ZusammenfassungIm Jahr 2019 haben deutsche Universitäten das Jubiläum einer 50-jährigen Informatikausbildung gefeiert. Die TU Dresden blickt dabei seit 1969 als erste Universität auf Erfahrungen in beiden deutschen Staaten zurück. Während die historische Entwicklung in der Informatikausbildung für die BRD gut dokumentiert ist, besteht in der Dokumentation durch dasInformatik Spektrumfür die DDR eine Lücke. Die Informatikausbildung in der DDR befand sich seit ihren Anfängen auf Augenhöhe mit den Entwicklungen in der BRD. Im vorliegenden Artikel wird die historische Entwicklung des Informatik-Studienganges an der TU Dresden unter Berücksichtigung der Entstehungs- und Entwicklungsphase in der DDR und der Wendezeit bis zur heutigen Fakultät Informatik dargestellt. Aus Anlass des 50-Jahre-Jubiläums im Jahr 2019 wurde die historische Entwicklung in einem webbasierten Zeitstrahl dokumentiert und mit Interviews von Zeitzeugen ergänzt. Dabei lag der Schwerpunkt auf der Lehre einschließlich der Einheit von Lehre und Forschung.

Rapid reconstruction of depth images from sparsely sampled data is important for many applications in machine perception, including robot or vehicle assistance or autonomy. Approximate computing techniques have been widely adopted to reduce resource consumption and increase efficiency in energy and resource constrained systems, especially targeted at FPGA and solid state implementation. Whereas previous work has focused on approximate, but static, representation of data in LiDAR systems, in this paper we show how the flexibility of an arbitrary precision accelerator with fine-grain tuning allows a better trade-off between algorithmic performance and implementation efficiency. A mixed precision framework of $$\ell _1$$ ℓ 1 solvers is presented, with compact ADMM and PGD, for the lasso problem, enabling compressive depth reconstruction by varying the precision scaling in single bit granularity during the iterative optimization process. Implementing mixed precision $$\ell _1$$ ℓ 1 solvers on an FPGA with a pipelined architecture for depth image reconstruction across various sensing scenarios, over 74% savings in hardware resources and 60% in power are achieved with only minor reductions in reconstructed depth image quality when compared to single float precision, while over 10% saving in hardware resources and power is achieved compared to relative consistently reduced precision solutions.

Reintroduction of grazing to counteract insect decline will lead to an increase of semi-open habitats. Semi-open habitats are highly heterogeneous, consisting of a mosaic of patches of trees or shrubs embedded in a matrix dominated by dwarf shrubs or grasses. Despite a lack of evidence, structural heterogeneity is expected to allow many species to co-occur, making semi-open habitats appear highly valuable for invertebrate conservation. We studied ground beetles in eight sites in two landscapes of Central Europe each encompassing semi-open, open, and forest habitats. Rapid response to environmental change and limited dispersal abilities make ground beetles an excellent model taxon to evaluate the effectiveness of such conservations measures. In both studied landscapes, ground beetle assemblages in semi-open habitats tended to be distinct and intermediate between those from the forest and open habitats. Species richness and functional diversity in semi-open habitats were similar to open habitats at site level. The majority of species entered the semi-open habitats, except for most threatened species, yet, few species were exclusively associated with semi-open habitats. We conclude that the continuous presence of many species in semi-open habitats likely results from mass effects rather than habitat heterogeneity per se. Our findings underline the conservation value of the existing forest, heathland, and grassland habitats over semi-open habitats which can, however, function as dispersal habitats and increase landscape connectivity. Strategies aiming at promoting semi-open habitats to counteract insect decline should target enhancing connectivity rather than the creation of habitats only.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic has significantly impacted the world and has driven many researchers into the pathophysiology of COVID-19. In the findings, there is a close association between purinergic signaling and the immune response. Then, this study aimed to evaluate alterations in the purinergic signaling in COVID-19 patients according to range severity. We divided the COVID-19 patients into moderate and severe cases following the guideless of NIH and WHO, together with clinical characteristics. The blood samples were collected to obtain PBMCs and platelets. We analyzed the ectonucleotidase activities through ATP, ADP, AMP, Ado hydrolysis, E-NTPDase1 (CD39), and 5'-NT (CD73) expression by flow cytometry in total leukocytes. The extracellular ATP was measured by bioluminescence, and cytokines were analyzed by flow cytometry. We observed a decrease in ATP hydrolysis and increased AMP hydrolysis in PBMCs for both groups. In severe cases, ATP hydrolysis was raised for the platelets, while ADP and AMP hydrolysis have risen significantly in both groups. Additionally, there was a significant increase in ADP hydrolysis in severe cases compared to moderate cases. In addition, we observed an increase in the ADA activity in platelets of moderate patients. Moderate and severe cases showed increased expression of CD39 and CD73 in total leukocytes. To finalize the purinergic signaling, extracellular ATP was increased in both groups. Furthermore, there was an increase in IL-2, IL-6, IL-10, and IL-17 in moderate and severe groups. Thus, for the first time, our findings confirm the changes in purinergic signaling and immune response in COVID-19, in addition to making it more evident that the severity range directly impacts these changes. Therefore, the therapeutic potential of the purinergic system must be highlighted and studied as a possible target for the treatment of SARS-CoV-2 disease. KEY MESSAGES: COVID-19 patients exhibit alterations in purinergic system and immune response. High levels of extracellular ATP lead to different inflammatory responses. CD39 and CD73 expression were increased in COVID-19 patients. Cytokines IL-2, IL-6, IL-10, and IL-17 also were altered in these patients. The purinergic system may be a possibility target to SARS-CoV-2 treatments.

