Attractive Toxic Bait (ATB) is a low risk practical method to cockroach control. This study aimed to evaluate efficacy of a newly modified eight-chamber-olfactometer for attractiveness experiments. Different stages of German cockroaches were exposed to carbohydrate-rich and protein-rich foods and their attractions were tested both at ambient conditions and in the modified olfactometer. In the latter method, cockroaches were released into the main chamber of olfactometer and their attraction to food compounds placed in test chambers were recorded as the number of captured cockroaches in test chambers. The direct attractiveness test was performed in plastic pans. Repellency of fipronil insecticide was performed with prepared bait powder using olfactometer. Data were subjected to analysis of variance with Tukey’s Test,P ≤ 0.05. Different notes should be considered when using introduced olfactometer like precision when opening the test chambers valves, the fan speed, placement of cardboard shelter in both test and main chambers, and initial starvation of cockroaches. Roasted peanut, banana and biscuits powder were the most attractive foods for cockroaches. The females significantly preferred roasted peanut and the males significantly preferred banana and roasted peanut. These results were similar to the results of direct attracted cockroaches. The cockroaches have repellency to fipronil but attractiveness of bait with 0.01% concentration was significantly higher than cockroach diet. Generally, three factors should be considered in formulation of ATB like un-repellency of the a.i, attractiveness of bait compositions, and palatability of bait texture. The olfactometer introduced in this study could be used by researchers to study cockroach behaviors.

Split-brood field studies using the biparental, subsocial wood-feeding cockroach Cryptocercus punctulatus indicate that parental absence in families of three different age classes of nutritionally independent juveniles results in significantly slower growth when compared to sib groups allowed to remain with adults, even when nymphs are more than 2 years old when parents are removed. In one family that was divided when nymphs were 15 months old and followed for 4 years after treatment, 69 % of nymphs allowed to remain with parents reached maturity in the third year of the study, while those isolated from parents remained in the subadult stage at the 3-year mark. In the fourth year, measurements of wet weights and head capsule widths of the newly mature adults in the two treatments of this family were not significantly different. A parental vs. non-parental social environment, then, resulted in different rates of growth to reproductive maturity, but nymphs in both treatments ultimately reached similar adult sizes. The results indicate that parental effects can modify offspring ontogeny independently of the direct transfer of resources in the form of trophallactic food and symbionts. Because cockroaches in this genus are sister group to the developmentally plastic and juvenilized termites, further exploration of the role of the social environment on development in Cryptocercus is warranted. Termites are eusocial cockroaches that exhibit weak or delayed post-embryonic development; the majority of colony members are arrested in the small, soft-bodied, altricial morphotype displayed by early juvenile stages of their subsocial sister group, Cryptocercus. Developmental dynamics in this cockroach genus can therefore provide insight into conditions that may have favored the origin of eusociality in the lineage. Here, we demonstrate that the parental social environment in Cryptocercus punctulatus modifies offspring development independently of the direct transfer of hindgut fluids and does so for more than 2 years after hatch. A system in which parent-offspring interactions modify juvenile ontogeny, whatever the mechanism, contributes to the unique set of circumstances that ultimately resulted in the evolution of termite eusociality.

Many insects find resources by means of the olfactory cues of general odors after learning. To evaluate behavioral responses to the odor of a particular chemical after learning with reward or punishment quantitatively, we developed a standardized odor-training method in the German cockroach, Blattella germanica (Linnaeus), an important urban pest species. A classical olfactory conditioning procedure for a preference test was modified to become applicable to a single odor, by which a (−)-menthol or vanillin odor was independently associated with sucrose (reward) or sodium chloride solution (punishment). The strength of the association with the odor was evaluated with the increase or decrease in visit frequencies to the odor source after olfactory conditioning. The frequency increased after (−)-menthol was presented with a reward, while it did not change with the rewarded vanillin odor. With both odors, the frequency decreased significantly after training with a punishment. These results indicate that cockroaches learn a single compound odor presented as a conditioned stimulus, although the association of the odor with a reward or punishment depends on the chemical. This olfactory conditioning method can not only facilitate the analysis of cockroach behavior elicited by a learned single chemical odor, but also quantify the potential attractiveness or repellency of the chemical after learning.

The mitochondrial genomes are one of the most information-rich markers in phylogenetics. The relationships within superorder Dictyoptera have been debated in the literature. However, the closely related termites (Isoptera) are retained as unranked taxon within the order Blattaria (cockroaches). In this work, we sequenced the complete mitogenomes of two cockroaches, reconstructed the molecular phylogeny and attempted to infer the phylogenetic position of termites in Blattaria more reliably. The complete mtDNA nucleotide sequences of the peridomestic American cockroach (Periplaneta americana L.) and the domestic German cockroach (Blattella germanica L.) are 15,025 and 15,584 bp in size, respectively. The genome shares the gene order and orientation with previously known Blattaria mitogenomes. Most tRNAs could be folded into the typical cloverleaf secondary structure, but the tRNA-Ser (AGN) of P. americana appears to be missing the dihydrouridine arm. Using nucleotide and amino acid sequences as phylogenetic markers, we proposed that termites should be treated as a superfamily (Termitoidea) of cockroaches. We suggested that Polyphagoidea was the sister group of Termitoidea in Blattaria and supported that the suborder Caelifera is more closely related to the Phasmatodea than to the suborder Ensifera of Orthoptera.

