Electrical stimulation is an important tool in medicine with applications like muscle rehabilitation, pain management and pacemakers. Portable stimulation devices for home use are divided mainly in two domains: low and middle frequency therapies. Due to the lack, in Germany, of a system to provide both therapies at the same equipment, the main objective of this project is the development of a current source not only capable of that, but also able to generate arbitrary stimulation patterns controlled by an external microcontroller. Moreover, the system should make possible the use of interferential current therapy. The result of the project is an electronic device that triggers current stimulation patterns based on three control signals, which could be easily generated by a microcontroller. The system provides LF stimulation with balanced current pulses up to 80mA through 1k? load impedance, pulse widths between 50 and 1000µs, and frequencies up to 150Hz. It also provides MF stimulation outputs up to 100mApp, with a modulation frequency up to 4kHz and a simple interface to control the envelope waveform. It presents also two galvanic isolated channels to allow interferential current therapy and, when no output current is intended, it is able to provide a short circuit on the load to remove possible net charges introduced by the system to the patient’s body. Additionally, a MATLAB application and a SPICE simulation were developed to assist the creation of control signals. As the system is able to deliver stimulation currents with an arbitrary pattern, it can be used to find the best pulse waveforms for a series of different applications involving electrical therapy, like TENS and MES. Tragbare Stimulatoren zum hauslichen Gebrauch warden hauptsachlich in zwei Gruppen unterteit: Niederfrequenz- und Mittelfrequenztherapiegerate. Bisher gibt es in Deutschland kein Great das beide Therapieformen durchfuhren kann, sodas das Hauptziel dieses Projekts die Entwicklung einer Stromquelle ist, die in der Lage ist, beide Therapieformen anzuwender. Das Great ist zusatzlich in der Lage beiebige, einstellbare Stimulationsmuster zu erzeugen, welche durch einen externen Mikrocontroller gesteuent warden. Auberdem soll disses System Interferenzstromtherapie ermoglichen. Das Ergebnis dieses Projekts ist ein elektronisches Great, das auf drei Steuersignalen basierende Reizstrommuster erzeugt. Diese Steuersignale konnen von einem Mikrocontroller einfach generiert warden. Das System ermoglicht NF-Stimulation mit ausbalancierten Stromimpulsen bis zu 80mA bei 1K? Lastimpedanz, Pulsbreiten zwischen 50 und 1000us und Frequezen bis 150Hz. Es beinhaltet MF-Stimulationsausgange fur bis zu 100mApp Ausgangsstrom, mit Modulationsfrequenzen bis zu 4k Hz und einem einfachen Interface zur Steuerung der Wellenform der Hullkurve. Es bietet zwei galvanische getrennte Kanale zur Interferenzstromtherapie. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) A agricultura brasileira assume papel contundente na intervenção sobre processos ecológicos, pois, com o advento da Revolução Verde, resultado da modernização, passa a adotar estratégias de desenvolvimento que afetaram significativamente os ecossistemas naturais, objetivando o aumento do rendimento da produtividade a busca pela sustentabilidade, que se intensifica cada vez mais nos últimos anos, passa a ser encarada como uma forma de promover o crescimento econômico na mesma medida que se estabelece justiça social e a preservação da natureza, ou seja, buscar a harmonização dessas três esferas. Nesse cenário, a Agricultura Familiar possui características que se diferem à agricultura convencional, fazendo com que seja citada na literatura como ambiente propício para estabelecer um desenvolvimento sustentável. Em contrapartida, atualmente, existem muitos obstáculos na manutenção, permanência e reprodução desta categoria social. Em virtude do contexto exposto, torna-se necessário conhecer cada vez mais as diferentes realidades que compõe a Agricultura Familiar brasileira, a fim de entender e buscar soluções, considerando sua heterogeneidade e viabilizando a execução de propostas de melhorias caso a caso. Sob essa