The phyla Nitrospirota and Nitrospinota have received significant research attention due to their unique nitrogen metabolisms important to biogeochemical and industrial processes. These phyla are common inhabitants of marine and terrestrial subsurface environments and contain members capable of diverse physiologies in addition to nitrite oxidation and complete ammonia oxidation. Here, we use phylogenomics and gene-based analysis with ancestral state reconstruction and gene-tree–species-tree reconciliation methods to investigate the life histories of these two phyla. We find that basal clades of both phyla primarily inhabit marine and terrestrial subsurface environments. The genomes of basal clades in both phyla appear smaller and more densely coded than the later-branching clades. The extant basal clades of both phyla share many traits inferred to be present in their respective common ancestors, including hydrogen, one-carbon, and sulfur-based metabolisms. Later-branching groups, namely the more frequently studied classes Nitrospiria and Nitrospinia, are both characterized by genome expansions driven by either de novo origination or laterally transferred genes that encode functions expanding their metabolic repertoire. These expansions include gene clusters that perform the unique nitrogen metabolisms that both phyla are most well known for. Our analyses support replicated evolutionary histories of these two bacterial phyla, with modern subsurface environments representing a genomic repository for the coding potential of ancestral metabolic traits.

Sexual selection on fitness-determining traits should theoretically erode genetic variance and lead to low heritability. However, many sexually selected traits maintain significant phenotypic and additive genetic variance, with explanations for this “lek paradox” including genic capture due to condition-dependence, and breaks on directional selection due to environmental sources of variance including maternal effects. Here we investigate genetic and environmental sources of variance in the intrasexually selected green badge of the sand lizard (Lacerta agilis). The badge functions as a cue to male fighting ability in this species, and male–male interactions determine mate acquisition. Using animal models on a pedigree including three generations of males measured over an extensive 9-year field study, we partition phenotypic variance in both badge size and body condition into additive genetic, maternal, and permanent environmental effects experienced by an individual over its lifespan. Heritability of badge size was 0.33 with a significant estimate of underlying additive genetic variance. Body condition was strongly environmentally determined in this species and did not show either significant additive genetic variance or heritability. Neither badge size nor body condition was responsive to maternal effects. We propose that the lack of additive genetic variance and heritability of body condition makes it unlikely that genic capture mechanisms maintain additive genetic variance for badge size. That said, genic capture was originally proposed for male traits under female choice, not agonistic selection. If developmental pathways generating variance in body condition, and/or the covarying secondary sex trait, differ between inter- and intrasexual selection, or the rate at which their additive genetic variance or covariance is depleted, future work may show whether genic capture is largely restricted to intersexual selection processes.

Liquid xenon time projection chambers are promising detectors to search for neutrinoless double beta decay (0$$\nu \beta \beta $$ ν β β ), due to their response uniformity, monolithic sensitive volume, scalability to large target masses, and suitability for extremely low background operations. The nEXO collaboration has designed a tonne-scale time projection chamber that aims to search for 0$$\nu \beta \beta $$ ν β β of $$^{136}$$ 136 Xe with projected half-life sensitivity of $$1.35\times 10^{28}$$ 1.35 × 10 28 yr. To reach this sensitivity, the design goal for nEXO is $$\le $$ ≤ 1% energy resolution at the decay Q-value ($$2458.07\pm 0.31$$ 2458.07 ± 0.31 keV). Reaching this resolution requires the efficient collection of both the ionization and scintillation produced in the detector. The nEXO design employs Silicon Photo-Multipliers (SiPMs) to detect the vacuum ultra-violet, 175 nm scintillation light of liquid xenon. This paper reports on the characterization of the newest vacuum ultra-violet sensitive Fondazione Bruno Kessler VUVHD3 SiPMs specifically designed for nEXO, as well as new measurements on new test samples of previously characterised Hamamatsu VUV4 Multi Pixel Photon Counters (MPPCs). Various SiPM and MPPC parameters, such as dark noise, gain, direct crosstalk, correlated avalanches and photon detection efficiency were measured as a function of the applied over voltage and wavelength at liquid xenon temperature (163 K). The results from this study are used to provide updated estimates of the achievable energy resolution at the decay Q-value for the nEXO design.

