AbstractIn this study, tea saponin was used as an emulsifier to construct tea saponin-cinnamaldehyde nanoemulsion (TS-CNE) system by high-pressure homogenization nanotechnology. Preparation conditions of TS-CNE were optimized using single factor and response surface methods. The results showed that optimal TS-CNE had a particle size of 88.81 nm, a polydispersity index (PDI) of 0.113 and 86.49% retention of cinnamaldehyde. TS-CNE was stored at 4 and 25 °C for 180 days and the nanosystem was stable. Antioxidant assay showed that TS-CNE had good radicals scavenging. Furthermore, antimicrobial assay showed that the minimum fungicidal concentration (MFC) of TS-CNE was 1.95 mL/L against three pathogenic fungi (Penicillium italicum, Penicillium digitatum, and Geotrichum citri-aurantii) of citrus. TS-CNE (MFC and 10 × MFC) effectively suppressed disease development in citrus inoculated with the three fungi. In this study cinnamaldehyde (CA) doped polyurethane cholesteryl pelargonate cholesteric liquid crystal composite membranes were produced and their properties were examined for use in biological applications. The relationship between the blood coagulation and protein adsorption was studied and the results showed that increasing CA concentration allowed an improvement in biological properties of them. Additionally, the effect of CA on the hyrophobicity of these membranes was investigated by determining water holding capacity, contact angles. The mechanical strength of these membranes were determined and it was found that their strength increased and the ability to elongate decreased with an increasing CA dopant concentration. The aim of this study was to prepare a solid self-microemulsifying drug delivery system (S-SMEDDS) of cinnamaldehyde (CA) by spray drying technique to improve the oral bioavailability of CA. The preparation of CA S-SMEDDS with maltodextrin as the solid carrier, a core-wall material mass ratio of 1:1, a solid content of 20% (w/v), an inlet air temperature of 150 °C, an injection speed of 5.2 mL/min, and an atomization pressure of 0.1 MPa was determined by using the encapsulation rate as the index of investigation. Differential scanning calorimetry (DSC) revealed the possibility of CA being encapsulated in S-SMEDDS in an amorphous form. The in-vitro release showed that the total amount of CA released by S-SMEDDS was approximately 1.3 times higher than that of the CA suspension. Pharmacokinetic results showed that the relative oral bioavailability of CA S-SMEDDS was also increased to 1.6-fold compared to CA suspension. Additionally, we explored the mechanism of CA uptake and transport of lipid-soluble drugs CA by S-SMEDDS in a Caco-2/HT29 cell co-culture system for the first time. The results showed that CA S-SMEDDS uptake on the co-culture model was mainly an energy-dependent endocytosis mechanism, including lattice protein-mediated endocytosis and vesicle-mediated endocytosis. Transport experiments showed that CA S-SMEDDS significantly increased the permeability of CA in this model. As a typical bioactive compound from the bark and leaves of the trees of the genus Cinnamomum, cinnamaldehyde (CIN) is natural and safe. Its excellent antibacterial activity against various foodborne microorganisms is growingly regarded as a promising additive for improving and enhancing the properties of bio-based packaging films/coatings. This review systematically summarized the bio-based food packaging films/coatings containing CIN developed recently. The effects of CIN incorporation on physical and chemical properties of the antibacterial food packaging films/coatings, including thickness, color index, transparency, water content, water solubility, water contact angle, mechanical performances, water barrier performances, and antibacterial performances, were discussed. Simultaneously, this work also concluded that an explanation of the antibacterial mechanism of CIN and preparation methods of bio-based packaging films/coatings containing CIN/CIN carriers. Notably, the incorporation of CIN into the films/coatings could enhance their antibacterial performance extend the shelf-life of various foods, such as fish, meats, vegetables, fruits, and other perishable food, while improving their physical and chemical properties. Although incorporating CIN into food packaging films/coatings has been extensively studied, long-term follow-up research on the human safety of active food packaging films/coatings containing CIN needs to be carried out. Wax-based solid lipid nanoparticles were prepared using beeswax (BW) and propolis wax (PW) by antisolvent precipitation. Response surface methodology was applied to optimize the production condition with respect to waxes concentrations, emulsifier content (sodium caseinate and polysorbate 80) and solvent to antisolvent ratios. The formulation of wax-based nanoparticles with higher yield, lower polydispersity index and smaller particle size for each wax was used to encapsulate cinnamaldehyde (CA). The results of applying different ratios of wax: CA (1:0.25–1:3 w/w) showed that the best ratio was 1:0.25 with the smallest particle size and the highest encapsulation efficiency (EE). CA-loaded nanoparticles had spherical morphology with β' form structure. Also, the release study confirmed a high capability of waxes to control release and desirable protection of CA. Cinnamaldehyde is an essential oil extracted from the leaves, bark, roots and flowers of cinnamon plants (genus Cinnamomum). Cinnamaldehyde has shown biological functions such as antioxidants, antimicrobials, anti-diabetic, anti-obesity and anti-cancer. However, poor solubility in water as well as molecular sensitivity to oxygen, light, and high temperature limit the direct application of cinnamaldehyde. Researchers are using different encapsulation techniques to maximize the potential biological functions of cinnamaldehyde. Different delivery systems such as liposomes, emulsions, biopolymer nanoparticles, complex coacervation, molecular inclusion, and spray drying have been developed for this purpose. The particle size and morphology, composition and physicochemical properties influence the performance of each delivery system. Consequently, the individual delivery system has its advantages and limitations for specific applications. Given the essential role of cinnamaldehyde in functional food and food preservation, appropriate approaches should be applied in the encapsulation and application of encapsulated cinnamaldehyde. This review systematically analyzes available encapsulation techniques for cinnamaldehyde in terms of their design, properties, advantages and limitations, and food application status. Background: Dental caries is a multifactorial disease, and the bacteria such as Streptococcus mutans (S. mutans) is one of the risk factors. The poor effect of existing anti-bacterial is mainly related to drug resistance, the short time of drug action, and biofilm formation. Methods: To address this concern, we report here on the cinnamaldehyde (CA) loaded chitosan (CS) nanocapsules (CA@CS NC) sustained release CA for antibacterial treatment. The size, ζ-potential, and morphology were characterized. The antibacterial activities in vitro were studied by growth curve assay, pH drop assay, biofilm assay, and qRT-PCR In