Seit einigen Jahren ist eine stetige Zunahme des Einsatzes von mechanischen extrakorporalen Herz-Kreislauf- und Lungenunterstutzungssystemen (ECLS/ECMO) zu verzeichnen. So wurden seit 2015 in Deutschland jahrlich bei etwa 3000 Patienten ECLS/ECMO-Systeme implantiert. Trotz dieser haufigen Anwendung der passageren Unterstutzungssysteme existieren zurzeit national und international nur Leitlinien zum Einsatz der ECMO/ECLS, die erkrankungsbezogene Teilaspekte komplexer Therapiekaskaden adressieren. Vor diesem Hintergrund erschien es notwendig, evidenzbasierte Empfehlungen zur ECLS/ECMO-Therapie im Hinblick auf Indikationen und das komplexe Patientenmanagement zu verfassen, in denen personelle, prozessuale und infrastrukturelle Anforderungen definiert werden. Aus diesem Grund erfolgte im Juli 2015 durch die Deutsche Gesellschaft fur Thorax‑, Herz- und Gefaschirurgie (DGTHG) die Anmeldung einer diesbezuglichen S3-Leitlinie bei der zustandigen Arbeitsgemeinschaft der Wissenschaftlichen Medizinischen Fachgesellschaften e. V. (AWMF). Im Mittelpunkt der Leitlinie steht die klinische Anwendung der ECLS/ECMO-Therapie; erganzend werden auch strukturelle und okonomische Aspekte adressiert. Unter Federfuhrung der DGTHG wurde mit Einbindung von Experten aus Deutschland, Osterreich und der Schweiz, delegiert aus 11 AWMF-Fachgesellschaften, 5 weiteren Fachgesellschaften sowie der Patientenvertretung, die S3-Leitlinie im Februar 2021 publiziert. Dieser Artikel stellt eine kurze Zusammenfassung des methodischen Konzepts sowie aller konsentierten Empfehlungen fur jede bearbeitete Fragestellung der Leitlinie dar.

Selection of the ideal surgical procedure for coronary revascularization in patients with severe cardiac dysfunction at times may represent a challenge. In recent years, with the advent of surgical large microaxial pumps, e.g., Impella 5.0 (Abiomed Inc., Boston, USA), specific support and effective unloading of the left ventricle has become available. In the interventional field, good results have been achieved with smaller microaxial pumps in the setting of so-called protected percutaneous coronary intervention. In this study, we would like to share our early experience with surgical coronary revascularization under the sole support of Impella 5.0, omitting the use of heart–lung machine in three cases of severe cardiac dysfunction due to complex ischemic heart disease. Effective circulatory support intraoperatively and postoperatively speaks in favor of this technique in selected patients.

Plant pathogenic fungi colonizing living plant tissue secrete a cocktail of effector proteins to suppress plant immunity and reprogramme host cells. Although many of these effectors function inside host cells, delivery systems used by pathogenic bacteria to translocate effectors into host cells have not been detected in fungi. Here, we show that five unrelated effectors and two membrane proteins from Ustilago maydis, a biotrophic fungus causing smut disease in corn, form a stable protein complex. All seven genes appear co-regulated and are only expressed during colonization. Single mutants arrest in the epidermal layer, fail to suppress host defence responses and fail to induce non-host resistance, two reactions that likely depend on translocated effectors. The complex is anchored in the fungal membrane, protrudes into host cells and likely contacts channel-forming plant plasma membrane proteins. Constitutive expression of all seven complex members resulted in a surface-exposed form in cultured U. maydis cells. As orthologues of the complex-forming proteins are conserved in smut fungi, the complex may become an interesting fungicide target., This study reports that five effectors and two transmembrane proteins from the plant pathogenic fungus Ustilago maydis form a stable cell surface-exposed protein complex required for virulence.

