2. Effect of CD4 count on Mycobacterium tuberculosis infection rates in people living with HIV: a comparative study in prison and community.

To evaluate the impact of the CD4 count on ESAT6-CFP10 (EC) skin test, QuantiFERON-TB Gold In-tube test (QFT) and tuberculin skin test (TST) in a large prison and communities in Jiangsu Province among people living with HIV (PLHIV). Participants in communities were tested with the QFT and EC skin test and in prison were tested with the QFT, TST and EC skin test. A 4-knotted restricted cubic spline fitted for Logistic models was used to explore the cutoff point of CD4 count and the associations between changes in CD4 count and Mycobacterium tuberculosis (M.tb) infection. Among 1,815 PLHIV included, 19.3% (350) were from prisons and 80.7% (1465) were identified through community screenings. M.tb infection rates were 12.2% and 8.3% through QFT and EC tests, respectively. Odd Ratios (ORs) for infection increased with CD4 counts, peaking at 618 cells/mm
3 for EC, 392 cells/mm3 for QFT, then plateaued. However, the pattern differed for EC between prison and screening scenarios. In prison settings, M.tb infection increased with CD4 count from 0 to 350 cells/mm3, plateaued until 500 cells/mm3 , and then persistently increased. Conversely, for community, ORs decreased until 300 cells/mm3 , followed by an increase between 300 and 729 cells/mm3 , after which they continued to rise persistently. Our cross-sectional study among PLHIV revealed a higher rate of M.tb infection in prison compared to the community. The relationship between CD4 count and infection became negative after a certain threshold. This pivotal point differed with detection methods, with QFT showing lower CD4 thresholds than EC. To ensure the successful implementation of the old community renewal project (OCRP), it is essential for the participants to allocate the project risks reasonably. Firstly, this study comprehensively identifies the 20 key risk factors of the OCRP. Secondly, an index system is established from three dimensions to evaluate the risk allocation ability of participants, including a total of nine evaluation indexes. Furthermore, a risk-sharing model based on TOPSIS method and bargaining game model is proposed to determine the optimal risk bearer and risk-taking ratio between the government and the private sector in OCRP. Finally, an OCRP in Chongqing is taken as a case study to verify the applicability of the developed model. The results indicate that in OCRP under PPP mode, the government need to independently bear 7 risks related to politics, law, policy, while the private sector needs to independently bear 8 risks mainly from project financing, design, construction, operation, and maintenance stages. In addition, the risk-taking ratios of 5 risks that require both parties to share are divided. Insulin resistance (IR) was found to be a critical element in the pathogenesis of Parkinson's disease (PD), facilitating abnormal α-synuclein (α-Syn) aggregation in neurons and thus promoting PD development. However, how IR contributes to abnormal α-Syn aggregation remains ill-defined. Here, we analyzed six PD postmortem brain transcriptome datasets to reveal module genes implicated in IR-mediated α-Syn aggregation. In addition, we induced IR in cultured dopaminergic (DA) neurons overexpressing α-Syn to identify IR-modulated differentially expressed genes (DEGs). Integrated analysis of data from PD patients and cultured neurons revealed 226 genes involved in α-Syn aggregation under IR conditions, of which 53 exhibited differential expression between PD patients and controls. Subsequently, we conducted an integrated analysis of the 53 IR-modulated genes employing transcriptome data from PD patients with different Braak stages and DA neuron subclasses with varying α-Syn aggregation scores. Protein tyrosine phosphatase receptor type O (PTPRO) was identified to be closely associated with PD progression and α-Syn aggregation. Experimental validation in a cultured PD cell model confirmed that both mRNA and protein of