Microbial electrosynthesis exploits the catalytic activity of microorganisms to utilize a cathode as an electron donor for reducing waste CO2 to valuable fuels and chemicals. Electromethanogenesis is the process of CO2 reduction to CH4 catalyzed by methanogens using the cathode directly as a source of electrons or indirectly via H2. Understanding the effects of different set cathode potentials on the functional dynamics of electromethanogenic communities is crucial for the rational design of cathode materials. Replicate enriched electromethanogenic communities were subjected to different potentials (− 1.0 V and − 0.7 V vs. Ag/AgCl) and the potential-induced changes were analyzed using a metagenomic and metatranscriptomic approach. The most abundant and transcriptionally active organism on the biocathodes was a novel species of Methanobacterium sp. strain 34x. The cathode potential-induced changes limited electron donor availability and negatively affected the overall performance of the reactors in terms of CH4 production. Although high expression of key genes within the methane and carbon metabolism pathways was evident, there was no significant difference in transcriptional response to the different set potentials. The acetyl-CoA decarbonylase/synthase (ACDS) complex were the most highly expressed genes, highlighting the significance of carbon assimilation under limited electron donor conditions and its link to the methanogenesis pathway.

Microbial uptake of free cathodic electrons presents a poorly understood aspect of microbial physiology. Uptake of cathodic electrons is particularly important in microbial electrosynthesis of sustainable fuel and chemical precursors using only CO2 and electricity as carbon, electron and energy source. Typically, large overpotentials (200 to 400 mV) were reported to be required for cathodic electron uptake during electrosynthesis of, for example, methane and acetate, or low electrosynthesis rates were observed. To address these limitations and to explore conceptual alternatives, we studied defined co-cultures metabolizing cathodic electrons. The Fe(0)-corroding strain IS4 was used to catalyze the electron uptake reaction from the cathode forming molecular hydrogen as intermediate, and Methanococcus maripaludis and Acetobacterium woodii were used as model microorganisms for hydrogenotrophic synthesis of methane and acetate, respectively. The IS4-M. maripaludis co-cultures achieved electromethanogenesis rates of 0.1–0.14 μmol cm−2 h−1 at −400 mV vs standard hydrogen electrode and 0.6–0.9 μmol cm−2 h−1 at −500 mV. Co-cultures of strain IS4 and A. woodii formed acetate at rates of 0.21–0.23 μmol cm−2 h−1 at −400 mV and 0.57–0.74 μmol cm−2 h−1 at −500 mV. These data show that defined co-cultures coupling cathodic electron uptake with synthesis reactions via interspecies hydrogen transfer may lay the foundation for an engineering strategy for microbial electrosynthesis.

Direct, shuttle-free uptake of extracellular, cathode-derived electrons has been postulated as a novel mechanism of electron metabolism in some prokaryotes that may also be involved in syntrophic electron transport between two microorganisms. Microbial electrocatalysis is a relatively new field of research in which the intrinsic metabolic capacities of various microbes are coupled with inorganic electrodes to carry out interesting chemical conversions. Given the great diversity in microbial metabolic pathways, a wide variety of processes are possible and have been demonstrated in principle. The generation of electrical currents coupled with the degradation of wastes or the capture of light energy is under extensive investigation. This area has seen the greatest development with an over tenfold increase in power densities in the past decade. A relatively new development is electrosynthesis, the electrically driven fixation of CO2 into various chemicals. Moreover, microbial electrochemical devices can be used to carry out desalination or “unbalanced” chemical conversions. Microbial electrocatalysis has the advantages of the exquisite specificity and regioselectivity of biochemical reactions coupled with the robustness and self-duplicating properties of living systems. Here, recent advances in this area are reviewed with significant achievements highlighted. As well, the major factors limiting practical application are discussed along with future directions for improvement.

To decompose carbon dioxide, which is a representative greenhouse gas, a 3-phase gliding arc plasmatron device was designed and manufactured to examine the decomposition of CO2, either alone or in the presence of methane with and without water vapour. The changes in the amount of carbon dioxide feed rate, the methane to carbon dioxide ratio, the steam to carbon dioxide ratio, and the methane to steam ratio were used as the parameters. The carbon dioxide conversion rate, energy decomposition efficiency (EDE), carbon monoxide and hydrogen selectivity, and produced gas concentration were also investigated. The maximum values of the carbon dioxide conversion rate, which is a key indicator of carbon dioxide decomposition, in different cases were compared. The maximum carbon dioxide conversion rate was 12.3 % when pure carbon dioxide was supplied; 34.5 % when methane was injected as a reforming additive; 7.8 % when steam was injected as a reforming additive; and 43 % when methane and steam were injected together. Therefore, this could be explained that the methane-and-steam injection showed the highest carbon dioxide decomposition, showing low EDE as 0.01 L/min W. Furthermore, the plasma produced carbon-black was compared with commercial carbon-black chemicals through Raman spectroscopy, surface area measurement and scanning electron microscopy. It was found that the carbon-black that was produced in this study has the high conductivity and large specific surface area. Our product makes it suitable for special electric materials and secondary battery materials applications.

Microbial electrocatalysis relies on microorganisms as catalysts for reactions occurring at electrodes. Microbial fuel cells and microbial electrolysis cells are well known in this context; both use microorganisms to oxidize organic or inorganic matter at an anode to generate electrical power or H(2), respectively. The discovery that electrical current can also drive microbial metabolism has recently lead to a plethora of other applications in bioremediation and in the production of fuels and chemicals. Notably, the microbial production of chemicals, called microbial electrosynthesis, provides a highly attractive, novel route for the generation of valuable products from electricity or even wastewater. This Review addresses the principles, challenges and opportunities of microbial electrosynthesis, an exciting new discipline at the nexus of microbiology and electrochemistry.

A coupling process of anaerobic methanogenesis and electromethanogenesis was proposed to treat high organic load rate (OLR) wastewater. During the start-up stage, acetate removal efficiency of the electric-biological reactor (R1) reached the maximization about 19 percentage points higher than that of the control anaerobic reactor without electrodes (R2), and CH4 production rate of R1 also increased about 24.9% at the same time, while additional electric input was 1/1.17 of the extra obtained energy from methane. Coulombic efficiency and current recorded showed that anodic oxidation contributed a dominant part in degrading acetate when the metabolism of methanogens was low during the start-up stage. Along with prolonging operating time, aceticlastic methanogenesis gradually replaced anodic oxidation to become the main pathway of degrading acetate. When the methanogens were inhibited under the acidic conditions, anodic oxidation began to become the main pathway of acetate decomposition again, which ensured the reactor to maintain a stable performance. FISH analysis confirmed that the electric field imposed could enrich the H2/H(+)-utilizing methanogens around the cathode to help for reducing the acidity. This study demonstrated that an anaerobic digester with a pair of electrodes inserted to form a coupling system could enhance methanogenesis and reduce adverse impacts.

The existing methods for ultrapurification of carbon dioxide and methane often suffer from a waste of time caused by the use of reagents requiring high temperatures and repeated regeneration.
