This paper describes the results of an experimental investigation on the coefficient of friction at the interface of a PVC–sand–PVC layer that is utilised as part of a low-cost geotechnical seismic isolation devised to be used in low-income countries. The PVC–sand–PVC configuration consists of two smooth PVC surfaces enclosing a single layer of sand grains, with surface densities between 0.5 kg/m2 and 3 kg/m2, which aim to facilitate relative sliding at friction resistance between 0.15 and 0.30 depending on the design acceleration, by acting like "non-perfectly rounded ball bearings". The latter isolation method has been extensively studied both numerically and experimentally by means of large-scale testing at the shaking table of the EQUALS Earthquake Laboratory of the University of Bristol. However, in the light of the construction of the first building worldwide to be designed and constructed in Nepal with the particular low-cost PVC–sand–PVC sliding interface, it was deemed necessary to reliably assess the mean and dispersion of the coefficient of friction as a function of vertical pressure, sand density and degree of saturation. The results of the tests performed using an improved direct shear apparatus are presented herein using sand samples and PVC sheets that were locally resourced in Nepal to be used in construction. The results indicate that the variation of friction is reasonably low and in any case within the desirable range, irrespectively of the parameters examined, thus establishing confidence to the forthcoming design of the novel isolated building. Polymorphism of transcription factor 7-like 2 (TCF7L2) has a link with type 2 diabetes mellitus (T2DM) through β cell dysfunction that causes defect in blood glucose homeostasis. This case–control study recruited 67 T2DM as cases and 65 age-matched healthy individuals as controls to determine whether the polymorphism rs12255372 (G > T) in the TCF7L2 gene have an association with T2DM in Bangladeshi population. Genomic DNA was purified from peripheral whole blood sample and direct Sanger sequencing was done for genotyping of SNP. Bivariate logistic regression was done to find out the association between genetic variant and T2DM. In our study, the minor T allele frequency was significantly more frequent in T2DM group than healthy controls (29.1% vs. 16.9%). After adjusting with confounding factors, heterozygous-genotype GT had higher odds of developing T2DM (OR 2.4; 95% CI: 1.0–5.5; p value = 0.04) and in dominant model, having SNP in TCF7L2 increased the risk of T2DM 2.3 times (95% CI: 1.0–5.2; p value = 0.04). In interaction model, genetic susceptible SNP cases interacted significantly with increasing age and BMI, female gender, and having family history of diabetes mellitus to develop T2DM (pinteraction < 0.001). Having minor T allele either in heterozygous or homozygous variant form of rs12255372 (G > T) TCF7L2 had significant association with T2DM. Clinical and pre-clinical work for a number of cancer types has demonstrated relatively positive outcomes and effective tumour regression when the level and function of p53, a well-established tumour suppressor, is restored. Human papillomavirus (HPV)-driven cancers encode the E6 oncoprotein, which leads to p53 degradation, to allow the carcinogenic process to proceed. Indeed, there have been several attempts to revive p53 function in HPV-driven cancers by both pharmacological and genetic means to increase p53 bioavailability. Here, we employed a CRISPR activation (CRISPRa) approach to overcome HPV-mediated silencing of p53 by hyperexpressing the p53 gene promoter. Our data show that CRISPRa-mediated hyperexpression of p53 leads to HPV+ cervical cancer cell killing and the reduction of cell proliferation. This proof-of-concept data suggest that increasing p53 bioavailability may potentially be a promising therapeutic approach for the treatment of HPV-driven cancers. [ABSTRACT FROM AUTHOR]

