Objectives: The goal of this study was to develop and evaluate an intervention aimed at increasing cognitive empathy, improving mental health, and reducing inflammation in dementia caregivers, and to examine the relevant neural and psychological mechanisms. Methods: Twenty dementia caregivers completed an intervention that involved taking 3–5 daily photographs of their person living with dementia (PLWD) over a period of 10 days and captioning those photos with descriptive text capturing the inner voice of the PLWD. Both before and after the intervention, participants completed questionnaires, provided a blood sample for measures of inflammation, and completed a neuroimaging session to measure their neural response to viewing photographs of their PLWD and others. Results: 87% of enrolled caregivers completed the intervention. Caregivers experienced pre- to post-intervention increases in cognitive empathy (i.e. Perspective-Taking) and decreases in both burden and anxiety. These changes were paralleled by an increased neural response to photographs of their PLWD within brain regions implicated in cognitive empathy. Conclusion: These findings warrant a larger replication study that includes a control condition and follows participants to establish the duration of the intervention effects. Background: Adolescent violence victimisation is associated with a spectrum of adult social and behavioural health outcomes, including adverse mental health symptoms. However, underlying social stress mechanisms linking adolescent victimisation to adult cardiometabolic health remains poorly understood. Aim: The current study aims to reveal how adolescent and adult interpersonal violence exposures each get "under the skin" to affect adult metabolic syndrome, including direct victimisation and, additionally, witnessing violence. Subjects and methods: We use a nationally representative longitudinal cohort, the National Longitudinal Study of Adolescent to Adult Health, and leverage a quasi-experimental approach, propensity score matching regression analysis (n = 14,267). Results: We find that adolescent violence exposure carries an enduring effect on young adult metabolic syndrome risk factor incidence and high-risk status, which is independent of young adult violence. Violence effects do not vary by sex or racial identity. Conclusion: In sum, adolescent exposure to direct interpersonal violence significantly affects young adult cardiometabolic health in ways suggesting adolescence is a sensitive period for the onset of harmful cardiometabolic effects in early adulthood. Epigenetic clocks quantify regular changes in DNA methylation that occur with age, or in relation to biomarkers of ageing, and are strong predictors of morbidity and mortality. Here, we assess whether measures of fetal nutrition and growth that predict adult chronic disease also predict accelerated biological ageing in young adulthood using a suite of commonly used epigenetic clocks. Data come from the Cebu Longitudinal Health and Nutrition Survey (CLHNS), a long-running cohort followed since birth in metropolitan Cebu, Philippines. Past work has shown that birth weight (BW) and the mother's arm fat during pregnancy (a measure of pregnancy energy status) relate inversely to health outcomes in the CLHNS but primarily in males. Genome-wide DNA methylation was assessed in whole blood using the Infinium EPIC array. Participants included males (n=895) and females (n=803) measured in 2005 (20.8–22.5 years). Clocks included the Hannum and Horvath clocks trained on chronological age, the DNAmPhenoAge and DNAmGrimAge clocks trained on clinical biomarkers, the Dunedin pace of ageing (DunedinPACE) clock trained on longitudinal changes in ageing biomarkers, and the DNAmTL clock trained on leukocyte telomere length. In males, lower BW predicted advanced biological ageing using the Hannum, DNAmPhenoAge, DunedinPoAm, and DNAmTL clocks. In contrast, BW did not predict any clock in female participants. Participants' mothers' pregnancy arm fat only predicted DNAmTL in males. These findings suggest that epigenetic clocks are a useful tool for gauging long-term outcomes predicted by fetal growth, and add to existing evidence in the CLHNS for sex differences in these relationships. By tracking a group of individuals through time, cohort studies provide fundamental insights into the developmental time course and causes of health and disease. Evolutionary life history theory seeks to explain patterns of growth, development, reproduction and senescence, and inspires a range of hypotheses that are testable using the longitudinal data from cohort studies. Here we review two decades of life history theory-motivated work conducted in collaboration with the Cebu Longitudinal Health and Nutrition Survey (CLHNS), a birth cohort study that enrolled more than 3000 pregnant women in the Philippines in 1983 and has since followed these women, their offspring and grandoffspring. This work has provided evidence that reproduction carries "costs" to cellular maintenance functions, potentially speeding senescence, and revealed an unusual form of genetic plasticity in which the length of telomeres inherited across generations is influenced by reproductive timing in paternal ancestors. Men in Cebu experience hormonal and behavioural changes in conjunction with changes in relationship and fatherhood status that are consistent with predictions based upon other species that practice bi-parental care. The theoretical expectation that early life cues of mortality or environmental unpredictability will motivate a "fast" life history strategy are confirmed for behavioural components of reproductive decision making, but not for maturational tempo, while our work points to a broader capacity for early life developmental calibration of systems like immunity, reproductive biology and metabolism. Inflammation is associated with increased risk for chronic degenerative diseases, as well as age-related