Hirschsprung disease (HSCR) is a congenital intestinal disease caused by the abnormal proliferation and migration of enteric nerve cells (ENCC). Research suggested critical roles for circular RNA (circRNA) itchy E3 ubiquitin protein ligase (ITCH) in gastrointestinal malignancies progression. However, the function of circ-ITCH in HSCR remains poorly defined.The related genes expression in 30 HSCR patients and 30 controls without HSCR were detected using qRT-PCR. Cell proliferation was assessed by CCK-8 assay and EdU assay. Cell migration was detected with wound-healing assay and transwell assay. The interactions among circ-ITCH, miR-146b-5p, and RET were confirmed by Dual luciferase reporter assay.Circ-ITCH and RET expressions were downregulated in HSCR patients and cells, while the miR-146b-5p expression was upregulated. Circ-ITCH overexpression facilitated cell proliferation, migration, and activated MAPK pathway, which were reversed by circRNA-ITCH knockdown. Circ-ITCH negatively regulated miR-146b-5p expression. MiR-146b-5p overexpression abolished the promoting effects of circ-ITCH overexpression on cell proliferation and migration. MiR-146b-5p inhibited RET expression. RET overexpression eliminated the inhibitory effects of miR-146b-5p overexpression on cell proliferation and migration.Circ-ITCH overexpression facilitated cell proliferation and migration in HSCR by regulating miR-146b-5p/RET/MAPK axis.The expressions of Circ-ITCH and RET were markedly reduced in HSCR, while miR-146b-5p expression was increased in HSCR. Circ-ITCH overexpression enhanced the proliferative and migratory abilities of SH-SY5Y and 293T cells. Circ-ITCH negatively regulated miR-146b-5p expression.

Background Intramuscular fat content, an important meat quality trait, strongly affects flavor, juiciness, and tenderness. Sex hormones regulate lipid metabolism, and female hormones stimulate fat deposition, thereby making the female chickens always fatter than males. In this study, the effect of sex on IMF deposition was screened following transcriptomics in chickens. Methods and results Results confirmed significantly higher IMF content of 150-day female chickens as compared to the male chickens. The female chickens manifested higher serum TG, LDL-C, and VLDL, and significantly lower HDL-C contents than male chickens. Moreover, differential expression of genes involved in lipid metabolism were obtained in the muscle and liver between female and male chicken, which could partly interpret the possible reasons for the sex-mediated differences of IMF content. Cellular results revealed that inhibition of PLIN2 significantly inhibited chicken preadipocyte proliferation and induces apoptosis of preadipocytes, as well as promoted adipocyte differentiation. Conclusions According to our results, PLIN2 may be considered as a molecular marker for poultry meat quality and applying this gene in early breed selection.

As Volatile Organic Compounds (VOCs) are one of the precursors of ozone, their distribution and variable concentrations are highly related to local ozone pollution control. In this study, we obtained vertical profiles of VOCs in Shanghai’s Jinshan district on 8 September and 9 September in 2016 to investigate their distribution and impact on local atmospheric oxidation in the near surface layer. Vertical samples were collected from heights between 50 m and 400 m by summa canisters using an unmanned aerial vehicle (UAV). Concentrations of VOCs (VOCs refers to the 52 species measured in this study) varied minimally below 200 m, and decreased by 21.2% from 100 m to 400 m. The concentrations of VOCs above 200 m decreased significantly in comparison to those below 200 m. The proportions of alkanes and aromatics increased from 55.2% and 30.5% to 57.3% and 33.0%, respectively. Additionally, the proportion of alkenes decreased from 13.2% to 8.4%. Toluene and m/p-xylene were the key species in the formation of SOA and ozone. Principal component analysis (PCA) revealed that the VOCs measured in this study mainly originated from industrial emissions.