This paper aims to explore the experiences of female leaders considering the interplay of gender, religion, and culture. Drawing on an inductive-qualitative study, the paper examines perceptions regarding the role of religion and cultural norms in women's ascension into leadership positions in Jordan. The results indicated that Jordanian women leaders adopted an Islamic feminist worldview and did not embrace a liberal nor a socialist/Marxist feminist worldview. Women leaders seemed wanting to claim their religion back from those forces that are reportedly holding their aspirations hostage to monolithic interpretations of religious texts. By constantly referring to their religion, female leaders wanted to be granted spaces of trust and responsibility in leadership positions that they did not see contradictory to the way they understood their faith. The paper provides insights into how women leaders understand prejudicial stereotypes and discrimination in their society, explaining how those are linked to patriarchal socio-cultural traditions emphasizing male control. Pyrimidine ring is an integral constituent of many natural biological systems, and pyrimidine derivatives have numerous synthetic and biological applications; therefore, there is great interest in the synthesis of pyrimidines for commercial purposes from pharmaceutical industry. In this work, a simple, efficient, and convenient methodology was developed for the synthesis of two series of bis-pyrimidine derivatives by the reaction of bis-chalcones with guanidine hydrochloride or thiourea under basic conditions. The synthesized compounds were evaluated for their antimicrobial and DNA photocleavage activity, and compounds of both series showed excellent potencies, especially those containing sulfur. It was also found that the antimicrobial activity strongly depended on the size and orientation of the olefinic/aromatic linker. Poly(ε-caprolactone) (PCL) is one of the promising polymers because it can be utilized as a substitute for petroleum-based polymers that are difficult to biodegrade in nature. PCL has non-toxic properties, high crystallinity, and biodegradable. Therefore, this polymer is widely used for medical purposes and environmentally friendly packaging. The purpose of this study was to compare the degree of polymerization, catalytic activity and thermal properties between PCL produced using bis(benzoylacetonato)zirconium(IV) catalyst (2a) with the PCL produced using other bis(β-diketonato)zirconium(IV) chloride catalyst (2b, 2c). In this research, ring opening polymerization (ROP) of ε-caprolactone (ε-CL) was performed by using complex 2a as a catalyst. The polymerization reaction lasts for 4 h at 100°C. The results of the research showed that ε-CL was successfully polymerized as proven in the results of its characterization. The molecular structure of the obtained PCL was analyzed by using FTIR, 1H NMR, 13C NMR, XRD, DSC, and TGA. The PCL produced in this study was a semicrystalline polymer. Meanwhile, a Mulliken charge of zirconium central atom in complex 2a–2c was also measured. Despite a vaccine being available, human papillomavirus virus (HPV)-driven cancers remain the ninth most prevalent cancers globally. Current therapies have significant drawbacks and often still lead to poor prognosis and underwhelming survival rates. With gene therapy becoming more available in the clinic, it poses a new front for therapeutic development. A characteristic of HPV-driven cancers is the ability to encode oncoproteins that aberrate normal p53 function without mutating this tumour-suppressor gene. The HPV E6 oncoprotein degrades p53 to allow the HPV-driven carcinogenic process to proceed. This review aimed to investigate the use of Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) gene-editing technology and how it may be used to overcome HPV-mediated silencing of p53 by hyper-expressing the p53 promoter. Increasing p53 bioavailability may have promising potential as a therapy and has been a goal in the context of HPV-driven cancers. Clinical trials and proof-of-concept pre-clinical work have shown positive outcomes and tumour death when p53 levels are increased. Despite previous successes of RNA-based medicines, including the knockout of HPV oncogenes, the use of CRISPR activation is yet to be investigated as a promising potential therapy. This short review summarises key developments on attempts that have been made to increase p53 expression in the context of HPV cancer therapy, but leaves open the possibility for other cancers bearing a p53 wild-type gene. By using Schiff base tricarboxylate ligand 5-(4-carboxybenzylideneamino)isophthalic acid (H3CIP), a new imine functionalized copper metal organic framework (MOF) has been synthesized solvothermally. It was fully characterized by Fourier Transform Infrared (FTIR) Spectroscopy, Powder X-Ray Diffraction (PXRD), Scanning Electron Microscopy (SEM), Energy Dispersive X-Ray Spectroscopy (EDS), and elemental mapping techniques. The as-synthesized MOF has been utilized as fluorescent probe for detection of nitro aromatic explosives (NAEs). The results show that the copper MOF can be developed into highly selective and sensitive sensor for detection of TNP in the aqueous medium via the "turn-off" quenching response. The linear fitting of the