functional declines across many systems and tissues. Current understandings of inflammation, aging, and human health are based on studies conducted almost exclusively in high-income nations that rely primarily on baseline measures of chronic inflammation. This analysis investigates the inflammatory response to vaccination as a predictor of cardiovascular disease (CVD) among older women in the Philippines, a lower-middle income nation with rising rates of overweight/obesity and relatively high burdens of infectious disease. Concentrations of C-reactive protein (CRP) were measured at baseline and 72 h following influenza vaccination in 530 women (mean age = 55.2 years). Ankle-brachial index (ABI) – an indicator of peripheral arterial disease and broader CVD risk – was measured approximately three years later. The magnitude of CRP response to vaccination was positively associated with ABI, indicating that a larger inflammatory response predicts lower CVD risk. Baseline CRP was negatively associated with CRP response to vaccination, and was not associated with ABI independently of CRP response. These results suggest that research across ecological settings, and with more dynamic measures of inflammatory response and regulation, may yield important insights into the associations among inflammation, aging, and disease. Aim: To determine if field-typical storage and collection conditions are related to salivary testosterone (T), cortisol (cort) and secretory immunoglobulin A (sIgA) as measured using commercially available kits. Subjects and methods: This study assessed whether storage time (∼6 months to 1.5 years) at −35 °C impacted levels of the measured biomarkers (n = 10,247 samples). For a sub-set of T samples (n = 2954), we also evaluated the impact of collection conditions, such as time spent at room temperature in participants' homes (0-39 hours) and time spent in coolers (0.3-10 hours) in transit. Results: T was unrelated to storage and collection variables and there was no evidence of reduced sample fidelity at longer storage times. Cort samples stored at −35 °C for longer had significantly lower values, but the effect was small (β = −0.003 nmol/L/day, SE <0.001, p = 0.005). sIgA trended higher with longer storage at −35 °C (β = 0.84 µg/mL/day, SE = 0.45, p = 0.063). Collection and storage time variables did not improve the fit of any of the models except the one that evaluated cortisol. Conclusions: Salivary T was unaffected by extended storage at −35 °C and only a weak relationship was found between storage time and salivary cort or sIgA. Genome-wide transcriptional profiling has emerged as a powerful tool for analyzing biological mechanisms underlying social gradients in health, but utilization in population-based studies has been hampered by logistical constraints and costs associated with venipuncture blood sampling. Dried blood spots (DBS) provide a minimally invasive, low-cost alternative to venipuncture, and in this article we evaluate how closely the substantive results from DBS transcriptional profiling correspond to those derived from parallel analyses of gold-standard venous blood samples (PAXgene whole blood and peripheral blood mononuclear cells [PBMC]). Analyses focused on differences in gene expression between African-Americans and Caucasians in a community sample of 82 healthy adults (age 18–70 years; mean 35). Across 19,679 named gene transcripts, DBS-derived values correlatedr = .85 with both PAXgene and PBMC values. Results from bioinformatics analyses of gene expression derived from DBS samples were concordant with PAXgene and PBMC samples in identifying increased Type I interferon signaling and up-regulated activity of monocytes and natural killer (NK) cells in African-Americans compared to Caucasian participants. These findings demonstrate the feasibility of DBS in field-based studies of gene expression and encourage future studies of human transcriptome dynamics in larger, more representative samples than are possible with clinic- or lab-based research designs. Our objective was to validate a commercially available ELISA to measure antibody titers against Epstein-Barr virus (EBV) in dried blood spots (DBS) to replace a previously validated assay for DBS that is no longer available. We evaluated the precision, reliability, and stability of the assay for the measurement of EBV antibodies in matched plasma, fingerprick DBS, and venous blood DBS samples from 208 individuals. Effects of hematocrit and DBS sample matrix on EBV antibody determination were also investigated, and the cutoff for seropositivity in DBS was determined. A conversion equation was derived to enable comparison of results generated using this method with the former DBS method. There was a high correlation between plasma and DBS EBV antibody titers (R2 = 0.93) with very little bias (−0.07 based on Bland-Altman analysis). The assay showed good linearity and did not appear to be affected by the DBS matrix, and physiological hematocrit levels had no effect on assay performance. There was reasonable agreement between DBS EBV titer estimates obtained using this assay and the previously validated assay (R2 = 0.72). The commercially available ELISA assay for EBV antibody titers that we validated for use with DBS will facilitate continued investigation of EBV antibody titers in DBS. [ABSTRACT FROM AUTHOR]