Over the past 10 years, lung cancer clinical and translational research has been characterised by exponential progress, exemplified by the introduction of molecularly targeted therapies, immunotherapy and chemo-immunotherapy combinations to stage III and IV non-small cell lung cancer. Along with squamous and small cell lung cancers, large cell neuroendocrine carcinoma (LCNEC) now represents an area of unmet need, particularly hampered by the lack of an encompassing pathological definition that can facilitate real-world and clinical trial progress. The steps we have proposed in this article represent an iterative and rational path forward towards clinical breakthroughs that can be modelled on success in other lung cancer pathologies.

The origin(s) of dissimilatory sulfate and/or (bi)sulfite reducing organisms (SRO) remains enigmatic despite their importance in global carbon and sulfur cycling since at least 3.4 Ga. Here, we describe novel, deep-branching archaeal SRO populations distantly related to other Diaforarchaea from two moderately acidic thermal springs. Dissimilatory (bi)sulfite reductase homologs, DsrABC, encoded in metagenome assembled genomes (MAGs) from spring sediments comprise one of the earliest evolving Dsr lineages. DsrA homologs were expressed in situ under moderately acidic conditions. MAGs lacked genes encoding proteins that activate sulfate prior to (bi)sulfite reduction. This is consistent with sulfide production in enrichment cultures provided sulfite but not sulfate. We suggest input of volcanic sulfur dioxide to anoxic spring-water yields (bi)sulfite and moderately acidic conditions that favor its stability and bioavailability. The presence of similar volcanic springs at the time SRO are thought to have originated (>3.4 Ga) may have supplied (bi)sulfite that supported ancestral SRO. These observations coincide with the lack of inferred SO42− reduction capacity in nearly all organisms with early-branching DsrAB and which are near universally found in hydrothermal environments.

Objective Our objective was to apply next-generation sequence-based DNA barcoding to identify the remnant larval bloodmeals in wild-caught host-seeking (unengorged) Ixodes scapularis nymphs (n = 216). To infer host species identification, vertebrate DNA was amplified using universal primers for cytochrome c oxidase subunit I (COI) and sequenced using next-generation sequencing (NGS) for comparison against known barcode references. Results Bloodmeal identification was unsuccessful in most samples (99% of 216 specimens) demonstrating a very low detection rate of this assay. Sequences that surpassed quality thresholds were obtained for 41.7% of nymphs (n = 90) and of those, confident species identification was obtained for 15.6% of nymphs (n = 14). Wild host identifications were only obtained from 2 specimens, where DNA from the eastern grey squirrel (Sciurus carolinensis) was identified. Human and bovine DNA was identified in remaining nymphs and considered to be contaminants. Further optimization of the technique is required to improve detection of remnant bloodmeals in host-seeking nymphs.

Summary We introduced an electronic triage system into our osteoporosis service to actively manage referral demand in a busy outpatient service. Our study demonstrated the effectiveness of e-triage in supporting alternative management pathways, through use of virtual advice and direct to investigation services, to improve patient access. Purpose Osteoporosis referrals are increasing with awareness of the potential for prevention of fragility fracture and with complex decision making around management with long-term bisphosphonate therapy. We examined whether active triage of referrals might improve referral management processes and patient access to osteoporosis services. Methods We implemented electronic triage (e-triage) of referrals to our osteoporosis service using the Northern Ireland electronic health care record. This included the option of ‘advice only’, direct to investigation with DXA or face-to-face appointments at the consultant-led complex osteoporosis service. We anticipated that there was scope to manage patient flow direct to investigation, or to provide referring clinicians with clinical advice without the need for a face-to-face assessment, at the consultant-led specialist service. Results We reviewed e-triage outcomes of 809 referrals (692 F; 117 M) to osteoporosis specialist services (mean age 65 ± 16.5 years) over a 12-month period. There was a high degree of agreement for the triage category between the referring clinician and specialist services (741/809). 73.3% attended a face-to-face appointment at the consultant-led clinic, while active triage enabled direct to investigation (18.4%) or discharge (8.3%) in the remainder. The mean time between receipt of an electronic referral and e-triage was 3 days over the 12-month period as compared with 2.1 days (median 1.1 days) when annual leave periods were excluded. Conclusion E-triage supports effective referral management in a busy osteoporosis service. Efficiency is limited by reliance on a sole clinician and 5 day working at present. There is scope to further improve systems access through multidisciplinary team working, virtual clinics and future information technology developments.