Stern-Volmer plot for TNP offered large quenching constant of 1.07 × 104 M−1 for Cu-MOF indicating the high sensitivity of the sensing process. Outstanding sensitivity of prepared material towards TNP detection was further validated by the low detection limit of 80 ppb (0.35 µM). The detailed mechanistic studies for their mode of action and density functional theory (DFT) calculations reveals that photo-induced electron transfer (PET) and fluorescence resonance energy transfer (FRET) processes, as well as electrostatic interactions (i.e. H-bonding) are the key factors for the turn-off response toward TNP by this fluorescent sensor. Thus, this new LMOF owing to their high water stability and remarkable functional features are potential candidates which can be developed into selective and sensitive TNP detection devices. An analysis of borehole logs with hydrophysical data from the Lagos coastal belt, Southwestern Nigeria, was carried out to evaluate the subsurface lithological units and geo-environmental impact of seawater intrusion into coastal freshwater. Forty-five (45) borehole logs, consisting of natural gamma, electrical resistivity log and hydrophysical parameters were integrated to delineate saline and fresh aquifer zones and propose simplified conceptual models for adequate groundwater characterisation and development. The qualitative interpretation of the borehole logs revealed a complex lithology of alternating sequences of sand and clay layers. The study also showed that the subsurface was variably intruded by saline water indicated by low resistivity values with freshwater/saline water boundaries occurrence at depths ranging between 14 and 157 m from west to east. The variation in depths of occurrence of freshwater/saline water interface is dependent principally on the proximity to saline/brackish water source as well as variation in lithology. Complementarily, the average electrical conductivity (EC) of 1770 μS/cm (GW1–7) and 1676 μS/cm (GW8–13) recorded in groundwater at Apapa in the west and Island at the central part, respectively, further confirmed saline incursion of the coastal aquifers in the region. Main conclusion: Present findings revealed a great variation in protein, oil, starch, tannin and phytic acid-like biochemical attributes of sorghum genotypes. GT bi-plot analysis was used for assessment of biochemical analysis results. Sorghum is quite resistant to negative environmental conditions. Thus, it has become the basic source of nutrient in majority of developing countries. It is also used as a supporting product against several diseases. In this study, biochemical composition of new 154 sorghum lines, selected from Turkish sorghum landraces through morphological, biochemical and molecular studies, was determined. Present findings revealed based on average of two years that crude protein content of 80 lines (> 9.65%), digestible protein content of 94 lines (> 56.25%), crude oil content of 75 lines (> 3.87%), resistant starch ratio of 32 lines (> 3.93%), starch content of two lines (> 77.07%) and amylose content of 10 lines (> %25.26) were greater than standard cultivar with the greatest values and amylopectin contents of 74 lines (< 47.75%) were lower than the standard cultivar with the lowest values. As the average of years, phytic acid contents and condense tannins contents of the lines, respectively, varied between 0.02 (IS 12850/1)—6.49% (PI 177161 03/2) and 0.09 (IS 12819/1)–5.38% (PI 255738 02/3). With this study, several sorghum lines superior than the standard cultivars were identified. Those lines can be used for direct cultivar registration in short run and can be used as parent materials in further breeding studies to be conducted for various purposes in long run. The organization of chromatin into higher-order structures and its condensation process represent one of the key challenges in structural biology. This is important for elucidating several disease states. To address this long-standing problem, development of advanced imaging methods has played an essential role in providing understanding into mitotic chromosome structure and compaction. Amongst these are two fast evolving fluorescence imaging technologies, specifically fluorescence lifetime imaging (FLIM) and super-resolution microscopy (SRM). FLIM in particular has been lacking in the application of chromosome research while SRM has been successfully applied although not widely. Both these techniques are capable of providing fluorescence imaging with nanometer information. SRM or "nanoscopy" is capable of generating images of DNA with less than 50 nm resolution while FLIM when coupled