Cadmium (Cd) pollution in paddy soils creates challenges in rice grain production, thereby threatening food security. The effectiveness of different base-tillering-panicle urea application ratios and the combined basal application of urea and Chinese milk vetch (CMV, Astragalus sinicus L.) in minimizing Cd accumulation in rice grains was explored in a Cd-contaminated acidic soil via a field experiment. The results indicated that under similar nitrogen (N) application rates, an appropriate amount of urea applied at the panicle stage or the combined basal application of urea and CMV decreased Cd absorption by rice roots and its accumulation in rice grains, as compared with that of conventional N application (control). Ubiquitination, a key and extensive posttranslational modification of proteins, has profound effects on a variety of physiological and pathological processes. The inherent complexity of ubiquitin conjugates makes it highly challenging to study the functional and structural mechanisms of ubiquitination. To address these challenges, accesses to sufficient poly-ubiquitin chains or ubiquitinated proteins are urgently needed. Over the last decade, synthetic protein chemists have developed several novel peptide ligation methods for the preparation of ubiquitin conjugates with precise control over the atomic structure. In this review, we summarize the recent breakthroughs and potential challenges in the chemical synthesis and semi-synthesis of ubiquitin conjugates with respect to the preparation of poly-ubiquitin-based proteins and ubiquitin-based probes.

Cr2O3@ZnO hetero-junction hierarchical nanostructures were designed to be enhanced xylene sensing material, and thereinto, flower-like ZnO hierarchical nanostructures were synthesized via a solution-based method, and then Cr2O3 particles were developed on the surface of ZnO petals via a solvothermal method. From the results of XRD patterns, SEM and TEM images, it can be observed that ZnO has a high-quallity crystallinity and Cr2O3 particles scatter uniformly on the suruface of ZnO. The products with different ratios of Cr2O3 were used to fabricate gas sensors, and the result indicates that the hetero-junction structures prompt the response to xylene, and the reason may be attributed to the decrease of main carriers concentration caused by the p-n junction between ZnO(n-type semiconductor) and Cr2O3(p-type semiconductor), as well as the catalytic oxidation effect on methyl groups of the xylene by Cr2O3.

The construction of mirror-image biological systems may open the next frontier for biomedical technology development and discovery. Here we have designed and chemically synthesized a mutant version of the thermostable Sulfolobus solfataricus P2 DNA polymerase IV (Dpo4) consisting of d-amino acids. With a total peptide length of 358 amino acid residues, it is the largest chemically synthesized d-amino acid protein reported to date. We show that the d-polymerase is able to amplify a 120-bp l-DNA sequence coding for the Escherichia coli 5S ribosomal RNA gene rrfB by mirror-image polymerase chain reaction, and that both the natural and mirror-image systems operate with strict chiral specificity. The development of efficient miPCR systems may lead to many practical applications, such as mirror-image systematic evolution of ligands by exponential enrichment for the selection of therapeutically promising nuclease-resistant l-nucleic acid aptamers.

Cyclotides constitute a fascinating family of circular proteins containing ca. 30 amino acid residues. They have a unique cyclic cysteine knot topology and exhibit remarkable thermal, chemical and enzymatic stabilities. These characteristics enable them to have a range of biological activities and promising pharmaceutical and agricultural applications. Here, we present a practical strategy for the chemical synthesis of cyclotides through the intramolecular ligation of fully unprotected peptide O-esters. This strategy involves the mild Fmoc solid-phase peptide synthesis of the peptide O-ester backbone, the head-to-tail cyclization of the cyclotide backbone by native chemical ligation, and the oxidative refolding to yield the natural knot protein. The simplicity and high efficiency of the strategy can be employed in the synthesis of artificial cyclotides for pharmaceutical applications.

Chemokine-guided lymphocyte positioning in tissues is crucial for normal operation of the immune system. Direct, real-time manipulation and measurement of single-cell responses to chemokines is highly desired for investigating the cell biology of lymphocyte migration in vivo. Here we report the development of the first two-photon-activatable chemokine CCL5 through efficient one-pot total chemical synthesis in milligram scale. By spatiotemporally controlled photoactivation, we show at the single-cell level that T cells perceive the directional cue without relying on PI3K activities, which are nonetheless required for persistent migration over an extended period of time. By intravital imaging, we demonstrate artificial T-cell positioning in cutaneous tissues and lymph nodes. This work establishes a general strategy to develop high-quality photo-activatable protein agents through tailor-designed caging of multiple residues and highlights the potential of photo-activatable chemokines for understanding and potential therapeutic manipulation of cell positioning and position-controlled cell behaviours in vivo., The precise spatiotemporal control of chemokine exposure would be an advantageous tool for immune cell research. Here, Chen et al. develop a two-photon-activatable chemokine CCL5 and use it to direct lymphocyte migration in vivo and to show that PI3-kinase is not required to sense a gradient in vitro.

The aim of this study is to shed light on the hydrogeochemical characteristics of karst underground waters at shallow depth in Guiyang City, Guizhou Province with an emphasis on the geochemistry of major elements. Guiyang City bears abundant underground waters and it is also an important representative of the karst areas throughout the world. Ca2+ and Mg2+ are the dominant cations, accounting for 81% – 99.7% of the total, and HCO3- and SO42- are the dominant anions. Weathering of limestones and dolostones is the most important factor controlling the hydrogeochemistry of underground waters, and weathering of sulfate and evaporite rocks is less important. Moreover, the precipitation and human activities also have a definite influence on the hydrogeochemistry of underground waters in the region studied.

For the product of two population means, the problem of constructing a fixed-width confidence interval with preassigned coverage probability is considered. It is shown that the optimal sample sizes which minimize the total sample size and at the same time guarantee a fixed-width confidence interval of desired coverage depend on the unknown parameters. In order to overcome this, a fully sequential procedure consisting of a sampling scheme and a stopping rule are proposed. It is then shown that the sequential confidence interval is asymptotically consistent and the stopping rule is asymptotically efficient, as the width goes to zero. Furthermore, a second order result for the difference between the expected stopping time and the (total) optimal fixed sample size is established. The theoretical results are supported by appropriate simulations.