PTPRO were reduced under IR conditions, and the downregulation of PTPRO significantly facilitated α-Syn aggregation and cell death. Collectively, our findings identified PTPRO as a key regulator in IR-mediated α-Syn aggregation and uncovered its prospective utility as a therapeutic target in PD patients with IR. Deschampsia antarctica Desv. and Poa annua L. are two Poaceae plants with enough endurance to successfully establish populations in the Antarctic region. Their adaptation to the Antarctic environment is closely linked to root-associated microbial communities. In this study, we obtained 16S rRNA sequencing data of the root-associated microbial communities of these two Poaceae plants from NCBI. Meta-analysis was used to investigate the similarities and differences between the root endosphere and rhizosphere-dwelling microbial communities in these two Poaceae plants. Here we report that two Poaceae-Poaceae plants' rhizospheric communities were found to be more species diversity than endospheric communities. The species diversity of P. annua was higher than that of D. antarctica in both endosphere and rhizosphere communities. Seven bacterial families form a core microbiome of two Antarctic Poaceae plants' root endosphere, in which Microbacteriaceae appears to be obligatory root endophytes of the two Antarctic Poaceae plants. The core microbiome of the two Poaceae plants' rhizosphere has six bacterial families. Chitinophagaceae, Burkholderiaceae, and Flavobacteriaceae are most likely to play a crucial role in Poaceae plants' adaptation to cold Antarctic conditions. Sphingobacteriaceae, Caulobacteraceae, Gemmatimonadaceae, and Flavobacteriaceae have a great influence on two Antarctic Poaceae plants. [ABSTRACT FROM AUTHOR]

With development of present energy-saving society, lightweight and green development in the automotive and aerospace industries have put forward urgent demands for heat-resistant cast aluminum alloys. Present cast aluminum alloys are of lightweight and have excellent mechanical properties when serving in ambient environment. However, when serving at temperatures of 200–300 ℃ or even higher, the alloys inevitably soften and the high-temperature mechanical properties decline rapidly, hardly meeting the requirements for high-performance equipments. This paper summarizes the development process of classic heat-resistant aluminum alloys, especially the microstructural characteristics of heat-resistant cast aluminum alloys in the past decade, and reviews the current microstructure tuning strategies of heat-resistant aluminum alloys from the aspects of intrinsic heat resistance of the second phases, phase interface modification, diffusion-controlled phase transition, grain boundary stabilization and composite material design. 7. FANs: fully attentional networks for image compression.

We propose a fully attentional networks model and achieve excellent results on top of both large-scale datasets and small-scale datasets to use as a codec for image compression. First, we convert the image to an underlying tensor using a lightweight image encoder with a convolutional neural network model. Then, a fully attentional networks model is designed to enhance image compression in combination with attentional channel processing. Combining the fully attentional networks model with associated feature learning can improve image compression performance. In addition, we design a channel processing module to compute the attention matrix in the channel dimension for improving the quality of the reconstructed images. Since compression networks are computationally and parametrically inefficient in channel processing, we design a high-efficiency channel self-attention for the FANs model that uses averaging of channel dimensions to generate token features for optimizing the reconstructed images at low rates. We evaluate our proposed method on the Kodak and CLIC datasets and compare it with recently published deep neural network-based methods. 8. Traffic forecasting on new roads using spatial contrastive pre-training (SCPT).