with energy transfer may provide less than 20 nm information. Here, we discuss the advantages and limitations of both methods followed by their contribution to mitotic chromosome studies. Purpose: Immunosuppressed (IS) patients are at increased risk for developing Merkel cell carcinoma (MCC) with worsened outcomes compared to immunocompetent (IC) patients. We sought to determine the effects of immune status on the efficacy of adjuvant RT regarding OS for patients with stage I, II or III (localized) MCC of the head and neck. Methods/patients: The National Cancer Database was queried for patients with resected, localized MCC of the head and neck with known immune status. Kaplan–Meier methods were used to describe OS. Log-rank tests, multivariable Cox regression models and interaction effect testing were used to compare OS by subgroup categorized by patient and treatment factors including immune status and adjuvant RT receipt. Results: A total of 892 (89.6%) IC and 104 (10.4%) IS patients with MCC of the head and neck were included. Adjuvant RT was associated with improved 3-year OS rate for both IS patients (49.4% vs. 35.5%, p = 0.0467) and stage I/II IC patients (72.4% vs. 62.9%, p = 0.0092). Adjuvant RT was associated with decreased hazard of death (HR 0.77, 95% CI 0.62–0.95). Interaction effect testing did not demonstrate a difference in the efficacy of adjuvant RT on OS between IC and IS status (p = 0.157). Conclusions: In this NCDB analysis, adjuvant RT was associated with decreased hazard of death for patients with localized MCC of the head and neck regardless of immune status and should be considered for both IS and IC patients. In this paper we develop two pivotal quantities to construct prediction intervals for future two-parameter exponential lifetimes based on a random number of generalized order statistics (gOs) under a general set-up including progressive type II censored order statistics (pOs) with general scheme. Moreover, the expected value as well as the mean square error for the upper limit of predictive confidence interval (PCI) are obtained. Furthermore, a maximum likelihood predictor (MLP) for future exponential lifetimes is derived when the sample size is deterministic or random. Hydrogen peroxide (HO) in minute quantity serves as a signalling molecule. However, the role of HO in combination with brassinosteroids (stress regulators) in plants under toxic levels of copper, is poorly understood. With an aim to explore and elaborate their role in plants subjected to abiotic stress, the surface sterilized seeds of mung bean ( Vigna radiata) were sown in earthen pots filled with soil and manure enriched with different levels of Cu (50 or 100 mg kg of soil) and allowed to grow under natural environmental conditions. At 15 and 20 days stage, the plants were sprayed with HO (2.5 mM) and/or 28-homobrassinolide (HBL, 10 mM), respectively. At 45 days stage, the analysis of the plants revealed that the presence of copper in the soil caused a significant decrease in growth characteristics, activity of carbonic anhydrase and nitrate reductase, relative water content, chlorophyll content and the rate of photosynthesis whereas, the activity of antioxidant enzymes (catalase, peroxidase and superoxide dismutase) and the proline accumulation in leaves increased in Cu stressed plants. However, the exogenously applied HBL and/or HO, in the absence of Cu-stress strongly favoured the growth, photosynthetic parameters and also improved the activity of antioxidant enzymes and the proline content. Furthermore, the combined application of HBL and HO to the foliage of the stressed plants neutralized the toxic impact of all copper regimes. Therefore, we are of the opinion that these chemicals somehow maintained the homeostasis of the metal in the plants that exhibit healthy growth. [ABSTRACT FROM AUTHOR]

Brassinosteroids have been extensively used to overcome various abiotic stresses. But its role in combined stress of salt and excess copper remains unexplored. Seeds of two cultivars (Rocket and Jumbo) of Cucumis sativus were grown in sand amended with copper (100 mg kg), and developed seedlings were exposed to salt stress in the form of NaCl (150 mM) at the 30-day stage of growth for 3 days. These seedlings were subsequently sprayed with 0 or 0.01 μM of 24-epibrassinolide (EBL) at the 35-day stage. The plants exposed to NaCl and Cu in combination exhibited a significant decline in fresh and dry mass of plant, chlorophyll content, activities of carbonic anhydrase, net photosynthetic rate and maximum quantum yield of the PSII primary photochemistry followed by NaCl and Cu stress