Recent phylogenetic studies indicating that termites are eusocial cockroaches closely related to the genus Cryptocercus have generated fresh interest in wood-feeding Blattaria. Here we summarize the social biology of the wood-feeding genus Salganea (Blaberidae: Panesthiinae). As in Cryptocercus, Salganea exhibits long term, biparental care that includes the defense and feeding of young nymphs. Unlike Cryptocercus, however, Salganea is iteroparous: most studied species reproduce periodically over their lifetime. These divergent reproductive strategies are likely related to parental costs associated with their differing reproductive modes: Cryptocercus is oviparous, while Salganea is ovoviviparous. The pattern of parental investment associated with ovoviviparous reproduction may be one reason why Salganea and other ovoviviparous wood-feeding cockroach lineages did not evolve eusociality.

The internal and external genitalia of both sexes of Zorotypus hubbardi Caudell 1918 were examined. A detailed description is provided, especially for the male genitalia. The results were compared with the conditions in potentially related neopteran groups. Most features of the internal genitalia of both sexes are plesiomorphic. Putative autapomorphies are: fusion of the distal parts of the vasa deferentia, division of the ductus ejaculatorius (de) into two sections by a distinct circular furrow, dorso-median processes on tergites VIII–X, an asymmetric aedoeagus with a cryptic slit-like phallotrema, reduction of the right outer paramere, the specific asymmetric shape of the endophallus with three conspicuous pouches, the endophallic sclerites scl1–scl6 and their specific arrangement, the specific arrangement of the endophallic muscularis and the unpaired genital muscles m2, m4, m5, m6 and m7. These features cannot be regarded as autapomorphies of the whole order Zoraptera with reliability, as the morphology of the external male genitalia seems to be highly variable within the genus Zorotypus and the intra-generic relationships are poorly resolved. Potential synapomorphies were found with several neopteran groups under comparison: with Embiidina a strengthened muscularis surrounding the de, reduction of the ovipositor and a shortened tergite IX, with Psocoptera the occurance of dense sperm bundles and with Hemiptera a strengthened muscularis of the de. No synapomorphies were found with Dictyoptera. Homologies of the muscles m1 and m3 found in Z. hubbardi with some genital muscles in Dictyoptera and Psocoptera are highly ambiguous.

(3S,11S)-3,11-Dimethylnonacosan-2-one is a major component of the courtship stimulating, contact sex pheromone of the female German cockroach. Although the four synthetic stereoisomers of this compound have been tested in behavioral assays, their relative activity remains unresolved. Using isolated male antennae dosed with synthetic test compounds to assay male behavior, we found that at high doses all four stereoisomers elicited responses from 100% of the males. However, at physiologically relevant doses similar to those found on the female antenna, the (3S,11S)-isomer was the least effective of the four stereoisomers at eliciting courtship responses in males. This is the first example of a natural stereoisomer having less bioactivity than related stereoisomers that do not occur naturally. Another component of the sex pheromone blend, 3,11-dimethylheptacosan-2-one, was previously purified from the female's epicuticule and behaviorally assayed, but its activity was not confirmed through synthesis. We now confirm that synthetic (3S,11S)-3,11-dimethylheptacosan-2-one elicits behavioral responses, but less so than its C29 homolog.

Members of the genus Cryptocercus are wood-feeding cockroaches that live in the temperate forests. Nine species are recognized in the genus worldwide: two in eastern Eurasia, two in China, and five in the United States. Within the United States, one species occurs in the Pacific Northwest and four occur in the Appalachian Mountains. Previous studies have revealed the presence of potential zones of overlap in distribution among the Appalachian species, raising the possibility of hybridization among them. Differences in mitochondrial DNA have previously been identified for the Appalachian species. However, to identify hybrid individuals one or more species-specific, codominant nuclear markers are required. Therefore, our objective was to undertake allozyme analysis of enzymatic loci to identify fixed, species-specific alleles for the four Appalachian species. We assayed a mean of 42 individuals each from 16 sites for allozyme variation for the four species. At 6 of the 33 loci examined, fixed alternate alleles were identified; a combination of 2 loci enabled the identification of all four species. To identify hybrids in the field, we examined 42 individuals each from 13 sites in which two or more of the above species occur in close proximity for presence of heterozygous individuals at one or more of the six fixed loci. No heterozygous individuals were found suggesting the lack of hybridization among the Appalachian species.

Males of the German cockroach, Blattella germanica, secrete a pheromonal substance from the abdominal tergal glands, which elicits a feeding response in females during the sequential courtship behavior. The nuptial secretion consists predominantly of a synergistic mixture of sugars and phospholipids. Cholesterol and a series of amino acids, which are also components of the male’s glandular secretion, significantly enhanced the phagostimulant activity of the sugar components. The nuptial feeding behavior of the female cockroach is therefore elicited by a complex assortment of nutritive components in the male tergal secretion, including sugars, phospholipids, cholesterol, and amino acids. These results indicate that a mixture of primary metabolites, and not of specific secondary metabolites, serves as a pheromonal cue that appeals to the female’s gustatory sense and effectively brings her to the precopulatory position. Although the male secretion consists of nutrients, we suggest that these compounds probably do not represent a significant nutrient investment in females and their progeny but rather function as a signal in the mating sequence of B. germanica.