New roads are being constructed all the time. However, the capabilities of previous deep forecasting models to generalize to new roads not seen in the training data (unseen roads) are rarely explored. In this paper, we introduce a novel setup called a spatio-temporal split to evaluate the models' capabilities to generalize to unseen roads. In this setup, the models are trained on data from a sample of roads, but tested on roads not seen in the training data. Moreover, we also present a novel framework called Spatial Contrastive Pre-Training (SCPT) where we introduce a spatial encoder module to extract latent features from unseen roads during inference time. This spatial encoder is pre-trained using contrastive learning. During inference, the spatial encoder only requires two days of traffic data on the new roads and does not require any re-training. We also show that the output from the spatial encoder can be used effectively to infer latent node embeddings on unseen roads during inference time. The SCPT framework also incorporates a new layer, named the spatially gated addition layer, to effectively combine the latent features from the output of the spatial encoder to existing backbones. Additionally, since there is limited data on the unseen roads, we argue that it is better to decouple traffic signals to trivial-to-capture periodic signals and difficult-to-capture Markovian signals, and for the spatial encoder to only learn the Markovian signals. Finally, we empirically evaluated SCPT using the ST split setup on four real-world datasets. The results showed that adding SCPT to a backbone consistently improves forecasting performance on unseen roads. The colloidal borescope, using colloidal particle motion, is used to monitor the flow velocities and directions of groundwater. It integrates advanced techniques such as microscopy, high-speed photography, and big data computing and enjoys high sensitivity at the micron level. However, In the same well, the groundwater flow velocity monitored by colloidal hole mirror is varies greatly from that obtained by conventional hydrogeological monitoring, such as pumping test. In order to solve this problem, the stability catcher and stratified packer are designed to control the interference of the vertical flow in drilling, and to monitor the flow velocity and direction of groundwater velocity at the target aquifer and target fracture. Five wells with different aquifers and different groundwater types were selected for monitoring in south-central China. The instantaneous velocity and direction are converted into east–west component and north–south component, the average velocity and direction is calculated according to the time of 10 min, and the particle trajectory diagram is established. Based on these results, it proposed a concept of cumulative flow velocity. Using curve-fitting equations, the limits of cumulative flow velocities as the monitoring time tends to infinity were then calculated as the actual flow velocities of the groundwater. The permeability coefficient of aquifer is calculated by using the fissure ratio of aquifer, hydraulic slope and flow velocity, and compared with the permeability coefficient obtained by pumping test. The results are as follows: (1) The variation coefficient of the instantaneous flow velocity measured at the same depth in the same well at different times is greater than that of the time average flow velocity and greater than that of the cumulative flow velocity. The variation coefficient of the actual velocity is the smallest, indicating that the risk of using the actual flow velocity is lower. (2) The variation coefficient of the flow rate monitored at different depths in the same well is mainly controlled by the properties of the aquifer. The more uniform water storage space in the aquifer, the smaller the variation coefficient. (3) The comparison between the permeability coefficient obtained by monitoring and the permeability coefficient obtained by pumping test shows that the flow of structural fissure water controlled by planar fissure is more surface flow, and the results are consistent. When the groundwater flow is controlled by pores and solution gaps, the flow channel is complicated, which is easy to produce turbulent flow, and the result consistency is poor. (4) According to different research accuracy requirements, different monitoring and calculation methods can be selected for different aquifers and groundwater types. Researches show that, the permeability coefficient calculated for the actual flow velocity in well DR01 is the same as that calculated for the pumping test. The aquifer characteristics reflected by the coefficient of variation of the actual flow velocity in the same aquifer are more realistic. The pumping test method obtains the comprehensive parameters of a certain aquifer, and this method can be used to monitor a certain fissure. In this paper, the new technology developed for monitoring, and the new algorithm established for data processing, can accurately obtain the flow velocity and direction of groundwater, using capsule hole mirror monitoring method. Mechanoluminescence (ML) sensing technologies open up new opportunities for intelligent sensors, self-powered displays and wearable devices. However, the emission efficiency of ML materials reported so far still fails to meet the growing application requirements due to the insufficiently understood mechano-to-photon conversion mechanism. Herein, we propose to quantify the ability of different phases to gain or lose electrons under friction (defined as triboelectric series), and reveal that the inorganic-organic interfacial triboelectricity is a key factor in determining the ML in inorganic-organic composites. A positive correlation between the difference in triboelectric series and the ML intensity is established in a series of composites, and a 20-fold increase in ML intensity is finally obtained by selecting an appropriate inorganic-organic combination. The interfacial triboelectricity-regulated ML is further demonstrated in multi-interface systems that include an inorganic phosphor-organic matrix and organic matrix-force applicator interfaces, and again confirmed by self-oxidization and reduction of emission centers under continuous mechanical stimulus. This work not only gives direct experimental evidences for the underlying mechanism of ML, but also provides guidelines for rationally designing high-efficiency ML materials. Mechanoluminescence enables sensing applications of mechanical stimuli. 11. Disulfidptosis features and prognosis in head and neck squamous cell carcinoma patients: unveiling and validating the prognostic signature across cohorts.

