Background: Laparoscopic sleeve gastrectomy (LSG) is the most popular bariatric surgery procedure in China. However, its cost-effectiveness in Chinese patients is currently unknown. Objectives: This study aims to assess the cost-effectiveness of LSG vs no surgery in Chinese patients with severe and complex obesity, taking into account both healthcare expenses and the potential improvement in health-related quality of life (HRQoL). Methods: A retrospective cohort study was conducted, encompassing 135 Chinese patients who underwent LSG between January 3, 2022 and December 29, 2022, at a major bariatric center. The study evaluated the cost-effectiveness from a healthcare service perspective, employing the incremental cost-effectiveness ratio (ICER) for quality-adjusted life years (QALYs) gained. The analyses compared LSG with the alternative of not undergoing surgery over a 1-year period, using actual data, and extended to a lifetime horizon by projecting costs and utilities at an annual discount rate of 3.0%. Subgroup analyses were undertaken to explore cost-effectiveness variations across different sex, age and BMI categories, and diabetes status, employing a one-way analysis of variance (ANOVA). To ensure the reliability of the findings, one-way and probabilistic sensitivity analyses were executed. Results: The results indicated that 1-year post-LSG, patients achieved an average total weight loss (TWL) of (32.7 ± 7.3)% and an excess weight loss (EWL) of (97.8 ± 23.1)%. The ICER for LSG compared to no surgery over a lifetime was $4,327/QALY, significantly below the willingness-to-pay (WTP) threshold for Chinese patients with severe and complex obesity. From a lifetime perspective, LSG proved to be cost-effective for all sex and age groups, across all BMI categories, and for both patients with and without diabetes. Notably, it was more cost-effective for younger patients, patients with higher BMI, and patients with diabetes. Conclusions: LSG is a highly cost-effective intervention for managing obesity in Chinese patients, delivering substantial benefits in terms of HRQoL improvement at a low cost. Background/Aim: Lenvatinib, a multikinase inhibitor, has become a second-line treatment option for unresectable liver cancer, while its monotherapy response rate is limited. Hence, we aim to investigate whether one of the epigenetic inhibitors will be synthetic lethal with Lenvatinib in liver cancer cells. Materials and Methods: We performed high-throughput drug screening in combination with Lenvatinib. And we employed CCK-8-based Bliss Synergy Score analysis, colony formation and western blotting to confirm our screening results in both HepG2 and HCCC9810 cells. Results: We identified that LSD1 inhibitor Pulrodemstat in combination with Lenvatinib dramatically suppressed the PI3K-AKT signaling and induced a more significant activation of Caspase3 compared to Lenvatinib monotherapy. The nervous system possesses the remarkable ability to undergo changes in order to store information; however, it is also susceptible to damage caused by invading pathogens or neurodegenerative processes. As a member of nucleotide-binding oligomerization domain-like receptor (NLR) family, the NLRP6 inflammasome serves as a cytoplasmic innate immune sensor responsible for detecting microbe-associated molecular patterns. Upon activation, NLRP6 can recruit the adapter protein apoptosis-associated speck-like protein (ASC) and the inflammatory factors caspase-1 or caspase-11. Consequently, inflammasomes are formed, facilitating the maturation and secretion of pro-inflammatory cytokines such as inflammatory factors-18 (IL-18) and inflammatory factors-1β (IL-1β). Precise regulation of NLRP6 is crucial for maintaining tissue homeostasis, as dysregulated inflammasome activation can contribute to the development of various diseases. Furthermore, NLRP6 may also play a role in the regulation of extraintestinal diseases. In cells of the brain, such as astrocytes and neurons, NLRP6 inflammasome are also present. Here, the assembly and subsequent activation of caspase-1 mediated by NLRP6 contribute to disease progression. This review aims to discuss the structure and function of NLRP6, explain clearly the mechanisms that induce and activate NLRP6, and explore its role within the central and peripheral nervous system. Kaolinite is a layered silicate material with a Janus character and has been directly used as the solid catalyst for the hydrolysis