perspectiva, o objetivo desse trabalho foi analisar as dimensões da sustentabilidade da agricultura familiar da Linha dos Alemães, em Dois Vizinhos, PR. A metodologia adotada caracteriza-se como exploratória analítica, sendo um estudo de caso de abordagem qualitativa, a coleta de dados ocorreu por meio do instrumento de entrevista semiestruturada, abordando 5 famílias da comunidade e 3 agentes (1 da Secretária de Agricultura Municipal, 1 do Sindicato dos Trabalhadores Rurais e 1 da Assistência Social). Dentre os resultados encontrados, destacamos: na dimensão social, problemas estruturais na educação, baixa qualidade ou falta de assistência técnica, extensão rural e servidos públicos básicos que colocam em risco, a médio a longo prazo, a extinção da comunidade; na dimensão econômica, encontramos uma variedade de estratégias para obtenção de renda, sendo que a predominância está na renda da produção leiteira e a pluriatividade se destaca como tática de arrecadação, a baixa adesão de políticas públicas e de programas de crédito também foi evidenciada, bem como a dificuldade de reprodução econômica entre alguns agricultores, revelando a necessidade de traçar novas medidas para melhoria das propriedades; já na dimensão ambiental, a falta ou baixa qualidade dos serviços de saneamento ambiental foi posta em destaque, os agricultores sofrem com a falta de água em épocas secas, revelando falhas no sistema de abastecimento de água e partilham da deficiência no tratamento da água e destinação do esgoto e resíduos sólidos. Brazilian agriculture assumes a decisive role in intervening in ecological processes, as, with the advent of the Green Revolution, as a result of modernization, it starts to adopt development strategies that significantly affected natural ecosystems, aiming at increasing productivity yields. The search for sustainability, which has intensified more and more in recent years, is now seen as a way of promoting economic growth in the same way that social justice and the preservation of nature are established, in other words, seeking to harmonize these three spheres. In this scenario, Family Farming has characteristics that differ from conventional farming, making it cited in the literature as a favorable environment for establishing sustainable development. On the other hand, currently, there are many obstacles in the maintenance, permanence and reproduction of this social category. Due to the above context, it becomes necessary to know more and more the different realities that make up the Brazilian Family Farming, in order to understand and seek solutions, considering its heterogeneity and enabling the implementation of improvement proposals on a case-by-case basis. From this perspective, the objective of this work was to analyze the dimensions of the sustainability of family farming in Linha dos Alemêes, in Dois Vizinhos, PR. The adopted methodology is characterized as exploratory analytical, being a case study with a qualitative approach, data collection occurred through the semi-structured interview instrument, approaching 5 families from the community and 3 agents (1 from the Municipal Agriculture Department, 1 from the Union of Rural Workers and 1 of Social Assistance). Among the results found, we highlight: in the social dimension, structural problems in education, low quality or lack of technical assistance, rural extension and basic public services that put at risk, in the medium to long term, the extinction of the community; in the economic dimension, we find a variety of strategies to obtain income, with the predominance being in the income from dairy production and pluriactivity stands out as a collection tactic, the low adherence to public policies and credit programs was also evidenced, as well. as the difficulty of economic reproduction among some farmers, revealing the need to devise new measures to improve the properties; in the environmental dimension, the lack or low quality of environmental sanitation services was highlighted, farmers suffer from a lack of water in dry seasons, revealing flaws in the water supply system and sharing the deficiency in water treatment and destination sewage and solid waste.