alone, more severely in Jumbo than in Rocket. However, the follow-up treatment with EBL to the stressed and nonstressed plant improved growth, chlorophyll content, carbonic anhydrase activity and photosynthetic efficiency, and further enhanced the activity of various antioxidant enzymes viz. catalase, peroxidase and superoxide dismutase and content of proline at the 40-day stage of growth, and the response of the hormone was more effective in Rocket than in Jumbo. The elevated level of antioxidant enzymes as well as proline could have conferred tolerance to the NaCl- and/or Cu-stressed plants resulting in improved growth, water relations and photosynthetic attributes. Floating knee injuries pose major challenges in management. There is little information on the patterns of these injuries and the challenges of managing them in our setting. Patients with floating knee injuries who were seen and treated over a 5-year period at our center were retrospectively reviewed. The patterns of the injuries, associated injuries, methods of management and factors that influenced the outcome were analyzed. Twenty-five patients, consisting of 19 males and 6 females with a mean age of 33.4 ± 16.04 years, satisfied the study criteria. All were caused by road traffic injuries, motor cycle crashes accounting for 19 (76 %). Twenty-two (88 %) injuries were Blake and McBryde type 1, while 3 (12 %) were of type 2A. Thirty-two percent of the patients had surgical fixation of both fractures, while 40 % of the patients had both fractures managed non-operatively. Non-operative management strongly correlated with development of malunion. The results of management of floating injuries in our setting are not satisfactory. Context: Magnesium sulphate is now the gold standard for the control of eclamptic fits. The place of low-dose magnesium sulphate for the control of eclamptic seizures is yet to be determined. Objectives: To determine the effectiveness of low-dose magnesium sulphate in controlling eclamptic fits. Study design: Randomized controlled trial comparing low-dose with standardized dosing regimen. Setting: Labour Unit of the department of Obstetrics and Gynecology Federal Medical Centre Azare, north-eastern Nigeria. Protocol: Thirty-nine patients randomized into the low-dose regimen group received 9 g loading dose (4 g iv and 5 g im) and im maintenance of 2.5 g four hourly for 24 h post-delivery or post last fit, while the 33 patients in the standard dose regimen group received loading dose of 14 g followed by im maintenance dose of 5 g four hourly. In both study groups, 2 g iv of magnesium sulphate is given for breakthrough fits and 10 ml of 10 % calcium gluconate (slowly iv) was administered in the event of toxicity. Outcome measures include recurrent fits, mode of delivery, mean Apgar Score at 5 min, perinatal death, maternal complications including death. Result: The mean age of the 72 patients was 22.3 ± 5.4 years and 60 % were primigravidas. Intrapartum eclampsia was encountered in 44 % of the patients followed by antepartum eclampsia (26 %). Overall 4.2 % recurrent convulsion rate was documented and it is not different among the study groups. There were also no differences in both foetal and maternal outcomes in the two study groups. Conclusion: The effectiveness of low-dose regimen of magnesium sulphate appeared comparable to the 'standard dose regimen'. Low-dose regimen may guarantee more safety and in an environment (such as ours) where cost is an important determinant of accessibility to qualitative health services, it is certainly attractive. Floral and faunal assemblages from rockshelter sites have provided data on the transition from a Late Stone Age way of life to an Iron Age way of life on the Shire highlands in southern Malawi. The data have come from the excavation of seven rockshelter sites located in the area. They show that while there were no noticeable changes in the exploitation of wild flora and macrofauna during the Late Stone Age period, exploitation of microfauna and use of domesticated plants by Later Stone Age hunter gatherers became prominent with the arrival of Iron Age agriculturalists in the area. The change appears to have been a direct response to a declining resource base of hunter-gatherers caused by Iron Age subsistence strategies that may have led to a hunter-gatherer dependence on Iron Age agriculturists. Nickel even though recognized as a trace element, its metabolism is very decisive for certain enzyme activities, maintaining proper cellular redox state and various other biochemical, physiological and growth responses. Study of the aspects related with uptake, transport