of cellulose in our previous work. Reducing sugar (RS) is one of the most important intermediates for the utilization of cellulose. In the present study, metal ions dispersed on kaolinite was easily prepared and further evaluated for the hydrolysis of cellulose to RS in water. The metal ions dispersed kaolinite was characterized by XRD, BET, SEM(EDS), and XPS. The catalytic results showed that copper was the best co-catalyst among the tested metal ions and the kaolinite treated with 4(wt)% copper nitrate by calcination at 623 K showed the highest total reducing sugar (TRS) yield of 51% under 478 K with the mass ratio of catalyst to cellulose of 0.2 and weight ratio of water to cellulose of 7 for 3 h. Moreover, the catalyst could be easily regenerated by calcination and the yield of TRS on the regenerated catalyst decreased from 51 to 43.2% after four times reuse. The characterization results revealed that copper was highly dispersed on the outer SiO4 tetrahedral layer. The surface cuprous species were the active sites and could make the C-O bond cleavage easily by attacking the positively charged carbon atoms of the β-(1,4)-glycosidic linkage in the cellulose. The leaching of the cuprous species was responsible for the deactivation of the catalyst. The copper dispersed kaolinite is the efficient catalyst for the hydrolysis cellulose to reducing sugars and worth further development. [ABSTRACT FROM AUTHOR]

Background: Non-invasive brain stimulation (NIBS) is a burgeoning approach with the potential to significantly enhance cognition and functional abilities in individuals who have undergone a stroke. However, the current evidence lacks robust comparisons and rankings of various NIBS methods concerning the specific stimulation sites and parameters used. To address this knowledge gap, this systematic review and meta-analysis seek to offer conclusive evidence on the efficacy and safety of NIBS in treating post-stroke cognitive impairment. Methods: A systematic review of randomized control trials (RCT) was performed using Bayesian network meta-analysis. We searched RCT in the following databases until June 2022: Cochrane Central Register of Controlled Trials (CENTRAL), PUBMED, and EMBASE. We compared any active NIBS to control in terms of improving cognition function and activities of daily living (ADL) capacity following stroke. Results: After reviewing 1577 retrieved citations, a total of 26 RCTs were included. High-frequency (HF)-repetitive transcranial magnetic stimulation (rTMS) (mean difference 2.25 [95% credible interval 0.77, 3.66]) was identified as a recommended approach for alleviating the global severity of cognition. Dual-rTMS (27.61 [25.66, 29.57]) emerged as a favorable technique for enhancing ADL function. In terms of stimulation targets, the dorsolateral prefrontal cortex exhibited a higher ranking in relation to the global severity of cognition. Conclusions: Among various NIBS techniques, HF-rTMS stands out as the most promising intervention for enhancing cognitive function. Alzheimer disease (AD) is an irreversible neurodegenerative disease, and astrocytes play a key role in its onset and progression. The aim of this study is to analyze the characteristics of neurotoxic astrocytes and identify novel molecular targets for slowing down the progression of AD. Single-nucleus RNA sequencing (snRNA-seq) data were analyzed from various AD cohorts comprising about 210,654 cells from 53 brain tissue. By integrating snRNA-seq data with bulk RNA-seq data, crucial astrocyte types and genes associated with the prognosis of patients with AD were identified. The expression of neurotoxic astrocyte markers was validated using 5 × FAD and wild-type (WT) mouse models, combined with experiments such as western blot, quantitative real-time PCR (qRT-PCR), and immunofluorescence. A group of neurotoxic astrocytes closely related to AD pathology was identified, which were involved in inflammatory responses and pathways related to neuron survival. Combining snRNA and bulk tissue data, ZEP36L, AEBP1, WWTR1, PHYHD1, DST and RASL12 were identified as toxic astrocyte markers closely related to disease severity, significantly elevated in brain tissues of 5 × FAD mice and primary astrocytes treated with Aβ. Among them, WWTR1 was significantly increased in astrocytes of 5 × FAD mice, driving astrocyte inflammatory responses, and has been identified as an important marker of neurotoxic astrocytes. snRNA-seq analysis reveals