Male cockroaches of the genus Blattella secrete courtshippheromones from abdominal tergal glands. Conspecific females mount courting males and feed on their tergal exudates. In the German cockroach, B. germanica, a complex mixture of oligosaccharides was previously identified as the major phagostimulants in the male tergal secretion. We compared the oligosaccharide compositions of three Blattella species from different habitats, B. germanica (house), B. lituricollis (field), and B.nipponica (forest), by gas chromatographic (GC) analysis of their trifluoroacetylated derivatives. The three possessed a series of maltooligosaccharides and oligoglucosyl trehaloses with α1→4 and α1→1 linkages in common. However, B. germanica was conspicuous for its accumulation of a oligoglucosyl trehalose with a α 1→6 linkage [O-α-D-glucopyranosyl-(1→6)-α-D-glucopyranosyl α-D-glucopyranoside] as the most prominent component of the secretion. Such diversification of the male pheromonal compositions within the genus might be reflected by biosynthetic systems in males and chemosensory properties in females suited for their natural habitat.

In the sequential courtship behavior of the German cockroach, Blattella germanica (Dictyoptera: Blattellidae), females feed on the tergal gland secretions from the male tergites. The phagostimulative components in the male secretions were composed of a complex mixture of oligosaccharides and phospholipids. Besides seven oligosaccharides previously identified, two new trisaccharides, O-α-D-glucopyranosyl-(1→6)-α-D-glucopyranosyl α-D-glucopyranoside and O-α-glucopyranosyl-(1→4)-O-α-glucopyranosyl- (1→1)-myo-inositol, were characterized as the sugar components of the secretions. The activity of the sugar components was compared in combination with the lipid fraction. Individual oligosaccharides exhibited the activity either alone or synergistically with the phospholipids. A complex mixture of these primary substances in the tergal gland secretions, thus, serves as a pheromonal phagostimulant in the precopulatory behavior, strongly eliciting feeding response in the female cockroach.

The volatile constituents of the supposed defensive secretions of the glandular pouches of the adults of both sexes of the cockroach Therea petiveriana have been shown to contain N-3-methylbutylacetamide (MBA) and N-3-methylbutylpropanamide (MBP), which represented 60% of the volatile fraction. The other 40% included acidic, aromatic, and aldehydic compounds. Behavioral experiments demonstrated that the secretion acts as an alarm pheromone for adults.

Insects, including cockroaches, have the ability to select a proper diet from different nutrient choices. We have showed previously that various neurotransmitters and neuromodulators appear to regulate certain aspects of feeding in the cockroach, Rhyparobia madera. In the current study, we examined the role of octopamine in feeding behavior of cockroach nymphs. By either injection or direct incorporation into the diet blocks, an octopamine agonist (octopamine or synephrine) or antagonist (phentolamine) was effective in altering feeding in R. madera nymphs. Both octopamine and synephrine increased feeding slightly, while phentolamine decreased feeding dramatically. Phentolamine was able to decrease feeding, but not motor activity, when injected directly into the nymphs. Octopamine appears to cause increased feeding in the cockroach.

Interactions between male Nauphoeta cinerea cockroaches are characterized by an elaborate ritual that leads to a stable dominant–subordinate hierarchy between two individuals. Chemical signals involving volatile sex pheromones and cuticular hydrocarbons play an important role in establishing and maintaining dominance status. The present study was performed to identify cuticular hydrocarbons in two- and three-times dominant or subordinate individuals obtained by forcing dyadic encounters. Two methods, i.e., solid-phase microextraction (SPME) and organic solvent extraction, were used to isolate cuticular hydrocarbons. A total of 23 peaks of cuticular hydrocarbons were identified. Analysis showed quantitative differences in hydrocarbon profiles of three-times dominant and subordinate individuals according to extraction method and dominance status. Dominant individuals were characterized by higher proportions of the monomethylalkanes 11- and 13-MeC36, 13- and 15-MeC38, and 11-, 13-, and 15-MeC35, while subordinate individuals had higher proportions of the following monomethylalkanes: 7-, 9-, and 11-MeC31, 5-MeC31, 3- and 8-MeC32, and 9-, 10- , 11-, and 12-MeC32. Discussion focuses on the reliability of hydrocarbons as indicators of dominance status and on their correlation with physiological processes.

We review literature on extant detritivores, including cockroaches and termites, and conclude that coprophagy was the key behavior leading to the evolution of hindgut fermentation systems in the stem group of Dictyoptera. Coprophagy exploits concentrated microbial consortia on cellulose based substrates. These microbes are potential mutualists and food, but they also initiate degradation of cellulose, detoxify allelochemicals and soften the substrate, a phenomenon known as the "external rumen". We suggest that the evolution of a sophisticated hindgut fauna is a process of internalizing this self-assembled microbial community, accompanied by changes in host-microbe interdependence, the source of microbial inoculum for neonates, and host social behavior. Proctodeal trophallaxis evolved from pre-existing intraspecific coprophagous behavior when termite ancestors became subsocial, because the physiology of encystment in oxymonad and hypermastigid flagellates precludes their transfer via cysts in adult feces. The behavior was reinforced by the benefits of using the trophic stages of flagellates as food. The association of these flagellates with the dictyopteran lineage is an ancient one, and may have originated as part of the external rumen in the Carboniferous coal swamps.

