Warmer climatic conditions have been associated with numerous hydrologic changes that may impact riverbank erosion in cold regions, but the net effect is not well understood. We used regression and correlation analyses to examine the relationships among subarctic riverbank erosion and seasonal hydrology, the impact of climate change and variability, and the societal implications. Geomorphic change (loss and gain of vegetated land) was mapped along several river reaches in the Yukon River Basin, Alaska, throughout 1984 and 2017 using Landsat satellite imagery. Annual erosion rates were estimated from these spatial data. At most study sites, erosion rates (km2/year) were either positively correlated (r = 0.68–0.84, p = 0.0085–0.061) with monthly mean discharge within the cold season or inversely correlated (r = − 0.74 – −0.62, p

Amplified climate change in the Arctic has altered interactions between rural communities and local wild resources. Shifting interactions warrant analysis because they can influence cultural practices and food security of northern societies. We collaborated with four indigenous communities in Alaska and conducted semi-directed interviews with 71 experienced harvesters to identify local perceptions of climate-driven trends in the environment, and describe the effects of those trends on the availability (i.e., abundance, distribution, accessibility) of subsistence resources. We then linked local perceptions with scientific climate projections to forecast how availability of subsistence resources may change in the future. Hunters identified 47 important relationships between climate-driven changes in the environment and availability of subsistence resources. Of those relationships, 60, 28, and 13 % focused on changes in harvester access, resource distribution, and resource abundance, respectively. Our forecast model indicated a net reduction in the availability of subsistence resources over the next 30 years. The reduction was caused primarily by climate-related challenges in access, rather than changes in abundance or distribution of resources. Our study demonstrates how giving insufficient attention to harvester access may produce misleading conclusions when assessing the impacts of climate change on future subsistence opportunities.

We synthesized an expert review of climate change implications for hydroecological and terrestrial ecological systems in the northern coastal temperate rainforest of North America. Our synthesis is based on an analysis of projected temperature, precipitation, and snowfall stratified by eight biogeoclimatic provinces and three vegetation zones. Five IPCC CMIP5 global climate models (GCMs) and two representative concentration pathways (RCPs) are the basis for projections of mean annual temperature increasing from a current average (1961–1990) of 3.2 °C to 4.9–6.9 °C (5 GCM range; RCP4.5 scenario) or 6.4–8.7 °C (RCP8.5), mean annual precipitation increasing from 3130 mm to 3210–3400 mm (3–9 % increase) or 3320–3690 mm (6–18 % increase), and total precipitation as snow decreasing from 1200 mm to 940–720 mm (22–40 % decrease) or 720–500 mm (40–58 % decrease) by the 2080s (2071–2100; 30-year normal period). These projected changes are anticipated to result in a cascade of ecosystem-level effects including: increased frequency of flooding and rain-on-snow events; an elevated snowline and reduced snowpack; changes in the timing and magnitude of stream flow, freshwater thermal regimes, and riverine nutrient exports; shrinking alpine habitats; altitudinal and latitudinal expansion of lowland and subalpine forest types; shifts in suitable habitat boundaries for vegetation and wildlife communities; adverse effects on species with rare ecological niches or limited dispersibility; and shifts in anadromous salmon distribution and productivity. Our collaborative synthesis of potential impacts highlights the coupling of social and ecological systems that characterize the region as well as a number of major information gaps to help guide assessments of future conditions and adaptive capacity.

