Flowering plants have evolved numerous intraspecific and interspecific prezygotic reproductive barriers to prevent production of unfavourable offspring1. Within a species, self-incompatibility (SI) is a widely utilized mechanism that rejects self-pollen2,3 to avoid inbreeding depression. Interspecific barriers restrain breeding between species and often follow the SI × self-compatible (SC) rule, that is, interspecific pollen is unilaterally incompatible (UI) on SI pistils but unilaterally compatible (UC) on SC pistils1,4–6. The molecular mechanisms underlying SI, UI, SC and UC and their interconnections in the Brassicaceae remain unclear. Here we demonstrate that the SI pollen determinant S-locus cysteine-rich protein/S-locus protein 11 (SCR/SP11)2,3 or a signal from UI pollen binds to the SI female determinant S-locus receptor kinase (SRK)2,3, recruits FERONIA (FER)7–9 and activates FER-mediated reactive oxygen species production in SI stigmas10,11 to reject incompatible pollen. For compatible responses, diverged pollen coat protein B-class12–14 from SC and UC pollen differentially trigger nitric oxide, nitrosate FER to suppress reactive oxygen species in SC stigmas to facilitate pollen growth in an intraspecies-preferential manner, maintaining species integrity. Our results show that SRK and FER integrate mechanisms underlying intraspecific and interspecific barriers and offer paths to achieve distant breeding in Brassicaceae crops.

Purpose The physics design of the High Energy Photon Source (HEPS) was finished after many times of iteration. Hereby, the typical equilibrium electron beam parameters corresponding to the proposed two baseline operation modes in the baseline design of HEPS are presented. Methods To compute the equilibrium parameters of the electron beam, the lattice parameters, RF parameters, and the parameters of the insertion devices (IDs) were determined first. Furthermore, it is more precise to use the full-current electron beam parameters in the estimations of the performance of the synchrotron light. Therefore, not only the single-particle dynamics but also the current-dependent collective effects need to be considered in the computations of the full-current, equilibrium parameters of the electron beam. Both analytic computations and multi-particle tracking simulations were carried out. Results The full-current, equilibrium parameters of the electron beams in the HEPS storage ring are presented in this paper. Moreover, the main beam parameters in the injector (the booster and the LINAC), corresponding to the two baseline operation modes of the storage ring, are also presented. Conclusion The typical electron beam parameters corresponding to the two baseline operation modes are given in detail in this paper.

Cancer stem cells or cancer stemness has been confirmed to a major obstacle for glioma progression and it has also been reported that circRNAs play an important part in cancer progression. This study mainly focuses on revealing the role of circRPPH1 and the underlying mechanisms in glioma cell stemness.In vitro experiment including RT-qPCR, Western blot, sphere-formation analysis, and ALDH1 activity, and in vivo tumorigenesis experiments were performed to evaluate the effects of circRPPH1 on glioma cell stemness. Luciferase reporter, ChIP, and DNA pull-down analysis were used to reveal the underlying mechanisms.It was found that circRPPH1 level was upregulated in glioma cell spheres and facilitated the stemness of glioma cells; C-FOS transcriptionally activated circRPPH1 expression via directly binding to circRPPH1 promoter in glioma cells. Moreover, circRPPH1 promoted the stemness of glioma cells dependent on c-FOS-mediated transcriptional activation.This study indicates that c-Fos-activated circRPPH1 contributes to glioma stemness and provides a potential target for glioma progression based on the c-FOS/circRPPH1 regulatory axis.

Self-assembly of particle-like dissipative solitons, in the presence of mutual interactions, emphasizes the vibrant concept of soliton molecules in varieties of laser resonators. Controllable manipulation of the molecular patterns, held by the degrees of freedom of internal motions, still remains challenging to explore more efficient and subtle tailoring approaches for the increasing demands. Here, we report a new phase-tailored quaternary encoding format based on the controllable internal assembly of dissipative soliton molecules. Artificial manipulation of the energy exchange of soliton-molecular elements stimulates the deterministic harnessing of the assemblies of internal dynamics. Self-assembled soliton molecules are tailored into four phase-defined regimes, thus constituting the phase-tailored quaternary encoding format. Such phase-tailored streams are endowed with great robustness and are resistant to significant timing jitter. All these results experimentally demonstrate the programmable phase tailoring and exemplify the application of the phase-tailored quaternary encoding, prospectively promoting high-capacity all-optical storage.