Using immunocytochemistry based on a monoclonal antibodyagainst Diploptera punctata allatostatin I and horseradishperoxidase-diaminobenzidine reaction, the presence of allatostatin-likeimmunoreactivity is demonstrated in the synganglion of Dermacentorvariabilis females. The immunoreactive cells are located in theprotocerebral, cheliceral, palpal, stomodeal, postesophageal, and opisthosomalregions of the synganglion. Strongly immunoreactive granules accumulate in theboundary area of the subganglia in the preesophageal part of the synganglion.This suggests that the immunoreactive materials may be released directly fromthere. In addition, a putative neurohemal area is found in the anterior area ofthe opisthosomal ganglion, where abundant immunoreactive materials are stored.Weak immunoreactivity and fewer immunoreactive cells are seen in newly moltedfemales compared with one month old, unfed females. Thus, the immunoreactiveproducts may be depleted during molting and synthesized in females beforefeeding.

In the present paper, we report on the occurrence of the cockroach Pycnoscelus surinamensis (Linnaeus, 1758) in Brachymyrmex cordemoyi Forel, 1895 nests, indicating a possible symbiosis between these two species. Also, the finding of intradomicile P. surinamensis nymphs may indicate this species is extending its habitat to human habitations, i.e. changing its ecological category from peridomestic to domestic.

The immature stages of some parasites live in prey animals (intermediate hosts) and only reach reproductive maturity when they are eaten by final host predators. Some of these parasites alter intermediate host behavior in ways that increase the likelihood of predation (parasite transmission). Using the acanthocephalan (Moniliformis moniliformis) in the cockroach (Periplaneta americana), we show that infected cockroaches experienced a decrease in wind-evoked escape responses, a predator avoidance behavior, that consisted of fewer escape responses, longer latency and higher threshold for escape behavior. We found no correlates of the impairment of the escape behavior in the abdominal portion of the escape neuronal circuitry. This study demonstrates a parasite-induced alteration of a behavior dedicated to predator avoidance.

Experiments were performed to test for nutrient-specific olfactory learning in the American cockroach, Periplaneta americana L. In a conditioning period, cockroaches were presented with two complementary foods, one of which contained protein and the other carbohydrate, this combination allowing them to select a nutritionally balanced diet. The foods were separated in space, and each was paired with one of two odors, citral or carvone. The cockroaches were then selectively deprived of one of the nutrients for 24 or 48 h. In the final (test) phase of the experiment the movement of the cockroaches toward the nutrient-associated odors was monitored. Associative learning was demonstrated with respect to protein, with protein-deprived cockroaches moving more frequently toward the protein-associated odor. No learned associations between carbohydrate and odor were demonstrated. These data are contrasted with similar experiments on an herbivorous insect, the locust Locusta migratoria.

Hydrocarbons (HC) are the most important waterproofing barrier on the cuticle of most terrestrial insects. Yet, the relationships among the type, amount, biophysical properties, and water retardation capacity of constituent HC are poorly understood. Melting temperatures and gas chromatographic profiles of HC of German cockroach tissues of various ages and stages were compared. The melting temperature (Tm) of oothecal HC was highest, Tm of epicuticular HC was substantially lower, and that of hemolymph HC was lowest. The epicuticular HC of older nymphs and adults had higher Tm than HC of the same sex and stage soon after the molt. The HC of females had higher Tm than did male HC. Principal components analysis suggested that normal and 3and 5-methylalkanes, which were more prevalent on the epicuticle, were associated with higher Tm, implicating these components of the HC blend in waterproofing roles. The cockroach ootheca is particularly well protected by an abundance of n-alkanes and its external HC exhibit the highest Tm of any HC blend. The methyl ketone sex pheromone components, which are derived from HC, appear to only slightly reduce the Tm of the epicuticular HC, probably because the methyl ketones represent only 1.12% of the mass of epicuticular HC. We suggest that the evolution of polar epicuticular chemical signals may be constrained by their tendency to increase water transpiration.

In the courtship behavior of the German cockroach, the male presents tergal glands to the female and feeds her with glandular secretions to place her in the appropriate precopulatory position. The phagostimulant activity of the secretions was quantitatively examined using the polyethylene glycol film method. The methanol extract of the glands on the eighth tergite induced a potent feeding response in 6-day-old virgin females (EC 50 = 0.0037 male equivalent/40 μg PEG spot). However, there was no temporal relation between the feeding response and the sexual receptivity of the females. Moreover, besides virgin females, the extract induced a feeding response in gravid or mated females, males, and the last-instar nymphs. These results strongly suggest that the secretions function as a dietary feeding stimulant in principle but as a courtship pheromone in the context of courtship behavior where the stimulants are offered as a nuptial gift.

