1. Production of warm aluminum cluster anions by femtosecond laser ablation

Abstract

We report on the production of warm aluminum cluster anions, Al − (1 ≤ n ≤ 26), after femtosecond laser ablation of an aluminum nitride substrate. Large cluster anions of n ≥ 8 suffer metastable dissociation after their production, which indicates the internal energy of the cluster anions is high enough for the dissociation of an Al atom. We find that the efficiency of metastable dissociation is dependent on the size of cluster anions and the dependence can be rationalized by the dissociation energy of an Al atom from the cluster anions calculated with thermochemical data of the clusters.

Abstract

Time development of Sm+ and Sm ablation plume produced by the femtosecond laser irradiation has been investigated. The two-dimensional spatial profiles of Sm and Sm+ emitted from oxidized and non-oxidized Sm surface were visualized using a planar laser-induced fluorescence method. It was observed that the flow velocity of Sm+ is much faster than that of Sm plume in both surfaces. The plumes from the oxidized Sm surface show higher velocity than that from non-oxidized surface, which is originated by the small electric conductivity at the surface. Expansion property observed for Sm+ and Sm plume in the oxidized Sm surface ablation implies the formation of the Knundsen layer nearby the surface. Meanwhile, continuous emission of Sm indicates the large contribution of heating effect to emission process at the non-oxidized surface. We conclude that the fsLA process strongly depends on the electric property of the ablated surface and the heating effect contributes to the particle emission process on the conductive material surface.

Abstract

We have observed the motion of Sm+ ions as well as Sm atoms produced by femtosecond laser ablation of a solidified samarium solution sample on substrates by using a planar laser-induced fluorescence method. Kinetic energies of both Sm+ ions and Sm atoms increase as the electrical conductivity of the substrate decreases, which suggests the effect of surface charging. The kinetic energy of Sm+ ions is larger than that of Sm atoms for a variety of substrates due to the further electrical acceleration by the surface charge. The knowledge of ion motion will be the key information for the optimization of femtosecond laser simultaneous atomization and ionization of organic and inorganic samples on substrates.

Abstract

By ablating solid C60 with a laser pulse, we observe various processes such as the prompt- and the delayed-ionization of C60, the fragmentation into molecular ions and the formation of cluster ions. We found these processes show distinct dependences on the temporal pulse width, the power and the wavelength of the ablation laser. From the observations, we could confirm efficient coupling of laser energy to C60 through the molecular absorption even with a laser pulse width less than the electron-phonon coupling time of the C60 molecule.

Abstract

FMS-like tyrosine kinase-3 (FLT3) is a new therapeutic target for acute myelocytic leukemia (AML), because FLT3 mutations are the most common genetic alterations in AML and are directly related to leukemogenesis. We studied cytotoxic interactions of a FLT3 inhibitor, PKC412, with eight conventional antileukemic agents (cytarabine, doxorubicin, idarubicin, mitoxantrone, etoposide, 4-hydroperoxy-cyclophosphamide, methotrexate and vincristine) using three leukemia cell lines carrying FLT3 mutations (MOLM13, MOLM14 and MV4-11) and five leukemia cell lines without FLT3 mutations (KOPB-26, THP-1, BALL-1, KG-1 and U937). PKC412 showed synergistic effects with all agents studied except methotrexate for FLT3-mutated cell lines in isobologram analysis. In contrast, PKC412 was rather antagonistic to most drugs, except for 4-hydroperoxy-cyclophosphamide and vincristine, in leukemia cell lines without FLT3 mutations. Cell-cycle analysis revealed that PKC412 induced G1 arrest in leukemia cell lines carrying FLT3 mutations, whereas it arrested cells in G2/M phase in the absence of FLT3 mutations, which may underlie the divergent cytotoxic interactions. These results suggest that the simultaneous administration of PKC412 and other agents except methotrexate is clinically effective against FLT3 mutation-positive leukemias, whereas it would be of little benefit for FLT3 mutation-negative leukemias. Our findings may be of help for the design of PKC412-based combination chemotherapy.