A cultura da soja é de extrema importância econômica para o estado do Paraná. No entanto, uma tendência dos produtores rurais do estado é a antecipação da semeadura da soja para os primeiros dias do zoneamento agrícola. Esta mudança comportamental acende o alerta sobre a necessidade da intensificação da pesquisa neste ponto, principalmente pesquisas em relação ao melhoramento genético, que objetivam obter cultivares adaptadas a semeadura antecipada. Desta forma, o trabalho objetivou caracterizar morfologicamente a dissimilaridade genética de nove cultivares elites em semeadura antecipada, promovendo o agrupamento destas, além de estimar parâmetros genéticos dos genótipos e das gerações F1 e F2 através da realização de cruzamentos dialélicos parcial, indicando assim as combinações e as cultivares mais promissoras para a obtenção de recombinações favoráveis para a semeadura antecipada objetivando o incremento da produtividade de grãos. As cultivares apresentaram significativa dissimilaridade genética, indicando a existência de variabilidade para os caracteres avaliados em semeadura antecipada, sendo possível separá-las em dois grupos distintos. O trabalho possibilitou a identificação de cultivares que podem ser utilizados em cruzamentos artificiais de elevada heterose. Os caracteres que mais contribuíram para a dissimilaridade genética foram dias da emergência ao florescimento e altura final de planta. As cultivares 53I54RSF IPRO, 5958RSF IPRO e 50I52RSF IPRO se destacaram das demais, combinando elevada média produtiva, capacidade especifica de combinação (CEC) e elevada capacidade geral de combinação (CGC) para rendimento de grãos (RG) em semeadura antecipada. A combinação destas cultivares indica ser promissor para compor blocos de cruzamentos por apresentar alto potencial produtivo em semeadura antecipada por meio da complementariedade alélica. The soybean crop is of extreme economic importance for the state of Paraná. However, a trend of the state's rural producers is the anticipation of sowing soybeans to the early days of agricultural zoning. This behavioral change raises the alert about the need for intensified research at this point, especially research related to genetic improvement, which aims to obtain cultivars adapted to early sowing. Thus, the objective of this work was to characterize morphologically the genetic dissimilarity of nine cultivars in early sowing, promoting their grouping, besides estimating genetic parameters of genotypes and F1 and F2 generations through partial diallel crosses, indicating the combinations and the most promising cultivars to obtain favorable recombination for early sowing aiming at increasing grain yield. The cultivars presented significant genetic dissimilarity, indicating the existence of variability for the traits evaluated in early sowing, being possible to separate them in two distinct groups. The work allowed the identification of cultivars that can be used in artificial crossings of high heterosis. The traits that most contributed to genetic dissimilarity were days from emergence to flowering and final plant height. The cultivars 53I54RSF IPRO, 5958RSF IPRO and 50I52RSF IPRO stood out from the others, combining high yield average, specific combining ability (CEC) and high overall combining capacity (CGC) for grain yield (RG) in early sowing. The combination of these cultivars indicates to be promising to compose blocks of intersections for presenting high productive potential in early sowing through allelic complementarity.

O surgimento da agricultura desencadeou a ocorrência de uma revolução social e cultural sobre a humanidade. Esta, que foi evoluindo ao longo dos séculos, passou por sucessivas transformações que afetaram as sociedades humanas em tempos e lugares distintos. No entanto os impactos socioambientais desencadeados pelas revoluções ocorridas, como a Revolução Verde, se tornam cada vez mais evidentes, tais como a brusca redução da diversidade de espécies e variedades de plantas cultivadas. A conservação desta agrobiodiversidade, no entanto, não implica somente em uma questão ambiental, mais sim na segurança alimentar e nutricional da população, desenvolvimento rural sustentável, inclusão social e o combate à fome e miséria estão direta ou indiretamente relacionados à conservação e ao uso dos recursos da agrobiodiversidade. Neste âmbito, conservar tamanha riqueza é imprescindível e uma das formas que asseguram tal biodiversidade são os “guardiões de sementes