- Abstract
Background: Head and neck squamous cell carcinoma (HNSCC) is a significant health concern with a variable global incidence and is linked to regional lifestyle factors and HPV infections. Despite treatment advances, patient prognosis remains variable, necessitating an understanding of its molecular mechanisms and the identification of reliable prognostic biomarkers. Methods: We analyzed 959 HNSCC samples and employed batch correction to obtain consistent transcriptomic data across cohorts. We examined 79 disulfidptosis-related genes to determine consensus clusters and utilized high-throughput sequencing to identify genetic heterogeneity within tumors. We established a disulfidptosis prognostic signature (DSPS) using least absolute shrinkage and selection operator (LASSO) regression and developed a prognostic nomogram integrating the DSPS with clinical factors. Personalized chemotherapy prediction was performed using the "pRRophetic" R package. Results: Batch corrections were used to harmonize gene expression data, revealing two distinct disulfidptosis subtypes, C1 and C2, with differential gene expression and survival outcomes. Subtype C1, characterized by increased expression of the MYH family genes ACTB, ACTN2, and FLNC, had a mortality rate of 48.4%, while subtype C2 had a mortality rate of 38.7% (HR = 0.77, 95% CI: 0.633–0.934, P = 0.008). LASSO regression identified 15 genes that composed the DSPS prognostic model, which independently predicted survival (HR = 2.055, 95% CI: 1.420–2.975, P < 0.001). The prognostic nomogram, which included the DSPS, age, and tumor stage, predicted survival with AUC values of 0.686, 0.704, and 0.789 at 3, 5, and 8 years, respectively, indicating strong predictive capability. In the external validation cohort (cohort B), the DSPS successfully identified patients at greater risk, with worse overall survival outcomes in the high-DSPS subgroup (HR = 1.54, 95% CI: 1.17–2.023, P = 0.002) and AUC values of 0.601, 0.644, 0.636, and 0.748 at 3, 5, 8, and 10 years, respectively, confirming the model's robustness. Conclusion: The DSPS provides a robust prognostic tool for HNSCC, underscoring the complexity of this disease and the potential for tailored treatment strategies. This study investigated the association between household characteristics, perceived family support (PFS), and the developmental outcomes (resilience, academic performance, and prosociality) among at-risk students. Our large sample included 1564 primary and secondary school students from poor rural China (M = 11.55 years old). Having a caregiver whose resilience score was in the top 50% of the sample was associated with a 0.48-point increase (or 0.31 d effect size), while having a migrant mother was correlated with a 0.26-point decrease (or 0.17 d effect size). PFS was a significant (p < 0.0001) mediator between household characteristics and developmental outcomes. Our study highlights the link between caregiver resilience and PFS, and the healthy functioning of disadvantaged students in a developing context. [ABSTRACT FROM AUTHOR]

The interaction between dislocation and solute atoms during uniaxial tensile-loading deformation was simulated based on molecular dynamics. It is proved from the microscopic view (atomic level) that the essence of Portevin–Le Chatelier (PLC) effect originates from the influence of the solute atmosphere on the dislocation motion. The formation of the solute atmosphere is caused by lattice distortion. The lattice distortion increases the energy of the system, which promote the spontaneous diffusion of solute atoms through vacancies to the vicinity of edge dislocations to reduce the energy of the system. Programmable metasurfaces present significant capabilities in manipulating electromagnetic waves, making them a promising candidate for simultaneous wireless information and power transfer (SWIPT), which has the potential to enable sustainable wireless communication in complex electromagnetic environments. However, challenges remain in terms of maximum power transmission distance and stable phase manipulation with high-power scattered waves. Additionally, waveform limitations restrict average scattered power and rectifier conversion efficiency, affecting data transmission rates and energy transmission distance. Here we show an amplifying programmable metasurface (APM) and a joint modulation method to address these challenges. The APM mitigates the peak-to-average power ratio and improves maximum power, phase response stability, average output power, and rectifier conversion efficiency. Through experimental validation, we demonstrate the feasibility of the SWIPT system, showcasing simultaneous LED array powering and movie video transmission. This innovative SWIPT system holds promise for diverse applications, including 6 G wireless communications, IoT, implanted devices, and cognitive radio networks. In this work, the authors demonstrate a 'jointly modulated' amplifying programmable metasurface (APM) for simultaneous wireless information and power transmission (SWIPT). Their technique outperforms existing methods, significantly improving power transmission and adaptability for conveying energy and data across various domains, including wireless implants, 6 G networks, and IoT systems. - Abstract
