Candidatus Methylomirabilis-related bacteria conduct anaerobic oxidation of methane (AOM) coupling with NO2− reduction, and Candidatus Methanoperedens-related archaea perform AOM coupling with reduction of diverse electron acceptors, including NO3−, Fe (III), Mn (IV) and SO42−. Application of nitrogen fertilization favors the growth of these methanotrophs in agricultural fields. Here, we explored the vertical variations in community structure and abundance of the two groups of methanotrophs in a nitrogen-rich vegetable field via using illumina MiSeq sequencing and quantitative PCR. The retrieved Methylomirabilis-related sequences had 91.12%-97.32% identity to the genomes of known Methylomirabilis species, and Methanoperedens-related sequences showed 85.49%-97.48% identity to the genomes of known Methanoperedens species which are capable of conducting AOM coupling with reduction of NO3− or Fe (III). The Methanoperedens-related archaeal diversity was significantly higher than Methylomirabilis-related bacteria, with totally 74 and 16 operational taxonomic units, respectively. In contrast, no significant difference in abundance between the bacteria (9.19 × 103–3.83 × 105 copies g−1 dry soil) and the archaea (1.55 × 104–3.24 × 105 copies g−1 dry soil) was observed. Furthermore, the abundance of both groups of methanotrophs exhibited a strong vertical variation, which peaked at 30–40 and 20–30 cm layers, respectively. Soil water content and pH were the key factors influencing Methylomirabilis-related bacterial diversity and abundance, respectively. For the Methanoperedens-related archaea, both soil pH and ammonium content contributed significantly to the changes of these archaeal diversity and abundance. Overall, we provide the first insights into the vertical distribution and regulation of Methylomirabilis-related bacteria and Methanoperedens-related archaea in vegetable soils. Accurate evaluation of the carbon release by stems is an important objective of many forest carbon cycling studies. However, few studies have assessed the applicability of different extrapolation methods of stem CO2 efflux. Therefore, a reliable estimation method that considers both temporal and vertical variations is required. To overcome this problem, we conducted a field experiment to measure the stem CO2 efflux rate of Abies fabri in subalpine forests, and evaluated the effect of three extrapolation methods (area-based estimation, volume-based estimation and integral method) on accurate estimation of stem respiration at the individual and stand levels. First, we calculated the stem carbon loss at the individual level using the three extrapolation methods, and discovered that the diameter at the breast height (DBH) played a major role in determining the magnitudes of stem respiration. Second, we summarized the relationship between the stem CO2 efflux rate (Es) and the DBH from previous studies. The Es for small diameter stems tended to increase with height, but remained constant in trees with large DBH, implying a role of DBH threshold in determination of the Es effect. Our calculations from three estimation methods supported the hypothesis that a threshold accounted for the different vertical variations in Es. Specifically, the vertical variation of Es primarily presented as two categories: Es decreased with the stem height for small diameters (DBH < 30 or 40 cm), or Es showed little change for stem height for large diameters (DBH > 60 cm). Based on these trends, we suggest a two-part approach to scale the individual tree stem CO2 efflux, in which a volume-based scaling method is recommended for trees with small DBH (0–30 or 40 cm) and an area-based estimation is suitable for stems with large DBH (> 30 or 40 cm). Using this approach, the annual amount of stem carbon release for an immature forest and a mature forest were estimated as 1.80 and 4.81 t C ha−1 year−1, respectively. [ABSTRACT FROM AUTHOR]

It has been suggested that the cryosphere is a new biome uniquely dominated by microorganisms, although the ecological characteristics of these cold-adapted bacteria are not well understood. We investigated the vertical variation with depth of the proportion of pigmented bacteria recovered from an ice core drilled in the Yuzhufeng Glacier, Tibetan Plateau. A total of 25,449 colonies were obtained from 1250 ice core sections. Colonies grew on only one-third of the inoculated Petri dishes, indicating that although the ice core harbored abundant culturable bacteria, bacteria could not be isolated from every section. Four phyla and 19 genera were obtained; Proteobacteria formed the dominant cluster, followed by Actinobacteria, Bacteroidetes and Firmicutes. The proportion of pigmented bacteria increased with depth from 79 to 95% and yellow-colored colonies predominated throughout the ice core, making up 47% of all the colonies. Pigments including α- and β-carotene, diatoxanthin, peridinin, zea/lutein, butanoyloxy, fucoxanthin and fucoxanthin were detected in representative colonies with α-carotene being the dominant carotenoid. Desert terrains in northern China are covered by widespread regolith sediments which mask geochemical signals from ore bodies and are major obstacles to mineral exploration. There is a critical need to study the