crioulas” que mantem as sementes/mudas ao longo do tempo em suas propriedades. Dentre as espécies de plantas de importância para a alimentação, temos o feijão-comum que está presente na dieta da maioria dos brasileiros e que devido sua importância para a segurança alimentar e nutrição humana está entre as principais culturas do país. Sob essa situação, o trabalho teve como objetivo identificar o contexto que os guardiões de sementes de feijão crioulo estão inseridos, bem como quais os principais desafios enfrentados por estes como mantenedores dessas cultivares. Após entrevista com os guardiões de sementes foi possível inferir que estes estão presentes em contextos bem diversificados entre si. Todavia a realidade encarada no processo de manutenção de sementes é bem semelhante, assim como as principais dificuldades e anseios destes. Como conclusão destacamos a necessidade de maior conscientização da população acerca do tema e apoio por parte do governo e das universidades a estes guardiões, assim como maiores investimentos em pesquisa, assistência técnica, curso entre outros. Além disso, a possibilidade de geração de renda através de manutenção destas sementes seria uma boa opção na tentativa de fortalecer a atividade, no entanto este é um ponto pouco conhecido e explorado pelos agricultores em estudo. The emergence of agriculture triggered the occurrence of a social and cultural revolution on humanity. This, which has evolved over the centuries, has undergone successive transformations that have affected human societies in different times and places. However, the socio-environmental impacts triggered by revolutions, such as the Green Revolution, have become increasingly evident, such as the sharp reduction in the diversity of species and varieties of cultivated plants. The conservation of this agrobiodiversity, however, does not only imply an environmental issue but a food and nutritional security of the population, sustainable rural development, social inclusion and the fight against hunger and misery are directly or indirectly related to the conservation and use of agrobiodiversity resources. In this context, conserving such wealth is imperative and one of the ways to ensure such biodiversity is the "guardians of creole seeds" that keeps seeds / seedlings over time on their properties. Among the species of plants of importance for food, we have the common bean that is present in the diet of most brazilians and that due to its importance for food security and human nutrition is among the main cultures of the country. Under this situation, the objective of the work was to identify the context that the guardians of bean seeds of Creole are inserted, as well as the main challenges faced by these as maintainers of these cultivars. After an interview with the seed guardians it was possible to infer that these are present in well diversified contexts. However the reality in the process of maintaining seeds is very similar, as well as the main difficulties and desires of these. As a conclusion we emphasize the need for greater awareness of the population about the theme and support by government and universities to these guardians, as well as greater investments in research, technical assistance, courses among others. In addition, the possibility of income generation through maintenance of these seeds would be a good option in trying to strengthen the activity, however this is a little known and exploited by the farmers under study.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Para a cultura do feijão existem poucos herbicidas registrados que exercem seletividade à cultura e sejam eficientes no controle de espécies daninhas de folhas largas. Um dos fatores que contribui para a escassez de herbicidas registrados é a elevada sensibilidade do feijão aos mesmos. Isso ressalta a grande importância de pesquisas investigando a tolerância do feijão a herbicidas que ainda não possuem registro à cultura, como novas alternativas para o controle das plantas daninhas. Estudos recentes comprovaram a existência de variabilidade de tolerância de cultivares feijão ao herbicida saflufenacil, inibidor da enzima protoporfirinogênio oxidase (Protox), quando aplicado na modalidade de pré-emergência da cultura. O presente trabalho teve por objetivo investigar a tolerância do feijão aos herbicidas inibidores da Protox. Os experimentos foram executados para determinar: (1) a tolerância diferencial do herbicida safluenacil nas modalidades de pré e