Aggregation behavior and reduced locomotory activity in the German cockroach is known to be caused by chemical compounds in the feces. The attractive and/or arrestant efficacy of three relevant substances was tested in first instars by a two-choice aggregation test and in adults with a locomotion compensator apparatus that allows quantification of taste-directed orientation and walking speed as a function of antennal stimulation. The three substances tested were a feces crude extract; a mixture of six carboxylic acids (mix G) out of a total of 29 that were identified in the feces extract and tested as single compounds and in various combinations; and a steroid glucoside denoted as blattellastanoside A, which has been suggested as an aggregation arrestant pheromone in Blattella germanica. With both of our test methods, feces extract and mix G proved to be very attractive, whereas the effects of blattellastanoside A were, if anything, very poor. Possible reasons for discrepancies are discussed.

Inoculation of a spore suspension of Aspergillus flavus in the haemocoel of the German cockroach (Blatella germanica) resulted in large-scale invasion of various internal organs as seen in tissue sections stained with a fungal stain. The organs affected were the alimentary canal, fat bodies, muscle fibres, malpighian tubules and the cerebral neurosecretory cells. No invasion of the trachea and tracheoles was noted.

We used bioassays to determine the sex pheromone production site in the female American cockroach (Periplaneta americana). Bioassayed extracts from various body parts aroused sexual responses from unmated males. However, the extract of hindgut (including both colon and rectum) induced strong responses. Colon extract stimulated the strongest sexual response. To compare the sex pheromone contents of atrial glands, pygidium, rectum, and colon, we recorded the EAG responses of male antennae to these extracts. Among the four tissues, colon extract caused the strongest EAG response. From gas chromatographic (GC) analysis, the quantities of periplanone-A (PA) and periplanone-B (PB) (the main sex pheromone components in female American cockroach) were 0.34 and 8.31 ng, respectively, in the colon of a virgin female. The ratio of PA to PB was ca. 1:24. The present results are different from those reported in a previous study, in which it was concluded that atrial glands are the sites of sex pheromone production in P. americana, and from GC analysis the quantity of PB was estimated to be 60 ng/female. There was no difference in sexual behaviors induced by colon and atrial glands of calling virgin females. It is unclear whether the colon is the sole sex pheromone source in the female American cockroach.

Natural infection of Aspergillus flavus was observed in adults of Blattella germanica. Though the adult insects exhibited no external symptoms, they became hypoactive and later died. The dead and experimentally infected insects repeatedly yielded Aspergillus flavus in culture on Czapek's medium. Direct microscopic observation of the tissues of infected insects revealed fungal material. The blood films stained with Giemsa stain showed granulocytes (GRs) engulfing fungal hyphae. A remarkable increase in GR and plasmatocyte (PL) counts occurred in differential haemocyte counts (DHCs) of the infected insects. Two main types of immunological responses of the insect noticed were phagocytosis and encapsulation. DHC showed maximum involvement of GRs and PLs in immune mechanism.

The sexual behavior of males and females ofEurycotis floridana was investigated and the various associated behavioral sequences are described. Olfactometer data proved that the male produces a volatile sex pheromone attractive at a distance to conspecific females. The male initiates courtship behavior by exposing the glandular areas on the anterior parts of abdominal tergites 2, 7, and 8. This male calling behavior was observed throughout the day. The males can mate when 8 days old, whereas virgin females are sexually receptive 18 days after becoming adults. Once attracted near the male, the female opens her genital atrium and climbs on the back of the male, where she feeds on the glandular secretions that oozed around a little tuft of setae on the first tergite. These setae are mechanoreceptors and they are stimulated when licked by the females, which informs the male that she is in a proper position for copulation.

We determined the food source and water balance properties of the hissing-cockroach mite, Gromphadorholaelaps schaeferi. The food source for mites was identified using Evans blue dye by direct injection into a fasting host cockroach, Gromphadorhina portentosa, or by incorporation into cockroach food. No coloration was observed in mites on dye-injected cockroaches, but coloration was present in mites when only the food for the cockroaches had been stained. Thus, the mites are scavengers of cockroach food, and are not parasitic as previously thought. Our results demonstrate that the mites can absorb water from the air anywhere between 0.84 and 0.93 a v (%RH/100), and wax-block experiments revealed that the mouth is the site of uptake. The mites are normally clumped together on the host, typically in between the cockroach's legs and around the spiracles. Water loss rates for mites in groups (0.16% h-1) were far lower than for isolated mites (0.30% h-1), suggesting a ‘group effect’ with regard to water balance. Above the transition temperature of 30°C rate of water loss was rapid. The sites occupied by mites on the cockroach's body seem to be highly specific for feeding and absorption of water vapour.

A new form of maternal provisioning of newly hatched nymphs is described in the ovoviviparous cockroach Gromphadorhina portentosa. Shortly after expelling the hatching egg case, the female exudes from her abdominal tip a whitish, translucent material on which neonates actively feed. Integumentary gland cells lining the brood sac are the most likely source of the secretion. This form of maternal provisioning may not be restricted to the Madagascar hissing cockroach; a glandular brood sac similar to that of G. portentosa is found in at least three additional ovoviviparous cockroaches.