The upcoming detector facility aims at developing new state-of-the-art particle detectors as well as providing hands-on training to postgraduate students using both analog and digital signal processing from nuclear radiation detectors. The project is two-fold and aims at developing: 1) ancillary detectors to be coupled with the new GAMKA array at iThemba LABS. Of particular interest to our group is the determination of nuclear shapes, which depend on the hyperfine splitting of magnetic substates; 2) PET scanners for cancer imaging using a cheaper technology. Performance of NaI(Tl) inorganic scintillator detectors has been evaluated using PIXIE-16 modules from XIA digital electronics. Gamma-ray energy spectra were acquired from 60Co and 137Cs radioactive sources to calculate the detector resolution as well as to optimize the digital parameters. The present study focuses on improving and optimizing the slow and fast filter parameters for NaI(Tl) detectors which can eventually be used in the list mode of data aquisition.

Focus Group.To develop a unified, regional spinal cord injury (SCI) research strategy for Australia and New Zealand.Australia.A 1-day structured stakeholder dialogue was convened in 2013 in Melbourne, Australia, by the National Trauma Research Institute in collaboration with the SCI Network of Australia and New Zealand. Twenty-three experts participated, representing local and international research, clinical, consumer, advocacy, government policy and funding perspectives. Preparatory work synthesised evidence and articulated draft principles and options as a starting point for discussion.A regional SCI research strategy was proposed, whose objectives can be summarised under four themes. (1) Collaborative networks and strategic partnerships to increase efficiency, reduce duplication, build capacity and optimise research funding. (2) Research priority setting and coordination to manage competing studies. (3) Mechanisms for greater consumer engagement in research. (4) Resources and infrastructure to further develop SCI data registries, evaluate research translation and assess alignment of research strategy with stakeholder interests. These are consistent with contemporary international SCI research strategy development activities.This first step in a regional SCI research strategy has articulated objectives for further development by the wider SCI research community. The initiative has also reinforced the importance of coordinated, collective action in optimising outcomes following SCI.

Despite being a recognized clinical entity for over 140 years, complex regional pain syndrome (CRPS) remains a difficult-to-treat condition. While there have been multiple therapies explored in the treatment of CRPS, NMDA antagonists such as ketamine continue to hold significant interest because of their potential ability to alter the central sensitization noted in chronic pain states. The objective of this review is to identify published literature for evidence of the efficacy and safety of ketamine in the treatment of CRPS. PubMed and the Cochrane Controlled Trials Register were searched (final search 26 May 2011) using the MeSH terms 'ketamine', 'complex regional pain syndrome', 'analgesia' and 'pain' in the English literature. The manuscript bibliographies were then reviewed to identify additional relevant papers. Observational trials were evaluated using the Agency for Healthcare Research and Quality criteria; randomized trials were evaluated using the methodological assessment of randomized clinical trials. The search methodology yielded three randomized, placebo-controlled trials, seven observational studies and nine case studies/reports. In aggregate, the data available reveal ketamine as a promising treatment for CRPS. The optimum dose, route and timing of administration remain to be determined. Randomized controlled trials are needed to establish the efficacy and safety of ketamine and to determine its long-term benefit in CRPS.

We describe a simple single-reaction technique for identifying the sex of white-tailed deer (Odocoileus virginianus) based on the PCR amplification of a zinc-finger intron using one pair of primers. Although Sry-coamplification confirmed sex identities, use of the Sry marker was unnecessary due to dimorphic alleles on the X and Y chromosomes at the zinc-finger locus. Insertions in intron 7 of the Y-linked allele (417 bp) make it nearly twice as long as the X-linked allele (236 bp) and thus the amplification products are easily discernable by simple agarose gel electrophoresis. The relatively short size of these products makes them useful for DNA-based sex identification from potentially low-yield tissue samples (e.g., hair, feces). This technique will provide ecologists, conservation geneticists and wildlife managers with a mechanism to readily and reliably identify the sex of un- known white-tailed deer tissue samples, and likely similar samples from other cervid species.