the biological functions of neurotoxic astrocytes. Background and Aims: Chromoendoscopy with Lugol's staining is used to screen for early esophageal squamous cell carcinoma (ESCC). Its efficacy is greatly limited by unstandardized defoaming preparation. This study aimed to confirm whether pre-procedure oral administration of pronase could improve the diagnostic performance of Lugol chromoendoscopy in high-risk patients being screened for early ESCC. Methods: A total of 955 patients at-risk were prospectively recruited for screening for ESCC. Patients were randomly assigned (1:1) to groups with or without (control group) pronase administration. Endoscopic diagnosis of early ESCC was based on the presence of pink-color sign in Lugol's unstained area, and a biopsy was routinely conducted if the Lugol's unstained lesion was larger than 0.5 cm. The early cancer detection rate was used as the primary endpoint. Results: Pre-procedure oral administration of pronase improved mucosal visibility during Lugol chromoendoscopy (P = 0.008). There were no differences in the number of Lugol's unstained lesions between the 2 groups (23.27% [111/477] vs. 25.11% [120/478], P = 0.508). Meaningfully, the detection rate of ESCC (confirmed by histopathology) was significantly higher in the pronase group than in the control group (27.03% [30/111] vs. 17.50% [21/120], P = 0.041), as well as the detection rate of lesions with pink-color sign during chromoendoscopy (35.14% [39/111] vs. 13.33% [16/120], P < 0.001). The diagnostic performance of Lugol chromoendoscopy had improved with the use of pronase (area under the curve = 0.85 vs. 0.69, P = 0.019), accompanied by an increased sensitivity (86.67% vs. 47.62%, P = 0.004). There was no difference in the adverse events between the 2 groups (P = 0.793). Conclusions: Pre-procedure oral administration of pronase significantly increased the detection rate of early ESCC and optimized the diagnostic performance of Lugol chromoendoscopy, which should be recommended during routine endoscopic screening for early ESCC in high-risk patients. In the study, silicon doped carbon quantum dots (Si-CQDs) was prepared by one-pot hydrothermal method with (3-aminopropyl) triethoxysilane (APTES) and o-phenylenediamine (OPD) as raw materials. Then a new ratiometric fluorescent probe (RF-probe) was successfully established for sensitively and selectively monitoring proanthocyanidins (PAs) with a linear range of 10–500 nM and limit of detection (LOD) of 5.6 nM. that is, the fluorescence (FL) intensity of Si-CQDs at 570 nm was used as the built-in reference, while dopamine (DA) reacting with 4-hexylresorcinol (4-HR) could produce a new fluorescent substance (named as azamonardine, AZMON), and its FL intensity at 480 nm was reduced because Cr(VI) could oxidize DA to generate quinone without fluorescence. In the presence of PAs, Cr(VI) was reduced to Cr(III), which caused that the amount of DA reacting with 4-HR was increased, thus the FL intensity of AZMON was recovered. Furthermore, the RF-probe was successfully used for the determination of PAs in black goji berry from two different areas and PAs capsule with satisfactory results compared to the standard HPLC method. Drought is influenced by numerous factors, and traditional studies have only considered meteorological factors, but human activities are also an important influencing factor. From 1980 to 2010, the reform and opening up and the project of returning farmland to forest have largely changed the land use type of Loess Plateau in Shaanxi. In order to study the influence of land use types on drought in the study area, 8 stations with different land use types were selected based on remote sensing data and meteorological data. Based on univariate linear regression, the response of the drought index SPI to land use types was analyzed sequentially in each of the five time stages. The results showed that from 1980 to 2000, forest had the greatest drought intensity, followed by arable land and town and country. However, the response of arable land to the drought index was the greatest, followed by town and country. Forest had the weakest response to the drought index. From 2000 to 2010, the greatest degree of drought was observed in forest, followed by town and country and arable land. And forest has the strongest response to the drought index, followed by town and country, and finally, arable land. The area of forest, grass and town and country keeps