vertical distribution of elements in this regolith and to establish optimum sampling and analytical methods. The aim of this study is to understand the dispersion and variation of elements throughout the cover in a vertical profile. The results demonstrate that the main elements show a distribution pattern of enrichment in clay layers in the vertical profile, i.e., most elements tend to be enriched in the most upper part of the profile above the orebodies except for some elements like Cu, Ca and Ag. Meanwhile, both fine (−160 mesh) and coarse (+20 mesh) fraction samples from clay-rich horizons are favorable samples and selective leaching of the elements absorbed on clays or oxide coatings is effective for localizing buried deposits. The distribution of active Cu and Mo is quite different because chalcopyrite is weathered into Cu sulfate and CuCO 3, and is precipitated in alkaline environments in a layer below close to the surface. Mo is soluble in the alkaline environment so it can penetrate the caliche layer and be enriched on the surface of soil enriched with weakly cemented clay. [ABSTRACT FROM AUTHOR]

Based on the molecular distribution of bound fatty acid (BFA) compound classes in core sediments of Lake Dianchi combined with the compound-specific δC values of the straight-chain BFAs, origin and vertical changes of organic matters in the sediments were investigated. The results indicated a significant change of BFA sources over the past 700 years. Contrast to the low concentrations of the terrestrial BFAs, the abundance of BFAs derived from the plankton/bacteria in the top sections (1944-recent) was more than 80 %. The increasing proportions of the branched and unsaturated BFAs in total fatty acids were closely correlated with the heavy eutrophication and the frequent algal blooms in the decades. Furthermore, the positive shift of δC of C16 and C18 (~2 ‰) in the upper section might be an indicator of the excess phytoplankton productivity. However, it was found that the plankton/bacteria-derived BFAs were more easily degraded during the early diagenetic process. The special compound carbon isotopic compositions of the long straight-chain BFAs (C24 and C26) in the sediments showed a depletion of heavier δC values (ca. −30 ‰) in the midsections (1559-1787), reflecting a relatively growing contribution of C3 plants to C4 plants or that C4 plant growth was inhibited in cold and arid climates during the period. The vertical variation and storage of nitrogen in the depth of 0–150 cm of an aquic brown soil were studied under 14 years of four land use patterns, i.e., paddy field, maize field, fallow field and woodland in Shenyang Experimental Station of Ecology, Chinese Academy of Sciences in November of 2003. The results showed that different land uses had different profile distributions of soil total nitrogen (STN), alkali N, ammonium (NH4 +-N) and nitrate (NO3 −-N). The sequence of STN storage was woodland>maize field>fallow field>paddy field, while that of NO3 −-N content was maize field>paddy field>woodland>fallow field, suggesting the different root biomass and biological N cycling under various land uses. The STN storage in the depth of 0–100 cm of woodland averaged to 11.41 t·hm−1, being 1.65 and 1.25 times as much as that in paddy and maize fields, respectively, while there was no significant difference between maize and fallow fields. The comparatively higher amount NO3 −-N in maize and paddy fields may be due to nitrogen fertilization and anthropogenic disturbance. Soil alkali N was significantly related with STN, and the correlation could be expressed by a linear regression model under each land use ( R 2≥0.929, p<0.001). Such a correlation was slightly closer in nature (woodland and fallow field) than in agro ecosystems (paddy and maize fields). Heavy N fertilization induced an excess of crop need, and led to a comparatively higher amount of soil NO3 −-N in cultivated fields than in fallow field and woodland. The pollen contents at different heights (1.5 and 15 m) of species of the Urticaceae family have been studied by sampling with Hirst type volumetric samplers. In order to achieve this, the two pollen types belonging to this family have been treated separately, Urtica urens-Parietaria sp. on the one hand and Urtica membranacea on the other, the latter having a smaller pollen grain. The results show that meteorological factors are bound to influence the behaviour of both these types of pollen in relation to height. With damp weather the pollen contents vary very slightly at different heights while when the weather is dry and calm, differences in pollen content at different heights become more significant. Nevertheless, when the atmosphere is stratified, the behaviour of each pollen type is different. The results show that, for most of the months considered, there is a higher pollen content of U. membranacea at upper heights, while U. urens-Parietaria sp. has higher levels of pollen content at a lower height. High temperatures, absence of rain and calm weather conditions favour the presence of convective phenomena which in turn create a favourable atmosphere for the vertical transportation of the small pollen grains of U. membranacea, which are better represented in the samplers placed at 15 m.
