Background and Aim: Brucella melitensis is responsible for brucellosis, a highly contagious, life-threatening disease that has a high impact in low- and middle-income countries. This study aimed to compare silica nanoparticles (SiO-NPs) loaded with ciprofloxacin with silver nanoparticles (AgNPs) loaded with ciprofloxacin to evaluate the possible replacement of silver by silica to enhance biological activity and reduce cytotoxicity. Materials and Methods: SiO-NPs and AgNPs loaded with ciprofloxacin were characterized using ultraviolet spectroscopy, scanning electron microscopy, and dynamic light scattering microscopy for size demonstration and loading efficiency. Both nanoparticles were treated with B. melitensis Rev 1 to evaluate their biological activity. Nanoparticle toxicity was also evaluated using cytotoxicity and hemolysis assays. Results: SiO-NP was found to have a smaller size (80 nm) and higher loading efficiency with polydispersity index and zeta potential of 0.43 and 30.7 mV, respectively, compared to Ag-NP (180 nm and 0.62 and 28.3 mV, respectively). SiO-NP was potent with a minimum inhibitory concentration of 0.043 μg/mL compared to Ag-NP (0.049 μg/mL), with a lower cytotoxicity and hemolysis activity. Conclusion: SiO-NP, as a drug delivery system for ciprofloxacin, has better antimicrobial activity against B. melitensis with lower cytotoxicity and hemolysis activity. These results can be attributed to the enhanced physical characterization and better loading efficiency when compared to Ag-NP.

Background and Aim: Brucellosis is a contagious livestock disease with a significant economic impact. This study aimed to compare the efficacy of antibiotics used alone or in combination with silver nanoparticles (AgNPs) against Brucella melitensis Rev 1 in vitro. Materials and Methods: AgNps conjugated with ciprofloxacin was synthesized and thoroughly characterized by ultraviolet visible spectrophotometry (UV-vis). The antimicrobial effect of ciprofloxacin alone and ciprofloxacin conjugated with AgNPs against B. melitensis Rev 1 was determined by minimum inhibitory concentration (MIC) and the erythrocyte hemolytic assay determined the capability of conjugation to cause hemolysis in human erythrocyte. Results: The UV-vis spectra of both silver-drug nanoconjugates showed a characteristic surface plasmon resonance band at 420 nm. The MIC assays showed that AgNPs conjugation to antibiotics enhanced the antibacterial potential of the selected antibiotics against B. melitensis Rev 1 relative to non-conjugated antibiotics. The results show that low concentrations of AgNPs can kill B. melitensis Rev 1. The MICs of ciprofloxacin and ciprofloxacin–AgNPs were 0.75 and 0.05 μM, respectively. Conclusion: The conjugation of ciprofloxacin with AgNPs enhanced the antibacterial effects against B. melitensis Rev 1. In addition, this conjugation appears to inhibit the capability of this bacterium to adapt to the presence of antibiotics, thereby inhibiting bacterial resistance. Further studies are required to examine its potential as an in vivo treatment.

Background and Aim: Clostridium perfringens type A is an anaerobic bacterium that produces four major toxins (alpha, beta, epsilon, and iota) that cause various diseases. Most of the important C. perfringens-associated diseases of farm animals are caused by alpha-toxin. This study aimed to produce a vaccine against alpha-toxin using C. perfringens type A (ATCC 13124) and investigate its potency, stability, and safety. Materials and Methods: The vaccine was formulated of its constituents for 1 h. Each milliliter of the final vaccine product contained alpha toxoid 15 lecithovitellinase activity (Lv) by adding (0.375 mL containing 40 Lv) and approximately 0.2 mL from 3% concentrated aluminum hydroxide gel, 0.05). Conclusion: This study describes a manufacturing process to obtain sufficient amounts of a vaccine against C. perfringens alpha-toxin. The formulated vaccine effectively elicited a higher level of neutralizing antibody response than the international standards. Furthermore, the vaccine was found to be stable, safe, and effective in preventing C. perfringens-related diseases in rabbits and cattle. Further studies are necessary to evaluate the efficacy of this vaccine in other farm animals.

