Cancer cells frequently evade apoptosis during tumorigenesis by acquiring mutations in apoptotic regulators. Chronic activation of the PI 3-kinase-Akt pathway through loss of the tumor suppressor PTEN is one mechanism by which these cells can gain increased protection against apoptosis. We report here that REDD1 (RTP801) can act as a transcriptional downstream target of PI 3-kinase signaling in human prostate cancer cells (PC-3). REDD1 expression is markedly reduced in PC-3 cells treated with LY294002 (LY) or Rapamycin and strongly induced under hypoxic conditions in a hypoxia-inducible factor-1 (HIF-1)-dependent manner. Loss of function studies employing antisense molecules or RNA interference indicate that REDD1 is essential for invasive growth of prostate cancer cells in vitro and in vivo. Reduced REDD1 levels can sensitize cells towards apoptosis, whereas elevated levels of REDD1 induced by hypoxia or overexpression desensitize cells to apoptotic stimuli. Taken together our data designate REDD1 as a novel target for therapeutic intervention in prostate cancer.

Twenty years ago a theoretical analysis showed that electron scattering by high-Z one-electron atoms might lead to interference of exchange and spin-orbit interactions at low electron energies, observable as a cross-section asymmetry if unpolarized electrons are scattered by polarized cesium atoms. By using a highly polarized cesium atomic beam, we studied exchange and spin-orbit effects at different electron energies, starting at 20 eV and going down. We observed the first distinctly non-zero interference asymmetry at 7 eV: Over the angular range of 35 to 145 degrees, it varies between +0.02 and -0.02 and goes through zero near 110 degrees being negative at larger angles.

We present a variational approach for treating the Hubbard Hamiltonian in one, two and three dimensions. It is based on 2M-fermion wavefunctions which are allowed to form correlated spin-singlet pairs. Expressions for the ground state energy and correlation functions are derived in terms of general pair coefficient functions. The presented approach offers a convenient starting point for improved variational treatments that allow to include different specific types of pair correlations. We present first applications to the attractive and to the extended Hubbard model using a very simple ansatz for the pair coefficient functions. The ground state energy, chemical potential, order parameter, momentum distribution as well as spin-spin and density-density correlation functions follow from a system of coupled nonlinear equations that has to be solved selfconsistently. All quantities are given for arbitrary band-filling in one, two and three dimensions. Our results are compared with those of other approximations and for the one-dimensional case with the exact results of Krivnov and Ovchinnikov.

We have set up an atomic beam of cesium for the study of spin-dependent electron-cesium scattering. The beam is produced by an effusive oven with continuous recirculation of the condensed metal. The beam is optically pumped by circularly polarized light from two laser diodes tuned to the 6(2)S1/2(F = 3) --> 6(2)P3/2(F' = 4) and 6(2)S1/2(F = 4) --> 6(2)P3/2(F' = 5) transitions, respectively. Nearly all atoms are transferred into the F = 4, m(F) = +4 or m(F) = -4 Zeeman level of the ground state, depending on the sense of circular polarization of the pumping light. The population distribution in the optically pumped beam is analyzed in terms of the m(J) = - 1/2 and m(J) = + 1/2 components with a Stern-Gerlach magnet. We find the atomic polarization to be very close to unity at a density of 8 x 10(8) atoms/cm3 in the scattering center. The polarization decreases slightly with increasing density of the cesium beam due to radiation trapping. A spin flipper serves as a means of polarization reversal, introducing no systematic errors in the spin asymmetry measurements. Lock-in technique is used to stabilize the atomic beam polarization by detecting fluorescence light signals.
