To gain further insight into the genomic features of border disease virus (BDV), we determined the nearly complete genome sequence of isolate TO/121/04 from an aborted ovine fetus. Its genome contains a single open reading frame (ORF), which comprises 11,681 nucleotides encoding a polyprotein of 3893 amino acids. Phylogenetic analysis of the near full-length genome sequence showed that the BDV isolate differed significantly from all ovine pestiviruses identified so far, thus re-affirming the presence in Italy of this novel genetic group, termed BDV-7.

Border disease virus (BDV) is a (+) single-stranded RNA pestivirus affecting mainly sheep and goats worldwide. Genetic typing of BDV has led to the identification of at least seven major genotypes. This study reports the detection of a BDV strain from a goat in northwestern Italy during routine investigations. Sequence analysis revealed mutations in the 5'-UTR of the virus with implications for BDV molecular diagnostics. Moreover, subsequent phylogenetic analysis based on the combined 5'-UTR and Npro/partial C genes, showed divergence from known BDV genotypes, revealing the detection of a novel pestivirus group, for which we propose the name BDV genotype 8.

Background In 2008, a program to eradicate bovine virus diarrhea (BVD) in cattle in Switzerland was initiated. After targeted elimination of persistently infected animals that represent the main virus reservoir, the absence of BVD is surveilled serologically since 2012. In view of steadily decreasing pestivirus seroprevalence in the cattle population, the susceptibility for (re-) infection by border disease (BD) virus mainly from small ruminants increases. Due to serological cross-reactivity of pestiviruses, serological surveillance of BVD by ELISA does not distinguish between BVD and BD virus as source of infection. Results In this work the cross-serum neutralisation test (SNT) procedure was adapted to the epidemiological situation in Switzerland by the use of three pestiviruses, i.e., strains representing the subgenotype BVDV-1a, BVDV-1h and BDSwiss-a, for adequate differentiation between BVDV and BDV. Thereby the BDV-seroprevalence in seropositive cattle in Switzerland was determined for the first time. Out of 1,555 seropositive blood samples taken from cattle in the frame of the surveillance program, a total of 104 samples (6.7%) reacted with significantly higher titers against BDV than BVDV. These samples originated from 65 farms and encompassed 15 different cantons with the highest BDV-seroprevalence found in Central Switzerland. On the base of epidemiological information collected by questionnaire in case- and control farms, common housing of cattle and sheep was identified as the most significant risk factor for BDV infection in cattle by logistic regression. Conclusion This indicates that pestiviruses from sheep should be considered as a source of infection of domestic cattle and might well impede serological BVD surveillance. The most important causal agent of border disease (BD), border disease virus (BDV), is a Pestivirus in the family Flaviviridae. In attention to the lack of molecular documented data on the prevalence of BDV in Iran, especially in East Azerbaijan Province, northwest of Iran, this research was designed to determine the prevalence of BDV in suspected ewes that have an antecedent of reproductive failure. Eighty-eight suspected ewes in the breed of Ghezel, Mogan, and mix (Ghezel × Mogan) in five various districts of East Azerbaijan Province (Azershahr, Lyqvan, Osku, Sofian, and Varzqan) were selected. Razi bovine kidney (RBK) cell lines were grown, and the buffy coat layer was separated and inoculated to the monolayer cell culture. All cell cultures were inspected for cytopathic effects after 5–7 days. BDV genomes were being checked using nested reverse transcription polymerase chain reaction method. Ten samples (10/88, 11.36 %) were positive for the genome of BDV. The prevalence of BDV infection was found to vary between different districts. The highest percentage of infected sheep was in animals at the 3–4.5-year-old range (15.38 %). There was no statistically significant difference in the prevalence among different age categories. The highest BDV infection rate was observed in full blood ewes in Mogan and Ghezel. Cytopathic effects of the virus in the culture on RBK cell line were not observed at all. This study proved that the infection of BDV is present in sheep in the East Azerbaijan, northwest of Iran, and cause some reproductive failure in the flocks. The survey is the first molecular evidence of the occurrence of the disease in the country.

