The nasal cavities of opossums prior to and shortly after birth were examined by scanning electron microscopy. Numerous morphologically mature olfactory receptor neurons are observed in the dorso-rostral-most extent of the olfactory epithelium positioned adjacent to the opening of the nares in all prenatal stages and newborn animals examined. The remainder of the olfactory epithelium, occupying a more dorso-caudal position within the nasal cavity, is undifferentiated, and lacks morphologically mature receptor neurons. A short transition zone of stratified squamous epithelium lies between the epithelium lining the nares and olfactory epithelium. It forms an abrupt junction with the latter. The remainder of the nasal cavity in this group of animals is lined by a non-ciliated pseudostratified type (undifferentiated respiratory) of epithelium. By the end of the second postnatal week the morphologically mature olfactory epithelium is no longer observed in the vestibular area of the nasal cavity, which is lined by stratified squamous epithelium at this time. Mature receptor neurons are now observed within the olfactory epithelium lining the roof of the nasal cavity and covering the turbinates. The greater part of the nasal cavity is lined by a ciliated respiratory epithelium. It is proposed that the precocious differentiation of mature olfactory receptor neurons within the rostral-most extent of the olfactory epithelium just prior to birth is important in guiding the newborn young to the pouch.

In the North American opossum, heat-stable specific binding sites for E. coli enterotoxin are observed (i) in epithelial cells lining the small intestine, colon, gall bladder, cystic duct, common bile duct and trachea, and (ii) in epithelial cells forming the duodenal (Brunner's) glands, liver, kidneys (metanephros, mesonephros) and testis, as demonstrated by autoradiography. Enterotoxin-specific binding sites in the intestinal tract are only found in intestinal epithelial cells with the highest concentration in the microvillus border. Enterotoxin-specific binding sites also occur in epithelial cells comprising the secretory tubules and ducts of the duodenal glands. In the kidneys (metanephros and mesonephros), enterotoxin-specific binding sites are confirmed primarily to the proximal tubules, whereas in the testis they are localized in seminiferous tubules. In the liver, enterotoxin-specific binding sites are confined primarily to hepatocytes. E. coli enterotoxin caused a 7-fold increase of cGMP in the liver and a 30-fold increase in the duodenal glands. The liver responded in about half of the animals studied, whereas the duodenal glands gave a consistent response in each case. Likewise, the duodenal glands consistently showed strong labelling for 125I-enterotoxin, whereas receptor labelling of hepatocytes was inconsistent in nearly half the incubations and corresponds to the observed cGMP measurements.

The effect on parietal cells of glucagon given prior to pentagastrin is unknown. Fifteen healthy volunteers were studied during constant intravenous infusion of pentagastrin (2 micrograms/kg body weight/hr) and during pentagastrin infusion initiated 20 min after intravenous injection of 2 mg glucagon. Three types of studies were performed: Gastric mucosal biopsies were obtained with a Quinton instrument. Electron micrographs of 320 parietal cells were analyzed by the Loud quantitative method, and intragastric pH (pH probe), or gastric potential difference (PD) were recorded continuously. Pentagastrin infusion produced a significant increase in canalicular and simultaneous reduction of tubulovesicular membrane area of parietal cells. Glucagon pretreatment did not inhibit canalicular and tubulovesicular membrane reaction to pentagastrin; unexpectedly this reaction was significantly greater when compared to that after pentagastrin alone. Initiation of pentagastrin infusion in subjects pretreated with glucagon produced a greater absolute value drop in gastric PD (23 +/- 2 mV) and an earlier drop in intragastric pH (3 min) than in subjects receiving pentagastrin infusion alone (13 +/- 1 mV and 10 min, respectively). In conclusion, at the doses studied, glucagon pretreatment increases parietal cell canalicular reaction to pentagastrin.

Biopsies from the fundic mucosa of healthy volunteers were examined by scanning electron microscopy following blunt dissection and freeze fracture. The mucosal surface exhibited a cobblestone appearance. With increased magnification, microvilli could be demonstrated on the luminal surface of individual surface cells. Blunt dissection of the gastric mucosa revealed tubular gastric pits descending from the surface until they opened into branched tubular gastric glands. The gastric glands are irregular in outline due to the knoblike projections of individual parietal cells. The internal structure of the component cells also was examined with the scanning electron microscope following freeze fracture. Mucous granules were observed in the apical portion of surface epithelial cells. Lumina of parietal cell canaliculi were found to be continuous with the lumen of the gastric glands. Pepsinogen granules could be seen throughout the cytoplasm of chief cells. The blunt dissection-freeze fracture technique utilizing the scanning electron microscope allows, for the first time, a three-dimensional view of human gastric mucosa, including the gastric pits and glands as well as some of the internal architecture of component cells.

The relative frequencies of endocrine cells and peptidergic nerve elements in the proximal small intestine of the adult platypus were studied by immunohistochemistry. Six kinds of endocrine cells - serotonin (5-HT)-, somatostatin-, gastrin-, motilin-, cholecystokinin (CCK)- and bovine pancreatic polypeptide (BPP)-immunoreactive cells - were identified in this study. These endocrine cells were found most frequently in the intestinal glands, in moderate numbers in the tubular ducts and were infrequent in the surface folds. 5-HT-immunoreactive cells were most numerous, somatostatin-, gastrin-, motilin- and BPP-immunoreactive cells were moderately numerous, whereas CCK-immunoreactive cells were rare. Five kinds of neuropeptides: substance P, vasoactive intestinal polypeptide (VIP), gastrin releasing peptide (GRP), somatostatin and leu-enkephalin, were detected in the intramural nerve elements. Substance P-, VIP- and GRP-immunoreactive nerve fibers were found most frequently in the lamina propria mucosae of the surface folds. The relationships between the possible functions of the peptides and amine detected in this study as well as the characteristic structure of the digestive tract of the adult platypus are discussed.

Cardiac muscle cells in the right ventricle of the postnatal opposum were studied ultrastructurally, with particular attention to the developmental stage of T-tubule formation. Animals ranging from 5.5 cm (day 29 of postnatal life) to 41 cm (young adult) in body length were used. Typical T-tubules were first recognized in a few myocardial cells of the 7.5 cm (day 43) opossum. T-tubules increased in number as cardiac muscle cells continued to differentiate until 22 cm in body length (about 105 days after birth). At this stage of development most of the T-tubules were completely established. The general mode of differentiation and development of cardiac muscle cells appeared essentially the same as in other mammals.

This study reports the presence of caveolated cells in the duodenal glands of the white-tailed deer. Caveolated cells have not been observed previously in the duodenal glands of other species studied to date.