With the advent of the era of big data, privacy computing analyzes and calculates data on the premise of protecting data privacy, to achieve data ‘available and invisible’. As an important branch of secure multi-party computation, the geometric problem can solve practical problems in the military, national defense, finance, life, and other fields, and has important research significance. In this paper, we study the similarity problem of geometric graphics. First, this paper proposes the adjacency matrix vector coding method of isomorphic graphics, and use the Paillier variant encryption cryptography to solve the problem of isomorphic graphics confidentiality under the semi-honest model. Using cryptography tools such as elliptic curve cryptosystem, zero-knowledge proof, and cut-choose method, this paper designs a graphic similarity security decision protocol that can resist malicious adversary attacks. The analysis shows that the protocol has high computational efficiency and has wide application value in terrain matching, mechanical parts, biomolecules, face recognition, and other fields.

Simultaneous localization and mapping (SLAM) is considered to be an important way for some smart devices to perform automatic path planning, and many successful SLAM systems have been developed in the past few years. Most existing approaches rely heavily on static world assumptions, and such strong assumptions limit the application of most vSLAM (visual SLAM) in complex dynamic reality environments, where dynamic objects often lead to incorrect data association in tracking, which reduces the overall accuracy and robustness of the system and causes tracking crashes. The dynamic objects in the map may change over time; thus, distinguishing dynamic information in a scene is challenging. In order to solve the interference problem of dynamic objects, most point-based visual odometry algorithms have concentrated on feature matching or direct pixel intensity matching, disregarding an ordinary but crucial image entity: geometric information. In this article, we put forward a novel visual odometry algorithm based on dynamic point detection methods called geometric prior and constraints. It removes the moving objects by combining the spatial geometric information of the image and depends on the remaining features to estimate the position of the camera. To the best of our knowledge, our proposed algorithm achieves superior performance over existing methods on a variety of public datasets.

The recurrence of atrial fibrillation (AF) after cryoablation still needs to be prioritized, including discriminating predictive indicators.Eighty-seven patients aged 43-83 years who underwent cryo-balloon ablation were divided into paroxysmal atrial fibrillation (PAF) and non-paroxysmal atrial fibrillation (non-PAF) groups. Baseline data, intraoperative index, and miRNA21, IL-18, NLRP3, and visfatin levels in peripheral venous blood and left atrial blood were assessed. Follow-up was performed for 6 months to observe the recurrence of AF. A Cox risk ratio model was used to analyze indicators for predicting AF recurrence.The non-PAF and PAF group recurrence rates of AF were statistically different (p 0.05) at 9/22 (40.9%) and 11/65 (16.9%), respectively. Biomarker levels in the left atrial blood were higher in the non-PAF group than in the PAF group (p 0.05). The effects of non-PAF and levels of miRNA21 and IL-18 in left atrial serum on the recurrence of AF after cryoablation statistically differed (p 0.05).The levels of miRNA21 and IL-18 were higher in left atrial blood than in peripheral blood, which may be related to the severity of AF and recurrence of AF after cryoablation.

To evaluate the validity of the Roussouly classification system for assessing distal junctional problems (DJP) after long instrumented spinal fusion in degenerative scoliosis.Sixty-four patients with degenerative scoliosis and long-segment fixation receiving treatment at our hospital between December 2012 and December 2018 were retrospectively analyzed. Patients were classified preoperatively and postoperatively (Roussouly classification) and divided into DJP and control groups. We observed whether patients restored to their preoperative Roussouly classification (based on pelvic incidence [PI]) postoperatively.The incidences of DJP were 11.11% and 50% in patients who did and did not match their sagittal Roussouly classification immediately postoperatively, respectively. The adjusted Chi-square test that showed whether the sagittal profile matched the Roussouly classification immediately after surgery was statistically significant (P = 0.012). PIs were 55.83 ± 4.94 and 47.21 ± 10.81 in the DJP and non-DJP groups, respectively (t' = 4.367, P 0.001). Distal junctional kyphosis angles were 6.33 ± 4.19° and 11.56 ± 5.02° in the DJP and non-DJP groups, respectively (t = - 2.595, P = 0.015). Preoperative PI-lumbar lordosis values were 29.14 ± 13.82 and 16.67 ± 11.39 in the DJP and non-DJP groups, respectively (t = - 2.626, P = 0.013). The logistic regression model showed that patients whose Roussouly classification did not match the postoperative PI value were more likely to have DJP (odds ratio [OR] = 4.01, 95% confidence interval [CI]: 0.51-31.61) and preoperative distal junctional kyphotic changes.If the postoperative sagittal profile can be restored to match the patient's own PI value, use of the Roussouly classification can greatly reduce the possibility of postoperative DJP.