Background and Aim: Diagnosis of fascioliasis depends on clinical symptoms and routine laboratory tests. Recently, antibodies and circulating antigens of Fasciola were used for detecting active infections. Therefore, this study aimed to identify Fasciola gigantica antigens in the sera of infected cattle using Western blotting and enzyme-linked immunosorbent assay (ELISA) for an accurate diagnosis of cattle infected with F. gigantica. Materials and Methods: Serum samples were obtained from 108, 23, and 19 cattle infected with Fasciola gigantica, Paramphistomum cervi, and Strongylids, respectively, including 57 non-infected cattle that were used as healthy cattle for the study. Western blotting and ELISA were then used to detect circulating Fasciola antigens at 27 kDa. Results: The target epitope was detected in an F. gigantica adult-worm antigen preparation, excretory/secretory products, and serum from cattle infected with F. gigantica. However, it was absent in sera from P. cervi, Strongylids, and healthy cattle. Background and Aim: Paracetamol (PCM) ingestion is one of the most frequent global causes of toxicity. Salvadora persica L. is a plant that among many other effects exhibits potent antioxidant, anti-inflammatory, antimicrobial, and anticancer effects. In this study, we investigated the possible protective effect of S. persica aqueous extract in the PCM overdose-induced liver and kidney injury and hematological changes in a mice model. Materials and Methods: Mice were given PCM with and without S. persica pretreatment. Blood cell counts and liver and kidney function biomarkers were measured. Liver and kidney samples were histologically examined. Results: A single overdose of PCM caused significant elevations of alanine and aspartate transaminases, alkaline phosphate, bilirubin, urea, uric acid, and creatinine compared with the control group. In addition, PCM toxicity significantly lowered red blood cell count but insignificantly increased both white blood cell and platelet counts in comparison to the control mice. Pretreatment with S. persica significantly prevented PCM-induced changes in hepatic and renal biomarkers. S. persica also caused marked reversal of hematological changes. Histologically, the liver and kidney showed inflammation and necrosis after PCM treatment, which were significantly reduced in mice pretreated with S. persica. Conclusion: Taken together, S. persica significantly inhibited PCM-induced renal, hepatic, and hematological toxicity, pointing to its possible use in the treatment of liver and renal disorders.

Background and Aim: The beta toxin is causing the most severe Clostridium perfringens-related diseases. This work was dedicated to developing a vaccine against beta toxin using C. perfringens type C (NCTC 3180). Materials and Methods: The crude toxoid harvest contained 710 limits of flocculation (Lf)/mL. The vaccine was formulated. Each 1 mL of the final vaccine product contained at least 50 Lf/mL of beta toxoids, 0.2 mL 3% aluminum hydroxide gel (equivalent to 5.18 mg of aluminum), 0.05). Conclusion: The research showed a procedure for the manufacturing process of the vaccine against C. perfringens beta toxins with a feasible quantity and the vaccine described here showed to be effective in eliciting levels of neutralizing antibodies higher than required by international standards. In addition, The vaccine was stable up to 30 months. Thus, it may represent an effective and safe for preventing C. perfringens-related diseases in rabbits and cattle, although further studies to prove its efficacy in the field on other farm animals are still needed.

Background and Aim: Exposure to pesticide mixtures used in agricultural practice poses a grave risk to non-target animals. This study aimed to determine whether red grape seed extract (RGSE, which is 95% bioflavonoids and equal to 12,000 mg of fresh red grape seed, and 150 mg of vitamin C) alleviated the changes in brain-derived neurotrophic factor (BDNF) level, acetylcholinesterase activity, oxidative stress, and apoptosis induced by orally administered malathion in a rat model of malathion-induced neurotoxicity. Materials and Methods: Thirty-two adult male Wistar albino rats were divided into four groups and exposed to malathion with or without 4 weeks of RGSE treatment, treated with RGSE alone, or left untreated as controls. The animals were euthanized 24 h after last treatment. Brain samples were collected to measure acetylcholinesterase, superoxide dismutase (SOD), and caspase 3 activity, total antioxidant capacity (TAC), and BDNF levels. Results: Malathion significantly reduced acetylcholinesterase and SOD activity and TAC and significantly increased caspase 3 activity. In comparison, acetylcholinesterase and SOC activity, BDNF level, and TAC were improved and caspase 3 activity was decreased in the malathion-RGSE group, indicating that RGSE corrected the alterations detected in these biochemical parameters. Conclusion: Oxidative stress and apoptosis in the brains of rats exposed to oral malathion were substantially controlled by RGSE treatment.