increasing. The area of arable land is decreasing year by year. Land use types are constantly undergoing complex transformations. The drought index responds strongly to the change of both arable land to grass and arable land to town and country, while it responds weakly to the mutual transformation of both grass and town and country. Building on the resource-based view (RBV) theory, this paper aims to shed light on how does the implementation of green strategies affect enterprises' performance. To distinguish the evolution of strategy implementation effect, we adopt a panel estimation strategy and gather data from 3869 listed companies in China from 2008 to 2019. Furthermore, we innovatively use the semi-supervised clustering algorithm to classify the companies according to whether they implement green strategies or not and then discuss long-term and short-term financial effects of implementing green strategies. Our study finds that the implementation of green strategy facilitates a company's long-term performance but hampers its short-term performance. According to the moderating analysis, a green strategy could negatively impact a company's financial performance by increasing debt ratios. The findings highlight the importance of implementing green strategies and the obstacles in the process of transforming enterprises to be green. [ABSTRACT FROM AUTHOR]

Kaolinite, a natural, layered phyllosilicate, has been used as the solid catalyst for the hydrolysis of cellulose in our previous work. In the present study, kaolinite was activated by mineral acid and further evaluated for the hydrolysis of cellulose in water. The acid-activated kaolinite was characterized by XRF, XRD, FT-IR, BET and TG. The effects of reaction temperature, reaction time, mass ratio and water amount were investigated in the system. It was found that the highest total reducing sugars (TRS) yield of 50.2% was obtained on the kaolinite activated by 20% HNO3 with the mass ratio of catalyst to cellulose of 0.2 and water to cellulose of 14 at 205 ℃ for 3 h. Moreover, the catalyst was easily regenerated by calcination and the yield of TRS on the regenerated catalyst changed between 50.2% and 45.2% after four times reuse. The results showed that the acid activation could influence the crystallinity and improve the specific surface area, but the high TRS yield of the activated kaolinite should be ascribed to the increasing of the interlayer Al–OH groups and the formation of inner hydrogen bonds between the octahedral sheets and tetrahedral sheets. Finally, it was suggested that this was the effective way for improving the catalytic performance by increasing the interlayer Al–OH groups on the kaolinite catalyst. [ABSTRACT FROM AUTHOR]

Background: COVID-19 is erupting globally. Mass quarantine had been implemented all around China which could influence the psychological status of patients with memory disorders and their caregivers. Aim: To investigate the psychological impact of mass quarantine on patients with memory disorders and their caregivers in China. Methods: We completed a cross-sectional study in 787 patients and their caregivers registered from 2010 to 2019 in Memory Clinic, The First Affiliated Hospital of Chongqing Medical University, by telephone interviews. The performance in neuropsychiatric symptoms (NPSs), sleep, nutrition, chronic diseases of patients, and the burden of care, anxiety and depression of caregivers was assessed by six assessment scales (MNA-SF, PSQI, NPI, RSS, PHQ-9 and GAD-7). Results: Only 68 (8.6%) patients worried about the outbreak of COVID-19. The prevalence of NPSs among all subjects was nearly 60.0%. Approximately 50.0% of the caregivers reported distress. More than 70.0% of patients remained stable in NPSs. However, anxiety, depression, aberrant motor disorder and delusion were exacerbated (22.9%, 18.6%, 17.1% and 16.8%, respectively). Appetite and eating disorder led alleviation rate by 25.8% while disappearing rate of agitation led by 5.8%. 