Since 1998, genetically engineered Bt maize varieties expressing the insecticidal Cry1Ab protein (i.e. event MON 810) have been grown in the European Union (EU), mainly in Spain. These varieties confer resistance against the European and Mediterranean corn borer (ECB and MCB), which are the major lepidopteran maize pests in the EU, particularly in Mediterranean areas. However, widespread, repeated and exclusive use of Bt maize is anticipated to increase the risk of Cry1Ab resistance to evolve in corn borer populations. To delay resistance evolution, typically, refuges of non-Bt maize are planted near or adjacent to, or within Bt maize fields. Moreover, changes in Cry1Ab susceptibility in field populations of corn borers and unexpected damage to maize MON 810, due to corn borers, are monitored on an annual basis. After two decades of Bt maize cultivation in Spain, neither resistant corn borer populations nor farmer complaints on unexpected field damage have been reported. However, whether the resistance monitoring strategy followed in Spain, currently based on discriminating concentration bioassays, is sufficiently sensitive to timely detect early warning signs of resistance in the field remains a point of contention. Moreover, the Cry1Ab resistance allele frequency to Bt maize, which has recently been estimated in MCB populations from north-eastern Spain, might exceed that recommended for successful resistance management. To ensure Bt maize durability in Spain, it is key that adequate resistance management approaches, including monitoring of resistance and farmer compliance with refuge requirements, continue to be implemented and are incorporated in integrated pest management schemes. To learn the spatial and temporal distribution of Bt proteins in Bt maize leaves, Bt protein levels of Bt maize leaves and their corresponding non-transgenic lines were detected and analysed by the ELISA method. The results showed that Bt proteins were expressed in the leaves of Bt maize varieties during all periods of their growth, while no such Bt proteins were detected in the leaves of non-transgenic maize varieties. The range of leaf Bt proteins was 1.08∼5.81 µg.g−1 FW, which could effectively protect Bt maizes from the corn borer pest during their growth periods. Furthermore, the younger leaves of Bt maizes had significantly lower levels of Bt proteins than their older leaves. In most periods, Bt protein content of Bt maize leaves under drought stress was lower than that under normal growth conditions, but Bt protein content under nutrition stress was higher than that under normal growth conditions. The influence of transgenic Bacillus thuringiensis (Bt) cotton (BtXincai1) and its corresponding nontransgenic isoline (Xincai1) on the microorganisms, enzyme activity, and nutrient content of rhizosphere soil was studied through experiments in potted plants. The calcareous drab soil samples were collected (0–15 cm deep) from an experimental field in Shanxi Agricultural University (China) in 2005. The pots were categorized in different groups with replicates for each variety (transgenic BtXincai1 and general Xincai1). The rhizosphere soil samples were collected at different growth periods (seedling, bud, flowering, peak boll, boll opening, and harvest). The Bt protein and other microbial properties in the soil samples were determined by using selected methods (material and methods session). The results demonstrated that the concentration of the Bt protein in the rhizosphere soil of BtXincai1 reached a peak at 56.14 ng g−1 during the flowering period. However, the Bt protein would not continuously accumulate in the soil. The rhizosphere soil of BtXincai1 was more suitable for the growth and proliferation of bacteria and fungi but it had no significant impact on the number of actinomycetes. BtXincai1 had some inhibitory effects on alkaline phosphatase activity in the rhizosphere soil, and it might promote dehydrogenase activity during the blooming period. However, it had no significant influence on protease, urease, or sucrase activities. Further, it had no significant impact on the contents of organic matter, total nitrogen, available nitrogen, or potassium in rhizosphere soil. It could significantly decrease the content of available phosphorus during the flowering period. Based on this study, the sensitive reactions of microorganisms and the activities of alkaline phosphatase and dehydrogenase might be considered as three potential indexes for assessing the risk posed by transgenic Bt cotton to soil ecology.