Abstract

Human leukemia-lymphoma (LL) cell lines represent an extremely important resource for research in a variety of fields and disciplines. As the cell lines are used as in vitro model systems in lieu of primary cell material, it is crucial that the cells in the culture flasks faithfully correspond to the purported objects of study. Obviously, proper authentication of cell line derivation and precise characterization are indispensable requirements to use as model systems. A number of studies has shown an unacceptable level of LL cell lines to be false. We present here the results of authenticating a comprehensively large sample (n = 550) of LL cell lines mainly by DNA fingerprinting and cytogenetic evaluation. Surprisingly, near-identical incidences (ca 15%) of false cell lines were observed among cell lines obtained directly from original investigators (59/395: 14.9%) and from secondary sources (23/155: 14.8%) implying that most cross-contamination is perpetrated by originators, presumably during establishment. By comparing our data with those published, we were further able to subclassify the false cell lines as (1) virtual: cross-contaminated with and unretrievably overgrown by other cell lines during initiation, never enjoying independent existence; (2) misidentified: cross-contaminated subsequent to establishment so that an original prototype may still exist; or (3) misclassified: unwittingly established from an unintended (often normal) cell type. Prolific classic leukemia cell lines were found to account for the majority of cross-contaminations, eg CCRF-CEM, HL-60, JURKAT, K-562 and U-937. We discuss the impact of cross-contaminations on scientific research, the reluctance of scientists to address the problem, and consider possible solutions. These findings provide a rationale for mandating the procurement of reputably sourced LL cell lines and their regular authentication thereafter.

Abstract

We present the establishment of a natural killer (NK) leukemia cell line, designated KHYG-1, from the blood of a patient with aggressive NK leukemia, which both possessed the same p53 point mutation. The immunophenotype of the primary leukemia cells was CD2+, surface CD3-, cytoplasmic CD3epsilon+, CD7+, CD8alphaalpha+, CD16+, CD56+, CD57+ and HLA-DR+. A new cell line (KHYG-1) was established by culturing peripheral leukemia cells with 100 units of recombinant interleukin (IL)-2. The KHYG-1 cells showed LGL morphology with a large nucleus, coarse chromatin, conspicuous nucleoli, and abundant basophilic cytoplasm with many azurophilic granules. The immunophenotype of KHYG-1 cells was CD1-, CD2+, surface CD3-, cytoplasmic CD3epsilon+, CD7+, CD8alphaalpha+, CD16-, CD25-, CD33+, CD34-, CD56+, CD57-, CD122+, CD132+, and TdT-. Southern blot analysis of these cells revealed a normal germline configuration for the beta, delta, and gamma chains of the T cell receptor and the immunoglobulin heavy-chain genes. Moreover, the KHYG-1 cells displayed NK cell activity and IL-2-dependent proliferation in vitro, suggesting that they are of NK cell origin. Epstein-Barr virus (EBV) DNA was not detected in KHYG-1 cells by Southern blot analysis with a terminal repeat probe from an EBV genome. A point mutation in exon 7 of the p53 gene was detected in the KHYG-1 cells by PCR/SSCP analysis, and direct sequencing revealed the conversion of C to T at nucleotide 877 in codon 248. The primary leukemia cells also carried the same point mutation. Although the precise role of the p53 point mutation in leukemogenesis remains to be clarified, the establishment of an NK leukemia cell line with a p53 point mutation could be valuable in the study of leukemogenesis.

Abstract

We describe two new human leukemia cell lines, MOLM-13 and MOLM-14, established from the peripheral blood of a patient at relapse of acute monocytic leukemia, FAB M5a, which had evolved from myelodysplastic syndrome (MDS). Both cell lines express monocyte-specific esterase (MSE) and MLL-AF9 fusion mRNA. Gene fusion is associated with a minute chromosomal insertion, ins(11;9)(q23;p22p23). MOLM-13 and MOLM-14 are the first cell lines with, and represent the third reported case of, MLL gene rearrangement arising via chromosomal insertion. Both cell lines carry trisomy 8 which was also present during the MDS phase, as well as the most frequent trisomies associated with t(9;11), ie, +6, +13, +19 variously present in different subclones. Despite having these features in common, differences in antigen expression were noted between the two cell lines: that of MOLM-13 being CD34+, CD13-, CD14-, CD15+, CD33+; whereas MOLM-14 was CD4+, CD13+, CD14+, CD15+, CD33+. Differentiation to macrophage-like morphology could be induced in both cell lines after stimulation with INF-gamma alone, or in combination with TNF-alpha, which treatment also induced or upregulated, expression of certain myelomonocyte-associated antigens, including CD13, CD14, CD15, CD64, CD65 and CD87. Together, these data confirm that both cell lines are likely to be novel in vitro models for studying monocytic differentiation and leukemogenesis.