Besides the important economical losses, pestiviruses may compromise the normal immune response to other pathogens and increase the severity of other infections in sheep. In this study, aborted foetuses (cattle and sheep) in either coastal or inland Black Sea region of Turkey were surveyed for the presence of RNA from pestiviruses (bovine viral diarrhoea virus (BVDV), border disease virus (BDV)). The presence of BVDV RNA was found in 6 of 21 aborted calves (28.57%), although BDV RNA was detected in 14 of 21 aborted lambs (66.66%) by reverse transcriptase polymerase chain reaction. This study also investigates the distribution of viral RNA within the brain, liver and lung of aborted foetuses. The viral RNA positivity rates for the organs varied and were as follows: brain 40.47% and liver and lung 38.09%. The results revealed that pestiviruses are important abort pathogen in the provinces of northern Turkey.

Background The Mekong Basin Disease Surveillance cooperation (MBDS) is one of several sub-regional disease surveillance networks that have emerged in recent years as an approach to transnational cooperation for infectious disease prevention and control. Since 2003 MBDS has pioneered a unique model for local cross-border cooperation. This study examines stakeholders’ perspectives of these MBDS experiences, based on a survey of local managers and semi-structured interviews with MBDS leaders and the central coordinator. Results Fifteen managers from 12 of 20 paired cross-border sites completed a written survey. They all monitor most or all of the 17 diseases agreed upon for MBDS surveillance information sharing. Fourteen agreed or strongly agreed with statements about the core MBDS values of cooperation, mutual trust, and transparency, and their own contributions to national and regional disease control (average score of 4.4 of 5.0). Respondents felt they implemented well to very well activities related to surveillance reporting (average scores 3.4 to 3.9 of 4.0), using computers for their work (3.9/4.0), and using surveillance data for action (3.8/4.0). Respondents reported that they did worst in implementing research (2.1/4.0) and somewhat poorly for local laboratory testing (2.9/4.0) and local coordination with cross-border counterparts (2.9/4.0), although all 15 maintain a list with contact information for these counterparts and many know their counterparts. Implementation of specified activities within their collective regional action plan was uneven across the cross-border sites. Most respondents reported positive lessons learned about local cooperation, information sharing and joint problem solving, based on trusting relationships with their cross-border counterparts. They recommend expansion of cross-border sites within MBDS and consideration of the cross-border cooperation model by other sub-regional networks. Conclusions MBDS has over a decade of experience with its model of local cross-border cooperation in disease surveillance and control. Frontline managers have documented success with this model, strongly support it and recommend its expansion within and beyond the MBDS network. The MBDS cross-border cooperation model is standing the test of time as a solid approach to building and sustaining the public health capabilities needed for disease surveillance and control from the local to national and global levels. Nine pestiviruses isolated from different batches of a contaminated Tunisian sheep pox vaccine and one Tunisian field ovine isolate of border disease virus (BDV) were studied at the antigenic and molecular levels. Seroneutralization tests were carried out on three vaccine isolates, the Tunisian field isolate and representative reference strains of the different pestivirus groups. The antigenic study showed that the Tunisian isolates were closer to the two BDV reference strains than to the Alfort-187 and the NADL reference strains. Comparison of the nucleotide sequences of the 5'-non coding regions of all the Tunisian isolates to those of other pestiviruses have shown that these isolates were distinct from the established pestivirus species. The entire N(pro)-E2 coding sequences of four Tunisian isolates were determined and compared to other pestiviruses. Segregation of these pestiviruses based on the N(pro)-E2 region was identical to that obtained with the 5'UTR sequences. The phylogenetic tree obtained with these sequences showed that the Tunisian isolates formed a separate branch between the BDV and CSFV groups, and consequently a possible new species within the pestivirus genus. However, as indicated by the antigenic study and the host origin of the isolates, the Tunisian isolates were assigned to a novel subgroup within the BDV species.