Over the past decade, the number of young people neither in employment, education, or training (NEET) has reached a seriously high level in many European countries. Previous studies have illustrated the heterogeneity of this group and that they differ considerably across Europe. However, the reasons of these cross-country differences have hardly been investigated so far. This study explores how the rates of different NEET subgroups are conditioned by various institutional configurations by applying fuzzy-set Quantitative Comparative Analysis for 26 European countries using aggregated EU Labour Force Survey data from 2018. The analysis reveals that institutional causes of being NEET are as diverse as the group itself. Thus, high levels of young NEETs with care responsibilities are found in countries with a lack of family-related services in conjunction with weak formalised long-term care as it is true in mostly Central Eastern European countries. In contrast, high rates of NEETs with a disability are prevalent mainly in Northern European countries where generous and inefficient disability benefit schemes exist that create false incentives to stay away from the labour market. Finally, high proportions of unemployed and discouraged young NEETs are found in those countries hit hardest by the crisis and with high labour market rigidities, low vocational specificity, and a lack of active labour market policies like in the Southern and some Central Eastern European countries. The results illustrate that young people face very different barriers across Europe and that country-specific measures must be taken to reduce the number of NEETs in Europe.

Background Physiological characteristics of climbers have been extensively studied, while the personality of climbers remains poorly understood. Personality research in sports is an established field, predicting long-term success as well as short-term behavior. The goal of our study was to examine recreational climbers’ personality and gain a better understanding of what makes them reach new heights. Methods We analyzed a cohort of 50 recreational climbers and 50 non-climbing athletes (control) regarding their personality characteristics. We assessed the BMI, the self-reported climbing grade, and the years of climbing experience. To evaluate the personality of recreational climbers and athletes, we used the German version of the Big Five Inventory (BFI-2), which summarizes the personality in five main categories. Results Recreational climbers scored significantly higher in conscientiousness than non-climbing athletes (p = 0.04), there was no significant difference between the other four main personality traits. Female climbers scored significantly higher in conscientiousness than male climbers (p = 0.02), while female athletes scored higher in openness than male athletes (p p Conclusion In conclusion, the personality of recreational climbers and recreational athletes differs in conscientiousness, with the climbers showing higher scores. Regarding gender, we found higher levels of conscientiousness in female climbers and higher openness in female athletes in comparison to their male counterparts. A negative correlation between BMI and conscientiousness of the climbers was detected.

BackgroundGenome-wide association studies (GWAS) aim to correlate phenotypic changes with genotypic variation. Upon transcription, single nucleotide variants (SNVs) may alter mRNA structure, with potential impacts on transcript stability, macromolecular interactions, and translation. However, plant genomes have not been assessed for the presence of these structure-altering polymorphisms or “riboSNitches.”ResultsWe experimentally demonstrate the presence of riboSNitches in transcripts of twoArabidopsisgenes,ZINC RIBBON 3(ZR3) andCOTTON GOLGI-RELATED 3(CGR3), which are associated with continentality and temperature variation in the natural environment. These riboSNitches are also associated with differences in the abundance of their respective transcripts, implying a role in regulating the gene's expression in adaptation to local climate conditions. We then computationally predict riboSNitches transcriptome-wide in mRNAs of 879 naturally inbredArabidopsisaccessions. We characterize correlations between SNPs/riboSNitches in these accessions and 434 climate descriptors of their local environments, suggesting a role of these variants in local adaptation. We integrate this information in CLIMtools V2.0 and provide a new web resource, T-CLIM, that reveals associations between transcript abundance variation and local environmental variation.ConclusionWe functionally validate two plant riboSNitches and, for the first time, demonstrate riboSNitch conditionality dependent on temperature, coining the term “conditional riboSNitch.” We provide the first pan-genome-wide prediction of riboSNitches in plants. We expand our previous CLIMtools web resource with riboSNitch information and with 1868 additionalArabidopsisgenomes and 269 additional climate conditions, which will greatly facilitate in silico studies of natural genetic variation, its phenotypic consequences, and its role in local adaptation.