addition, cytotoxicity assay, organ index, body weight, and histopathology results were analyzed to evaluate the safety and biocompatibility in a rat model. Results: CA@CS NC can adsorb the bacterial membrane due to electronic interaction, releasing CA slowly for a long time. At the same time, it has reliable antibacterial activity against S. mutans and downregulated the expression levels of QS, virulence, biofilm, and adhesion genes. In addition, it greatly reduced the cytotoxicity of CA and significantly inhibited dental caries in rats without obvious toxicity. Conclusion: Our results showed that CA@CS NC had antibacterial and antibiofilm effects on S. mutans and inhibit dental caries. Besides, it showed stronger efficacy and less toxicity, and was able to adsorb bacteria releasing CA slowly, providing a new nanomaterial solution for the treatment of dental caries. To prove the internal connection, we deciphered the effect of cinnamaldehyde on kidney senescence through establishing animal and cell models. In vivo, a rat senescence model was constructed using D-galactose (D-gal), and the modeled rats were further treated with cinnamaldehyde. In vitro, rat renal tubular epithelial cells (NRK-52E) were transfected with miR-155 mimic or inhibitor and then treated with cinnamaldehyde, D-gal or PI3K inhibitor (LY294002). The serum levels of blood urea nitrogen (BUN) and serum creatinine (Scr) of the rats were measured by an automatic biochemical analyzer. Pathological changes of kidney were determined by hematoxylin-eosin staining. The senescence and viability of NRK-52E cells were assessed by SA-β-gal staining and CCK-8 assay, respectively. The levels of miR-155, p-PI3K/PI3K, p-Akt/Akt, LC3B (LC3-II and LC3-I) and Beclin1 were detected by qRT-PCR, immunohistochemistry, or western blot. D-gal elevated the levels of BUN, Scr and miR-155 in the kidney, induced the renal pathological damage, inhibited the cell viability, increased the numbers of SA-β-gal-, LC3B- and Beclin1-positive cells and upregulated the levels of LC3-II/LC3-I and Beclin1 both in the kidney and cells. Cinnamaldehyde reversed D-gal-induced effects on the kidney and cells, and moreover, the cinnamaldehyde-induced anti-D-gal effects on cells could be suppressed by miR-155 mimic but promoted by miR-155 inhibitor. LY294002 potentiated D-gal-induced effects, and reversed cinnamaldehyde- and miR-155 inhibitor-caused impacts on the PI3K/Akt pathway and LC3-II/LC3-I level in D-gal-induced cells. Cinnamaldehyde has an effective antimicrobial activity and therefore it received interest as a feed additive in swine nutrition. However, its high reactivity to amino acid residues might affect its efficacy and digestive processes. As an alternative, 20 chemical derivatives were evaluated. The in vitro antimicrobial activities were tested in an in vitro fermentation model. Thereof three compounds were selected and fed to newly weaned piglets. Five dietary treatments were replicated in six pens of four pigs per pen; i.e. control, cinnamaldehyde at 100 and 400 mg/kg, and 2-methoxycinnamaldehyde and 4-methoxycinnamaldehyde, both equimolar to 400 mg/kg cinnamaldehyde. In vitro results showed that 4-nitrocinnamaldehyde has the highest antimicrobial activity; however, this compound is carcinogenic and was not further issued. Cinnamaldehyde had the second-highest activity, particularly against coliform bacteria and Escherichia coli (E. coli), followed by 4-methoxycinnamaldehyde, 2-methoxycinnamaldehyde and hydrocinnamaldehyde. All other derivatives showed lower potency, but they were consistently more bactericidal against coliform bacteria and E. coli as compared to Gram-positive bacteria. At pH 7, aldehydes showed stronger bactericidal activity than their corresponding carboxylic acids, which was not the case at pH 5, suggesting a different mode of action. In the in vivo trial, no significant improvements in animal performance or antimicrobial effects were observed. To conclude, apart from 4-nitrocinnamaldehyde, none of the derivatives showed higher antimicrobial potency than cinnamaldehyde. Three selected compounds from in vitro trials failed to demonstrate major positive outcomes in the in vivo trial. 4-Nitrocinnamaldehyde has the highest antimicrobial activity in vitro. Cinnamomum verum essential oil and its main component, cinnamaldehyde, were incorporated into nanoemulsions to improve their stability and activity. Oil-in-water (O/W) nanoemulsions were prepared using the high-energy ultra-sonication method. Cinnamon oil nanoemulsion exhibited a more vigorous DPPH radical scavenging activity (IC50 = 1.81 mg/L) than the standard α-tocopherol (IC50 = 9.16 mg/L). Cinnamaldehyde nanoemulsion exhibited the highest antibacterial action against Salmonella typhimurium and Staphylococcus aureus (MIC = 175 and 150 mg/L, respectively). It was also displayed the highest inhibition against Candida albicans (EC50 = 19.92 mg/L). Cinnamaldehyde nanoemulsion was the most active against mycelial growth of Aspergillus flavus and A. niger (EC50 = 38.20 and 134.80 mg/L, respectively). It also presented the highest activity against spore germination. The purpose of this experiment was to explore the effect of Solid lipid nanoparticles (SLNs) on improving the oral absorption and bioavailability of cinnamaldehyde (CA). CA-SLNs were prepared by high-pressure homogenization and characterized by particle size, entrapment efficiency, and morphology, thermal behavior and attenuated total reflection Fourier transform infrared (ATR-FTIR). In vitro characteristics of release, stability experiments, cytotoxicity, uptake and transport across Caco-2 cell monolayer of CA-SLNs were studied as well. In addition, CA-SLNs underwent pharmacokinetic and gastrointestinal mucosal irritation studies in rats. CA-SLNs exhibited a spherical shape with a particle size of 44.57 ± 0.27 nm, zeta potential of −27.66 ± 1.9 mV and entrapment efficiency of 83.63% ± 2.16%. Differential scanning calorimetry (DSC) and ATR-FTIR confirmed that CA was well encapsulated. In vitro release of CA-SLNs displayed that most of the drug (90.77% ± 5%) was released in the phosphate buffer, and only a small amount of drug (18.55% ± 5%) was released in the HCl buffer. CA-SLNs were taken up by an energy-dependent, endocytic mechanism mediated by caveolae mediated endocytosis across Caco-2 cells. The CA permeation through Caco-2 cell was facilitated by CA-SLNs. The outcome of the gastrointestinal irritation test demonstrated that CA-SLNs had no irritation to the rats' intestines. Compared with CA dispersions, incorporation of SLNs increased the oral bioavailability of CA more than 1.69-fold. Population growth directly affects the global food supply, demanding a higher production efficiency without farmland expansion - in view of limited land resources and biodiversity loss worldwide. In such scenario, intensive agriculture practices have been widely used. A commonly applied method to maximize yield in animal production is the use of subtherapeutic doses of antibiotics as growth promoters. Because of the strong antibiotic selection pressure generated, the intense use of antibiotic growth promoters (AGP) has been associated to the rise of antimicrobial resistance (AMR). Also, cross-resistance can occur, leading to the emergence of multidrug-resistant