Conservation genetic theory suggests that small and isolated populations should be subject to reduced genetic diversity i.e., heterozygosity and allelic diversity. Our 34 years study of an isolated island population of adders (Vipera berus) in southern Sweden challenges this notion. Despite a lack of gene flow and a yearly mean estimated reproductive adult population size of only 65 adult adders (range 12-171), the population maintains high levels of heterozygosity and allelic diversity similar to that observed in two mainland populations. Even a 14-year major "bottleneck " i.e., a reduction in adult adder numbers, encompassing at least four adder generations, did not result in any reduction in the island adders' heterozygosity and allelic diversity. Female adders are polyandrous, and fertilisation is non-random, which our empirical data and modelling suggest are underpinning the maintenance of the population's high level of heterozygosity. Our empirical results and subsequent modelling suggest that the positive genetic effects of polyandry in combination with non-random fertilisation, often overlooked in conservation genetic analyses, deserve greater consideration when predicting long-term survival of small and isolated populations.

Telomeric repeat binding factor 2 (TRF2) binds to telomeres and protects chromosome ends against the DNA damage response and senescence. Although the expression of TRF2 is downregulated upon cellular senescence and in various aging tissues, including skeletal muscle tissues, very little is known about the contribution of this decline to aging. We previously showed that TRF2 loss in myofibers does not trigger telomere deprotection but mitochondrial dysfunction leading to an increased level of reactive oxygen species. We show here that this oxidative stress triggers the binding of FOXO3a to telomeres where it protects against ATM activation, revealing a previously unrecognized telomere protective function of FOXO3a, to the best of our knowledge. We further showed in transformed fibroblasts and myotubes that the telomere properties of FOXO3a are dependent on the C-terminal segment of its CR2 domain (CR2C) but independent of its Forkhead DNA binding domain and of its CR3 transactivation domain. We propose that these non-canonical properties of FOXO3a at telomeres play a role downstream of the mitochondrial signaling induced by TRF2 downregulation to regulate skeletal muscle homeostasis and aging.

By clearing GABA from the synaptic cleft, GABA transporters (GATs) play an essential role in inhibitory neurotransmission. Consequently, in vivo visualization of GATs can be a valuable diagnostic tool and biomarker for various psychiatric and neurological disorders. Not surprisingly, in recent years several research attempts to develop a radioligand have been conducted, but so far none have led to suitable radioligands that allow imaging of GATs. Here, we provide an overview of the radioligands that were developed with a focus on GAT1, since this is the most abundant transporter and most of the research concerns this GAT subtype. Initially, we focus on the field of GAT1 inhibitors, after which we discuss the development of GAT1 radioligands based on these inhibitors. We hypothesize that the radioligands developed so far have been unsuccessful due to the zwitterionic nature of their nipecotic acid moiety. To overcome this problem, the use of non-classical GAT inhibitors as basis for GAT1 radioligands or the use of carboxylic acid bioisosteres may be considered. As the latter structural modification has already been used in the field of GAT1 inhibitors, this option seems particularly viable and could lead to the development of more successful GAT1 radioligands in the future.

The IRMM-1027 Large-sized dried (LSD) spikes are certified reference materials applied to measure the uranium and plutonium content of dissolved fuel solutions using isotope dilution mass spectrometry. High quality starting metals of uranium and plutonium are dissolved to produce a stock solution, which is dispensed into individual vials and dried down. The present spikes are mixtures of typically 50 mg 20% enriched U and 2 mg enriched 239Pu as dried nitrates, conditioned in an organic substance for stability. Each vial of an IRMM-1027 LSD spike comes with certified masses of uranium (235U and 238U) and 239Pu and isotopic composition with associated uncertainty. This article will discuss major developments since the production of the first batch of LSD spikes and will reflect on the current preparation and certification approaches.

International audience; The stiction phenomenon which results in the adhesion of the brake pad to the disc brake of a vehicle has been investigated from a corrosion point of view. Asbestos-free organic pad that contains copper associated with a cast iron disc was investigated and compared to a model system which consisted in a ceramic pad (chemically inert) associated with the same cast iron disc. The whole system was described as a thin-layer cell of electrolyte and was studied using different electrochemical methods including polarization curves and impedance spectroscopy. The influence of the cell geometry was pointed out, but the corrosion of the system is enhanced due to the presence of copper in the pad. Indeed, the copper dissolves from the pad and redeposits on the disc. This was confirmed by scanning electron microscopy observations and Raman spectroscopy. A mechanism taking into account the thin-layer geometry was then proposed to account for the role of metallic additive (in this case copper) on the corrosion of the disc.