A residence time of approximately 60 s for lubricant in the piston ring pack of a gasoline engine, running at 1500 rpm, 50% throttle, has been measured by adding a hydrocarbon marker to the oil in the sump and monitoring its build up in oil extracted from the top ring groove. Comparison of the rate of increase of oxidised products in the sump with the level of oxidation in the ring pack allows a rate of flow of oil entering the ring pack from the sump to be calculated as approximately 0.4 cm3 min−1 cylinder−1, with at least 88% returning to the sump and the remainder lost into the combustion chamber. The volume of oil in the ring pack was determined as approximately 0.4 cm3 cylinder−1, approximately one quarter of the free volume available between the top of the oil control ring and the top piston ring.

Measurement of plasma ACTH levels by RIA was originally used to identify adrenal causes (AA) of Cushing's syndrome (CS) and for the differential diagnosis of ACTH-dependent CS with ovine corticotropin-releasing hormone (CRH) stimulation testing and inferior petrosal sinus sampling (IPSS). It is unclear whether diagnostic criteria developed with RIA also work for IRMA. Methods: ACTH was measured by RIA and IRMA (Covance Laboratories, Vienna VA) in paired samples i.) basally (63 Cushing's disease (CD), 5 AA, 2 ectopic CS (EAS) and 37 non-CS patients) ii.) in 44 CD patients following CRH (1 μ g/kg i.v.) iii) in 6 EAS and 17 CD patients during IPSS. Results were analyzed using Spearman's rank correlations, the Wilcoxon signed-rank test, linear regression and McNemar's test. Results: i)Basal RIA and IRMA results were highly correlated (r = 0.9, p 2.0 before or ≥3.0 after CRH, IRMA had the same sensitivity (100%) but higher specificity (100% vs 83%, p = 1.0) vs. RIA. Conclusion: ACTH measured by IRMA correlates closely to RIA values and offers similar diagnostic accuracy during the CRH and IPSS testing. However in ACTH samples < 15 pg/ml IRMA probably offers better diagnostic accuracy.

Somatostatin analogues are an established treatment in acromegaly. This study was designed to evaluate whether the acute serum growth hormone (GH) response to a test dose of octreotide in acromegaly predicts longer-term response to the drug at 3 years.In 23 patients, GH responses across 8 h to a subcutaneous test dose (50 microg) of octreotide were compared with GH levels after 3 years of therapy. The majority had pituitary surgery as primary therapy and at 3 years were receiving at least 600 microg octreotide daily subcutaneously or 20 mg LAR monthly intramuscularly.Seven had a test day GH Nadir of 5 mU/l or less of whom 4 achieved GH5 mU/l at 3 years. Sixteen had a test day nadir GH of 10 mU/l or less and of these 8 achieved GH5 mU/l at 3 years. Seven of the 23 had a GH Nadir10 mU/l and of these 3 had achieved GH5 mU/l at 3 years. However all of these 3 had received external pituitary irradiation within 4 years of the 3 year assessment, as compared with 3 of the5 mU/l nadir group and 5 of the10 mU/l nadir group.In patients on optimal long-term doses of octreotide for acromegaly, absence of a nadir GH10 mU/l in the 8 h after a test dose was associated with failure to achieve GH levels associated with a normal life expectancy (5 mU/l or less) unless adjunctive external pituitary irradiation was given. As well as testing tolerability a test dose of octreotide may help in determining which patients should be offered early external pituitary irradiation or therapy with a GH receptor antagonist if surgery has failed to achieve 'safe' GH levels.