pathogens and limiting treatment options in both human and animal health. Thereon, alternatives have been studied to replace AGP in animal production. Among such alternatives, essential oils and essential oil components (EOC) stand out positively from others due to, besides antimicrobial effectiveness, improving zootechnical indexes and modulating genes involved in resistance mechanisms. This review summarizes recent studies in essential oils and EOC for zoonotic bacteria control, providing detailed information about the molecular-level effects of their use in regard to AMR, and identifying important gaps to be filled within the animal production area. Aim: The cardiovascular toxicity of unheated and heated flavorants and their products as commonly present in electronic cigarette liquids (e-liquids) was evaluated previously in vitro. Based on the results of in vitro assays, cinnamaldehyde, eugenol, menthol, and vanillin were selected to conduct a detailed chemical analysis of the aerosol generated following heating of each compound both at 250 and 750 °C. Materials and Methods: Each flavoring was heated in a drop-tube furnace within a quartz tube. The combustion atmosphere was captured using different methods to enable analysis of 308 formed compounds. Volatile organic compounds (VOCs) were captured with an evacuated Summa canister and assayed via gas chromatography interfaced with mass spectrometry (GC–MS). Carbonyls (aldehydes and ketones) were captured using a 2,4-dinitrophenylhydrazine (DNPH) cartridge and assayed via a high-performance liquid chromatography-ultra-violet (HPLC-UV) assay. Polyaromatic hydrocarbons (PAHs) were captured using an XAD cartridge and filter, and extracts were assayed using GC–MS/MS. Polar compounds were assayed after derivatization of the XAD/filter extracts and analyzed via GC–MS. Conclusion: At higher temperature, both cinnamaldehyde and menthol combustion significantly increased formaldehyde and acetaldehyde levels. At higher temperature, cinnamaldehyde, eugenol, and menthol resulted in increased benzene concentrations. At low temperature, all four compounds led to higher levels of benzoic acid. These data show that products of thermal degradation of common flavorant compounds vary by flavorant and by temperature and include a wide variety of harmful and potentially harmful constituents (HPHCs). The conversion of cinnamaldehyde (CALD) to Cinnamyl alcohol (CALC) over a Pt/SiO2 catalyst was investigated. To achieve maximum CALC production, the historical data design of using a response surface methodology (RSM) experimental design was applied to Correlate and optimize the operation process variables (i.e., catalyst amount, 0.1-0.3 g; reaction temperature, 70-110 °C; pressure, 20-40 bar; and reaction time, 30-360 min) for the CALD conversion and selectivity for CALC. The obtained experimental values of the conversion and selectivity were in good agreement with those predicted by a second-order quadratic model, indicating the suitability of the model to estimate the conversion and selectivity. The maximum CALD conversion and selectivity for CALC were 98.905% and 80.295%, respectively, at the following conditions: temperature, 75.96 °C; pressure, 20.11 bar; catalyst amount, 0.19 g; and reaction time, 296.59 min. Diabetes is a metabolic disorder related to blood insulin deficiency and high glucose level. Cinnamaldehyde is an important component of cinnamon and has an effect on blood glucose. It was aimed to investigate the the effect of cinnamaldehyde on the liver glutathione (GSH), glucose-6-phosphate dehidrogenase (G6PD) activity, blood glucose, protein, lipid and erythrocyte parameters, live weight in diabetic rats. Rat used for this research were divided 4 group as control, diabetic, cinnamaldehyde and diabetic + cinnamaldehyde group. The live weight and fasting blood glucose level, taken from tail vein were recorded every ten days. End of the trail the blood samples were taken from rats. Biochemical parameters with autoanalyzer and hematological parameters with blood cell counter were determined in blood. The activity of G6PD and GSH amounts were measured with ELISA in the liver tissues. Blood sugar, triglyceride, total cholesterol, VLDL, LDL, and urea levels increased in diabetic rats, and cinnemaldehyde significantly decreased these parameters. Cinnemaldehyde also showed a positive effect on body weight, blood total protein, and mean corpuscular volume in diabetes. A decrease in HbA1c and an increase in liver G6PD, GSH activity were found in treatment group, but these changes were not statistically significant. In conclusion, the antidiabetic, hypolipidemic and antioxidant effects of cinnamaldehyde were determined. It has also been shown to improve anaemia, ürea levels and weight loss. Long-term morphine use for therapeutic approaches may lead to serious side effects. Several studies have suggested opioid antagonist and antioxidant therapy for reducing adverse effects of morphine. Cinnamaldehyde has a potent anti-oxidant property. In this study, separate and combined effects of cinnamaldehyde and naloxone (an opioid receptor antagonist) on behavioral changes and cerebellar histological and biochemical outcomes were investigated after long-term morphine administration. Seventy-eight rats were divided into two major morphine-treated and morphine-untreated groups. Morphine-treated group was subdivided into seven subgroups for receiving vehicle, normal saline, cinnamaldehyde (1.25, 5, and 20 mg/kg), naloxone, and cinnamaldehyde plus naloxone before morphine. Morphine-untreated group was subdivided into six subgroups and treated with vehicle, cinnamaldehyde (1.25, 5, and 20 mg/kg), naloxone, and their combination. Chemical compounds were administered for 28 consecutive days. Behavioral tests including footprint, rotarod, and beam balance tests were employed. Histopathological and biochemical alterations of cerebellum were determined. Body and cerebellum weights, stride width, time spent on the rotarod, Purkinje cell number, thickness of molecular and granular layers, superoxide dismutase (SOD), and total antioxidant capacity (TAC) decreased as a result of administrating morphine. Morphine increased beam transverse time, malondealdehyde (MDA), tumor necrosis factor-α (TNF-α), and caspase-3 levels. Histopathological changes such as cellular vacuolation and loss were also produced as a result of treatment with morphine. Cinnamaldehyde, naloxone, and their combination treatments improved all the above-mentioned alterations induced by morphine. We concluded that cinnamaldehyde produced a neuroprotective effect through anti-oxidant, anti-inflammatory, apoptotic, and probably naloxone-sensitive opioid receptor interaction mechanisms. This study was conducted to evaluate the growth performance and carcass characteristics of finishing steers fed diets containing different feed additives: control (CTL; no feed additives); monensin [26 mg/kg dry matter (DM); MON]; a blend of cinnamaldehyde, eugenol, and capsicum (120 mg of a commercial product/kg DM; XT), or the combination of monensin and the blend of cinnamaldehyde, eugenol, and capsicum (26 mg/kg DM and 120 mg of a commercial product/kg DM, respectively; MON + XT). A total of 860 steers [initial body weight (BW) = 334 kg] were