and distributive localization of Ni is very important in various cellular metabolic processes particularly under increased nitrogen metabolism. This review article, in core, encompasses the dual behavior of Ni in plants emphasizing its systemic partitioning, essentiality and ill effects. However, the core mechanism of molecules involved and the successive physiological conditions required starting from the soil absorption, neutralization and toxicity generated is still elusive, and varies among the plants. As an alternative to current bone grafting strategies, a poly-lactide- co-glycolide/calcium phosphate composite microsphere-based scaffold has been synthesized by the direct formation of calcium phosphate within forming microspheres. It was hypothesized that the synthesis of low crystalline calcium phosphate within forming microspheres would provide a site-specific delivery of calcium ions to enhance calcium phosphate reprecipitation onto the scaffold. Both polymeric and composite scaffolds were incubated in simulated body fluid (SBF) for 8 weeks, during which time polymer molecular weight, scaffold mass, calcium ion concentration of SBF, pH of SBF, and calcium phosphate reprecipitation was monitored. Results showed a 20% decrease in polymeric scaffold molecular weight compared to 11–14% decrease for composite scaffolds over 8 weeks. Composite scaffold mass and SBF pH decreased for the first 2 weeks but began increasing after 2 weeks and continued to do so up to 8 weeks, suggesting interplay between pH changes and calcium phosphate dissolution/reprecipitation. Free calcium ion concentration of SBF containing composite scaffolds increased 20–40% over control values within 4 h of incubation but then dropped as low as 40% below control values, suggesting an initial burst release of calcium ions followed by a reprecipitation onto the scaffold surface. Scanning electron micrographs confirm calcium phosphate reprecipitation on the scaffold surface after only 3 days of incubation. A synthesis of 3-(4-methoxycarbonyl-2,6-dinitrophenyl)indole, its 2,6-diamino analog, and 3-(2-amino-4-trifluoromethyl-6-nitrophenyl)indole is described. 4-(Trifluoromethyl)phenyl derivatives exhibit higher antibacterial potency than the former 4-(methoxycarbonyl)phenyl homologs, while 3-(4-trifluoromethyl-2-nitrophenyl)indole was the most active agent in the series, with MIC ≈ 7 μg/cm3 against E. coli and S. aureus. Background: Zingiber officinale is a rhizome commonly consumed as a delicacy, medicine or spice. It is considered as a safe spice with several medicinal properties. The aim of this study was to evaluate in-vitro antioxidants, antimicrobial and safety of Nigerian Z. Officinale.Methods: Methanol extract of Nigerian Z. Officinale (MEZO) was evaluated for phytochemical composition, total flavonoids and phenol contents using standard procedures. Antibacterial study was carried out via the agar well diffusion method. Antioxidant activities were carried out using 2, 2′- diphenyl-1- picrylhydrazyl (DPPH), and ferric reducing antioxidant properties (FRAP) assay. Twenty five (25) wister rats were randomly grouped into five (A-E) of five animals each. Animals in Groups A-D were orally administered 75,150, 300 and 600 mg/kg bwt of the extract on daily basis for 4 weeks while those in Group A (control) received distilled water.Results: Total phenolic and flavonoids contents of the extract were 15.24 ± 0.02 mg GAE/g and 19.84 ± 0.32 mg/g CE respectively. The extract promoted an inhibition of free radicals with IC50 values of 47.05 ± 2.03 μg/mL and 89.15 ± 0.29 μg/mL in DPPH and FRAP assay. At extract concentration of 100 μg/mL, K. pneumoniae showed the highest susceptibility of 29.04 ± 0.35 mm, followed by P. aeuruginisa (26.03 ± 0.41 mm), while S. aureus (15.08 ± 0.20 mm) was least susceptible. The serum concentrations of alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), sodium, albumin, total proteins and the computed organs/body weight ratios compared favorably (p > 0.05) with control at all extract doses tested. The bilirubin, urea and creatinine levels significantly (p < 0.05) increase while chloride decreases in rats dosed 600 mg/kg bwt. However, potassium level increases significantly (p < 0.05) in rats dose 300 mg/kg of the extract when compare with the control.Conclusion: This study revealed the strong antioxidant and antimicrobial potentials of methanol extract of Nigerian Zingiber officinale. It was also found to be relatively safe for consumption and thus could serve as a source of candidate for the development of new antioxidants and antimicrobial drugs. [ABSTRACT FROM AUTHOR]