Dried blood spot (DBS) methodology offers significant advantages over venipuncture in studies of vulnerable populations or large-scale studies, including reduced participant burden and higher response rates. Uncertainty about the validity of cardiovascular disease (CVD) risk biomarkers remains a barrier to wide-scale use. We determined the validity of DBS-derived biomarkers of CVD risk versus gold-standard assessments, and study-specific, serum-equivalency values for clinical relevance of DBS-derived values. Concurrent venipuncture serum and DBS samples (n= 150 adults) were assayed in Clinical Laboratory Improvement Amendments–certified and DBS laboratories, respectively. Time controls of DBS standard samples were assayed single-blind along with test samples. Linear regression analyses evaluated DBS-to-serum equivalency values; agreement and bias were assessed via Bland-Altman plots. Linear regressions of venipuncture values on DBS-to-serum equivalencies provided R2values for total cholesterol, high-density lipoprotein cholesterol (HDL-C), and C-reactive protein (CRP) of 0.484, 0.118, and 0.666, respectively. Bland-Altman plots revealed minimal systematic bias between DBS-to-serum and venipuncture values; precision worsened at higher mean values of CRP. Time controls revealed little degradation or change in analyte values for HDL-C and CRP over 30 weeks. We concluded that DBS-assessed biomarkers represent a valid alternative to venipuncture assessments. Large studies using DBS should include study-specific serum-equivalency determinations to optimize individual-level sensitivity, the viability of detecting intervention effects, and generalizability in community-level primary prevention interventions. Background: Although maternal infection and inflammation during pregnancy can adversely affect offspring birth weight (BW), whether low grade inflammation in the non-pregnant state predicts BW is unknown. Aim: To evaluate relationships between offspring BW and pro- and anti-inflammatory factors measured in parous but non-pregnant women. Subjects and methods: Data come from 234 parous Filipino females (21.5 ± 0.3 years) in the Cebu Longitudinal Health and Nutrition Survey, a population-based birth cohort in Metropolitan Cebu, Philippines. Pro-inflammatory [Interleukin-6 (IL-6), Interleukin-1 beta (IL-1β), tumour necrosis factor alpha (TNFα), C-reactive protein (CRP)] and anti-inflammatory [Interleukin-10 (IL-10)] factors were measured in fasting plasma when the women were not pregnant, and related to recalled offspring BW. Results: BW in female offspring was lower only among women with high IL-1β. Although pro-inflammatory cytokines did not predict BW in male offspring, women with higher anti-inflammatory IL-10 gave birth to larger males. Women with a combination of low inflammatory (IL-6) and high anti-inflammatory (IL-10) factors (interaction p < 0.104) gave birth to the largest males. Conclusion: Immune factors measured outside of pregnancy predict offspring BW in these young women. Stable variation in inflammatory phenotype could impact the gestational environment of offspring, thus pointing to potential inter-generational effects of chronic low-grade inflammation. In industrial economies schooling produces positive non-market returns but do traditional forms of human capital also produce such returns, and do schooling and traditional human capital act as complements or substitutes in their association with well-being? Drawing on data from 450 adults (16+ years of age) from an indigenous Amazonian society in Bolivia, we estimate the association between traditional plant knowledge and nutritional status as measured by body-mass index. After conditioning for many covariates, we find that doubling an adult's traditional knowledge is associated with a mean improvement in BMI of 6.3 per cent; the association is stronger for unschooled adults and for those living far from the market town. Though schooling bore a negative association with traditional knowledge, those two forms of human capital had independent associations with BMI. Understanding why traditional cultures weaken matters because they embody humanity's heritage. Schooling has been singled out as an abrader of traditional culture. We assess whether schooling erodes one aspect of traditional culture: social capital as shown by generosity to people outside the household. In industrial nations researchers find positive associations between social capital and schooling. We hypothesize that in pre-industrial societies schooling will produce ambiguous effects on social capital: it will reduce social capital expressed through labour help but increase social capital expressed through gifts given. Furthermore, we hypothesize that schooling will lower the likelihood of displaying social capital to kin. To test the hypotheses we use 2004 data from a highly autarkic society of farmers and foragers in the Bolivian Amazon, the Tsimane'. To strengthen the inferences we make about causality we use instrumental variables. Participants in the study included all people of 16+ years of age (n=574) in 13 villages. Social capital included (a) gifts given to people of other households and (b) labour help offered to people of other households and participation in communal work during the week before the day of the interview. Instrumental variables included childhood village of residence and father's writing ability. Contrary to expectations, schooling weakened all forms of social capital, including expressions of generosity toward kin, but results were statistically insignificant. Parameters estimated with standard techniques and with instrumental variables differed in sign and magnitude, suggesting that researchers should correct for the endogeneity of schooling when assessing schooling's effect on social capital. Benign prostatic hyperplasia (BPH) is clinically diagnosed in approximately three‐fourths of American men, a prevalence two to three times higher than that currently reported in Scotland and Japan. Prostatectomy is the most common operation performed on American men over 65. A critical medical anthropological analysis reveals that American urologists rely on prostatectomy to maintain their practices, and the growth of urology as a specialty is significantly related to the 50% increase in the rate of surgery since 1965. Marketing campaigns and studies funded by pharmaceutical companies promote the perception of urinary difficulties as problematic, encourage men to medicalize their symptoms, and create demand for drug therapy and urologist treatment.