fed the basal diet (7% alfalfa hay and 93% concentrate; DM basis) containing the treatments during 154 days. Feed additives did not affect final BW, DM intake, average daily gain, and feed efficiency of finishing steers (P ≥ 0.12). Dressing percentage was lower for MON + XT than MON and XT (P = 0.01). Feeding XT tended to increase the percentage of the carcass with a small marbling score compared to MON (P = 0.09). The proportion of carcass grading premium choice tended to be greater for steers fed MON compared to XT (P = 0.07). It is a challenge for many researchers to separate volatile compounds. In this study, we introduce a rapid and efficient method of separating target compound from the twigs of Cinnamomum cassia by high performance counter-current chromatography. Under the bioassay guidance, the total extract exhibited a potential activity against NO production in RAW 264.7 macrophages and the total extract was further separated by high performance counter-current chromatography. Cinnamaldehyde (1) was enriched by counter-current chromatography (CCC) with reversed-phase mode using n-hexane–ethyl acetate–methanol–water (1:1:1:1,v/v/v/v) as the solvent system. Fusarium spp. are able to cause a wide range of opportunistic infections in both immunocompromised and immunocompetent individuals. Thus, new therapeutic options should be considered. We evaluated the antifungal activity of three phytocompounds (cinnamaldehyde, CIN; carvacrol, CRV; and thymol, THY), as well as their activities when combined with amphotericin B (AMB), caspofungin (CAS), itraconazole (ITC) and voriconazole (VRC) against 20 clinical isolates of Fusarium. Broth microdilution (M38-A2) and checkerboard methodologies were used to obtain MICs (Minimum inhibitory concentration) and FICIs (Fractional Inhibitory Concentration Index), respectively. The MICs observed for the phytocompounds were: 20–80 for CIN, 40-> 320 for CRV and 40–320 µg/mL for THY. The most effective combinations (synergism rates) were: THY + CAS (96%), CRV + CAS (88%), THY + AMB (76%), CRV + AMB (68%), THY + VRC (68%) and CRV + VRC (60%). Cinnamaldehyde (CA) is one of the major active pharmaceutical ingredient of cinnamon bark. Hydrodistillation (HD) is usually used in CA extraction, however, the extraction yield is lower. The cell wall is a key factor limiting the extraction of essential oils. In-situ reactive heat breaking cell wall (RHB) could destroy the cell wall, which was conducive to the diffusion of CA. The aim of this work was to examine the effect of RHB pretreatment to HD extraction. Response surface methodology (RSM) was used to optimize RHB pretreatment parameters, and Box-Behnken Design (BBD) method was performed to evaluate the effects of different operating parameters. The maximum yield was increased to 3.31 ± 0.11% (w/w) from 2.08 ± 0.042% (w/w) after RSM optimization. Scanning electron microscopic (SEM) analysis showed that RHB destroyed and disrupted the cell wall of cinnamon bark. The GC analysis demonstrated that the purity of cinnamaldehyde was improved and no new components were presented in the extraction product from the cinnamon via RHB pretreatment. The present study deals with the antifungal and antiaflatoxigenic potential of Cinnamomum cassia essential oil (CCEO) against aflatoxigenic food borne Aspergillus flavus strain (AF-LHP-PE-4), and probable mode of action including the safety profile on male mice. CCEO completely inhibited fungal growth and aflatoxin B1 secretion at its minimum inhibitory concentration (0.06 µL/mL). CCEO inhibited ergosterol biosynthesis in cell membranes and also disturbed the membrane fluidity. Significant enhancement in leakage of cellular ions and 260 and 280 nm absorbing cellular materials in response to increased concentrations of CCEO indicated the plasma membrane as the probable site of antifungal toxicity. The antiaflatoxigenic potency of CCEO was confirmed in terms of reduction in the level of cellular methylglyoxal (MG), the inducer of aflatoxin. Antioxidant activity of CCEO was confirmed through DPPH free radical scavenging activity as well as total phenolic content. Chemical profiling of CCEO by gas chromatography-mass spectrometry (GC-MS) analysis revealed the presence of cinnamaldehyde (84.01%) as the most abundant compound. CCEO exhibited a high LD50 (10410.75 µL/kg) on male mice, strengthening its favorable safety profile. The objective of the study was to evaluate the cytotoxicity and effect of curcumin-cinnamaldehyde hybrids (CCHs) on the biofilm of oral pathogens. Of the 18 hybrids tested, nine had an inhibitory effect on at least one of the bacterial species tested, with minimal inhibitory and bactericidal concentrations ranging from 9 to 625 μg ml−1. CCH 7 promoted a potent inhibitory effect against all the bacterial species tested and better compatibility than chlorhexidine (CHX). CCH 7 also presented a similar or improved effect over that of CHX, causing a reduction in bacterial metabolism and viability in single and dual-species biofilms. CCH 7 reduced by 86% and 34% the viability of multispecies biofilms formed by collection and clinical strains. It can be concluded that CCH 7 was cytocompatible at the minimal inhibitory concentration, presented anti-biofilm action against oral pathogens, and could act as an antimicrobial agent for application in endodontics. As a principal compound of cinnamon essential oil, cinnamaldehyde has been used as a marker for the quality control of cinnamon and its essential oil. During the HPLC analysis, a reaction product was observed in methanol solution of cinnamaldehyde reference standard. This product was isolated and identified to be cinnamaldehyde dimethyl acetal based on the spectroscopic analysis. Although dimethylacetalization of cinnamaldehyde in MeOH has been recorded in many literatures, this is the first report about this reaction in normal analysis procedure. Further studies suggested that this reaction could be affected by many parameters, including cinnamaldehyde concentration, storage temperature, and light illumination. Those results suggested the stock solution of cinnamaldehyde reference standard should not be prepared with methanol, and the extraction procedure by heating the sample with methanol might accelerate the acetalization of cinnamaldehyde. An environmentally friendly method has been proposed to synthesize silver nanoparticles using polyvinyl alcohol (PVA)-cinnamaldehyde (CIN) microemulsion. Ternary composite films with different concentration of Ag-NPs were prepared by solution casting method. The formation of Ag-NPs was confirmed by measuring surface plasmon resonance (SPR) peak. The SPR peak of Ag-NPs did not change after three months later, suggesting that the Ag-NPs stability without aggregation. Scanning electron microscopy (SEM) demonstrated that the Ag-NPs were uniformly dispersed in the film. The tensile strength and water vapor transmission rate of the composite film enhanced (p < 0.05) with the proportion of nano-silver increased, whereas the elongation at break unchanged. The composite film exhibited a strong antibacterial activity against both E. coli and S. aureu food-borne pathogenic bacteria. The aim was to evaluate the acaricidal effects of pure active components of essential oils against poultry red mite (Dermanyssus gallinae) as an alternative to chemical acaricides (organophosphates and pyrethroids). 