In a Swedish sheep flock comprising 202 ewes and 13 rams, a pair of twin lambs born in the spring of 1990 demonstrated signs of border disease (BD) and were persistently infected (PI) with border disease virus (BDV). Investigation showed that BDV had been introduced in the preceding tupping period 5-6 months earlier by a bought-in ewe which, on the basis of immunoperoxidase- and polymerase chain reaction techniques, was shown to be PI with BDV. Only 7 of the ewes, all of which had been in close contact with the PI ewe, seroconverted during the subsequent gestation. Apart from the PI twin lambs the losses caused by BDV were restricted to 2 barren ewes. The twin lambs, the PI ewe and lambs from the other 4 ewes that seroconverted were removed from the flock. The flock was thereafter free from an ongoing infection with BDV as shown by the absence of seroconversion. In addition, 5 heifers in late pregnancy most probably seroconverted to bovine virus diarrhoea virus (B VDV) when kept in close contact with the same PI ewe during the winter of 1989-90. When these heifers were reintroduced to the BVDV-free dairy herd from which they originated, their serum antibody titres ranged between 1:250 and 1:1250. Neither these heifers - nor their calves–caused any spread of the infection in the herd, as indicated by the absence of seroconversion in 70 cows. The present investigation shows that in the control of both BDV in sheep and BVDV in cattle, it is important to ensure that the risk of transmission of pestivirus between the 2 species is minimized.

Serum samples from 704 animals from 54 Swedish sheep flocks were analysed by ELISA twice during 1 breeding season for antibodies to Toxoplasma gondii and border disease virus (BDV). An ELISA, originally developed for the detection of antibodies to bovine viral diarrhoea virus (BVDV) in cattle, was assessed on sheep sera and the results were compared with those obtained in a virus neutralization test. The correlation between the 2 assays proved good. Before breeding, 132 (19%) sheep in 42 flocks had antibodies to T. gondii and 7 (1%) sheep in 5 flocks were seropositive to BDV. During the observation period 4 sheep seroconverted to T. gondii and 13 to BDV, giving an incidence rate of 0.7% and 1.9% respectively. No clinical signs due to the infections were observed. In 5 flocks the frequency of barrenness, abortion or stillbirths exceeded 5%, 5% and 8%, respectively, but there was no evidence that this was attributable to the agents studied. The proportion of BDV-positive flocks was significantly higher among flocks that had been in contact with cattle than among those that had not.

Border disease of sheep is a transplacental viral infection of the foetus. In medium and fine fleeced breeds, the most consistent of the pathological changes is permanent primary follicle enlargement, first apparent at 115 days gestation and accompanied by increased fibre size and frequency of fibre medullation. A tendency for fewer secondary follicles to develop in Border disease is probably an indirect consequence of infection.

The sedimentation coefficient and buoyant density of hog cholera, bovine viral diarrhea and Border disease viruses, have been compared with those of representative members of the family togaviridae. It appears that the pestivirus genus is a homogeneous group which is not only antigenically but also structurally, unrelated to the other genera of the togavirus family.

A fetal lamb muscle cell line has been isolated in which two strains of Border disease virus replicate well and induce a clear-cut cytopathic effect, thus providing a sensitive and practical assay system for both the virus and neutralizing antibodies. This system enabled us to study some characteristics of Border disease virus in comparison with two other agents responsible for hog cholera and bovine viral diarrhea. Our results indicate that the last virus and the Border disease agent are indistinguishable in vitro.

Serological examination of 4 Border disease affected flocks of sheep using the neutralization test showed antibody prevalences between 14 and 96 %. Prevalence in yearlings in 3 of the 4 flocks was 37 %, it increased with age to 72 % in 5-year-old sheep. Possible reason for low prevalence (2 %) in yearlings in one of the flocks is discussed.

Congenital myelin dysgenesis in lambs and piglets can be characterised by the analysis of whole spinal cord for extractable lipid, cerebroside (measured as lipid hexose) and total phospholipid contents.

Cryostat sections of lambs with Border disease (BD) were stained with the direct fluorescent antibody technique and examined by fluorescent microscopy. In young lambs fluorescence was observed in almost every organ, demonstrating a generalised infection with BD-virus. Although the number of fluorescing tissues decreased with age, fluorescence was still present in a lamb examined at 22 weeks of age. BD-virus was isolated in tissue culture from six out of seven lambs and was shown to persist for as long as five months.