Glycation of insulin, resulting in impaired bioactivity, has been shown within pancreatic beta cells. We have used a novel and specific radioimmunoassay to detect glycated insulin in plasma of Type 2 diabetic subjects.Blood samples were collected from 102 Type 2 diabetic patients in three main categories: those with good glycaemic control with a HbA(1c) less than 7%, moderate glycaemic control (HbA(1c) 7-9%) and poor glycaemic control (HBA(1c) greater than 9%). We used 75 age- and sex-matched non-diabetic subjects as controls. Samples were analysed for HbA(1c), glucose and plasma concentrations of glycated insulin and insulin.Glycated insulin was readily detected in control and Type 2 diabetic subjects. The mean circulating concentration of glycated insulin in control subjects was 12.6+/-0.9 pmol/l ( n=75). Glycated insulin in the good, moderate and poorly controlled diabetic groups was increased 2.4-fold ( p0.001, n=44), 2.2-fold ( p0.001, n=41) and 1.1-fold ( n=17) corresponding to 29.8+/-5.4, 27.3+/-5.7 and 13.5+/-2.9 pmol/l, respectively.Glycated insulin circulates at noticeably increased concentrations in Type 2 diabetic subjects.

Visual improvement following octreotide for growth hormone secreting pituitary macroadenomas is uncommon without tumour shrinkage. A 45-year old lady presented with blurred vision for 12 months. Visual assessment revealed a bitemporal hemianopia and CT scan demonstrated a large pituitary tumour with lateral and suprasellar extension. Acromegaly was confirmed by 75 g glucose tolerance testing. Primary transsphenoidal surgery was performed with normalisation of visual acuity and fields of vision. Post-operatively she had anterior pituitary hormone deficiency. As GH and IGF-1 levels remained elevated she underwent external pituitary irradiation. CT scanning demonstrated tumour shrinkage associated with a modest fall in GH levels. IGF-1 levels remained elevated falling to the age-related upper limit of normal after 5 years. At regular review she had stable visual acuity and fields of vision. She presented as an emergency 7 years from presentation with reduced vision and recurrence of bitemporal hemianopia. An MRI demonstrated a large pituitary adenoma. We therefore undertook a carefully monitored trial of octreotide with great caution with daily reassessment of acuity and fields. A decision was made to proceed to surgery in the event of deterioration or lack of improvement after a short trial over 5-7 days. We observed normalisation of visual acuity and perimetry within 3 days. She then commenced long-acting octreotide (Sandostatin LAR) 20 mg every 28 days. MRI after 1 week showed shrinkage of the tumour by a few millimetres. Five months later repeat MRI failed to show any further improvement in tumour size. However she remains well 29 months from treatment with normal vision and is being monitored carefully as her chosen form of therapy. Somatostatin analogues may be effective as therapy in a selected group of patients with acromegaly and visual loss who are not suitable for pituitary surgery. If used in this way the drug must be given cautiously with frequent detailed ongoing visual assessments. In this present case there has been a restoration of vision but the long-term outlook remains guarded without significant tumor shrinkage.

The influence of wave exposure and of tidal height on mussel (Perna perna Linnaeus) population structure (size, density, biomass and adult/juvenile correlations) was examined at 18 sites along the south coast of South Africa. Sites were classified as exposed or sheltered prior to sampling, without reference to the biota, on the basis of aspect, topography and wave regime. A single set of samples was collected from each site during three spring tide cycles. Adult mussels on these shores almost always attach directly to the rocks, and layering of mussels is virtually absent. Shore height always had a strong influence on population structure, but exposure had significant effects only lower on the shore, and almost exclusively on mussel sizes. Principal component analysis (PCA), based on size distribution data for each population, revealed a general upshore decrease in the modal size of the adult cohort. The effects of exposure on size distribution, however, varied with tidal height. PCA separated exposed zones, with larger mussels, from sheltered zones on the low-shore. Farther upshore the two shore types were increasingly confounded. The maximum size of mussels showed a similar pattern, with significant differences (ANOVA, p 15 mm) densities decreased up the shore, with low-, mid- and high-shore zones being significantly different from one another (ANOVA, p 15 mm) densities in all zones and for both exposure regimes ( p

The complex [LMnIV(O)3MnIVL](PF6)2 (1), where L is 1,4 7-trimethyl-1,4,7-triazacyclononane, catalyzes a highly efficient stereoselective oxygenation of saturated hydrocarbons in the presence of H2O2. A carboxylic acid is an obligatory component of the reaction mixture, while acetonitrile or acetone can be used as solvent. The reaction occurs, forming alkyl hydroperoxide, ketone, and alcohol. Substitution at the tertiary carbon atom proceeds more easily than that at the secondary carbon atom, whereas primary C−H bonds are rather inactive. Oxidation of alkanes and alcohols with peroxy acids catalyzed by complex 1 occurs with lower efficiency.