2. The toxicities of five pure active components of essential oils (eugenol from clove bud, eucalyptol from rosemary, limonene from citrus fruits, linalool from lavender and cinnamaldehyde from cinnamon) were tested on D. gallinae females in an impregnated paper assay. 3. The active substances were dissolved in water and Tween 20 and applied at concentrations ranging from 0.002 to 0.06 µl/cm2. Toxicity was expressed as a lethal dose (LD50 or LD90). 4. The highest mortality was observed with eugenol. The LD90 was estimated to be 5.1 µg/cm2 for this substance, followed by cinnamaldehyde, the LD90 of which was estimated to be 11.0 µg/cm2. Traditional herbal monomers (THMs) are widely distributed in many traditional Chinese formulas (TCFs) and decoctions (TCDs) and are frequently used for the prevention and treatment of fungal infections. The antifungal activities of five common THMs, including sodium houttuyfonate (SH), berberine (BER), palmatine (PAL), jatrorrhizine (JAT) and cinnamaldehyde (CIN), and their potential for inducing cell wall remodeling (CWR), were evaluated against Candida albicans SC5314 and Candida auris 12372. SH/CIN plus BER/PAL/JAT showed synergistic antifungal activity against both Candida isolates. Furthermore, SH-associated combinations (SH plus BER/PAL/JAT) induced stronger exposure of β-glucan and chitin than their counterparts, while CIN triggered more marked exposure compared with CIN-associated combinations (CIN plus BER/PAL/JAT). Collectively, this study demonstrated the anti-Candida effect and the CWR induction potential of the five THMs and their associated combinations, providing a possibility of their in vivo application against fungal-associated infections. Objectives: Cholinergic system dysfunction was found to play a key role in Alzheimer's disease (AD) pathogenesis. Therefore, the animal model of scopolamine-induced amnesia has been widely used in AD researches. Cinnamon, as a spice commonly used in cuisine, has been shown to exert some therapeutic effects. The most abundant compound in cinnamon is cinnamaldehyde which recently was shown to exert several neuroprotective effects in animal models. Therefore, this study aimed to assess whether cinnamaldehyde has the potency to prevent memory retrieval impairment and hippocampal protein kinase B (Akt) and MAPK (extracellular signal-regulated kinase (ERK)) alterations induced by scopolamine in mice. Methods: Adult male mice were pretreated with cinnamaldehyde (12.5, 25, 40 and 100 mg/kg/oral gavage) 10 days before training. The training of passive avoidance task was performed on the 10th day and a memory retention test was done 24 h later. Scopolamine (1 mg/kg) was injected intraperitoneally, 30 min before the retention test to induce memory retrieval deficit. At the complement of the behavioral experiments, the hippocampi were isolated for western blot analysis to assess the phosphorylated and total levels of hippocampal MAPK and Akt proteins. Results: The results showed that cinnamaldehyde pretreatment at the dose of 100 mg/kg significantly prevented the amnesic effect of scopolamine. Furthermore, cinnamaldehyde prevented scopolamine induced dysregulations of hippocampal MAPK and Akt. Discussion: The results of the present study revealed that oral sub-chronic cinnamaldehyde administration has the capability to prevent memory retrieval deficit induced by cholinergic blockade and restores hippocampal MAPK and Akt dysregulations. Context:Cinnamomum verum J. Presl. (Lauraceae) has a high number of polyphenols with insulin-like activity, increases glucose utilization in animal muscle, and might be beneficial for diabetic patients. Objective: This study evaluated the effectiveness of an ointment prepared from Cinnamomum verum hydroethanolic extract on wound healing in diabetic mice. Materials and methods: A total of 54 male BALB/c mice were divided into three groups: (1) diabetic non-treated group mice that were treated with soft yellow paraffin, (2 and 3) mice that were treated with 5 and 10% C. verum. Two circular full-thickness excisional wounds were created in each mouse, and the trial lasted for 16 d following induction of the wound. Further evaluation was made on the wound contraction ratio, histopathology parameters and mRNA levels of cyclin D1, insulin-like growth factor 1 (IGF-1), glucose transporter-1 (GLUT-1), total antioxidant capacity, and malondialdehyde of granulation tissue contents. HPLC apparatus was utilized to identify the compounds. Results: The HPLC data for cinnamon hydroethanolic extract identified cinnamaldehyde (11.26%) and 2-hydroxyl cinnamaldehyde (6.7%) as the major components. A significant increase was observed in wound contraction ratio, fibroblast proliferation, collagen deposition, re-epithelialization and keratin biosynthesis in the C. verum-treated groups in comparison to the diabetic non-treated group (p < 0.05). The expression level of cyclin D1, IGF1, GLUT 1 and antioxidant capacity increased in the C. verum-treated groups in comparison to the diabetic non-treated group (p < 0.05). Anxiety and stress are considered as universal psychiatric exhibitions of the present societies and lifestyles. Several experiments have been conducted to examine natural anxiolytic agents to find out an alternative to synthetic anxiolytic drugs. The present study investigated the anxiolytic effects of cinnamaldehyde (Cin) on mice behavior in the elevated plus maze (EPM) and open field (OF) tests. Sixty male Swiss mice, weighing 20–30 g, were divided into six groups including: acute stress + mazola oil; chronic stress + oil; acute stress + Cin (20 mg/kg); chronic stress + Cin; non-stress + oil; and non-stress + Cin groups. The groups were administered for seven days (once a day). The acute stress + Cin group showed a meaningful rise in the percentage of entries into the open arms compared to the acute stress + oil group (p <.05). The percentage of time spent in the open arms in the chronic stress + Cin group was significantly higher compared to the chronic stress + oil group (p <.01). The percentage of entries into the open arms increased significantly (p <.01) in the chronic stress + Cin group in comparison with the chronic stress + oil group. The Cin treated groups showed significant increases in the time spent in the center area and in the number of entries into the center area compared with the oil treated groups in OF test. The number of entries into the arms (total activity), as well as locomotor activity was not significant among groups. The results of the present study indicated that Cin, as a natural product, might have anxiolytic effects in mice behavior in the EPM and OF tests. The results demonstrated that the administration of cinnamaldehyde (Cin) produced anxiolytic effects in mice. Natural antioxidant products have been reported effective for anxiety. Synthetic medications have notable adverse effects. Therefore, these natural substances with broad therapeutic applicability are able to reduce anxiety-related behavior with rare side effects. Context: The resistance of bacteria to antibiotics is raising serious concern globally. Asian medicinal plants could improve the current treatment strategies for bacterial