Background The Arctic tundra is subject to the greatest climate change-induced temperature rises of any biome. Both terrestrial and freshwater biota are responding to recent climate warming through variability in their distribution, abundance, and richness. However, uncertainty arises within models of future change when considering processes that operate over centennial timescales. A systematic evidence synthesis of centennial-scale variability in biodiversity does not currently exist for the Arctic biome. Here, we sought to address the primary research question: what evidence exists for temporal variability in Arctic terrestrial and freshwater biodiversity throughout the Holocene (11,650 years before present (yBP)—0yBP)? Methods Consultation with stakeholders informed key definitions, scoping and the appropriateness of the research question. The research question was structured using a PECO framework—Arctic biota (P), a timestamped year in the Holocene (E), another year in the Holocene (C), and the dimensions of biodiversity that have been measured (O)—to inform the search strategy. Search strings were benchmarked against a test list of 100 known sources to ensure a specific and comprehensive return of literature. Searches will occur across 13 bibliographic databases. The eligibility criteria specify that sources must: (a) use ‘proxy’ methods to measure biodiversity; (b) fall within the spatial extent of the contemporary Arctic tundra biome; and (c) consist of a time-series that overlaps with 11,650yBP to 0yBP (1950AD). Information coded from studies will include proxy-specific information to account for both temporal uncertainty (i.e., the characteristics of age-depth models and dating methods) and taxonomic uncertainty (i.e., the samples and processes used for taxonomic identification). We will assess temporal uncertainty within each source by determining the quality of dating methods and measures; this information will be used to harmonise dates onto the IntCal20 calibration curve and determine the available temporal resolution and extent of evidence through space. Key outputs of this systematic map will be: (1) a graph database containing the spatial–temporal properties of each study dataset with taxonomic harmonisation; and (2) a geographical map of the evidence base.

At the CERN Large Hadron Collider (LHC), the knowledge and control of the available aperture is crucial because the nominal proton beams carry an energy of 362 MJ stored in a superconducting environment. Even a tiny fraction of beam losses could quench the superconducting magnets or cause severe material damage. Furthermore, in a circular collider, the performance in terms of peak luminosity depends to a large extent on the aperture of the inner triplet quadrupoles, which are used to focus the beams at the interaction points. In the LHC, this aperture represents the smallest aperture at top-energy with squeezed beams and determines the maximum potential reach of the peak luminosity. Beam-based aperture measurements in these conditions are difficult and challenging. ZusammenfassungDer Priapismus als klinische Manifestation einer hämatologischen Erkrankung ist selten. In diesem Fall liegt ein sowohl urologischer als auch internistischer Notfall vor, der einer umgehenden Therapie bedarf. Dieser Artikel beschreibt den klinischen Fall eines Priapismus als Erstmanifestation einer bis dahin nicht diagnostizierten chronisch-myeloischen Leukämie (CML) und erläutert die Resultate einer Literaturrecherche zu dieser Thematik.

Die optimierte Biokompatibilitat ist entscheidend fur die Haltbarkeit von Herz-Kreislauf-Implantaten. Bisher wurde gezeigt, dass eine kombinierte Beschichtung mit Fibronektin (FN) und Stromal cell-derived factor 1α (SDF1α) die In-vivo-Zellularisierung von synthetischen Gefastransplantaten beschleunigt und die Verkalkung von biologischen Pulmonalarterienstammtransplantaten verringert. In dieser Studie wurde die Wirkung einer seitenspezifischen Beschichtung mit SDF1α und FN auf die In-vivo-Rebesiedlung und -degeneration von dezellularisierten Aortenklappen-Conduit-Transplantaten untersucht. Spenderaorten wurden mit einem detergenzienbasierten Verfahren dezellularisiert. Die endoluminale Oberflache wurde mit SDF1α beschichtet, wahrend die Adventitiaoberflache mit FN beschichtet wurde. Mit SDF1α beschichtete und unbeschichtete Transplantate wurden Empfangerratten infrarenal implantiert (n = 20) und bis zu 8 Wochen nachuntersucht. ZusammenfassungIm zukünftigen taktilen Internet wachsen die physische und virtuelle Welt auch über mehrere Standorte hinweg immer weiter zusammen. Robotic Co-working – das gemeinsame Kollaborieren von Robotern und Menschen – gewinnt dabei zunehmend im Kontext des Internet der Dinge (IoT) und cyber-physischer Systeme (CPS) an Bedeutung. Mit dieser Arbeit präsentieren wir eine Fallstudie, die anlässlich des 50-jährigen Jubiläums des Informatikstudiums in Deutschland durchgeführt wurde. In dieser arbeiten Menschen und Roboter über mehrere Standorte verteilt in einer virtuellen Co-Working-Zelle zusammen, um einen „physischen und virtuellen Informatik-Deutschland-Campus 2069“ zu konstruieren. Unter Nutzung von Sensorik, Aktuatorik und Software wird der cyber-physische Campus, von einem Workflow gesteuert, schrittweise errichtet. Mithilfe einer Mixed-Reality-Anwendung kann dieser Prozess immersiv, d. h. eintauchbar und interaktiv, unabhängig von einem konkreten Standort erlebt werden.