Background The use of magnetic resonance imaging (MRI) in forensic age estimation has been explored extensively during the past decade. Objective To synthesize the available MRI data for forensic age estimation in living children and young adults, and to provide a comprehensive overview that can guide age estimation practice and future research. Materials and Methods MEDLINE, Embase and Web of Science were searched. Additionally, cited and citing articles and study registers were searched. Two authors independently selected articles, conducted data extraction, and assessed risk of bias. Study populations including living subjects up to 30 years were considered. Results Fifty-five studies were included in qualitative analysis and 33 in quantitative analysis. Most studies suffered from bias, including relatively small European (Caucasian) populations, varying MR-approaches and varying staging techniques. Therefore, pooling of the age distribution data was not appropriate. Reproducibility of staging was remarkably lower in clavicles than in any other anatomical structure. Age estimation performance was in line with the gold standard, which uses radiographs, with mean absolute errors ranging from 0.85 to 2.0 years. The proportion of correctly classified minors ranged from 65% to 91%. Multi-factorial age estimation performed better than based on a single anatomical site. Conclusion More multi-factorial age estimation studies are necessary, together with studies testing if the MRI data can safely be pooled. The porcine Helicobacter suis and canine-feline H. heilmannii are gastric Helicobacter species with zoonotic potential. However, little is known about the pathogenesis of human infections with these Helicobacter species. To gain more insight into the interactions of both zoonotic Helicobacter species with human gastric epithelial cells, we investigated bacterial genes that are differentially expressed in a H. suis and H. heilmannii strain after adhesion to the human gastric epithelial cell line MKN7. In vitro Helicobacter-MKN7 binding assays were performed to obtain bacterial RNA for sequencing analysis. H. suis and H. heilmannii bacteria attached to the gastric epithelial cells (i.e. cases) as well as unbound bacteria (i.e. controls) were isolated, after which prokaryotic RNA was purified and sequenced. Differentially expressed genes were identified using the DESeq2 package and SARTools pipeline in R. A list of 134 (83 up-regulated and 51 down-regulated) and 143 (60 up-regulated and 83 down-regulated) differentially expressed genes (padj ≤ 0.01; fold change ≥ 2) were identified for the adherent H. suis and H. heilmannii strains, respectively. According to BLASTp analyses, only 2 genes were commonly up-regulated and 4 genes commonly down-regulated in both pathogens. Differentially expressed genes of the H. suis and H. heilmannii strains belonged to multiple functional classes, indicating that adhesion of both strains to human gastric epithelial cells evokes pleiotropic adaptive responses. Our results suggest that distinct pathways are involved in human gastric colonization of H. suis and H. heilmannii. Further research is needed to elucidate the clinical significance of these findings.

This paper provides a metaphysical framework which enables the possibility of the hypostatic union. More specifically, social ontology will be used to philosophically ground the distinction between nature or substance on the one hand, and person on the other hand, which is crucial to that debate. There are some historical precedents for a juridical approach in christological debates, but the main sections develop a systematic metaphysical account. Relying on a generic version of dispositional realism, and the distinction between the ability space and the liberty space of a person, a person is the locus or nexus of contingent activations of alternative possibilities. This is distinct from the set or range of alternative possibilities open to a person. Persons can therefore function as artificial persons, paradigmatically exemplified in sovereign states. Extending that framework to the supernatural level enables the possibility of a divine person uniting a divine and a human nature.

In this study, we have investigated the phylogeny and the antagonistic interactions of culturable bacteria isolated from the sea urchin Paracentrotus lividus collected from Aber and Morgat, both located in Crozon peninsula, France. Bacteria were isolated from the gastrointestinal tracts of ten specimens by using conventional culture-dependent method and then investigated by using phylogenetic analysis based on 16S rRNA gene sequence comparisons. Assays for antagonistic interactions among the bacterial strains were performed; bacteria (including at least one strain representative of each OTU identified) were screened for antimicrobial substance production. So, 367 bacterial strains were isolated on marine-agar. On the basis of morphological characteristics, 180 strains were sequenced and 94 OTUs were classified. The dominant phyla were Proteobacteria, Firmicutes and Actinobacteria, with a high abundance of the strains belonging to the genus Psychrobacter. From the antagonistic interactions assays, it could be determined that 22.7% strains were positive for at least one antagonism interaction, 18.3% of them isolated from the sea urchins collected in Morgat. We hypothesize that the bacteria isolated in this study may represent the transitory microbiota of the gastrointestinal tract of P. lividus, and that this microbiota may be related to the diet of this marine invertebrate. Furthermore, our results suggest that chemical antagonism could play a significant role in shaping the bacterial communities within gastrointestinal tract of the sea urchins. In addition, most isolated bacteria may have promising biotechnology applications.