Recent climatic disasters have shown the vulnerability of transportation infrastructures against natural hazards. To understand the risk of coastal hazards on urban travel activities, this study presents an activity-based modeling approach to evaluate the impacts of storm surge on the transportation network under sea-level rise in Miami-Dade County, FL. A Markov-Chain Monte Carlo (MCMC) based algorithm is applied to generate population attributes and travel diaries in the model simulation. Flooding scenarios in 2045 are developed based on different adaptation standards under the 100-year storm surge and population projections are from the land-use conflict identification strategy (LUCIS) model. Our analysis indicates that about 29.3% of the transportation infrastructure, including areas of the US No. 1 highway, roadways in the south and southwest of the county, and bridges connecting Miami Beach area, will be damaged under the storm surge when a low-level adaptation standard is chosen. However, the high-level adaptation standard will reduce the vulnerable infrastructures to 12.4%. Furthermore, the total increased travel time of the low-level adaptation standard could be as high as twice of that in the high-level adaptation standard during peak morning hours. Our model results also reveal that the average increased travel time due to future storm surge damage ranges between 14.2 and 62.8 min per trip.

Adaptation has become the major approach to reduce the adverse effects of storm surge and sea-level rise. However, maladaptation can happen when adaptation actions unintentionally increase community vulnerability. To evaluate the adequacy and efficacy of adaptation policies under uncertain sea-level rise, this study presents an agent-based model by integrating the random nature of storm surges, private adaptation decisions, and real estate market valuation. We evaluated the evolving flood damage of different adaptation strategies under two bounding cases of real estate market change. Our model results quantitatively illustrate the accelerating damages of storm surges under climate-induced sea-level rise. A reform in flood insurance to risk-based rates with a means-tested voucher program and a government-subsidized “twice and out” buyout program could both substantially improve coastal resilience. However, community adaptation with a public seawall may deliver false risk perception to high-risk property owners and result in maladaptation when sea-level rise rate is high. The modeling approach developed in this study can be used as a policy analysis tool to measure the impacts of sea-level rise and the effectiveness of adaptation strategies in coastal communities.

In this paper, carbon nanotubes (CNTs) reinforced Ti–12.8Mo–2.1Nb–2.7Al–0.2Si (TB8) matrix composites were synthesized by low-energy ball milling and cold pressing sintering. Based on theoretical calculation and thermodynamic simulation method to obtain phase transition points, CNTs/TB8 composites were subjected to a solid-solution and aging treatment. The effect of heat treatment and CNTs content on microstructure and mechanical properties of the composites was investigated. The results indicated that 0.5 wt% CNTs/TB8 composite formed α + β phase structure after different heat treatment processes, α phase dispersed in the β phase, grain refinement was distinct, and the microhardness of composite was significantly improved. It had the effect of solution strengthening and dispersion strengthening. The composites’ microhardness was the highest, reaching 615.3HV after 850 °C/3 h/AC and 550 °C/6 h/AC heat treatment. Under these conditions, with the CNTs content increasing, the microhardness of composite increased initially and then decreased. The microhardness of the composite containing 1.0 wt% CNTs reached 635.1HV, which was an increase of 21.9% compared to without executing heat treatment. In addition, the compression property of the titanium matrix composite at room temperature was excellent; the compressive strength and the fracture strain were 1263 MPa and 18.5%, respectively.

Many coastal communities around the world are increasingly faced with pressure from sea level rise (SLR). Adapting to the impacts of SLR is now considered inevitable. Many coastal communities in the USA have started to invest in vulnerability assessments that seek to identify the degree of future risk induced by SLR and key vulnerable sectors as well as to provide a sound factual basis for designing and implementing adaptations. However, no study has systematically analyzed the content of these emerging assessments and their quality. This study aims to address this gap by evaluating 64 SLR vulnerability assessments in the USA using an established multi-criteria evaluation framework and by identifying governance factors that affect assessment quality. This study is the first baseline study to understand how the vulnerability of SLR has been assessed by US governments and, more importantly, shed light on pathways for their future improvement. Our analysis finds that the content and quality of the existing assessments vary widely. One-sided assessments that only consider physical exposure are common, and most assessments do not include plans for adaptations. The wealth of the area, amount of funding, mainstreaming efforts, and public awareness are all positively correlated with the assessment quality. We also find that assessments primarily authored by planning staff are lower in quality than those conducted through collaboration. Our findings offer comparative, empirical knowledge for urban planners and coastal managers to improve future vulnerability assessments and adaptation planning for SLR.