This paper describes data on bone mineral levels in the proximal femur of US adults based on the nationally representative sample examined during both phases of the third National Health and Nutrition Examination Survey (NHANES III, 1988-94), and updates data previously presented from phase 1 only. The data were collected from 14,646 men and women aged 20 years and older using dual-energy X-ray absorptiometry, and included bone mineral density (BMD), bone mineral content (BMC) and area of bone scanned in four selected regions of interest (ROI) in the proximal femur: femur neck, trochanter, intertrochanter and total. These variables are provided separately by age and sex for non-Hispanic whites (NHW), non-Hispanic blacks (NHB) and Mexican Americans (MA). NHW in the southern United States had slightly lower BMD levels than NHW in other US regions, but these differences were not sufficiently large to prevent pooling of the data. The updated data provide valuable reference data on femur bone mineral levels of noninstitutionalized adults. The updated data on BMD for the total femur ROI of NHW have been selected as the reference database for femur standardization efforts by the International Committee on Standards in Bone Measurements.

This paper describes bone mineral levels in the proximal femur of US adults based on a nationally representative sample of 7116 men and women aged 20 years and older. The data were collected in phase 1 of the third National Health and Nutrition Examination Survey (NHANES III, 1988-1991) using dual-energy X-ray absorptiometry, and included bone mineral density (BMD), bone mineral content (BMC) and area of bone scanned in five selected regions of interest (ROI) in the proximal femur: femur neck, trochanter, intertrochanter, Ward's triangle and total. These variables are provided separately by age and sex for non-HIspanic whites (NHW), non-Hispanic blacks (NHB) and Mexican Americans (MA). BMD and BMC in the five ROI tended to decline with age, whereas area did not. BMD and BMC were highest in NHB, intermediate in MA and lowest in NHW, but areas were highest in NHW, intermediate in NHB and lowest in MA. Men had greater BMD, BMC and area than women in all three race/ethnic groups. Differences by age, sex or race/ethnicity tended to be the largest in Ward's triangle, followed by the femur neck; patterns in the trochanter, intertrochanter and total ROI were reasonably similar to each other. This report provides extensive data on femur bone mineral levels of adults from one of the largest samples available to date and should be valuable as reference data for other studies which examine this skeletal site in adults.

On heating, a MnIV complex initiates oxidation of cyclohexanecarbaldehyde with atmospheric oxygen in MeCN resulting in the formation of cyclohexanecarboxylic acid and considerable amounts of oxidative decarboxylation products (cyclohexane, cyclohexanol, and cyclohexanone). Cyclooctanol and cyclooctanone are also formed in the presence of cyclooctane.

Estrogens clearly have beneficial effects on the skeleton of postmenopausal women. The development of the agents that act like estrogens in the skeleton but may have a different pattern of effect in other tissues, suggests the possibility that tissue-selective estrogens may become important in the management of post-menopausal women in the future, especially those who may be at risk of osteoporosis. Greater understanding of the action of these tissue-selective estrogens will allow the development of a novel compound with profiles that may target more definitively the needs of the postmenopausal population.

As miniaturisation proceeds, the electrical properties of conductive films used in modem IC's are increasingly influenced by the grain sizes and the texture of the films. There is a need therefore to devise techniques which can examine these properties. Described in this work are two new cross-sectional TEM techniques for use on fully-processed IC's to determine quantitatively grain size distributions and the degree of texture in a film. The technique which investigates texture is used to determine how quickly the texture develops through a polysilicon film.