Aromatase inhibitors are standard of care in women with hormone receptor-positive early breast cancer. Published evidence demonstrates that adverse effects may have an impact on drug compliance, with arthralgias being one of the most commonly reported adverse effects. Eligible patients were postmenopausal women who had experienced arthralgia following initiation of an aromatase inhibitor. Patients who experienced arthralgias following a minimum of a 3-month treatment on the aromatase inhibitor were randomized to emu oil or placebo oil. The primary endpoint was to assess for a reduction in pain as measured by a visual analogue score after 8 weeks of treatment. Seventy-three patients comprised the intent-to-treat population, and there was no statistically significant benefit with use of EO. We show that for any standard Turing machine, it is possible to compute in polynomial time on input x a list of polynomial size guaranteed to contain a $${\operatorname{O}\bigl({\mathrm{log}}|x|\bigr)}$$ -short program for x. We also show that there exists a computable function that maps every x to a list of size |x|2 containing a $${\operatorname{O}\bigl(1\bigr)}$$ -short program for x. This is essentially optimal because we prove that for each such function there is a c and infinitely many x for which the list has size at least c|x|2. Finally we show that for some standard machines, computable functions generating lists with 0-short programs must have infinitely often list sizes proportional to 2|x|.

Nearly 15 years following the attacks on September 11, 2001, much learning has been acquired regarding the psychological affects of mass violence. Less is known, however, about the long-term impact of 9/11 on those with family members who died in the attacks. A few quantitative studies were conducted specific to grief and terrorism and these findings suggest that symptoms may be more severe and persist over a longer period than for those facing death due to other violent circumstances. While only a few studies examined the long-term experiences of family members aggrieved by 9/11, several qualitative studies were conducted years following the attacks but these investigations are found in the grey literature. Therefore, this review included a sample of five qualitative studies, which met inclusion criteria, and had not been published in the peer-review literature. The purpose of this review was to understand if any common themes existed between investigations that were conducted from 3 to 12 years after 9/11/01. Ten noteworthy themes were found that were common across investigations. Several themes were endorsed by participants in three to four studies, and included, sharing the tragedy and personal grief publicly, taking action, and financial compensation. The remaining themes were endorsed by family members across all five studies and included discussions about bodily remains, persistent and comparative grief, relationship changes, support groups and services, remembering their family member, and healing and growth after loss. While the findings in this review cannot be generalized to all families affected by 9/11, given the nature of the data, there are a number of important implications from this review discussed that are relevant for shaping future research and having clinical awareness about the long-term affects of mass violence.

Proteomic approach was applied to identify total proteins, particularly the enzymatic content, from wild cardoon flowers. As the selection of an appropriate sample preparation method is the key for getting reliable results, two different extraction/precipitation methods (trichloroacetic acid and phenol/ammonium acetate) were tested on fresh and lyophilized flowers. After two-dimensional electrophoresis (2D–E) separations, a better protein pattern was obtained after phenol extraction from lyophilized flowers. Only 46 % of the total analyzed spots resulted in a protein identification by mass spectrometry MALDI-TOF. Four proteases (cardosins A, E, G, and H), which have become a subject of great interest in dairy technology, were identified. They presented molecular weights and isoelectric points very close and high levels of homology between matched peptides sequences. The absence of the other cardosins (B, C, D, and F) could be an advantage, as it reduces the excessive proteolytic activity that causes bitter flavors and texture defects, during cheese making.

Although antibiotic-resistant pathogens pose a significant threat to human health, the environmental reservoirs of the resistance determinants are still poorly understood. This study reports the detection of resistance genes (ermB, mecA, mupA, qnrA, qnrB and tetL) to antibiotics among certain culturable and unculturable bacteria associated with the marine sponge Petromica citrina. The antimicrobial activities elicited by P. citrina and its associated bacteria are also described. Van Lambalgen's theorem states that a pair (ź, β) of bit sequences is Martin-Lof random if and only if ź is Martin-Lof random and β is Martin-Lof random relative to ź. In [Information and Computation 209.2 (2011): 183-197, Theorem 3.3], Hayato Takahashi generalized van Lambalgen's theorem for computable measures P on a product of two Cantor spaces; he showed that the equivalence holds for each β for which the conditional probability P(ź|β) is computable. He asked whether this computability condition is necessary. We give a positive answer by providing a computable measure for which van Lambalgen's theorem fails. We also present a simple construction of a computable measure for which conditional measure is not computable. Such measures were first constructed by Ackerman et al. ([1]).