7.5% of caregivers manifested depressive symptoms while 4.9% expressed anxiety symptoms, and 40.8% showed care burden. The coefficients of RSS and PHQ-9, RSS and GAD-7, RSS and NPI-D, PHQ-9 and GAD-7 were 0.7, 0.5, 0.5 and 0.6, respectively (p < 0.01). Conclusion: Changes in NPSs during COVID-19 were observed in some patients with memory disorders and their caregivers, and adherence to medication contributed to the stabilization of NPSs. Sodium alginate (SA) is a linear biopolymer, which is the nontoxic, biodegradable, and rich in carboxyl and hydroxyl groups. In the paper, the SA-based hydrogel bead was prepared by the cationic hectorite clay and anionic sodium alginate with a simple ionic gelation method under freeze-drying, and the adsorption properties were evaluated by the removal of copper ions from aqueous solutions. The composites were characterized by X-ray diffraction (XRD), nitrogen adsorption-desorption isotherm (BET), thermal analysis (TG), and Fourier transform infrared spectroscopy (FT-IR). The pseudo-first-order and pseudo-second-order kinetic models were used to describe the kinetic data and the Langmuir, Freundlich, Dubinin–Radushkevich (D-R), and Temkin models were applied to describe the adsorption isotherms. The results showed that the adsorption process was found to follow the Freundlich isotherm model and the maximum sorption capacity was observed to be 160.28 mg/g under the initial concentration from 10 to 700 mg/L at 45 °C. Adsorption kinetics data fitted well with pseudo-second-order rate model. The porous structure of the composite was responsible for the adsorption of Cu2+ ions. Recent studies have shown that the non-enzymatic function of CD73 plays a key role in tumor progression, but this function of CD73 in pancreatic cancer cells has not been studied. Furthermore, little is known about the mechanism involved in CD73 regulation in tumors. Here, we found that CD73 expression was upregulated in pancreatic ductal adenocarcinoma (PDAC) and that its expression correlated with poor prognosis. CD73 knockdown inhibited cell growth and induced G1 phase arrest via the AKT/ERK/cyclin D signaling pathway. We also found that tumor necrosis factor receptor (TNFR) 2 was involved in CD73-induced AKT and ERK signaling pathway activation in PDAC. Further, miR-30a-5p overexpression significantly increased the cytotoxic effect of gemcitabine in pancreatic cancer by directly targeting CD73 messenger RNA (mRNA), suggesting that regulation of the miR-30a-5p/CD73 axis may play an important role in the development of gemcitabine resistance in pancreatic cancer. In summary, this regulatory network of CD73 appears to represent a new molecular mechanism underlying PDAC progression, and the mechanistic interaction between miR-30a-5p, CD73, and TNFR2 may provide new insights into therapeutic strategies for pancreatic cancer. Clinically, superimposed systemic inflammation generally has significant deleterious effects on the Alzheimer's disease (AD) progression. However, the related molecular mechanisms remain poorly understood. Microglial toll-like receptor 4 (TLR4) and triggering receptor expressed on myeloid cells 2 (TREM2) are two key regulators of inflammation that may play an essential role in this complex pathophysiological process. In this study, intraperitoneal injection of lipopolysaccharide (LPS) into APP/PS1 transgenic AD model was used to mimic systemic inflammation in the development of AD. Initial results from the cortex showed that compared with wild-type mice, APP/PS1 mice exhibited elevated gene and protein expression levels of both TLR4 and TREM2 with different degree. Interestingly, after LPS treatment, TLR4 expression was persistently up-regulated, while TREM2 expression was significantly down-regulated in APP/PS1 mice, suggesting that the negative regulatory effect of TREM2 on inflammation might be suppressed by LPS-induced hyperactive TLR4. This imbalance of TLR4/TREM2 contributed to microglial over-activation, followed by increased neuronal apoptosis in the cortex of APP/PS1 mice; these changes did not alter the expression level of Aβ1−42. Similar alterations were observed in our in vitro experiment with β-amyloid1–42 (Aβ1–42)-treated N9 microglia. Further, Morris water maze (MWM) testing data indicated that LPS administration acutely aggravated cognitive impairment in APP/PS1 mice, suggesting that the addition of systemic inflammation can potentially accelerate the progression of AD. Collectively, we conclude that an imbalance of TLR4/TREM2 may be a potential link between AD and systemic inflammation. Smad anchor for receptor activation (SARA) is an important regulator of transforming growth factor β (TGF-β) signaling by recruiting Smad2/3 to TGF-β receptors. We recently demonstrated that the expressions of SARA and level of downstream phospho-Smad3 (p-Smad3) were upregulated in the brain in the epileptic rat model, but were never examined