Abstract

In order to clarify neutronic performance of a pulsed cold-neutron source, scattering properties of some typical hydrogenous moderators are studied. Namely, cross-section models for liquid ortho- and para-H2 at temperatures between 14 and 20.4 K and solid and liquid CH4 at temperatures down to 20 K are developed. A set of group constants for liquid H2 is also generated: 80 groups between 0.1 μeV and 10 eV in a P3 approximation. The major results are as follows. (a) Cold-neutron production due to a para-to-ortho transition and up-scattering of lower-energy neutrons by ortho-H2. (b) Neutron slowing-down by rotational excitation of a CH4 molecule, especially at 20 K. And (c) an optimum condition of a liquid-H2 source model for cold-neutron production.

Abstract

T-cell prolymphocytic leukemia (T-PLL) is a postthymic T-cell neoplasm with a characteristic morphology and heterogeneous immunophenotype. Most cases of T-PLL express membrane T-cell receptors (TCRs) of the alphabeta phenotype. We experienced a 30-year-old man suffering from TCRgammadelta T-cell leukemia with morphology compatible to T-PLL with a postthymic phenotype. He was admitted with skin eruption and pancytopenia. Peripheral blood and bone marrow were occupied with medium-sized lymphocytes, which had moderately condensed chromatin with a single nucleolus and sparse, nongranular basophilic cytoplasm. The immunophenotype was CD1a-, CD2-, CD3+, CD4-, CD5+, CD7+, CD8-, and terminal deoxynucleotidyl transferase negative. Hepatosplenomegaly was absent. He was diagnosed as having T-PLL and was treated with combination chemotherapy. Six months later the leukemic cell became chemoresistant. Although the patient showed transient improvement in response to pentostatin, he died 13 months after the diagnosis. To our knowledge, this is the first case of T-PLL with a TCRgammadelta phenotype.

Abstract

An in situ hybridization experiment showed that the gene transcripts of P-450IID subfamily were uniformly distributed in bovine liver cells and at low levels in bovine adrenal cortex and medulla. There was no clear localization of transcripts of P-450IID genes in three adrenocortical zones, zona glomerulosa, zona fasciculata and zona reticularis. The intensity of signal obtained in the adrenal cortex and medulla showed approximately 2.5:1 ratio. The presence of gene transcripts of P-450IID subfamily in adrenocortex was confirmed by amplifying cDNA using PCR and hybridizing the PCR products with P-450IID cDNA.

The nuclear electric quadrupole moment of41Sc(Iπ=7/2−) was experimentally determined by use of the NMR detection in which the asymmetric β-ray distribution from spin polarized nuclei was monitored. The magnetic interaction of the state with high external magnetic field and the nuclear quadrupole interaction with the electric field gradient obtained in TiO2 crystal were studied. The field gradient seen to the implanted41Sc was measured independently by the high field NMR detection on the stable isotope45Sc located in the equivalent41Sc site with. |Q(41Sc;Iπ=7/2−)|=(0.120±0.006) b was determined.

The lifetime and magnetic moment of43Ti (Iπ-7/2−, T1/2=0.50 sec), one of the mirror beta-radioactive nuclei in the f7/2-shell, have been measured with a fragment separator built at the Bevalac of the LBL. By means of the beta-NMR method on43Ti, a preliminary value of the magnetic moment of43Ti was determined to be μ=(0.70±0.07) μπ.