pós-emergência; (2) a existência de níveis diferenciados de tolerância entre cultivares de feijão; (3) a tolerância a herbicidas de diferentes grupos químicos inibidores da Protox; (4) a seletividade de diferentes doses de saflufenacil e sulfentrazone em em cultivares de feijão e o efeito no controle de plantas daninhas; e (5) mecanismos de tolerância baseados na metabolização dos herbicidas. Os resultados permitem inferir que os níveis de tolerância do feijoeiro ao saflufenacil são muito superiores na modalidade de pré-emergência, em relação à pós-emergência, em que doses muito reduzidas de saflufenacil causam a morte das plantas. Dentre as cultivares testadas, a BRSMG Talismã e a IAC Milênio apresentaram, respectivamente, a maior e menor tolerância ao saflufenacil (20,5 g ha-1). A tolerância aos herbicidas saflufenacil, sulfentrazone, flumioxazin e fomesafen aplicados em pré-emergência depende da cultivar e da dose utilizada, não havendo um padrão de tolerância cruzada aos inibidores da Protox nas cultivares. Em condições de campo, saflufenacil apresentou controle satisfatório das plantas daninhas apenas na dose de 52,5 g ha-1, a qual causou elevada fitotoxicidade para o feijão. Sulfentrazone (400 g ha-1) apresentou nível satisfatório de controle das plantas daninhas e seletividade para as cultivares BRSMG Talismã e IPR Tuiuiú, demonstrando bom potencial para ser empregado em pré-emergência em solo do tipo latossolo. Em ensaios em casa-de-vegetação, o aumento da tolerância da cultivar sensível com uso do protetor mefenpyr-diethyl e a redução da tolerância da cultivar tolerante com o inibidor clorpirifós, sugerem que a metabolização está envolvida no mecanismo de tolerância aos inibidores da Protox. Conclui-se que o nível de tolerância manifestada em pré-emergência é dependente da cultivar e do herbicida empregado, sugerindo o envolvimento da metabolização do herbicida como um dos mecanismos de tolerância do feijão aos inibidores da enzima Protox. For bean crop there are few registered herbicides that exert crop selectivity and are efficient in controlling broadleaf weeds. This underscores the great importance of research investigating the tolerance of the bean to herbicides that do not yet have a record to the crop, as new alternatives for the control of weeds. Recent studies have confirmed the existence of variability of tolerance of bean cultivars to saflufenacil herbicide, inhibitor of the protoporphyrinogen oxidase (Protox) enzyme, when applied in the pre-emergence mode of the culture. The present work aimed to investigate the tolerance of beans to herbicides inhibitors of Protox enzyme. The experiments were performed to determine: (1) the differential tolerance of safluenacil herbicide in pre and post emergence modalities; (2) the existence of different tolerance levels among bean cultivars; (3) herbicide tolerance of different Protox inhibitor chemical groups; (4) the selectivity of different doses of saflufenacil and sulfentrazone in bean cultivars and the effect on weed control; and (5) tolerance mechanisms based on herbicide metabolism. The results allow us to infer that the bean tolerance levels to saflufenacil are much higher in the preemergence modality than the postemergence, in which very low doses of saflufenacil cause plant death. Among the cultivars tested, BRSMG Talismã and IAC Milênio presented, respectively, the highest and lowest tolerance to saflufenacil (20.5 g ha-1). The tolerance to the saflufenacil, sulfentrazone, flumioxazin and fomesafen herbicides applied in pre-emergence depends on the cultivar and dose used, and there is no cross tolerance pattern to Protox inhibitors in the cultivars. In field conditions, saflufenacil presented satisfactory control of weeds only at the dose of 52.5 g ha-1, which caused high phytotoxicity to the bean. Sulfentrazone (400 g ha-1) showed satisfactory level of weed control and selectivity for the cultivars BRSMG Talismã and IPR Tuiuiú, showing good potential to be used in pre-emergence on latosol type soil. In greenhouse trials, increased tolerance of the sensitive cultivar with mefenpyr-diethyl protector and the reduction of tolerance of the tolerant cultivar to the chlorpyrifos inhibitor, suggest that the metabolism is involved in the mechanism of tolerance to Protox inhibitors. It is concluded that the tolerance level manifested in pre-emergence is dependent on the cultivar and herbicide employed, suggesting the involvement of herbicide metabolism as one of the mechanisms of bean tolerance to Protox enzyme inhibitors.