Nav1.8, a tetrodotoxin-resistant voltage-gated sodium channels (VGSCs) subtype encoded by SCN10A, which plays an important role in the production and transmission of peripheral neuropathic pain signals. Studies have shown that VGSCs may be key targets of MicroRNAs (miRNAs) in the regulation of neuropathic pain. In our study, bioinformatics analysis showed that the targeting relationship between miR-3584-5p and Nav1.8 was the most closely. The purpose of this study was to investigate the roles of miR-3584-5p and Nav1.8 in neuropathic pain. The effects of miR-3584-5p on chronic constriction injury (CCI)-induced neuropathic pain in rats was investigated by intrathecal injection of miR-3584-5p agomir (an agonist, 20 μM, 15 μL) or antagomir (an antagonist, 20 μM, 15 μL). The results showed that over-expression of miR-3584-5p aggravated neuronal injury by hematoxylin–eosin (H&E) staining and mechanical/thermal hypersensitivity in CCI rats. MiR-3584-5p indirectly inhibited the expression of Nav1.8 by up-regulating the expression of key proteins in the ERK5/CREB signaling pathway, and also inhibited the current density of the Nav1.8 channel, changed its channel dynamics characteristic, thereby accelerating the transmission of pain signals, and further aggravating pain. Similarly, in PC12 and SH-SY5Y cell cultures, miR-3584-5p increased the level of reactive oxygen species (ROS) and inhibited mitochondrial membrane potential (Δψm) in the mitochondrial pathway, decreased the ratio of apoptosis-related factor Bcl-2/Bax, and thus promoted neuronal apoptosis. In brief, over-expression of miR-3584-5p aggravates neuropathic pain by directly inhibiting the current density of Nav1.8 channel and altering its channel dynamics, or indirectly inhibiting Nav1.8 expression through ERK5/CREB pathway, and promoting apoptosis through mitochondrial pathway. 17. White dwarf binary modulation can help stochastic gravitational wave background search.

- Abstract
For the stochastic gravitational wave backgrounds (SGWBs) search centred at the milli-Hz band, the galactic foreground produced by white dwarf binaries (WDBs) within the Milky Way contaminates the extra-galactic signal severely. Because of the anisotropic distribution pattern of the WDBs and the motion of the space-borne gravitational wave interferometer constellation, the time-domain data stream will show an annual modulation. This property is fundamentally different from those of the SGWBs. In this article, we propose a new filtering method for the data vector based on the annual modulation phenomenon. We apply the resulted inverse variance filter to the LISA Data Challenge. The result shows that for the weaker SGWB signal, such as energy density Ω
astro = 1 × 10−12 , the filtering method can enhance the posterior distribution peak prominently. For the stronger signal, such as Ωastro = 3 × 10−12 , the method can improve the Bayesian evidence from “substantial” to “strong” against null hypotheses. - Abstract
The electric field-assisted (EFA) heat treatment is proposed to solve the problem of excessive grain growth of β phase during the heat treatment of ZrTiAlV alloy. The effect of the EFA heat treatment process on the phase transformation and microstructure of the as-forged and severe plastic deformation (SPD) alloys are studied. For the as-forged 47Zr alloy, the pure β phase can be obtained by holding at 700 °C for 5 min. After SPD, the increase in defect concentration in the alloy reduces the effectiveness of the EFA heat treatment, and required an extension of the holding time to 15 min to obtain a pure β phase. Furthermore, a new mechanism for the transition of needle-like α phase to β phase is proposed. Dislocations in the β-phase matrix shear the needle-like α phase, resulting in the formation of a large number of stacking faults inside it, which are then transformed into the β phase through atomic plane slippage. The mechanism of the phase separation of polymer blends in the presence of a static electric field was investigated