The musculature of the mushroom-shaped accessory gland receives innervation from trunks 5C1 of the phallic nerves, which arise from the posterior part of the terminal abdominal ganglion of the male cockroach Periplaneta americana. Anterograde cobalt filling through trunks 5C1 with the subsequent precipitating procedure has shown the fine innervation of the accessory gland. By retrograde cobalt filling through the same trunks, different types of cells have been mapped in the terminal abdominal ganglion. About 25 dorsal unpaired median (DUM) neurons have been identified among them. About 36 octopamine-like immunoreactive DUM neurons with large somata have been characterized in whole-mount preparations of the terminal abdominal ganglion. The combination of the cobalt-filling technique with immunohistochemical mapping of cells suggests an octopaminergic innervation of the musculature of the accessory gland by DUM neurons.

Antisera were raised against leucokinin IV, a member of the leucokinin peptide family. Immunohistochemical localization of leucokinin immunoreactivity in the brain of the cockroach Nauphoeta cinerea revealed neurosecretory cells in the pars intercerebralis and pars lateralis, several bilateral pairs of interneurons in the protocerebrum, and a group of interneurons in the optic lobe. Several immunoreactive interneurons were found in the thoracic ganglia, while the abdominal ganglia contained prominent immunoreactive neurosecretory cells, which projected to the lateral cardiac nerve. The presence of leucokinins in the abdominal nerve cord was confirmed by HPLC combined with ELISA. Leucokinin-immunoreactive neurosecretory cells were also found in the pars intercerebralis of the cricket Acheta domesticus and the mosquito Aedes aegypti, but not in the locust Schistocerca americana or the honey bee Apis mellifera. However, all these species have leucokinin-immunoreactive neurosecretory cells in the abdominal ganglia. The neurohemal organs innervated by abdominal leucokinin-immunoreactive cells were different in each species.

We studied agonistic interactions among male Madagascar hissing cockroaches,Gromphadorhina portentosa, in groups of five (low-density) or 10 (high-density) males. Consistent with previous studies of male pairs, we observed aggression (Abdomen Flick, Abdomen Push, Butt, Lunge), submission (Crouch, Retreat), and noncontact behavior (Abdominal Extension, Abdomen Thrash, Agonistic Hiss, Stilt). Males at both densities performed all acts. However, males in the high-density group performed Abdomen Push significantly more often than males at a low density. The rate of each remaining act was unaffected by density. Regardless of density, males within social groups varied in aggression. More aggressive males utilized frontal assaults (Butt and Lunge) during interactions, while males displaying lower levels of aggression preferentially used the abdomen during interactions. More aggressive males performed Abdomen Flick more frequently, while males displaying lower levels of aggression performed Abdomen Push. We also investigated the relationship between male aggression and the four noncontact behaviors. We found that Abdominal Extension, Abdomen Thrash, and Agonistic Hiss were positively correlated with our aggregate measure of male aggression suggesting these are aggressive displays. Stilt was positively correlated with measures of both aggression and submission, leaving its function unclear. None of the behavioral acts examined in this study were highly correlated with male weight. Our results are discussed in light of possible hypotheses addressing the function of specific behavior during male-male competition.

Summary Nymphs and adult females ofBlatta orientalis and nymphs ofEurycotis floridana produce a proteinaceous sticky secretion which accumulates on the last abdominal tergites. The proteic patterns do not differ between individuals of the same species. HPLC analyses show that all the common amino acids are found in both species, aspartic and glutamic acids representing 24 to 37% of the total amount of amino acids. InB. orientalis, glutamic acid is the more abundant amino acid whereas inE. floridana it is the aspartic acid. The secretion appears and accumulates rapidly on isolated insects. Behavioural assays revealed that these secretions have a defensive role.

Two chemical signals are essential in all cockroach sexual behavioral sequences: the sex pheromone released by one partner, generally the female (for long distance attraction), and an aphrodisiac sex pheromone produced exclusively by male tergal glands (for female mounting and tergal contact or “feeding” behavior). Unlike the other cockroach groups, the males of the Oxyhaloinae species produce both chemical signals: the pheromone and the aphrodisiac. The occurrence of three patterns of mating behavior (A, B, and C), the production of male sex pheromones, and the existence in the male of developed sternal and tergal glands in seven related Oxyhaloinae species, make these cockroaches a useful model for studying the evolution of mating behavior patterns. The various types of mating behavior were not classified in the previous studies by Roth and Barth. In this report, they have been named type A (female in upper position), B (male in upper position), and C (male and female end to end). In type A mating, the male tergal glands, which are licked by the females, are well developed, whereas in types B and C, there is no licking of the male's tergal secretion by the females and the tergal glands are much less developed; the aphrodisiacs secreted by the tergal glands may no longer act in this case through contact chemoreception, but through an olfactory process involving volatile components. One common sex pheromone component seems to be acetoin. I suggest that the mating behavior tends from A toward B and C during the evolutionary process with a concomitant regression of the tergal glands and changes in the aphrodisiac emission levels. The mating behavioral sequences of cockroaches (Dictyoptera) and crickets (Orthoptera) show a striking degree of similarity and are probably examples of convergent evolution.