The spatial extent of Sitka black-tailed deer (Odocoileus hemionus sitkensis) populations below the regional scale is relatively unknown, as is dispersal between populations. Here, we use noninvasive samples to genotype 221 Sitka black-tailed deer in three watersheds on Prince of Wales Island, Alaska, separated by a maximum of 44 km, using traditional and spatial genetic approaches. We find that despite geographic proximity, multiple lines of evidence suggest fine-scale genetic structure among the three study sites. The 2 most geographically distant watersheds differed significantly in genetic composition, suggesting an isolation-by-distance pattern. Within study sites, deer exhibited spatial genetic structure within a radius of 1,000 m. Based on a reduced sample of known-sex individuals, females exhibited positive spatial genetic structure within a radius of 500 m but males showed no structure. Moreover, females were more likely to be related to their 5 nearest female neighbors, regardless of distance, than were males. Evidence indicates dispersal by both sexes although it may be more common, or dispersal distances are greater, in males. Nonetheless, analysis of assignment indices and comparison of sex-specific correlograms found no evidence of sex-biased dispersal between watersheds. Patterns of spatial relatedness and connectivity suggest limited dispersal among Sitka black-tailed deer, creating genetic structure on a fine spatial scale, perhaps as small as the watershed.

We tested a protocol for extracting DNA from fecal pellets from Sitka black-tailed deer (Odocoileus hemionus sitkensis) and evaluated genotyping performance of previously developed microsatellite markers as well as a suite of new markers designed specifically for this study. We screened 30 microsatellites, and identified 7 (23%) loci including 4 new markers, that fit well into a single multiplex and consistently genotyped deer with low error rates. DNA was extracted from 2,408 fecal-pellet samples. Of those, 1,240 (52%) were genotyped successfully at all 7 markers allowing identification of 634 genetically unique deer. Using DNA from fecal pellets collected in the field was an effective technique for identifying and distinguishing among deer.

Non-invasive wildlife research using DNA from feces has become increasingly popular. Recent studies have attempted to solve problems associated with recovering DNA from feces by investigating the influence of factors such as season, diet, collection method, preservation method, extraction protocol, and time. To our knowledge, studies of this nature have not addressed DNA degradation over time in wet environments, and have not been performed on fecal pellets of ungulates. Therefore, our objective was to determine the length of time a fecal pellet from a Sitka black-tailed deer (Odocoileus hemionus sitkensis) could remain in the field in a temperate rainforest environment before the DNA became too degraded for individual iden- tification. Pellets were extracted from the rectum of recently killed deer and placed in an environment protected from rainfall and in an environment exposed to rainfall. Pellets from each treatment group were sampled at intervals of 2, 7, 14, 21, and 28 days after deer harvest. DNA was extracted from sampled pellets and individual samples were geno- typed using microsatellite markers. Amplification failure and errors (dropout and false alleles) were recorded to determine extent of DNA degradation. Eighty percent of samples in the protected environment and 22% of samples in the exposed environment were successfully genotyped during the 28-day experiment. With no samples being suc- cessfully genotyped in the exposed environment after 7 days, our study showed that rainfall significantly increases degradation rates of DNA from ungulate pellets.

We evaluated PCR primer sets to determine the most effective technique for identifying sex of northern ungulates. We sought markers that required only a single pair of primers to amplify both X- and Y-linked alleles; that amplified X- and Y-linked products that were easily distinguishable using agarose gel electrophoresis; and that produced short amplicons amenable to amplification using DNA of poor quality and low quantity, as is often found in non-invasively collected samples such as feces. Primer pairs KY1/KY2 and SE47/SE48, which amplify X- and Y-specific alleles of the amelogenin gene, met our criteria and were tested for moose (Alces alces), mountain goat (Oreamnos americanus), Sitka black-tailed deer (Odocoileus hemionus sitkensis), and caribou (Rangifer tarandus). KY primers amplified shorter PCR products than did SE primers; moreover, SE primers inconsistently amplified certain Y-chromosome products, creating potential for misidentification of sex. DNA fragments amplified using KY primers were sequenced for each species, allowing us to characterize a 45-bp deletion for Y-linked alleles (136-bp product) relative to X-linked alleles (181-bp product) in all species and a 9-bp deletion in the X-linked allele of moose relative to other species. This is the first sex-determination technique using PCR reported for several ungulate species of Alaska. Although other protocols exist for cervids and bovids, this is the first report of markers meeting the aforementioned criteria for Odocoileus, the most abundant and intensively managed genus of large mammals in North America.
