Perfluoroalkyl substances (PFAS) are highly persistent organic pollutants that have been detected in a wide array of environmental matrices and, in turn, biota including humans and wildlife wherein they have been associated with a multitude of toxic, and otherwise adverse effects, including ecosystem impacts. In the present study, we developed a toxicity assay for embryonic stages of mahi-mahi (Coryphaena hippurus), as an environmentally relevant pelagic fish species, and applied this assay to the evaluation of the toxicity of “legacy” and “next-generation” PFAS including, respectively, perfluorooctanoic acid (PFOA) and several perfluoroethercarboxylic acids (PFECA). Acute embryotoxicity, in the form of lethality, was measured for all five PFAS toward mahi-mahi embryos with median lethal concentrations (LC50) in the micromolar range. Consistent with studies in other similar model systems, and specifically the zebrafish, embryotoxicity in mahi-mahi generally (1) correlated with fluoroalkyl chain length and hydrophobicity, i.e., log P, of PFAS, and thus, aligned with a role of uptake in the relative toxicity; and (2) increased with continuous exposure, suggesting a possible role of developmental stage specifically including a contribution of hatching (and loss of protective chorion) and/or differentiation of target systems (e.g., liver). Compared to prior studies in the zebrafish embryo model, mahi-mahi was significantly more sensitive to PFAS which may be related to differences in either exposure conditions (e.g., salinity) and uptake, or possibly differential susceptibility of relevant targets, for the two species. Moreover, when considered in the context of the previously reported concentration of PFAS within upper sea surface layers, and co-localization of buoyant eggs (i.e., embryos) and other early developmental stages (i.e., larvae, juveniles) of pelagic fish species to the sea surface, the observed toxicity potentially aligns with environmentally relevant concentrations in these marine systems. Thus, impacts on ecosystems including, in particular, population recruitment are a possibility. The present study is the first to demonstrate embryotoxicity of PFAS in a pelagic marine fish species, and suggests that mahi-mahi represents a potentially informative, and moreover, environmentally relevant, ecotoxicological model for PFAS in marine systems.

Maintaining energy balance over a wide range of temperatures is critical for an active pelagic fish species such as the mahi-mahi (Coryphaena hippurus), which can experience rapid changes in temperature during vertical migrations. Due to the profound effect of temperature on mitochondrial function, this study was designed to investigate the effects of temperature on mitochondrial respiration in permeabilized heart and red skeletal muscle (RM) fibres isolated from mahi-mahi. As RM is thought to be more anatomically isolated from rapid ambient temperature changes compared to the myocardium, it was hypothesized that heart mitochondria would be more tolerant of temperature changes through a greater ability to match respiratory capacity to an increase in temperature and to maintain coupling, when compared to RM mitochondria. Results show that heart fibres were more temperature sensitive and increased respiration rate with temperature increases to a greater degree than RM. Respiratory coupling ratios at the three assay temperatures (20, 26, and 30 °C), revealed that heart mitochondria were less coupled at a lower temperature (26 °C) compared to RM mitochondria (30 °C). In response to an in vitro acute temperature challenge, both tissues showed irreversible effects, where both heart and RM increased uncoupling whether the assay temperature was acutely changed from 20 to 30 °C or 30 to 20 °C. The findings from this study indicate that mahi-mahi heart mitochondria were more temperature sensitive compared to those from RM.

At the University of Basel (Switzerland), a multidisciplinary team was established for pre-operative selection and treatment of patients with gender dysphoria. As a result, the indications for surgical gender reassignment could be judged with considerably greater accuracy than previously possible. In the 9-year period of this prospective study only 14 of 57 patients with gender dysphoria were selected for surgical treatment. At the time of this survey, six patients are still under psychiatric preoperative evaluation, and six further male-to-female transsexuals are under hormonal treatment awaiting surgery. Following the operation, only one of nine male-to-female patients is socially unstable and that patient’s quality of life is worse than prior to gender reassignment. Of the female-to-male transsexuals, all four are stable in their professional and family relations. In conclusion, a comprehensive evaluation of patients with gender dysphoria and the conclusive indications established within the team considerably improved the postoperative outcome of gender reassignment.

