1. Market Analysis of Food and Feed Products for Detection of Horse DNA by a TaqMan Real-Time PCR

Abstract

Food-labelling regulations require that meat species in food and feed are accurately declared to the consumer. Traceability systems based on species identification through DNA analysis are potent tools for the supervision of food adulteration. In this study, a TaqMan real-time polymerase chain reaction (PCR) assay targeting a short mitochondrial 12S ribosomal RNA (rRNA) gene fragment of 73 base pair (bp) was developed for detection of horse DNA in different commercial meat products for human and pet consumption. The method was found to be specific for horse and did not show any cross-reactivity with different species of mammals, birds, fish and plants. The assay complies with the acceptance criteria required for real-time PCR methods in terms of applicability, linear dynamic range, accuracy and PCR efficiency and showed to be sensitive allowing the detection of 1 pg of horse DNA. A range of food and feed products (n = 171) was screened with the horse-specific real-time PCR to determine whether a correct labelling had been employed at the market level. Results obtained when testing the meat products for human consumption were in agreement with the labelling description provided by the suppliers. In the case of the pet foods, undeclared horse meat was detected in 21 % of the samples at low levels, suggesting unintentional cross-contamination during processing. The reported real-time PCR methodology may represent a suitable tool for the detection of food mislabelling.

Abstract

Animal species identification in food and feed has gained increasing interest in recent years due to public health, economic and legal concerns. In the present work, a TaqMan real-time PCR assay targeting the mitochondrial 12S rRNA gene was developed for detection of a specific DNA fragment (87 bp) common to the most economically important fish families in fish-derived ingredients used in feed formulations. Performance of the assay was evaluated against stringent acceptance criteria in terms of specificity and sensitivity. The TaqMan real-time fish-specific system proved to be highly sensitive, allowing the detection of 0.1 pg of fish DNA. The assay was successfully applied to the authentication of two types of real-world compound feeds: industrial farm animal feeds and commercial pet foods, allowing detection of small fish percentages in the samples. The quantitative potential of the real-time PCR assay was further performed, concluding that, although results were well reproducible under the experimental conditions tested, an accurate estimation of the target content in feeds is not feasible in practice. The reported methodology may be useful as a molecular analytical tool to support fisheries control and enforcement, as well as the verification of authenticity of fish and fish-derived products in the food and feed sectors.

Abstract

In the last few years, species of the genus Arcobacter have emerged as etiologic agents of food-borne diarrhoeal illness in humans. The routes of Arcobacter spp. infection are not yet elucidated, but consumption of contaminated food and water seem as the most likely vehicles for the transmission to persons. Food commodities from which arcobacters are more frequently isolated are meats, especially poultry followed by pork, beef and others. The growing number of reports alluding to the association of Arcobacter spp. with animal foods and human illness has concentrated efforts of researchers towards the development of improved molecular methods for the identification and characterisation of these emerging pathogenic bacteria. In this review paper, an update of the occurrence of arcobacters in foods and their association with human illness is provided. Special emphasis is given to the main DNA-based methods applied for the detection and identification of Arcobacter species, as well as to the molecular typing methods currently in use for these emerging food-borne pathogens.

Abstract

A polymerase chain reaction (PCR) procedure has been applied for the detection of caprine milk in ovine cheeses by using primers targeting the mitochondrial 12S ribosomal RNA gene. The use of a primer pair specific for goat in PCR analysis of cheese samples, enabled the amplification of a goat 122 bp fragment with a sensitivity threshold of approximately 1%, whereas no amplification signal was achieved with sheep's, cow's and water buffalo's milk DNA. This study demonstrates the usefulness of the proposed PCR assay for the qualitative detection of goats’ milk in ewes’ cheeses, and may therefore provide a simple and accurate approach applicable to the authentication of cheese or other dairy products in routine monitoring programs.

Abstract

Assuming that the mechanism proposed by Gell-Mann and Hartle works as a mechanism for decoherence and classicalization of the metric field, we formally derive the form of an effective theory for the gravitational field in a semiclassical regime. This effective theory takes the form of the usual semiclassical theory of gravity, based on the semiclassical Einstein equation, plus a stochastic correction which accounts for the back reaction of the lowest order matter stress-energy fluctuations.

Abstract

We present a new measurement of the total photoproduction cross section performed with the H1 detector at HERA. For an average centre of mass energy of 200 GeV a value of σtotλp=165±2±11µb has been obtained. A detailed analysis of the data in adequate kinematic regions enabled a decomposition of the total cross section in its elastic, single diffractive dissociation and remaining non-diffractive parts, based on safe assumptions on the double diffractive dissociation contribution.