Background The gibberellic acid-stimulated Arabidopsis (GASA) gene encodes a class of cysteine-rich functional proteins and is ubiquitous in plants. Most GASA proteins are influence the signal transmission of plant hormones and regulate plant growth and development, however, their function in Jatropha curcas is still unknown. Results In this study, we cloned JcGASA6, a member of the GASA family, from J. curcas. The JcGASA6 protein has a GASA-conserved domain and is located in the tonoplast. The three-dimensional structure of the JcGASA6 protein is highly consistent with the antibacterial protein Snakin-1. Additionally, the results of the yeast one-hybrid (Y1H) assay showed that JcGASA6 was activated by JcERF1, JcPYL9, and JcFLX. The results of the Y2H assay showed that both JcCNR8 and JcSIZ1 could interact with JcGASA6 in the nucleus. The expression of JcGASA6 increased continuously during male flower development, and the overexpression of JcGASA6 was associated with filament elongation of the stamens in tobacco. Conclusion JcGASA6, a member of the GASA family in J. curcas, play an important role in growth regulation and floral development (especially in male flower). It is also involved in the signal transduction of hormones, such as ABA, ET, GA, BR, and SA. Also, JcGASA6 is a potential antimicrobial protein determined by its three-dimensional structure.

Plasma cell-free DNA (cfDNA) are small molecules generated through a non-random fragmentation procedure. Despite commendable translational values in cancer liquid biopsy, however, the biology of cfDNA, especially the principles of cfDNA fragmentation, remains largely elusive. Through orientation-aware analyses of cfDNA fragmentation patterns against the nucleosome structure and integration with multidimensional functional genomics data, here we report a DNA methylation – nuclease preference – cutting end – size distribution axis, demonstrating the role of DNA methylation as a functional molecular regulator of cfDNA fragmentation. Hence, low-level DNA methylation could increase nucleosome accessibility and alter the cutting activities of nucleases during DNA fragmentation, which further leads to variation in cutting sites and size distribution of cfDNA. We further develop a cfDNA ending preference-based metric for cancer diagnosis, whose performance has been validated by multiple pan-cancer datasets. Our work sheds light on the molecular basis of cfDNA fragmentation towards broader applications in cancer liquid biopsy.

Pressure relief borehole is one of the most effective methods to treat coal burst in deep-buried coal seams. However, due to the large amount of randomly distributed fractures in the coal body and the varying stress environments in underground coal mines, there is no practical method to assess relief boreholes by far, which brings problems for borehole design and construction. This study focuses on the assessment of relief boreholes in a single driving entry wherein the impact of mining panels on relief boreholes is eliminated. A practical method, focusing on field monitoring of borehole deformation with a new device, is developed to evaluate the single driving entry’s relief boreholes. This method is applied to a Chinese coal mine. The application results showed that the convergence process of the boreholes showed a three-phase characteristic. Phase one involves the fastest deformation, reaching up to 8% in the first 3 to 4 days. Phase two lasts longer and converges more, reaching up to 30% in 4 months. Phase three stays balanced until the excavating face moves. The three-phase characteristic indicates that a large diameter of the borehole ensures relief efficiency because the borehole provides adequate space for deformation throughout its life span. In addition, it is verified that long boreholes and slow footage improve relief efficiency by providing adequate time for the boreholes to work. The application proposes that boreholes in the test coal mine need to be drilled larger and longer for optimization.

Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is the ongoing global pandemic that poses substantial challenges to public health worldwide. A subset of COVID-19 patients experience systemic inflammatory response, known as cytokine storm, which may lead to death. Receptor-interacting serine/threonine-protein kinase 1 (RIPK1) is an important mediator of inflammation and cell death. Here, we examined the interaction of RIPK1-mediated innate immunity with SARS-CoV-2 infection. We found evidence of RIPK1 activation in human COVID-19 lung pathological samples, and cultured human lung organoids and ACE2 transgenic mice infected by SARS-CoV-2. Inhibition of RIPK1 using multiple small-molecule inhibitors reduced the viral load of SARS-CoV-2 in human lung organoids. Furthermore, therapeutic dosing of the RIPK1 inhibitor Nec-1s reduced mortality and lung viral load, and blocked the CNS manifestation of SARS-CoV-2 in ACE2 transgenic mice. Mechanistically, we found that the RNA-dependent RNA polymerase of SARS-CoV-2, NSP12, a highly conserved central component of coronaviral replication and transcription machinery, promoted the activation of RIPK1. Furthermore, NSP12 323L variant, encoded by the SARS-CoV-2 C14408T variant first detected in Lombardy, Italy, that carries a Pro323Leu amino acid substitution in NSP12, showed increased ability to activate RIPK1. Inhibition of RIPK1 downregulated the transcriptional induction of proinflammatory cytokines and host factors including ACE2 and EGFR that promote viral entry into cells. Our results suggest that SARS-CoV-2 may have an unexpected and unusual ability to hijack the RIPK1-mediated host defense response to promote its own propagation and that inhibition of RIPK1 may provide a therapeutic option for the treatment of COVID-19.