Growth rates of larval and juvenile bigeye scad Selar crumenophthalmus reared in captivity were studied. The results are presented, discussed, and compared to wild S. crumenophthalmus and other pelagic fish. S. crumenophthalmus are a small pelagic carangid fish of circumtropical distribution. Larvae were reared in a modified mesocosm system and sampled daily for growth. Larvae grew to a mean size of 4.74 cm (Standard Length) and 1.30 g by 45 days post hatch (dph). Larval length-at-age was best described by the exponential equation Y = 1.966e(0.0704t) . For juvenile growth trials, 1940 fish were stocked into four 2.5 m(3) cylindro-conical tanks at two different densities (262 fish m(-3) and 120 fish m(-3)) and reared from 45 dph to subadult stage. Fish were sampled daily for growth. No statistically significant differences in growth or survival were detected between tanks. Mean length and weight at 141 dph was 13.24 cm (Total Length) and 25.20 g. Juvenile length-at-age was best described by the Von Bertalanffy Growth Model equation L t = 27.75(1 - e(-0.03(t-1.57))). Weight-at-age was best described by a linear equation W t = 1.7313x + 12.4662. The exponent of the length-weight equation was 3.14. In addition to providing the first published description of larviculture and juvenile growout techniques for S. crumenophthalmus, this study contains the first published data on this species' larval growth and directly confirms estimates of S. crumenophthalmus juvenile growth done by other researchers using indirect techniques such as otolith daily growth increment and frequency distribution analysis.

There is little agreement about the methodology of clinical trials of antipsychotic drugs in patients with negative symptoms. A literature review revealed wide variation in experimental design, rating scales and study duration. This reflects differing views as to the definition and response to treatment of negative symptoms. Some degree of standardization would improve comparability of studies and aid the development of new compounds. Patients included in such studies should have displayed negative symptoms for at least 6 months. Depressive symptoms, positive schizophrenic symptoms and extrapyramidal signs may all influence or be confused with negative symptoms and may respond to treatment; they should be at a low level at baseline and should be measured during the study period. Studies should last at least 8 weeks. Several scales are available for measuring negative symptoms and are reviewed; a global impression score should be used additionally.

The subjects were 34 acutely ill in-patients who met the RDC criteria of schizophrenic psychosis, and 4 EEGs were recorded from each patient before, 2 h and 24 h after oral intake of a single dose of 150 mg perazine, and on the 28th day of the neuroleptic treatment period. As a criterion of clinical response a decrease of at least 66% in the schizophrenia-specific sum score of the Brief Psychiatric Rating Scale on day 28 relative to the baseline value was decided upon. The EEGs were assessed using a newly developed procedure which takes into consideration 4 derivations simultaneously. As we tried to search out EEG variables with predictive value the statistical data analysis underlying our findings should largely by regarded as exploratory. Independent of day, responders (R) showed a tendency towards more low voltage desynchronized epochs (non-A stage) than non-responders (NR). Thus, R exhibited a higher degree of dynamic variability or a broader range of control of the spontaneous vigilance fluctuation (dynamic lability) than NR (dynamic rigidity). Furthermore, R and NR differed with respect to their time-dependent changes of non-A epoch frequencies before medication. While R showed a monotonous increase which is typical for normals, NR did not. Because of considerable inter-individual variability these group differences could not be used for individual prediction of the therapy response. By means of a qualitative data analysis R could be distinguished from NR with regard to various test dose-induced changes of the topographical distribution of absolute alpha power. All the group differentiating variables showed a time course of the same kind: R showed a prompt and ample deflection and the same recovery of baseline; NR, in contrast, showed no significant deflections at all. These findings are in line with the results concerning the dynamics of vigilance and certain claims of earlier authors according to which EEG changeability should be decisive for therapeutic outcome. The prediction power could be enhanced considerably by means of a classification procedure for qualitative data, which allows a combination of two variables. Allowing the three qualities increase, decrease and no change a combination of two variables rendered 32 = 9 possible answer patterns, the individual patient showing only one of these patterns, of course. A correct classification of 26 out of the 34 patients (76.5%) was achieved by 12 different 2-fold combinations of variables. Eventually, we tried to base an individual prediction upon a multitude of selected, clearly response predicting answer patterns. We were governed by the idea that a patient who shows the total number of the response predicting answer patterns, can be regarded as R with much higher probability than a patient who shows only half the number or even none of them.

In psychiatry the relationship between rating by others and self-rating has been discussed again and again. The question has been raised as to the influence of complete data material (all patients with self-rating and rating by others) on results, in comparison to results where patients without self-rating are not taken into evaluation. This question was answered by conducting a double-blind study which included two pharmacologically active substances (amitriptylinoxide and trazodone) and two sub-samples of patients (patients with rating by others and self-rating, and patients with rating by others alone). The trial was performed in a total of 57 in-patients suffering from endogenous depression (ICD 296.1) who received drug treatment for 21 days. The effect was assessed multi-methodically. While no differences between the two drugs were seen in the sub-sample "self-rating and rating by others", amitriptylinoxide proved to be superior to trazodone in the sub-sample "rating by others". On the basis of these findings the use of rating by others and self-rating procedures will be considered.