infections. The antibacterial properties of medicinal plants used by the Khyang tribe in Bangladesh have not been investigated. Objective: The present study examines the antibacterial properties of 18 medicinal plants used by the Khyang tribe in day-to-day practice against human pathogenic bacteria. Materials and methods: Leaves, bark, fruits, seeds, roots and rhizomes from collected plants were successively extracted with hexane, ethyl acetate and ethanol. The corresponding 54 extracts were tested against six human pathogenic bacteria by broth microdilution assay. The antibacterial mode of actions of phytoconstituents and their synergistic effect with vancomycin and cefotaxime towards MRSA was determined by time-killing assay and synergistic interaction assay, respectively. Results and discussion: Hexane extract of bark of Cinnamomum cassia (L.) J. Presl. (Lauraceae) inhibited the growth of MRSA, Enterococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Acinetobacter baumannii with MIC values below 100 µg/mL. From this plant, cinnamaldehyde evoked at 4 × MIC in 1 h an irreversible decrease of MRSA count Log10 (CFU/mL) from 6 to 0, and was synergistic with vancomycin for MRSA with fractional inhibitory concentration index of 0.3. Recent research has showed that Aspergillus flavus and Aspergillus parasiticus are aflatoxigenic species that can become very competitive in the framework of climate change. Aflatoxins show carcinogenic, mutagenic, immunotoxic and teratogenic effects on human and animals. Effective and sustainable measures to inhibit these species and aflatoxins in food are required. Origanum vulgare and Cinnamomum zeylanicum essential oils (EOs) and their major active constituents, carvacrol and cinnamaldehyde, respectively, were assayed for inhibiting these species and aflatoxin production in maize extract medium under different environmental conditions. Doses of 10-1000 mg l−1 were assayed and the effective doses for 50 (ED50) and 90% (ED90) growth inhibition were determined. The ED50 of cinnamaldehyde, carvacrol, oregano EO, and cinnamon EO against A. flavus were in the ranges 49-52.6, 98-145, 152-505, 295-560 mg l−1 and against A. parasiticus in the ranges 46-55.5, 101-175, 260-425 and 490-675 mg l−1, respectively, depending on environmental conditions. In A. flavus treatments ED90 were in the ranges 89.7-90.5, 770-860 and 820->1000 mg l−1 for cinnamaldehyde, carvacrol and cinnamon EO, and in A. parasiticus treatments in the ranges 89-91, 855->1000 and 900->1000 mg l−1, respectively. ED90 values for oregano EO against both species were >1000 mg l−1. Growth rates of both species were higher at 37 than at 25°C and at 0.99 than at 0.96 aw. Aflatoxin production was higher at 25 than at 37°C. Stimulation of aflatoxin production was observed at low doses except for cinnamaldehyde treatments. The effectiveness of EOs and their main constituents to inhibit fungal growth and aflatoxin production in contact assays was lower than in vapour phase assays using bioactive EVOH-EO films previously reported. Cinnamon (Cinnamomum zeylanicum, Lauraceae) is a food additive greatly used for its taste. However, recently this medicinal plant has been brought to attention due to its medical effects. Cinnamon has constituents such as cinnamaldehyde and cinnamic acid that offers some health benefits including antioxidant and free-radical scavenging properties, lowering of blood glucose, anti-cholesterolemic, analgesic, antimicrobial, anti-inflammatory, anti-yeast, anti-secretagogue, and anti-gastric ulcer effects. This review summarizes various in vitro and animal studies on the protective effects of cinnamon against natural and chemical toxins. These studies consider the antidotal and/or protective effects of cinnamon and its major constituents against natural toxins and chemical-induced toxicities. The presence of coumarin, a hepatotoxic phenyl propanoid, in samples of the spice, cinnamon, procured from the market has raised food safety concerns world over. A validated, ultra-performance liquid chromatography- linear ion trap triple quadrupole mass spectrometry method was developed in the present study for the estimation of coumarin and other phenolics in authentic Cinnamomum verum bark samples and market samples from south India. Coumarin contents in authentic bark samples were in the range of 12.3 to 143.0 mg/kg, whereas the coumarin contents in samples procured from the market were up to 3462.0 mg/kg. The high content of coumarin and cinnamaldehyde in cinnamon samples procured from the market suggested possible blending with substitutes like Cinnamomum cassia barks, which possess substantial amounts of coumarin and cinnamaldehyde. Hydrodistillation (HD), supercritical fluid extraction (SFE) and reflux extraction (RE) were applied to obtain Cinnamomi ramulus extracts. The yields, chemical compositions and antiviral activities of the extracts were investigated. Extracts were analysed using gas chromatography–mass spectrometry and the antiviral activities were evaluated using cytopathic effect inhibition assay. HD, SFE and RE afforded 0.376, 1.227 and 5.914% yields, respectively. Cinnamaldehyde (CA), SFE and ethanol extracts exhibited antiviral activities against herpes simplex virus type 1. Context:Cinnamaldehyde (CA) has a protective effect in endotoxin poisoning of mice, but there is no direct evidence for the protective effect of CA through inhibition of NLRP3 inflammasome activation in endotoxin poisoning of mice. Objective:We aimed to investigate the protective mechanism of CA in endotoxin poisoned mice through NLRP3 inflammasome. Materials and methods:First, we evaluated the anti-inflammatory effect of CA in phorbol-12-myristate acetate–differentiated THP-1 cells through the NLRP3 inflammasome. Second, in a mouse model of lipopolysaccharide (LPS)-induced endotoxin poisoning, CA was administrated for 5 d (once a day) before the 15 mg/kg LPS challenge. Then, the levels of IL-1β in serum were measured, and the effect of CA on the NLRP3 inflammasome activation and the expression of cathepsin B and P2X7R proteins in lung were explored. Results:In vitro, CA decreased the levels of p20, pro-IL-1β and IL-1β in cell culture supernatants, as well as the expression of NLRP3 and IL-1β mRNA in cells.In vivo, CA decreased IL-1β production in serum. Furthermore, CA suppressed LPS-induced NLRP3, p20, Pro-IL-1β, P2X7 receptor (P2X7R) and cathepsin B protein expression in lung, as well as the expression of NLRP3 and IL-1β mRNA. Conclusions:CA has a protective effect in the endotoxin poisoned mice through the inhibition of NLRP3 inflammasome activation. Furthermore, CA suppresses the NLRP3 inflammasome activation by inhibiting the expression of cathepsin B and P2X7R protein expression. This study was conducted to explore the potential benefits of using cinnamaldehyde (CIN), resveratrol (RES) separately or in combination on cyadox (CYA)-induced alterations in isolated rabbit erythrocytes. Erythrocytes suspensions were partitioned into 7 groups (5 replicates/group), 1st kept as control treated with phosphate buffered saline (PBS) with dimethyl sulphoxide (DMSO); 2nd group was subjected to CYA (40 μg/ml), 3rd group was incubated with CIN (40 μM), 4th group was subjected to RES (40 μM), 5th group was co-exposed to CYA (40 