Abstract

Background: The combination of chemotherapy with the vascular endothelial growth factor (VEGF) antibody bevacizumab is a standard of care in advanced colorectal cancer (CRC). However, biomarkers predicting outcome of bevacizumab-containing treatment are lacking. As angiopoietin-2 (Ang-2) is a key regulator of vascular remodelling in concert with VEGF, we investigated its role as a biomarker in metastatic CRC. Methods: Serum Ang-2 levels were measured in 33 healthy volunteers and 90 patients with CRC. Of these, 34 had metastatic disease and received bevacizumab-containing therapy. To determine the tissue of origin of Ang-2, quantitative real-time PCR was performed on microdissected cryosections of human CRC and in a murine xenograft model of CRC using species-specific amplification. Results: Ang-2 originated from the stromal compartment of CRC tissues. Serum Ang-2 levels were significantly elevated in patients with metastatic CRC compared with healthy controls. Abstract

The integral membrane protein M2 of influenza virus forms pH-gated proton channels in the viral lipid envelope. The low pH of an endosome activates the M2 channel before haemagglutinin-mediated fusion. Conductance of protons acidifies the viral interior and thereby facilitates dissociation of the matrix protein from the viral nucleoproteins--a required process for unpacking of the viral genome. In addition to its role in release of viral nucleoproteins, M2 in the trans-Golgi network (TGN) membrane prevents premature conformational rearrangement of newly synthesized haemagglutinin during transport to the cell surface by equilibrating the pH of the TGN with that of the host cell cytoplasm. Inhibiting the proton conductance of M2 using the anti-viral drug amantadine or rimantadine inhibits viral replication. Here we present the structure of the tetrameric M2 channel in complex with rimantadine, determined by NMR. In the closed state, four tightly packed transmembrane helices define a narrow channel, in which a 'tryptophan gate' is locked by intermolecular interactions with aspartic acid. A carboxy-terminal, amphipathic helix oriented nearly perpendicular to the transmembrane helix forms an inward-facing base. Lowering the pH destabilizes the transmembrane helical packing and unlocks the gate, admitting water to conduct protons, whereas the C-terminal base remains intact, preventing dissociation of the tetramer. Rimantadine binds at four equivalent sites near the gate on the lipid-facing side of the channel and stabilizes the closed conformation of the pore. Drug-resistance mutations are predicted to counter the effect of drug binding by either increasing the hydrophilicity of the pore or weakening helix-helix packing, thus facilitating channel opening.

Abstract

A novel method for the directional cloning of native PCR products was developed. Abasic sites in DNA templates make DNA polymerases stall at the site during synthesis of the complementary strand. Since the 5' ends of PCR product strands contain built-in amplification primers, abasic sites within the primers result in the formation of 5' single-stranded overhangs at the ends of the PCR product, enabling its direct ligation to a suitably cleaved cloning vector without any further modification. This "autosticky PCR" (AS-PCR) overcomes the problems caused by end sensitivity of restriction enzymes, or internal restriction sites within the amplified sequences, and enables the generation of essentially any desired 5' overhang.

Abstract

This document describes the technical layout and the expected performance of the Straw Tube Tracker (STT), the main tracking detector of the $\bar{P}$ANDA target spectrometer. The STT encloses a Micro-Vertex-Detector (MVD) for the inner tracking and is followed in beam direction by a set of GEM stations. The tasks of the STT are the measurement of the particle momentum from the reconstructed trajectory and the measurement of the specific energy loss for a particle identification. Dedicated simulations with full analysis studies of certain proton-antiproton reactions, identified as being benchmark tests for the whole $\bar{P}$ANDA scientific program, have been performed to test the STT layout and performance. The results are presented, and the time lines to construct the STT are described.

Abstract

In a strain of the mutantw (yellow eye colour, sex-linked and sex-limited to the females) ofCalliphora erythrocephala yellow-eyed males have been found. In connexion with the mechanism of sex-determination in this fly, their origin is explained by an alteration on the Y-chromosome (one of the IIIrd large chromosomes) in form of either a pre-meiotic cross-over or a spontaneous mutation on thew+-locus. For being recessive, it can become manifest only in thew strain.

Abstract

Malgre ses enclaves de savance et la prairie, riches en especes tropicales xerophiles, le territoire etudie appartient incontestablement au domaine forestier guineo-equatorial. Plusieurs climax, tous forestiers, appartenant, suivant les conditions locales, aux ordres des Triplochitetalia, Lophiretalia et Parinarietalia, y sont representes. En fait, cette vegetation primitive est profondement degradee, aux basses altitudes par les defrichements, et sur les cretes par les feux. C'est dire tout l'interet que presente, dans ce territoire, la protection de la Nature; la mise en reserve naturelle integrale du massif du Nimba, promulguee en juillet 1944 sur l'initiative de M. le Professeur Heim et de M. le Professeur Monod, Directeur de l'Institut Francais d'Afrique Noire, preservera de la destruction les richesses botaniques de cette region montagneuse.