We read the recent article by Erro et al. [1] ‘‘The eye of the tiger sign in pure akinesia with gait freezing’’ with great interest. The eye of the tiger sign is a neuroradiologic sign that reflects accumulated iron. This sign has been associated with neurodegeneration with brain iron accumulation type 1 (NBIA1), and mutations in the gene encoding pantothenate kinase 2 (PANK2) are known to be involved [2]. We recently experienced a patient with atypical parkinsonism who showed the eye of the tiger sign upon T2-weighted magnetic resonance imaging (MRI) of the brain. However, T2*-weighted MRI showed no abnormal lesions, and the eye of the tiger sign in our case turned out to be false. To our knowledge, our case is the first case of pseudo eye of the tiger sign and we need to consider the significance of T2*-weighted MRI findings in judging the validity of the eye of the tiger sign. A 76-year-old woman was admitted to our hospital because of progressive gait disturbances for more than 2 years. On neurological examination, she walked with short shuffling steps with stooped posture and presented difficulty while turning. She also showed postural instability and bradykinesia. In addition to her parkinsonian symptoms, she presented with urinary incontinence, severe constipation and gastric dysmotility. She did not show resting tremor, rigidity, dysphagia, dysarthria, ataxia, sensory disturbance, gaze palsy or orthostatic hypotension. In addition, her muscle strength and deep tendon reflex were normal. The results of a Mini-Mental State Examination (MMSE) revealed intact cognitive function. Although she was treated with L-dopa (up to 600 mg/day), her neurological symptoms persisted. T2-weighted MRI of the brain revealed high signal intensity in the globus pallidus surrounded by a ring of low signal intensity, the so-called eye of the tiger sign (Fig. 1a, b). However, she had no family history of neurological disorders and the results of hematological examinations, including iron, ferritin and ceruloplasmin, were all normal. Moreover, T2*-weighted MRI showed no accumulated iron in the affected regions (Fig. 1c). Therefore, we diagnosed the patient with atypical parkinsonism with pure akinesia, but not NBIA1. The eye of the tiger sign has been infrequently reported in neurodegenerative diseases other than NBIA1, including multiple system atrophy (MSA), pure akinesia and corticobasal degeneration (CBD) [1, 3, 4]. These reports suggest that the eye of the tiger sign is not always specific for NBIA1. On the other hand, the authors of these reports did not evaluate the deposition of iron as we did in our case. Therefore, whether neurodegenerative diseases other than NBIA1 manifest the eye of the tiger sign is not obvious, and the signs in previous reports may have been pseudo signs, as in our case. Although the cause of the pseudo eye of the tiger sign is unknown, it may reflect degeneration of the globus pallidus. Regardless, it is important to consider the existence of the pseudo eye of the tiger sign and confirm the deposition of iron.

Abstract

A polymer foil of polyethylene-terephthalate was successfully used for the tilted foil to create nuclear polarization of beta-emitters. The foil was carefully prepared to produce atomic flatness on the surfaces. The resultant smoothness was confirmed with beta-NMR measurements on12B and found to be as good as those of carbon foils used previously in the measurements.

Abstract

The peak broadening and peak positions of Bragg reflections in a single-phase decagonal Al70Ni10Co20 quasicrystal has been studied by the synchrotron X-ray diffraction technique at SPring-8. Full-widths at half-maximum (FWHM) of the Bragg reflections along the longitudinal direction (L) and transverse direction (TI) which is perpendicular to L-direction in an aperiodic plane have linear Q⊥, dependence. This means that the Al70Ni10Co20 decagonal phase has random phason strains. On the other hand, another transverse direction (T2), which is perpendicular to the aperiodic plane, have linear Q, dependence. So the single crystal has some distortion of mosaics to the periodic layer stacking. Notable peak shifts from ideal Bragg peak positions were not observed.

The current-voltageI-V characteristics of amorphous W monolayers and W/Si multilayers have been measured at different temperatures belowTc in the absence of magnetic field. For the W monolayer, we observed the power lawV ∝ Iα dependence and a sudden, increase of α from 1 to 3 at the Kosterlitz-Thouless transition temperature. However, in the W/Si multilayer, theI-V characteristics obey the modified power lawV ∝I(I−Ic)α−1 with a non-vanishing critical currentIc. This result is ascribed to the effect of interlayer coupling between the W layers on the thermal excitations of vortex-antivortex pairs.

Abstract

The influence of a duplex microstructure on fatigue crack growth in the near threshold region has been studied for AISI types 1018, 1045, and 10B35, as well as 2.25 Cr-1 Mo steels. For a duplex microstructure which consists of a continuous martensitic phase encapsulating ferrite, an increase in threshold level and yield strength in both the AISI 1018 and 2.25 Cr-1 Mo steels was observed. However, with increase in carbon content and consequent decrease in the volume of ferrite, the threshold levels were not as significantly affected, although the yield strengths were higher.