in patients with temporal lobe epilepsy (TLE). In this study, we examined the expressions of SARA and level of p-Smad3 in brain tissues of TLE patients using immunohistochemistry and western blot to demonstrate that SARA activation in neurons is sufficient to facilitate TGF- β pathway in patients to regulate epilepsy. We found that the expressions of SARA and level of p-Smad3 were significantly upregulated in neurons of the temporal cortex of TLE patients compared to controls. Moreover, SARA and p-Smad3 were strongly stained in the cytoplasm in the temporal cortex of TLE patients. Our results indicate that upregulation of SARA and p-Smad3 in cortex neurons might be involved in the development of intractable temporal lobe epilepsy. [ABSTRACT FROM AUTHOR]

Inhibition of endocytosis in an Alzheimer’s disease (AD) model has been shown to be able to prevent amyloid β (Aβ)-induced damage and to exert a beneficial effect in treating AD. Adaptor-associated kinase 1 (AAK1), which binds to the adaptor protein complex 2 (AP-2), regulates the process of clathrin-mediated endocytosis. However, how AAK1 expression varies over the course of AD is unknown. In this study, we investigated AAK1 levels in AD model mice over time. Aβ1-42 was used to establish a mouse AD model, and the Morris water maze test was used to characterize the time course of Aβ1-42-induced cognition changes. ELISA was used to determine AAK1 levels in plasma and Aβ1-42 levels in brain tissues. Subsequently, the protein or gene levels of AAK1, AP-2, and Rab5 (an early endosome marker) were tested in each group. The cognitive function of Aβ1-42-induced mice was significantly declined compared to control group, and the deficits reached a peak on day 14, but partly recovered on day 30. Moreover, the level of Aβ1-42 detected with ELISA was highest on day 14, but reduced on day 30, paralleling the cognitive changes in the mice in our study. AAK1, AP-2, and Rab5 expression showed the same periodic variation as the changes in cognition. Thus, periodic variation in AAK1 expression is closely correlated to the decline in cognition, and AAK1 might be a suitable indicator for Alzheimer’s disease. [ABSTRACT FROM AUTHOR]

It is reported that chitinase1 increases in Alzheimer's disease (AD). However, the alteration of chitinase1 in the progress of AD is still unclear. Thus, we designed the present study to detect chitinase1 level in different stages of APP/PS1 double transgenic mice. Experimental models were APP/PS1 double transgenic mice with 4, 12 and 22 months. Cognitive function was detected by Morris water maze test in APP/PS1 mice as well as controls. ELISA and the quantitative RT-PCR were used to detect chitinase1 level in different groups. The study displayed that expression of chitinase1 gradually increased in a time-dependent manner in APP/PS1 mice, while there were no statistical differences among the wild-type mice in varies ages. Moreover, chitnase1 increased significantly in APP/PS1 mice aged 12 and 22 months compared with the age matched wild-type group, respectively. However, no difference of chitnase1 was found between 4 months-old APP/PS1 mice and wild-type mice. Comparing with the age matched wild type group, the consequences of mRNA on the increase in chitnase1 is in accordance with protein in APP/PS1 mice. Furthermore, Morris water maze showed that 4 months-old APP/PS1 mice have normal spatial learning and impaired spatial memory; both spatial learning and spatial memory in 12 and 22 months-old APP/PS1 mice were declined. Time-dependent increase of chitnase1 in APP/PS1 double transgenic mice indicates that the level of chitinase1 is associated with decline of cognition. Emerging data suggests that mitochondrial dysfunction is prominently involved in Alzheimer disease (AD) progression. Sirtuin-3 (Sirt3) is a member of the sirtuin family of nicotinamide adenine dinucleotide dependent deacetylases that regulates a variety of mitochondrial functions and suppresses mitochondria-related physiology. Here, we determined sirt3 expression in a mouse model of AD. Spatial learning and memory were tested by Morris water maze in APP/PS1 double transgenic mice. The expression of sirt3 was assayed by real-time quantitative PCR and western blotting. Age-and gender-matched wild-type (WT) littermates were used as controls. Cortical sirt3 localization was assessed using immunohistochemistry. The expression of sirt3 mRNA was significantly lower in the cortex of