There are numerous possible implications of nanotechnology in cancer therapy. To improve the therapeutic benefits of anticancer drugs nano-delivery systems (NDS) have been established with significant solubility, degradability and cytocompatibility. These NDS are the result of several advantages that high-performance nanoparticles hydroxyapatite (HA) advance rapidly in specific cancer treatments. Salinomycin is a pharmaceutical drug that serves to suppress tumour cells and the growth of many cancers as a chemical inhibitor. Nevertheless, Salinomycin’s low bioavailability is the main challenge to be used. In this work, HA chitosan-coated nanoparticles were utilised to enhance the efficiency and availability of Salinomycin as a pH-sensitive biopolymer. HA nanoparticles were initially produced using the sol-gel technique for this purpose. Then a coating of chitosan was coated with the nanocarrier under regulated circumstances, and the salinomycin medicine was encapsulated. For the characterization of nanocarriers were employed techniques such as scanning electron microscopy, X-ray diffraction, and Fourier transformations infrared spectroscopy. Different bioassays have tested nano-drugs developed in the second part. In this connection, we examined the cytotoxicity rate of the hepatocellular carcinoma HepG2 and Caco-2 by the MTT assay and the apoptosis induction rate by ethidium bromide and acridine orange staining method. It has been established that histological analysis is very suitable for nanomedicine, which does not cause apparent stress like swelling and inflammation organs of the mouse. The findings could offer solid evidence for nanoparticles’ apoptotic cell death mechanism and a novel cancer therapy approach.

In order to reveal the evolution of ceiling jet temperature characteristics beneath curved ceiling in two-dimensional view, experimental and theoretical studies were conducted to analyze the vertical temperature profile and longitudinal temperature attenuation beneath a curved ceiling of a semicircular tunnel. The temperature profile was first measured from experiments carried out in the model-scale semicircular tunnel. The relationship between temperature and the horizontal positions is described by an empirical model. A formula describing the thickness of the thermal boundary layer is proposed, based on the vertical distribution of smoke temperature. An engineering model for the longitudinal attenuation of the gas temperature with different fire plume types at impingement region is established in terms of conservation equations. After validation, model predictions show a good agreement with trend proposed by former literature and the relative error of prediction is found to be within 10%. Two-dimensional prediction for temperature profile beneath semicircle ceiling centerline was proposed. It was noted that two-dimensional prediction model could predict majority of measured data within 20% relative error even though some fluctuation, which could be applied to the fire protection engineering in semicircular tunnel.

The production of electricity is important, suitable and secure for human living, yet electricity is actually generated mainly from fossil fuels and nuclear energy, calling for renewable energies such as solar, wind and tidal renewable energies such as solar, wind and tidal. Solar energy is broadly harvested by various types of solar cells. Three-dimensional perovskite solar cell exhibits high power conversion efficiency of 25.2% with low stability, whereas two-dimensional perovskite solar cell exhibits better stability with moderate power conversion efficiency. Hence, we review two-dimensional perovskite solar cells fabricated with varying numbers of hybrid two-dimensional perovskite layers, organic cations, deposition techniques, the addition of additives and capping layers to improve power conversion efficiency with long-term better stability.

Abstract Carascynol A, an unprecedented 4/9/8 ring system hybrid with a peroxide bridge, was characterized from Hypericum ascyron. The architecture contains a caryophyllane-type moiety and a C6 unit derived from polyprenylated acylphloroglucinols. Its structure and absolute configuration were determined by comprehensive spectroscopic and X-ray diffraction data. Biologically, compound 1 inhibited cell proliferation in LoVo, SW480, and HCT116 cell lines (IC50 = 12.30–24.57 µM). Graphical Abstract