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Sabendo da existência de guardiões de sementes crioulas na região Sudoeste do Paraná, mas não tendo acesso a um mapeamento dos mesmos, a presente pesquisa através de uma metodologia baseada na indicação a partir de informantes-chave (bola de neve) buscou por guardiões de sementes crioulas com a finalidade de compreender o universo inerente a esses agricultores. No total foram mapeados oito guardiões e destes, quatro foram entrevistados, destes, três pertencem a região Sudoeste do Paraná e um a região Oeste de Santa Catarina. Os guardiões de sementes crioulas são agricultores que recebem esse “título” informal por dedicarem-se a conservar sementes crioulas em suas propriedades (conservação on farm). Analisando a relação homem-natureza de quatro guardiões de sementes foi possível perceber que a agrobiodiversidade presente nessas propriedades se manifesta de forma natural, visto que a presença da diversidade é obrigatória nesse cenário. Estes agricultores possuem características que os configuram como agricultores familiares, entre elas se destacam: mão de obra familiar, policultivo, autoconsumo e participação social. Quanto ao universo inerente a esses agricultores, estes reproduzem os conhecimentos da agricultura tradicional repassados pela família, através de gerações. Os quatro guardiões entrevistados manejam suas propriedades de forma holística e privilegiam formas sustentáveis de produção e possuem pouca dependência a insumos externos. Ainda sobre as sementes crioulas: a colheita é manual, a secagem ao sol, o armazenamento é em sua maioria em litros de plástico e as trocas são em festas de sementes. Somente aqui somam-se mais de 500 variedades de sementes crioulas que segundo os mesmos precisam de suporte do Estado, da sociedade e das instituições de ensino. A manutenção e reprodução dos saberes tradicionais herdados por esses agricultores se configura na principal forma de resistência deles frente ao agronegócio e também resulta em algo buscado por todos eles, a autonomia. A agrobiodiversidade intrínseca a forma como esses agricultores se relacionam com a natureza sugere que ações relacionadas a introdução de agrobiodiversidade no meio rural não deve se limitar a esfera ambiental mas também considerar aspectos culturais e simbólicos. Knowing the existence of creole seeds guardians in the Southwest region of Paraná, but not having access to a mapping of guardians, the present research through a methodology based on the indication from key informants (snowball) searched by guardians of creole seeds with the purpose of understanding the universe inherent in these farmers. In total eight mapped guardians and four were interviewed, of which three belong to the Southwest region of Paraná and one to the West region of Santa Catarina. Creole seed guardians are farmers who receive this informal title by dedicating themselves to preserving creole seeds on their properties (conservation on farm), analyzing the man-nature relationship of four seed guardians, it was possible to perceive that the agrobiodiversity present in these properties is manifested in a natural way, since the presence of diversity is mandatory in this scenario. These farmers have characteristics that make them family farmers, among them the following: family labor, polyculture, self-consumption and social participation. As for the universe inherent to these farmers, these reproduce the knowledge of traditional agriculture passed on by the family, through generations. The four guardians interviewed manage their properties holistically and favor sustainable forms of production and have little dependence on external inputs. Still about the creole seeds: the harvest is manual, drying in the sun, the storage is mostly in liters of plastic and the exchanges are in seed parties. Only here are added more than 500 varieties of creole seeds that according to them need support from the state, society and educational institutions. The maintenance and reproduction of traditional knowledge inherited by these farmers is the main form of resistance to agribusiness and also results in something sought after by all of them, autonomy. Agrobiodiversity intrinsic to how these farmers relate to nature suggests that actions related to the introduction of agrobiodiversity in rural areas should not be limited to the environmental sphere but also consider cultural and symbolic aspects.