Background Trunk accelerations during running provide useful information about movement economy and injury risk. However, there is a lack of data regarding the key biomechanical contributors to these accelerations. The purpose was to establish the biomechanical variables associated with root mean square (RMS) accelerations of the trunk. Methods Eighteen healthy males (24.0 ± 4.2 yr; 1.78 ± 0.07 m; 79.7 ± 14.8 kg) performed treadmill running with high resolution accelerometer measurement at the lumbar spine and full-body optical motion capture. We collected 60 sec of data at three speeds (2.22, 2.78, 3.33 m∙s−1). RMS was calculated for medio-lateral (ML), anterio-posterior (AP), vertical (VT), and the resultant Euclidean scalar (RES) acceleration. From motion capture, we calculated 14 kinematic variables, including mean sagittal plane joint angles at foot contact, mid-stance, and toe-off. Principal components analysis (PCA) was used to form independent components comprised of combinations of the original variables. Stepwise regressions were performed on the original variables and the components to determine contributions to RMS acceleration in each axis. Although this was not associated with significant metabolic effects in the fasting state, metformin-induced inhibition of DPP IV may contribute to glycaemic control by promoting insulin-releasing activity of the enteroinsular hormones actively secreted following feeding.

The prevalence of LJM has decreased most likely as a result of improved blood glucose control during the past 20 years. LJM remains a useful clinical risk indicator of microvascular disease.

The effect of renal transplantation is, in most instances, to restore a functioning kidney which can respond to the metabolic status of the patient. The serum phosphate returns to normal, the serum calcium rises and the hyperparathyroid state, during maintenance dialysis, resolves in many cases. In a variable proportion of those patients who have received a renal allograft, structural bone failure may occur, within the next few years (2). This condition, aseptic necrosis, is a major influence in determining how quickly a recipient of a successful graft can return to a completely normal way of life. Its aetiology is still obscure; of importance may be continued hyperparathyroidism, steroid therapy or both (2).

A comparison is presented of insomniacs' self-reported sleep pattern and objectively monitored sleep using the Sleep Assessment Device (SAD; Kelley and Lichstein, 1980). Analysis of 110 subject nights revealed modest but significant subjective overestimates of both sleep latency and sleep duration which were accounted for by the demands of the task (i.e., longer time periods are harder to estimate accurately). Correlations between self-report and SAD measures were, however, consistently high, suggesting that self-report may be a valid index of sleep disturbance. Results are discussed with reference to the clinician's task in evaluating sleep complaints. © 1989 Plenum Publishing Corporation.

Results from Dual Photon Absorptiometry, a common tool in the assessment of bone mass, vary as a function of source life, increasing by as much as 0.6% per month. This rate of increase may vary with source, and its cause is not clear. However, this long-term "drift" must be considered a significant problem for longitudinal studies and regular standardization of DPA equipment, in addition to calibration to machine standards, may be required for stringent quality control.

ON the basis of results obtained with testosterone administered, together with pregnant mare serum gonadotrophin, to anœstrous ewes Cole1 suggested that androgen may be responsible for the induction of œstrous behaviour in the sheep. Robinson2, however, has shown that œstrogen given following a period of progesterone priming will induce both behavioural and vaginal œstrus in spayed ewes, and it is probable that normal cyclic œstrous behaviour is due to an interaction of these two hormones. In the course of work on the interaction of several steroid hormones on behavioural and vaginal œstrus in spayed ewes, a preliminary trial showed that a single dose of testosterone or of testosterone propionate resulted in mating behaviour in a number of animals. Consequently, a factorial design test, using 72 spayed ewes, was conducted to examine this phenomenon and to define the nature of the apparent œstrous response. The variables introduced (with n = 12) were priming treatment (no progesterone or 10 mgm. per day for 12 days)3, and dose of androgen (1.0, 3.2 or 10.0 mgm.). The androgen was testosterone propionate. Sexual receptivity of the ewes was determined using 8 intact rams fitted with ‘Sire-sine’ harnesses and crayons4 and the vaginal response was determined by smears taken for four days following injection of androgen5. Results are presented in Table 1.

INVESTIGATIONS of the hydroxylating activity of A. niger have been carried out mainly in connexion with the metabolism of the phenoxy- and naphthyloxy-alkyl-carboxylic acids1,2, at least in part because of the importance of these compounds as herbicides. We have been examining the action of this organism, using van Tiegh strain, on various benzenoid compounds, and since our results for the hydroxylation of phenoxyacetic acid differ fundamentally from those obtained with van Tiegh (Mulder) strain1, we wish to report our findings for both this compound and the related aromatic ether, anisole.
