Oncidium orchids are important ornamental flowers for export in Taiwan, but their production is limited by bacterial soft rot disease. The common pathogens of bacterial soft rot include Pectobacterium carotovorum subsp. carotovorum (Pcc) and P. chrysanthemi (Pch). Pch is a unique pathogen that can be isolated from Oncidium. Thus, the possible reasons for that are need to be studied. In this study, we sought to determine the possible factors affecting the pathogenicity of Pcc and Pch in Oncidium Gower Ramsey. Results showed that both Pcc and Pch are pathogenic to Oncidium orchid and two other common hosts of soft rot bacteria, potato and radish. However, Pch is more virulent in Oncidium orchid than Pcc. Moreover, temperature is not the dominant factor contributing to differences in soft rot development in Oncidium caused by Pcc and Pch, suggesting that host selection is more important than temperature. We further found that Pcc growth is not inhibited by Oncidium leaf extract and that the inhibitory effect of Oncidium on Pcc may be attributed to the defense response of living cells. In addition, stronger reactive oxygen species accumulation was triggered by Pcc rather than Pch. This defense response against Pcc is related to plant immune signal pathways, including calcium influx and NADPH oxidase activation. Through this study, we have provided evidences that Pch, not Pcc, is the dominant soft rot pathogen in Oncidium. The immune response triggered by bacterial recognition explains the inhibition of Pcc growth in Oncidium Gower Ramsey.

The NGM medium developed in a previous study was used for differential isolation of Pectobacterium chrysanthemi, Burkholderia gladioli, and B. glumae. P. chrysanthemi developed blue colonies, and all B. gladioli and B. glumae strains tested produced diffusible yellow pigments on the NGM medium, easily distinguishable from other Burkholderia spp. and plant pathogenic bacteria. The produced yellow pigments contained a toxoflavin determined by the TLC and orchid leaf chlorosis tests. A specific oligonucleotide primer pair was designed for the detection of toxA, which is involved in toxoflavin biosynthesis. All B. gladioli and B. glumae strains tested contained toxA as determined by PCR amplification. No amplification was observed with other plant pathogenic bacteria. In addition, the toxA-based species-specific PCR assays, based on the nucleotide sequence differences in the promoter region of toxA, were developed for identification of B. gladioli and B. glumae, respectively. The NGM medium and the toxA-based PCR assays were used to determine the causal agents of leaf rot of Phalaenopsis and Oncidium orchids at three cultivation areas in Taiwan. It was found that both P. chrysanthemi and B. gladioli are important pathogenic bacteria of orchid leaf rot in Taiwan. The results indicate that the combination of NGM medium with toxA-based PCR assays is a newly designed and efficient method for isolation and identification of leaf rot pathogenic bacteria especially from plant hosts on which P. chrysanthemi and B. gladioli (or B. glumae) could cause symptoms.

Bacterial pathogens need to scavenge iron from their host for growth and proliferation during infection. They have evolved several strategies to do this, one being the biosynthesis and excretion of small, high-affinity iron chelators known as siderophores. The biosynthesis of siderophores is an important area of study, not only for potential therapeutic intervention but also to illuminate new enzyme chemistries. Two general pathways for siderophore biosynthesis exist: the well-characterized nonribosomal peptide synthetase (NRPS)-dependent pathway and the NRPS-independent siderophore (NIS) pathway, which relies on a different family of sparsely investigated synthetases. Here we report structural and biochemical studies of AcsD from Pectobacterium (formerly Erwinia) chrysanthemi, an NIS synthetase involved in achromobactin biosynthesis. The structures of ATP and citrate complexes provide a mechanistic rationale for stereospecific formation of an enzyme-bound (3R)-citryladenylate, which reacts with L-serine to form a likely achromobactin precursor. AcsD is a unique acyladenylate-forming enzyme with a new fold and chemical catalysis strategy.

Erwinia chrysanthemi 3937 is a soft-rotting plant pathogen in Enterobacteriaceae. It attacks a wide range of plant host species. Previously, we identified dozens of E. chrysanthemi 3937 genes induced during plant infection by microarray differential display. Here, we have mutated plant-upregulated and putatively plant-upregulated genes in E. chrysanthemi 3937 using a transposon insertion method. Of 57 mutants produced, 8 were significantly reduced in maceration in African violet leaves. These 8 E. chrysanthemi genes are similar to Escherichia coli purU (formyltetrahydrofolate deformylase; ASAP20623) and wcaJ (undecaprenylphosphate glucosephosphotransferase; ASAP18556), Bacillus subtilis dltA (d-alanine-d-alanyl carrier protein ligase; ASAP19406), Pseudomonas syringae PSPTO2912 (ABC transporter, periplasmic glutamine-binding protein; ASAP15639), Pseudomonas aeruginosa pheC (cyclohexadienyl dehydratase; ASAP19773), P. syringae syrE (peptide synthase; ASAP19989), Vibrio vulnificus VV12303 (unknown protein; ASAP18555), and Yersinia pestis speD (S-adenosylmethionine decarboxylase; ASAP20536). In some of the genes, possible roles in virulence could be postulated based on the functions of their homologues. This work demonstrated that a low proportion of pathogenicity-related genes were among the plant-upregulated genes of E. chrysanthemi 3937. This study and further dissection of these putative virulence genes should lead to new insights into infection mechanisms in pathogens.

Pectobacterium chrysanthemi PY35 secretes the endoglucanase Cel5Z, an enzyme of the glycoside hydrolase family 5. Cel5Z is a 426 amino acid, signal peptide (SP)-containing protein composed of two domains: a large N-terminal catalytic domain (CD; 291 amino acids) and a small C-terminal cellulose binding domain (CBD; 62 amino acids). These two domains are separated by a 30 amino acid linker region (LR). A truncated cel5Z gene was constructed with the addition of a nonsense mutation that removes the C-terminal region of the protein. A truncated Cel5Z protein, consisting of 280 amino acid residues, functioned as a mature enzyme despite the absence of the SP, 11 amino acid CD, LR, and CBD region. In fact, this truncated Cel5Z protein showed an enzymatic activity 80% higher than that of full-length Cel5Z. However, cellulase activity was undetectable in mature Cel5Z proteins truncated to less than 280 amino acids.

Tachyplesin I is a 2.3 kDa antimicrobial peptide isolated from Southeast Asian horseshoe crabs. Bacterial suspensions containing 1×106 colony-forming units/ml of six isolates of pectolytic Erwinia spp., the causal pathogens of potato soft rot and blackleg, were killed in vitro by 1.4 to 11.1 μg/ml of tachyplesin I. In an attempt to enhance resistance to Erwinia spp., each of the potato cultivars Bintje, Karnico and Kondor were transformed with two gene constructs encoding different precursor tachyplesin I proteins under the control of a cauliflower mosaic virus 35S promotor. Northern and western blot analysis showed that the tachyplesin I gene was expressed in transgenic plants. Small tubers of 17 transgenic clones were screened twice for soft rot resistance to Erwinia carotovora ssp. atroseptica. Under aerobic or anaerobic conditions, transgenic clones showed slightly less rot than control tubers.

Under iron-depleted conditions, bacteria produce siderophores that bind iron and are then actively taken up by the cell. New structural and biochemical insights are reported for the synthetic pathway of achromobactin, a siderophore from the plant pathogen Pectobacterium chrysanthemi.
