulfan (FB2A2) RIC regimen for allogeneic stem cell transplantation. Between 2009 and 2014, 355 allotransplanted cases were identified from the SFGM‐TC registry as having received either the FB2A2 (n = 316, 56% males, median age: 59.2 years, AML 78.5%, first complete remission [CR1] 72%, median follow‐up: 20 months) or the CloB2A2 (n = 39, 62% males, median age: 60.8 years, AML 62%, CR1 69%, median follow‐up: 22.4 months) RIC regimen. In multivariate analysis, FB2A2 was associated with significant lower overall survival (OS, HR: 2.14; 95%CI: 1.05–4.35, P = 0.04) and higher relapse incidence (RI, HR: 2.17; 95%CI: 1.02–4.61, P = 0.04) and a trend for lower leukemia‐free survival (LFS, HR: 1.75; 95%CI: 0.94–3.26, P = 0.08). These results were confirmed using a propensity score‐matching strategy. However, when considering AML and MDS patients separately, the benefit of the CLOB2A2 regimen was restricted to AML patients (2‐year OS FB2A2: 38% [14.5–61.6] vs. CloB2A2: 79.2% [62.9–95.4], P = 0.01; 2‐year LFS FB2A2: 38% [16–59.9] vs. CloB2A2: 70.8% [52.6–89], P = 0.03). The better survivals were due to the lower risk of relapse in this CloB2A2 AML subgroup (2‐year RI FB2A2: 41.2% [19–62.4] vs. CloB2A2: 16.7% [5–34.2], P = 0.05). This retrospective comparison suggests that the CloB2A2 RIC regimen can likely provide longer survival than that awarded by a FB2A2 RIC regimen and may become a new standard of care RIC regimen for allotransplanted AML patients. A prospective phase 3 randomized study is warranted.