Xenograft bone scaffolds have certain advantages such as mechanical strength, osteoinductive properties, sufficient source and safety. This study aimed to compare osteogenesis of the two main bovine bone xenografts namely true bone ceramics (TBC) and decalcified bone matrix (DBM), and TBC or DBM combined with bone morphogenetic protein (BMP)-2 (TBC&BMP-2 and DBM&BMP-2). The characteristics of TBC and DBM were investigated by observing the appearance and scanning electron microscopic images, examining mechanical strength, evaluating cytotoxicity and detecting BMP-2 release after being combined with BMP-2 in vitro. The femoral condyle defect and radial defect models were successively established to evaluate the performance of the proposed scaffolds in repairing cortical and cancellous bone defects. General observation, hematoxylin and eosin (HE) staining, mirco-CT scanning, calcein double labeling, X-ray film observation, three-point bending test in vivo were then performed. It indicated that the repair with xenograft bone scaffolds of 8 weeks were needed and the repair results were better than those of 4 weeks whatever the type of defects. To femoral condyle defect, TBC and TBC&BMP-2 were better than DBM and DBM&BMP-2, and TBC&BMP-2 was better than TBC alone; to radial defect, DBM and DBM&BMP-2 were better than TBC and TBC&BMP-2, and DBM&BMP-2 was better than DBM alone. This study has shown that TBC and DBM xenograft scaffolds can be more suitable for the repair of cancellous bone and cortical bone defects for 8 weeks in rats, respectively. We also have exhibited the use of BMP-2 in combination with DBM or TBC provides the possibility to treat bone defects more effectively. We thus believe that we probably need to select the more suitable scaffold according to bone defect types, and both TBC and DBM are promising xenograft materials for bone tissue engineering and regenerative medicine.

The Coronavirus Disease 2019 (COVID-19) caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) is a global pandemic that seriously threatens health and socioeconomic development, but the existed antiviral drugs and vaccines still cannot yet halt the spread of the epidemic. Therefore, a comprehensive and profound understanding of the pathogenesis of SARS-CoV-2 is urgently needed to explore effective therapeutic targets. Here, we conducted a multiomics study of SARS-CoV-2-infected lung epithelial cells, including transcriptomic, proteomic, and ubiquitinomic. Multiomics analysis showed that SARS-CoV-2-infected lung epithelial cells activated strong innate immune response, including interferon and inflammatory responses. Ubiquitinomic further reveals the underlying mechanism of SARS-CoV-2 disrupting the host innate immune response. In addition, SARS-CoV-2 proteins were found to be ubiquitinated during infection despite the fact that SARS-CoV-2 itself didn’t code any E3 ligase, and that ubiquitination at three sites on the Spike protein could significantly enhance viral infection. Further screening of the E3 ubiquitin ligases and deubiquitinating enzymes (DUBs) library revealed four E3 ligases influencing SARS-CoV-2 infection, thus providing several new antiviral targets. This multiomics combined with high-throughput screening study reveals that SARS-CoV-2 not only modulates innate immunity, but also promotes viral infection, by hijacking ubiquitination-specific processes, highlighting potential antiviral and anti-inflammation targets.

Intertwining quantum order and non-trivial topology is at the frontier of condensed matter physics1–4. A charge-density-wave-like order with orbital currents has been proposed for achieving the quantum anomalous Hall effect5,6 in topological materials and for the hidden phase in cuprate high-temperature superconductors7,8. However, the experimental realization of such an order is challenging. Here we use high-resolution scanning tunnelling microscopy to discover an unconventional chiral charge order in a kagome material, KV3Sb5, with both a topological band structure and a superconducting ground state. Through both topography and spectroscopic imaging, we observe a robust 2 × 2 superlattice. Spectroscopically, an energy gap opens at the Fermi level, across which the 2 × 2 charge modulation exhibits an intensity reversal in real space, signalling charge ordering. At the impurity-pinning-free region, the strength of intrinsic charge modulations further exhibits chiral anisotropy with unusual magnetic field response. Theoretical analysis of our experiments suggests a tantalizing unconventional chiral charge density wave in the frustrated kagome lattice, which can not only lead to a large anomalous Hall effect with orbital magnetism, but also be a precursor of unconventional superconductivity. An unconventional chiral charge order is observed in a kagome superconductor by scanning tunnelling microscopy. This charge order has unusual magnetic tunability and intertwines with electronic topology.