μg/ml) and CIN (40 μM), 6th group was co exposed to CYA (40 μg/ml) and RES (40 μM), and 7th group was exposed to CYA in combination with both CIN and RES at the same indicated concentrations. The reaction mixtures of different groups were incubated at 37 °C for 3 h with gentle shaking every 15 minutes. Our results revealed that exposure to CYA caused a significant decrease (linear and quadratic) in superoxide dismutase (SOD) and catalase (CAT) activities and the contents of reduced glutathione (GSH) and glutathione transferase (GST). Incubation of erythrocytes with CYA increased GSSG content, GSSG/GSH ratio, malonaldehyde (MDA) and protein carbonyl (PrC) concentrations while it decreased the total protein (TP). CYA also lead to hemolysis and energy depletion of erythrocytes beside activation of caspase cascades, suggesting the pro-oxidant effect CYA that could be implicated in eryptosis. CIN and RES were able to inverse these hazardous effects of CYA. The aim of the study was to evaluate the pattern of content (%) of major compounds inCinnamomum tamaladuring three months (September, October and November) as affected by various tree girth (cm) of the tree. The average oil yields (%) varied between 0.20±0.05 to 1.20±0.20% during three months. GC-fingerprinting analysis revealed different trends in the concentration of principal constituents. A decreasing trend was shown in the concentration of linalool with the increase in girth sizes. Initially, linalool concentration increased from 26.36–30.18% (September), 22.52–23.29% (October) and 20.97–31.81% (November) upto the tree girth size of 66 cm, beyond which the contents were decreased. Cinnamaldehyde has shown throughout an increasing trend with the increase in tree girth. The contents increased from 10.56–69.62% (September), 24.34–70.43% (October) and 32.39–68.64% (November) with increasing tree girth from 26 to 186 cm, while an irregular pattern of occurrence of cinnamyl acetate (1.03–31.0%) was observed in the essential oils isolated from the leaves samples of different girth sizes (cm). The statistical analysis revealed that contents of linalool and cinnamaldehyde were significantly influenced by various tree girth. Cinnamyl alcohol (COL) is one of the important perfumery chemicals and is obtained from cinnamaldehyde (CAL). This work presents one of the two steps of intensifying the hydrogenation of CAL to COL. Critical analysis of published results was carried out to identify most promising catalyst system (5% Pt/C catalyst and aqueous KOH as a promoter). Appropriate experimental methodology, which ensures adequate material balance and eliminates influence of mass transfer, etc., was established. Influence of key reaction parameters such as solvents, catalyst loading, hydrogen pressure and temperature of the reaction on the activity and selectivity towards COL was investigated. The experimental results were analysed to clearly bring out desired operating parameters which optimise reaction rate and selectivity. Recyclability of catalyst was also verified by carrying out couple of recycle experiments. Pt catalysts and Co doped Pt catalysts with TiO2as the support were prepared by the sol immobilization method based on biological synthesis process, whereby Pt nanoparticles (PtNPs) were reduced from Na2PtCl4usingCacumen Platycladiextract (CPE). The catalytic performance of the catalysts was studied using hydrogenation of cinnamaldehyde as the model reaction. For comparison, Co doped Pt/catalysts were also prepared by traditional impregnation method, TEM observation, and XRD measurement were carried out to characterize the catalysts. The effects of different Pt loadings and Co doping amounts were investigated. The results showed that the PtNPs could be well dispersed onto the TiO2support with narrow size distribution between 4 and 6 nm. 1.0% was observed as the optimal Pt loading amount in our experiments showing a cinnamaldehyde conversion of 73.3% with 68.8% selectivity to cinnamyl alcohol. A proper Co doping amount was also essential to the catalytic performance of the prepared Pt/catalysts as well as a favorable for the reaction process, 1.0% doping amount showed the best activity in which a 70.4% conversion and 88.3% selectivity was achieved. Recyclability tests carried out revealed that the 1.0 Co–1.0 Pt/TiO2catalysts possessed possessing good durability. In this work, Co-Pt/TiO2catalysts prepared by the sol immobilization method based on biological reduction process, showed better selectivity than that from traditional impregnation method. Natural antioxidants recently have gained popularity since synthetic ones have toxic and carcinogenic effects. In the present study, effect of temperature (120, 150 and 180°C) and cinnamaldehyde on oxidative stability of several oils (olive, hazelnut and palm oils) and fats (milkfat and butter) was examined. In order to compare the results with the synthetic antioxidant, butylated hydroxy toluene (BHT) was used at a concentration of 200 ppm. This level is the legal maximum limit allowed. Experiments were conducted by using a PetroOxy device, a rapid small scale oxidation stability test. According to induction time values obtained by PetroOxy device, the stability of oils drastically decreased with increasing temperature. Cinnamaldehyde had no significant effect (p > 0.05) on all fat and oil samples compared to control (no antioxidant added) and BHT added samples. BHT significantly increased induction times of all fat and oil samples at lower temperatures. However, it was not effective at 180°C (p < 0.05). The yield of benzaldehyde in the 2-hydroxypropyl-β-cyclodextrin (2-HP-β-CD), with a low degree of substitution (DS = 3.9), catalytic system was 70%, which was higher than that of the system with 8.8 DS under the optimised conditions (323 K, 2% NaOH (w/v), cinnamaldehyde:2-HP-β-CD = 1:1 (molar ratio)). Due to the structural complementary effects of the host and guest, cyclodextrins (CDs) played the catalysis role in the reaction, which was confirmed by kinetic studies and solubilisation. The characterisation of NMR and fluorescence measurement suggested that the extent of hydroxypropyl substitution affected the cavity structure of the CD molecule. Staphylococcus epidermidisis reported to be the main causative agent of nosocomial infections. It has become increasingly difficult to treat this micro-organism because of the emergence of new antibiotic-resistant strains and its ability to form biofilm on medical associated devices. Phytochemicals acting in synergy are effective in killing the micro-organisms by lowering the doses, and synergistic compounds evade the development of resistance due to different mechanism of action. This study aims to determine the synergistic antimicrobial potential of curcumin with cinnamaldehyde, eugenol, and ellagic acid againstS. epidermidis.Curcumin with ellagic acid as well as eugenol were found to have additive antimicrobial effect, whereas, in combination, curcumin and cinnamaldehyde were found to have synergistic effect againstS. epidermidis(fractional inhibitory concentration index (FICI) = 0.5). Synergy between curcumin and cinnamaldehyde was established by time–kill kinetics and was further evaluated for antibiofilm activity. The dose required to inhibit biofilm formation was