A germinação pré-colheita em trigo, têm sido responsável por perdas consideráveis de produtividade e qualidade em diversas regiões produtoras a nível mundial. Diversos programas de melhoramento têm buscado selecionar genótipos com elevados níveis de resistência, mas em função da complexidade da característica, os avanços obtidos são pouco expressivos, e técnicas mais eficientes para identificação de genótipos superiores são necessárias. Dessa forma, o objetivo do presente estudo foi realizar uma análise de associação genômica, a fim de identificar marcadores SNPs e regiões genômicas associadas com a resistência a germinação pré-colheita em trigo, para posterior utilização em seleção assistida por marcadores moleculares (SAM). Para isso, foram utilizados 344 genótipos de trigo de diferentes empresas obtentoras e épocas de lançamento, que representam a base genética do trigo cultivado no Brasil. O experimento foi constituído por três etapas principais: a genotipagem, a fenotipagem e a análise de associação genômica. Para a genotipagem, foi realizada a extração e quantificação do DNA, e as amostras foram genotipadas com um chip de DNA para detecção de SNPs contendo 35 mil SNPs para o genoma do trigo. Na etapa de fenotipagem, foi conduzido um experimento em Cascavel-PR, em delineamento experimental de blocos cazualizados com três repetições. Os genótipos foram avaliados na maturidade fisiológica, a partir da coleta de 20 espigas por genótipo, as quais foram submetidas a um simulador de chuva para germinação, sendo então atribuídas notas de acordo com o índice de germinação apresentado. Após as determinações genotípicas e fenotípicas, foi realizada a análise de associação genômica a fim de identificar marcadores moleculares associados com a resistência à germinação pré-colheita. Foram identificados dez marcadores significativos nos cromossomos 3B, 4A, 5B, 5D, 6D, 7B e 7D, onde estão presentes QTLs (Quantitative Trait Loci) associados com a resistência a germinação pré-colheita. Os SNPs foram localizados em genes codificadores de enzimas envolvidas em vias de sinalização de giberelinas e ácido abscísico, hidrólise de maltose e alongamento e divisão celular. A explicação da variação fenotípica pelos marcadores significativos variou de 4,2 a 5,4%, o que é esperado para caracteres quantitativos. Os marcadores significativos identificados no presente estudo, após validados, poderão ser utilizados em programas de SAM, para a seleção precoce de genótipos mais resistentes a germinação pré-colheita, propiciando a obtenção de genótipos desejáveis de maneira mais ágil e precisa. Pre-harvest sprouting (PHS) in wheat, has been responsible for considerable losses in productivity and quality in several growing regions worldwide. Several breeding programs have sought to select genotypes with high levels of resistance, but due to the complexity of the trait, the advances achieved are inexpressive, and most efficient techniques to identify superior genotypes are demanded. Thus, the objective of this study was to perform a genome-wide association analysis, for identify SNPs markers and genomic regions associated with resistance to PHS in wheat, for later use in marker-assisted selection (MAS). For this, 344 wheat genotypes of different times and companies that represents the genetic basis of wheat grown in Brazil were used. The experiment consisted of three main steps: genotyping, phenotyping and genome-wide association analysis. In the genotyping step, the DNA extraction and quantification was conducted, and the samples were genotyped with a DNA chip for detection of SNPs containing 35,000 SNPs for the wheat genome. In the phenotyping step, an experiment was conducted in Cascavel-PR, in a randomized block design with three replicates. The genotypes were evaluated at physiological maturity, with the collection of 20 spikes per genotype, and these were subjected to a simulated rainfall for germination, being then allocated scores according to the germination rate observed. After the genotypic and phenotypic determinations, it was conducted a genome-wide association analysis to identify molecular markers associated with resistance to PHS. Ten significant markers were identified on chromosomes 3B, 4A, 5B, 5D, 6D, 7B and 7D, where QTLs (Quantitative Trait Loci) associated with pre-harvest sprouting resistance are present. The SNPs were localized in genes encoding enzymes involved in the gibberellin and abscisic acid signaling pathways, maltose hydrolysis and cell division and stretching. The explanation of the phenotypic variation by the significant markers ranged from 4.2 to 5.4%, which is expected for quantitative traits. Significant markers identified in the present study, after being validated, could be used in MAS programs for the early selection of genotypes more resistant to PHS in wheat, allowing the obtainment of desirable genotypes in the more agile and precise way.