Virgin German cockroach females, Blattella germanica(L.), were observed, for the first time, to exhibit a characteristic calling behavior during which females emit a volatile sex pheromone. Under a photoperiod of 12L∶12D, the percentage of 7-day-old virgin females that exhibited this behavior peaked before the end of the scotophase in a similar pattern to the diel periodicity of mating. A clear relationship was evident between calling and stages of sexual receptivity during successive gonotrophic cycles. Females initiated calling 5–6 days after the imaginal molt, when their basal oocytes were 1.6 mm long. If not mated, females continued to exhibit bouts of calling during the next 3–4 days until 24 h before ovulation. Calling was completely suppressed by mating as well as the presence of an egg case in the genital atrium in both virgin and mated gravid females. We suggest that calling and the emission of a volatile sex pheromone serve to attract males from a distance as well as to potentiate responses to contact sex pheromone in aggregations.

In 6-day-old females ofBlattella germanica, the activity of corpora allata (CA) was inhibited in vitro by juvenile hormone III (JH III). Effective doses (281.5 and 375.4 μM in the medium) were somewhat higher than (although of the same order of magnitude as) the estimated intraglandular concentration of JH III at this age, and they induced about 45% inhibition of hormonal release and a significant intraglandular accumulation of JH III and methyl farnesoate. The results suggest that autoinhibitory mechanisms operate in the CA to constrain the upper limit of JH III production at the end of the gonadotrophic cycle.

Females ofBlattella germanica andBlaberus craniifer produce a volatile sex pheromone attractive at a distance for conspecific males. During the emission of the pheromone, females ofB. craniifer adopt a typical calling posture; we never observe such a stance inB. germanica females. For both species, the glandular structures responsible for pheromonal production are located on the female pygidium. InB. germanica, the thickness of the glandular epithelium is clearly correlated with the attractiveness of females, being maximum for 13 day old females. The functions of the pygidial glands are discussed in both species.

Parasites are known to alter the behavior of their hosts, but little is known about their effects on responses to sexual stimuli. Periplaneta americanainfected with the acanthocephalan Moniliformis moniliformiswere compared to uninfected animals in their behavioral and electroantennogram responses to a synthetic P. americanapheromone component, periplanone-B, and a pheromone mimic, bornyl acetate. In a t-maze there was no significant difference between infected animals' responses to periplanone-B and a random binomial distribution; the responses of uninfected animals were significantly nonrandom. The electroan-tennogram responses of infected and uninfected animals to bornyl acetate or periplanone-B did not differ significantly, however, indicating that the alteration probably does not occur at the peripheral level but at a central nervous system level.

A monoclonal antibody against allatostatin I was used to demonstrate the allatostatin-immunoreactive pathways between the brain and the corpus cardiacum-corpus allatum complex in the adult cockroach Diploptera punctata. The antibody was two to three orders of magnitude more sensitive to allatostatin I than to the other four known members of the allatostatin family. Whole and sectioned brains in which immunoreactivity was localized with horseradish peroxidase-H2O2-diaminobenzidine reaction showed strongly immunoreactive cells in the pars lateralis of the brain with axons leading to and arborizing in the corpus cardiacum and the corpus allatum. Although many neurosecretory cells of the pars intercerebralis project to the corpora allata only, four strongly immunoreactive cells were evident here (two pairs on either side), and these did not project to the corpus cardiacum and corpus allatum but rather terminated within the protocerebrum in areas in which lateral cells also formed arborizations. Immunoreactivity was found in many other cells in the brain, especially in the tritocerebrum.

Effects of dehydration on the behavioral response to humidity have been examined in the cockroach, Periplaneta americanaL. Under normal conditions cockroaches prefer lower humidity, but the preference is reversed by dehydration. Dehydrated cockroaches can distinguish smaller differences between two air streams compared with normal individuals. Cockroach behavioral responses to humidification include swing of antennae and locomotion. The period from onset of humidification to behavioral response decreases with increasing dehydration, indicating that the humidity threshold required for behavioral response decreases. Observed modulations of the behavioral response are discussed in relation to the contribution of the antennal hygroreceptors.

The mechanism of long-distance flight in insects was investigated by comparing lipid mobilization and transport in gregarious- and solitary-phase locusts and in the American cockroach. Unlike the gregarious-phase locust, both the American cockroach and the solitary locust were unable to form low-density lipophorin (loaded with increased amount of diacylglycerol) even when injected with adipokinetic hormone (AKH). The cockroach fat body responded to AKH. However, not only does the American cockroach lack apolipophorin-III (apoLp-III) in the haemolymph, but the fat body contains only an extremely small amount of diacylglycerol and a relatively large triacylglycerol pool. By contrast, the solitary-phase locust had apoLp-III in the haemolymph, but the fat body was only one-seventh or less in weight of the fat body of the gregarious locust. Furthermore, the fat body of the solitary locust contains a very small amount of triacylglycerol (1/20 or less of that of the gregarious locust) with only a trace of diacylglycerol. It was concluded that in the American cockroach and the solitary locust, the stores of fuel in the fat body are insufficient to maintain prolonged flight.

The cockroaches Blatella germanica and Supella longipalpa can act as intermediate hosts of Trichospirura leptostoma as demonstrated by experimental infestation. The parasite developed from the embryonated egg into the infective larval stage (L3) in cockroaches within 5-6 weeks. After experimental infection of marmosets (Callithrix jacchus), eggs were first found in faecal samples (prepatency) at 8-9 weeks post-infection. Patency lasts about 2 years. Despite the presence of living adult worms in the marmosets' pancreas, no additional eggs were observed in their faeces after the patent period.