reduced to half than that needed to inhibit its planktonic culture (minimal inhibitory concentration (MIC) of curcumin = 3.12 μg/ml; MIC of cinnamaldehyde = 15.62 μg/ml; FICI = 0.248). The plant disease produced by Sclerotinia sclerotiorum is a big problem in agriculture. This paper tested the effects of cinnamon oil and its components on the growth of S. sclerotiorum. The cinnamon oil was first separated into carbonyl and noncarbonyl parts by treatment with sodium bisulfite. Then two main cinnamaldehyde derivatives, that is 2′-methoxycinnamaldehyde and coniferyl aldehyde, were further separated by column chromatography from the carbonyl parts. Cinnamon oil demonstrated a significant antifungal effect against S. sclerotiorum with a minimum inhibitory concentration (MIC) of 256 μg/mL in agar and 64 μg/mL air, respectively. trans-Cinnamaldehyde exhibited the highest antifungal activity among all the three cinnamaldehydes tested. Efficient oxidative cleavage of cinnamaldehyde to benzaldehyde by hydrogen peroxide in the presence of β-cyclodextrin (β-CD) and sodium bicarbonate in water has been developed. β-CD showed excellent activity for the oxidation under mild conditions. The effects of different reaction conditions, e.g. reaction temperature, the amount of β-CD, sodium bicarbonate and hydrogen peroxide concentrations, etc. have been investigated. The yield of benzaldehyde could reach 78% under the optimum conditions (333 K, 2 h, molar ratio for cinnamaldehyde:β-CD:sodium bicarbonate = 1:1:12). This study was conducted to evaluate the effect of wheat dried distillers' grains with solubles (DDGS) and cinnamaldehyde (CIN) on in vitro fermentation and microbial profiles using the rumen simulation technique. The control substrate (10% barley silage, 85% barley grain and 5% supplement, on dry matter basis) and the wheat DDGS substrate (30% wheat DDGS replaced an equal portion of barley grain) were combined with 0 and 300 mg CIN/l of culture fluid. The inclusion of DDGS increased (p < 0.05) the concentration of volatile fatty acids (VFA) and the molar proportion of acetate and propionate. Dry matter disappearance (p = 0.03) and production of bacterial protein (p < 0.01) were greater, whereas the disappearances of crude protein (CP) and neutral detergent fibre were less (p < 0.01) for the DDGS than for the control substrate. With addition of CIN, concentration of total VFA decreased and fermentation pattern changed to greater acetate and less propionate proportions (p < 0.01). The CIN reduced (p < 0.01) methane production and CP degradability. The copy numbers of Fibrobacter, Prevotella and Archaea were not affected by DDGS but were reduced (p < 0.05) by CIN. The results indicate that replacing barley grain by DDGS increased nutrient fermentability and potentially increase protein flows to the intestine. Supplementation of high-grain substrates with CIN reduced methane production and potentially increased the true protein reaching the small intestine; however, overall reduction of feed fermentation may lower the feeding value of a high-grain diet. Cinnamaldehyde (1) is a pharmacologically active ingredient isolated from cassia twig (Ramulus Cinnamomi), which is commonly used in herbal remedies to treat fever-related diseases. Both TRPV1 and TRPM8 ion channel proteins are abundantly expressed in sensory neurons, and are assumed to act as a thermosensor, with the former mediating the feeling of warmth and the latter the feeling of cold in the body. Both of them have recently been reported to be involved in thermoregulation. The purpose of this paper is to further uncover the antipyretic mechanisms of 1 by investigating its effects on the mRNA expression levels and functions of both TRPV1 and TRPM8. The results showed that 1 could up-regulate the mRNA expression levels of TRPV1 at both 37 and 39°C, and its calcium-mediating function was significantly increased at 39°C, all of which could not be blocked by pretreatment of the neuronal cells with ruthenium red, a general transient receptor potential (TRP) blocker, indicating that the action of 1 was achieved through a non-TRPA1 channel pathway. A thorough comparative analysis of cytotoxic effect of an aqueous cinnamon extract (ACE) from the bark of Cinnamomum zeylanicum L. (Lauraceae) with that of commercially available cinnamaldehyde was performed on various human as well as mouse cell lines and primary cells. The aqueous cinnamon extract (ACE) proved to be more cytotoxic to cancerous cells at concentrations just above 0.16 mg/mL (containing 1.28 μM cinnamaldehyde) around which the commercial cinnamaldehyde (1.6 μM) had no cytotoxic effect. At a critical concentration of 1.28 mg/mL (containing 10.24 μM cinnamaldehyde), ACE treatment resulted in 35-85% growth inhibition of the majority of the cancerous cells, whereas at a similar concentration (10 μM) commercial cinnamaldehyde treatment resulted in 30% growth inhibition of only SK-N-MC cells with no effect on other cell lines. The present study was conducted to isolate the most important bioactive compound from Cinnamomum zeylanicum L. (Lauraceae) bark oil. The plant essential oil was extracted via steam distillation. Cinnamaldehyde was separated using a separating funnel and identified according to Tollen’s test followed by detection on TLC plates in comparison with standard cinnamaldehyde that served as positive control. Moreover, FTIR spectrometry and HPLC analysis were used to confirm the purity and identity of cinnamaldehyde. The isolated material was investigated for its antibacterial activity against six selected pathogenic bacteria. The Gram-positive bacteria were Staphylococcus aureus and Bacillus cereus; Gram-negative bacteria included Escherichia coli, Proteus mirabilis, Klebsiella pneumonia, and Pseudomonas aeruginosa. Cinnamaldehyde at different concentrations (1:1, 1:5, 1:10 and 1:20) was active against all tested bacteria and the highest inhibitory effect was observed against B. cereus (zone of inhibition: 25.3 mm) using the disk diffusion method. The minimal inhibitory concentration (MIC) of cinnamaldehyde was determined using a broth microdilution method in 96-well microtiter plates. MIC values ranged from 31.2 to 125.0 μg/mL. The most promising result was observed against B. cereus, while S. aureus, E. coli, and K. pneumonia ranked next (MIC: 62.5 μg/mL) followed by P. mirabilis and P. aeruginosa with a MIC of 125.0 μg/mL. This manuscript describes the antioxidant activity of essential oil of Cinnamon (Cinnamomum zeylanicum) bark extracted by supercritical fluid extraction (SCFE), hydro distillation and steam distillation. The cinnamon bark essential oil exhibited a wide range of total phenolic contents, total flavonoid contents, reducing power, inhibition of linoleic acid peroxidation and DPPH radical-scavenging activity (IC50). Bioactivity of cinnamon essential oil was assayed against various bacterial strains including Bacillus subtilis, Escherichia coli, Pastrurella multocida and Straphylococcus aureus and fungal strains including Aspergillus niger and Aspergillus flavus. More essential oil yield was obtained using SCFE in comparison to other methods. The oil extracted by SCFE was dominated by cinnamaldehyde, limonene, copaene, naphthalene, heptane, bicyclo[4.2.0]octa-1,3,5-triene and 2-propenal.