The timing and transcriptomic changes during the C3 to CAM transition of common ice plant support the notion that guard cells themselves can shift from C3 to CAM. Crassulacean acid metabolism (CAM) is a specialized type of photosynthesis: stomata close during the day, enhancing water conservation, and open at night, allowing CO2 uptake. Mesembryanthemum crystallinum (common ice plant) is a facultative CAM species that can shift from C3 photosynthesis to CAM under salt or drought stresses. However, the molecular mechanisms underlying the stress induced transition from C3 to CAM remain unknown. Here we determined the transition time from C3 to CAM in M. crystallinum under salt stress. In parallel, single-cell-type transcriptomic profiling by 3′-mRNA sequencing was conducted in isolated stomatal guard cells to determine the molecular changes in this key cell type during the transition. In total, 495 transcripts showed differential expression between control and salt-treated samples during the transition, including 285 known guard cell genes, seven CAM-related genes, 18 transcription factors, and 185 other genes previously not found to be expressed in guard cells. PEPC1 and PPCK1, which encode key enzymes of CAM photosynthesis, were up-regulated in guard cells after seven days of salt treatment, indicating that guard cells themselves can shift from C3 to CAM. This study provides important information towards introducing CAM stomatal behavior into C3 crops to enhance water use efficiency.

Das hepatozellulare Karzinom (HCC) wird zu einer immer groser werdenden soziookonomischen Herausforderung. Internationale Leitlinien sprechen sich vorwiegend gegen die pratherapeutische Bestimmung der α‑Feto-Protein(AFP)-Konzentration aus. Bezuglich der prognostischen Aussagekraft des AFP ist die europaische Studienlage nicht aussagekraftig genug. Diese Arbeit soll mogliche Aspekte des AFP-Spiegels aufzeigen und die prognostische Vorhersagekraft des AFP-Spiegels untersuchen sowie Anregungen fur zukunftige prospektive Studien geben. Eine prospektive Leberdatenbank (n = 1382) wurde im Hinblick auf HCC-Patienten, die einen histologisch gesicherten Tumor aufweisen (n = 92), ausgewertet. Es lagen Informationen zu T‑, N‑, M‑, G‑Stadium und R‑Status bzw. zu Geschlecht, Alter, Genese vor. Fur die Analyse teilten wir das Patientengut anhand von 3 Cut-off-Werten in 6 Gruppen ein. Des Weiteren fuhrten wir eine Uberlebensanalyse mittels Kaplan-Meier und eine multifaktorielle Analyse der Einflussfaktoren auf das Outcome durch. Es gibt einen statistisch signifikanten Zusammenhang zwischen dem AFP-Spiegel, dem Tumordurchmesser (T1/2 vs. T3/4) und dem Grading (G1/G2 vs. G3/G4). In allen Gruppen fiel die Uberlebensprognose in der jeweiligen Gruppe mit dem hoheren AFP-Spiegel signifikant schlechter aus (p 8 μg/l betrug 35 Monate; bei Patienten mit AFP-Spiegeln von >200 μg/l bzw. >400 μg/l betrug das mediane Uberleben 15 Monate bzw. 11 Monate. Hohe AFP-Spiegel sind unabhangig vom T‑Stadium, Alter und R‑Status des Patienten im Vergleich zu niedrigen AFP-Spiegeln ein signifikanter Einflussfaktor fur das Outcome. Unter Berucksichtigung der vorliegenden Ergebnisse kann dem AFP-Spiegel eine therapeutische Nutzlichkeit zugeschrieben werden. Es konnten sich aus der Hohe der gemessenen AFP-Konzentration therapeutische Konsequenzen im Hinblick auf die Therapiestrategie ergeben. Demnach empfehlen wir bei allen HCC-Patienten die pra- und postoperative Bestimmung des AFP-Spiegels.