Background 21q22 amplification is a rare cytogenetic aberration in acute myeloid leukemia (AML). So far, the cytogenomic and molecular features and clinical correlation of 21q22 amplification in AML have not been well-characterized. Case presentation Here, we describe a case series of three AML patients with amplified 21q22 identified by fluorescence in situ hybridization using a RUNX1 probe. Two of these patients presented with therapy-related AML (t-AML) secondary to chemotherapy, while the third had de novo AML. There was one case each of FAB M0, M1 and M4. Morphologic evidence of dysplasia was identified in both t-AML cases. Phenotypic abnormalities of the myeloblasts were frequently observed. Extra copies of 21q22 were present on chromosome 21 and at least one other chromosome in two cases. Two showed a highly complex karyotype. Microarray analysis of 21q22 amplification in one case demonstrated alternating levels of high copy number gain split within the RUNX1 locus at 21q22. The same patient also had mutated TP53. Two patients died at 1.5 and 11 months post-treatment, while the third elected palliative care and died within 2 weeks. Conclusions Our results provide further evidence that 21q22 amplification in AML is associated with complex karyotypes, TP53 aberrations, and poor outcomes. Furthermore, we demonstrate that 21q22 amplification is not always intrachromosomally localized to chromosome 21 and could be a result of structural aberrations involving 21q22 and other chromosomes.

We mainly proceed from the view of biological effect to study the acellular bovine bone matrix (ABBM) by the low concentration of hydrogen oxidation. After cleaning the bovine bone routinely, it was cleaned with different concentrations of NaOH and stained with hematoxylin-eosin (HE) to observe the effect of decellulization. The effect of bovine bone matrix treated with NaOH were observed by optical microscopy and scanning electron microscopy (SEM), and compared by DNA residue detection. Cell toxicity was also evaluated in MC3T3-E1 cells by CCK-8. For the in vitro osteogenesis detection, alkaline phosphatase (ALP) staining and alizarin red (AR) staining were performed in MC3T3-E1 cells. And the in vivo experiment, Micro CT, HE and Masson staining were used to observe whether the osteogenic effect of the materials treated with 1% NaOH solution was affected at 6 and 12 weeks. After the bovine bone was decellularized with different concentrations of NaOH solution, HE staining showed that ultrasonic cleaning with 1% NaOH solution for 30 min had the best effect of decellularization. The SEM showed that ABBM treated with 1% NaOH solution had few residual cells on the surface of the three-dimensional porous compared to ABBM treated with conventional chemical reagents. DNA residues and cytotoxicity of ABBM treated with 1% NaOH were both reduced. The results of ALP staining and AR staining showed that ABBM treated with 1% NaOH solution had no effect on the osteogenesis effect. The results of micro-CT, HE staining and Masson staining in animal experiments also showed that ABBM treated with 1% NaOH solution had no effect on the osteogenesis ability. The decellularization treatment of ABBM with the low concentration of NaOH can be more cost-effective, effectively remove the residual cellular components, without affecting the osteogenic ability. Our work may provide a novelty thought and a modified method to applicate the acellular bovine bone matrix clinically better.

Diabetes is an urgent health issue characterized by ethnic and regional variations, and is inseparable from the different dietary habits. It is worthy to note that the incidence of diabetes in Bai nationality has been reported to be much lower than Han in China. As a daily vegetable of Bai, the phytochemical and antidiabetic study of Ottelia acuminata var. acuminata had not been carried out. In this study, 41 metabolites with diverse diarylheptanoid (six new ones, Otteacumienes A–F), flavone, sesquiterpenoid, coumarin, lignan, polyacetylene, and alkaloid skeletons were characterized from O. acuminata var. acuminata. Among them, the racemic nature of 3 was characterized by chiral resolution and calculated ECD methods. The biological study revealed diarylheptanoids showed significant α-glucosidase inhibitory activities with 5 as the most effective one (60-fold stronger than acarbose). Molecular docking studies indicated that these structures have different binding cavities with acarbose. This study demonstrated that O. acuminata var. acuminata might correlated with the low incidence diabetes of Bai and the diarylheptanoids may have potential therapeutic value for diabetes mellitus. The degradation of pyrazinamide (PZA) by electron beam irradiation (EBI) was studied. It was found that the degradation of PZA was efficient with the removal rate of 99% for 0.2 mM PZA under 5 kGy, and the degradation kinetics followed pseudo-first-order kinetics. The presence of CO32−, HCO3−, NO3−, and fulvic acid inhibited the degradation of PZA. The addition of H2O2 or K2S2O8 effectively enhanced the degradation and mineralization of PZA. The scavenger experiments and quantum chemical calculations showed that ·OH was the primary reactive species in the degradation. The identification of intermediates and quantum chemical calculations illustrated the mechanism of degradation.