APP/PS1 double transgenic mice than in WT littermates (0.83 ± 0.24 vs. 1.10 ± 0.21, P < 0.05). A comparable reduction was found in sirt3 protein levels using western blotting. The ratio of mean optical density (MOD) of total sirt3/β-actin in the cortex was 0.77 ± 0.11 in APP/PS1 double transgenic mice and 1.34 ± 0.17 in the WT littermates ( P < 0.01). Immunohistochemistry showed the same change as western blotting. The ratio of MOD of integral optical density/total area in APP/PS1 and WT littermates was 0.58 ± 0.02 and 0.71 ± 0.05 ( P < 0.01). These data show that sirt3 was depleted in APP/PS1 double transgenic mice. The results suggest that mitochondrial sirt3 might participate in the development of AD via mitochondrial dysfunction. Transforming growth factor beta (TGFβ) signaling participates in pathogenesis of epilepsy. TGFβ1, as a transmitter of TGFβ signaling, might be a useful marker for predicting the prognosis of patients with epilepsy. The present study aimed to measure TGFβ1 level in the cerebrospinal fluid (CSF) of patients with drug-resistant epilepsy and non-resistant epilepsy. A total of 43 patients with epilepsy were recruited, 28 were non-resistant epilepsy subgroup, 15 drug-resistant epilepsy subgroup. 11 patients with intracranial infection and 11 individuals with primary headache were used as controls. The concentration of CSF and serum TGFβ1 was measured by enzyme-linked immunosorbent assay. The concentration of CSF-TGFβ1 was 209.26 ± 81.07 pg/ml in the drug-resistant epilepsy subgroup, 121.80 ± 40.32 pg/ml in the non-resistant epilepsy subgroup, 552.17 ± 456.20 pg/ml in intracranial infection control, 133.80 ± 68.55 pg/ml in headache control, respectively. TGFβ1 level was significantly increased in the drug-resistant epilepsy subgroup compared to the non-resistant epilepsy subgroup. TGFβ1 level in intracranial infection control was higher than that in the non-resistant epilepsy subgroup. There was no statistically difference of CSF-TGFβ1 between the non-resistant epilepsy subgroup and headache controls, between the resistant epilepsy subgroup and intracranial infection controls. TGFβ levels are increased in the CSF of patients with drug-resistant epilepsy. High CSF-TGFβ1 levels may be a potential screening biomarker of antiepileptic drug resistance in patients with epilepsy. Our previous study on proteomic analysis has shown that clusterin (CLU) is significantly decreased in the cerebrospinal fluid (CSF) of patients with epilepsy. Therefore, the present study aimed to confirm CLU concentration reduction in the CSF of patients with drug-resistant epilepsy and drug-responsive epilepsy. Fifty-two patients with epilepsy (23 drug resistance and 29 drug effectivity) and 20 control individuals were recruited. The concentrations of CSF and serum CLU were detected. Moreover, alteration of CLU was detected in the rat hippocampus over time after pilocarpine-induced status epilepticus (SE). Our results showed that human CSF-CLU levels were decreased in patients with both drug-resistant epilepsy and drug-responsive epilepsy compared to controls, and concentration of CSF-CLU was obviously lower in drug-resistant epilepsy than in drug-responsive epilepsy. In the pilocarpine-induced seizure rats, expression of neuronal CLU was gradually decreased in a time-dependent manner from acute phase to chronic phase after the onset of SE. In conclusion, CLU level is decreased in the CSF of human epilepsy and the similar alteration is confirmed in a rat model with epilepsy. The emission of gasoline vapors is becoming a significant environmental problem especially for the population-dense area and also results in a significant economic loss. In this study, adsorption equilibrium and dynamics of gasoline vapors onto macroporous and hypercrosslinked polymeric resins at 308 K were investigated and compared with commercial activated carbon (NucharWV-A 1100). The results showed that the equilibrium and breakthrough adsorption capacities of virgin macroporous and hypercrosslinked polymeric resins were lower than virgin-activated carbon. Compared with origin adsorbents, however, the breakthrough adsorption capacities of the regenerated activated carbon for gasoline vapors decreased by 58.5 % and 61.3 % when the initial concentration of gasoline vapors were 700 and 1,400 mg/L, while those of macroporous and hypercrosslinked resins decreased by 17.4 % and 17.5 %, and 46.5 % and 45.5 %, respectively. Due to the specific