O presente trabalho teve por objetivo estudar a herança genética, determinar o melhor protocolo de extração de DNA para esta espécie, e identificar marcadores moleculares associados à resistência de leiteiro (Euphorbia heterophylla L.) aos herbicidas inibidores da ALS e da PROTOX. A herança genética da resistência foi determinada a partir de cruzamentos entre os biótipos de E. heterophylla suscetível (S) e resistente (R), retrocruzamentos e avanço de geração para F2. A dominância completa da resistência foi comprovada com curvas de dose resposta. Foram testados dez protocolos de extrações de DNA adaptados de métodos descritos na literatura. Os iniciadores específicos para os genes ALS e PROTOX foram desenhados a partir da sequência de DNA consenso destes genes, obtida pelo alinhamento das espécies Manihot esculenta e Ricinus communis. Adicionalmente, foi testada a transferibilidade de vinte marcadores SSRs (sequências simples repetidas) desenhados para o genoma de Manihot esculenta, pois dentre espécies de Euphorbiaceae com maior número de marcadores SSRs desenvolvidos, é a espécie filogeneticamente mais próxima de E. heterophylla. Em relação à herança genética, as frequências observadas nas gerações F1, F2, RCs e RCr não diferiram estatisticamente das frequências esperadas para característica controlada por dois genes dominantes para resistência múltipla e um gene dominante para resistência simples aos inibidores da ALS e PROTOX. Os níveis similares de resistência observados para o heterozigoto F1 e o biótipo homozigoto R, para doses de até 2000 g i.a. ha-1 de fomesafen e doses de até 800 g i.a. ha-1 de imazethapyr, confirmam a dominância completa da resistência aos inibidores da PROTOX e ALS, respectivamente. O protocolo 0,2%BME possibilitou a extração de 7,083 ng μL-1 de DNA, sendo estatisticamente (P=0,05) superior aos demais protocolos. Os compostos fenólicos contaminaram o DNA extraído pelos protocolos FENOL e 3%BME+TB, mas a adição de polivinilpirrolidona (PVP40) no tampão de extração do protocolo 3%BME+TA solucionou este problema. Os iniciadores desenhados para os genes ALS e PROTOX não amplificaram ou não apresentaram polimorfismo visível em gel de agarose entre os biótipos S e R de E. heterophylla. Dez marcadores SSR foram transferidos para E. heterophylla e destes, seis iniciadores apresentaram polimorfismo entre os biótipos S e R. This study aimed to assess the genetic inheritance, determine the better DNA isolation protocol for this species and to identify molecular markers associated with the Wild Poinsettia (Euphorbia heterophylla L.) resistance ALS- and PROTOX- inhibiting herbicides and. The genetic inheritance of resistance was determined from crosses between E. heterophylla biotypes susceptible (S) and resistant (R), backcrosses and F2 generation. The complete dominance of resistance was confirmed with dose response curves. Ten adjusted methods for DNA isolation described in the literature were tested. The specific primers for ALS and PROTOX genes were designed from the consensus DNA sequence of these genes, obtained by aligning the gene sequences of the species Manihot esculenta and Ricinus communis L. Additionally, it was assessed the transferability of twenty SSR (simple sequence repeat) markers designed for Manihot esculenta, because among the species of Euphorbiaceae with more developed SSRs markers, because it is the closest relative phylogenetic species of E. heterophylla. Regarding genetic inheritance, the frequencies observed in the F1, F2, RCs and RCr did not differ significantly from the expected frequencies for a trait controlled by two dominant genes for multiple resistance and a single dominant gene for simple resistance to ALS- and PROTOX-inhibiting herbicides. The similar levels of resistance to dosage up to 2000 g i.a. ha-1 of fomesafen and dosage up to 800 g i.a. ha-1 of imazethapyr observed in F1 (heterozygous) and homozygous R biotype confirm the complete dominance of resistance to PROTOX- and ALS-inhibiting herbicides, respectively. The 0.2%BME protocol allowed the isolation of 7,083 ng μL-1 DNA, significantly (P=0.05) higher than other methods. Co-isolation of phenolic compounds was observed in FENOL and 3%BME+TB methods, but the addition of polyvinylpyrrolidone (PVP40) in the protocol extraction buffer 3%BME+TA solved this problem. The primers designed for ALS and PROTOX genes amplified but not showed no visible polymorphism in agarose gel between the S and R biotypes of E. heterophylla. Regarding the SSR transferability, ten markers were transferred to E. heterophylla, however, these six primers showed polymorphism among S and R biotypes.