Background Neuropeptide ligands have to fit exactly into their respective receptors and thus the evolution of the coding regions of their genes is constrained and may be strongly conserved. As such, they may be suitable for the reconstruction of phylogenetic relationships within higher taxa. CAPA peptides of major lineages of cockroaches (Blaberidae, Blattellidae, Blattidae, Polyphagidae, Cryptocercidae) and of the termite Mastotermes darwiniensis were chosen to test the above hypothesis. The phylogenetic relationships within various groups of the taxon Dictyoptera (praying mantids, termites and cockroaches) are still highly disputed. Results Tandem mass spectrometry of neuropeptides from perisympathetic organs was used to obtain sequence data of CAPA peptides from single specimens; the data were analysed by Maximum Parsimony and Bayesian Interference. The resulting cladograms, taking 61 species into account, show a topology which is in general agreement with recent molecular and morphological phylogenetic analyses, including the recent phylogenetic arrangement placing termites within the cockroaches. When sequence data sets from other neuropeptides, viz. adipokinetic hormones and sulfakinins, were included, the general topology of the cladogram did not change but bootstrap values increased considerably. Conclusion This study represents the first comprehensive survey of neuropeptides of insects for solely phylogenetic purposes and concludes that sequences of short neuropeptides are suitable to complement molecular biological and morphological data for the reconstruction of phylogenetic relationships.

International audience; Males and females of Cryptocercus punctulatus possess tergal glands, but they differ in position, size, morphology, and secretion chemistry. Compound A (linalyl acetate) is the most abundant of the 21 compounds found only in the secretion of these glands. Compound B, 4,6,8-trimethyl-7,9-undecadien-5-ol, is specific to the tergal secretion of females.C. punctulatus lives only in the United States; its distribution is disjunct. Compound A is found in samples from the eastern population but is absent in samples from the western population. The amount of compound B per gland in samples from the western population is at least twice as high as in the samples from the eastern populations.

When juvenile praying mantids (Tenodera sinensis)were exposed to unpalatable prey (the milkweed bug Oncopeltus fasciatus),they attacked, sampled, and then usually rejected the prey. About 70% of the handling time was spent feeding. When offered a second milkweed bug, the mantids usually attacked the prey. However, the overall time required for the mantids to sample, recognize, and then reject the unpalatable prey decreased by half. The proportion of handling time that was spent feeding remained the same as in the first encounter. In contrast, when the second prey individuals encountered by mantids were Drosophila melanogaster,the flies were completely consumed and the proportion of handling time that was spent feeding significantly increased. When praying mantids were exposed to the milkweed bugs for the first time, up to 33% of the bugs survived attack by the mantids. Survival of milkweed bugs increased to 55% when mantids had been previously exposed to the bugs. In contrast, flies that were caught never survived.

The biological activities of synthetic periplanone analogs, including four candidates of periplanone-A (P-A), were evaluated by behavioral and electroantennogram (EAG) assays. Among 16 periplanone analogs, six compounds evoked pheromonal activity from the male American cockroaches. The threshold dosages of these biological active analogs were 10-10(5) times lower than that of the known periplanone mimic, germacrene-D. The conformation required for eliciting the pheromonal activity is discussed in terms of the structure-activity relationship of these analogs. Hauptmann's P-A elicited the strongest pheromonal activity among four candidates of P-A in our bioassay, suggesting that Hauptmann's P-A is a natural P-A produced from female cockroaches.

The diel pattern of pheromone-releasing behavior (calling) of the adult female brown-banded cockroach, Supella longipalpa (F.),was examined. Calling occurs discontinuously throughout most of the scotophase in a 12L: 12D photoperiodic regime. Females exhibited a free-running calling rhythm after transfer to continuous light or dark conditions. Shifts in the temporal pattern of calling following changes in the timing of the photoperiodic cues indicated that “lights-off” is the Zeitgeber. Studies of insects under long- and short-day conditions suggest that, although insects within a population may call synchronously or asynchronously, respectively, the duration of an individual's calling bout is innately limited.

In cockroaches infected with Moniliformis moniliformis, the melanogenic enzyme phenoloxidase (PO) was histochemically localized in the posterior midgut and in haemocytes. Midguts were incubated with either 3-hydroxytyramine-HCl (dopamine) or 3-(3,4-dihydroxyphenyl)-L-alanine (dopa), and the resulting electron-dense reaction products of PO activity were found to be homogeneously distributed in the cytoplasm of both midgut cells and haemocytes. Following experimental infection, the first acanthors that reached the outer surface of the gut elicited a haemocyte response similar to that observed during wound healing. Larvae that remained attached to the gut became melanized or developed successfully. PO activity gradually decreased as the course of infection proceeded (10-50 days post-infection) but was apparently not inhibited in either midgut cells or haemocytes that were closely associated with the parasites. PO was lacking in the midgut cells of uninfected cockroaches. The results of the present study are discussed with respect to the defence reactions of the host and the survival mechanisms of the parasite.