bimodal property in the region of micropore (0.5−2.0 nm) and meso-macropore (30-70 nm), the regenerated hypercrosslinked polymeric resin exhibited the comparable breakthrough adsorption capacities with the regenerated activated carbon at the initial concentration of 700 mg/L, and even higher when the initial concentration of gasoline vapors was 1,400 mg/L. In addition, 90 % of relative humidity had ignorable effect on the adsorption of gasoline vapors on hypercrosslinked polymeric resin. Taken together, it is expected that hypercrosslinked polymeric adsorbent would be a promising adsorbent for the removal of gasoline vapors from gas streams. [ABSTRACT FROM AUTHOR]

Background: Topiramate is a newer generation antiepileptic drug with a wide range of antiepileptic efficacy as monotherapy or as adjunctive therapy, and which has shown positive activity in intractable epilepsy and newly diagnosed epilepsy. Topiramate has also been shown to exert good seizure control with a low incidence of adverse effects in brain tumour-associated epilepsy. However, there have been few reports on the efficacy of topiramate in the treatment of symptomatic epilepsy of varying aetiologies.Objective: The aim of the present study was to evaluate the efficacy of topiramate in the treatment of adult patients with symptomatic epilepsy of various aetiologies.Methods: This was an open-label, long-term, retrospective observation. 227 patients with symptomatic epilepsy (110 male, 117 female) were enrolled into this study. The underlying aetiologies included low-grade brain tumour, head trauma, cerebrovascular diseases, infection, diabetes mellitus, hydrocephalus and parasitosis. Topiramate was titrated up to a target dosage of 200 mg/day and maintained for at least 1 year. Response to topiramate was defined as > or = 50% reduction in seizure frequency compared with baseline. Seizure free was defined as no seizure occurring during 1 year of topiramate therapy.Results: 157 (69.2%) patients were responders and 124 (54.6%) patients were seizure free with topiramate administration. Responders by subgroup included 40 patients (74.0%) with low-grade brain tumour, 32 (55.2%) with trauma, 30 (90.9%) with cerebrovascular disease, 21 (55.3%) with infection, 18 (81.8%) with diabetes, 12 (85.7%) with parasitosis and 4 (50.0%) with hydrocephalus. The percentage of seizure-free patients by subgroup was 61.0% with brain tumours, 31.0% with trauma, 78.8% with cerebrovascular disease, 44.7% with infection, 59.0% with diabetes, 85.7% with parasitosis and 50.0% with hydrocephalus. The incidence of adverse effects was 36.1%. The most commonly reported adverse effects were weight loss, memory impairments, paraesthesia, headache and dizziness; most were mild to moderate in severity and transient. Sixty-eight (30.0%) patients withdrew from topiramate treatment in this study: topiramate was discontinued in 56 patients because of lack of efficacy and in 12 patients because of adverse effects. At the end of the study, 109 patients received topiramate monotherapy, including 52 newly diagnosed patients and 57 subjects who transferred to topiramate monotherapy successfully; another 118 patients received add-on topiramate therapy. The percentage of patients responding to topiramate was 85.3% in the monotherapy group and 54.2% in the topiramate add-on therapy group; the percentage of seizure-free patients was 68.8% in the topiramate monotherapy group and 41.5% in the topiramate add-on therapy group.Conclusion: When administered either as a single drug or as an add-on drug, topiramate is effective and well tolerated in adult patients with symptomatic epilepsy of various aetiologies. Voltage-gated sodium channel genes are associated with idiopathic generalized epilepsy. Our group preciously identified a suggestive new locus on chromosome 11q22.1-23.3 in a five-generational Chinese epileptic family with generalized tonic-clonic seizure. SCN2B, SCN3B and SCN4B, which located at 11q22.1-23.3 locus, were chosen as candidate genes for this family. In the present study, genomic DNA was extracted in six affected family members. All exons of SCN2B, SCN3B and SCN4B were sequenced using direct DNA sequence analysis. The results showed that no mutation or polymorphism of coding regions of SCN2B, SCN3B and SCN4B was detected in the tested family members. Therefore, SCN2B, SCN3B and SCN4B are not major susceptibility genes contributed to our large family. [ABSTRACT FROM AUTHOR]