using the molecular dynamics with a modified coarse-grained polarizable model. Two electric field effects, i.e., the promotion and hindrance of the phase separation, were found in the system of the upper critical solution temperature (UCST) by varying the dielectric properties of polymer components. Our simulation directly demonstrated that the electric field effects originate from the energy changes of intramolecular unit polarization, intermolecular van der Waals (VDW) and Coulomb interactions induced by the external electric field. The relationship of the energy change and the dielectric constant for the system under electric field is obtained. The interface orientation under electric field was found to happen even before the transition point of phase separation. By assuming that the relative order parameter of the phase separation undergoes the same fluctuations for a system with and without an electric field, the transition temperature under an electric field can be predicted, and quantitative consistency with simulations is obtained. This paper proposed a hybrid system that combined dielectric barrier discharge plasma with catalysis (DPC) for toluene degradation. To improve the performance of DPC, photocatalysts TiO
2 were doped by Mn and Fe, respectively. All prepared photocatalysts were characterized using UV-Visual DRS., SEM, XPS, BET, and XRD. The effects of the doping ratio, AC frequency, electric field intensity, gas flow rate, and initial concentration on toluene degradation efficiency, ozone decomposition capacity, and COx selectivity have been investigated. The best doping ratios of Mn and Fe were both 1.0 at%. The increase of electric field intensity in the range of 6.9–10.3 kV/cm could favor the synergism for DPC significantly, but the ascending of AC Frequency failed to do that. Fe-DPC showed slightly better performance than Mn-DPC in degradation efficiency and COx selectivity, while Mn-DPC was ahead of Fe-DPC for the ozone decomposition. Mn-DPC and Fe-DPC both could maintain the high toluene degradation efficiency, when gas flow rate and initial concentration increase from 2.5 to10.1 cm/s and from 700 to 2300 mg/cm3 , respectively. - Abstract
Gliomas are the most common central nervous system tumours; they are highly aggressive and have a poor prognosis. RGS16 belongs to the regulator of G-protein signalling (RGS) protein family, which plays an important role in promoting various cancers, such as breast cancer, pancreatic cancer, and colorectal cancer. Moreover, previous studies confirmed that let-7c-5p, a well-known microRNA, can act as a tumour suppressor to regulate the progression of various tumours by inhibiting the expression of its target genes. However, whether RGS16 can promote the progression of glioma and whether it is regulated by miR let-7c-5p are still unknown. Here, we confirmed that RGS16 is upregulated in glioma tissues and that high expression of RGS16 is associated with poor survival. Ectopic deletion of RGS16 significantly suppressed glioma cell proliferation and migration both in vitro and in vivo. Moreover, RGS16 was validated as a direct target gene of miR let-7c-5p. The overexpression of miR let-7c-5p obviously downregulated the expression of RGS16, and knocking down miR let-7c-5p had the opposite effect. Thus, we suggest that the suppression of RGS16 by miR let-7c-5p can promote glioma progression and may serve as a potential prognostic biomarker and therapeutic target in glioma. - Abstract
Background: The microenvironment of hypoxia is an important factor contributing to the development of glioblastoma (GBM). MicroRNA-588 and its potential target Roundabout-directed receptor 1 (ROBO1) have been reported to promote tumor invasion and proliferation in diseases such as gastric, pancreatic and hepatocellular carcinoma, while their function in GBM and response to hypoxic states remain elusive. Methods: A microarray was leveraged to identify differentially expressed microRNAs in U251 glioma cells cultured under normoxic and hypoxic conditions. The expression of miR-588 was assessed using quantitative real-time PCR (qRT‒PCR). Gain- and loss-of-function studies were used to evaluate the role of miR-588 under hypoxic and normoxic conditions. Cell invasion, migration, proliferation, and vasculogenic mimicry (VM) formation experiments were performed. The relationship between miR-588 and ROBO1 was confirmed using western blot and luciferase reporter assays. Intracranial xenograft tumor mouse models were used to study the function of miR-588 in vivo. Results: The expression of miR-588 was significantly upregulated in hypoxic glioma cells relative to normoxic glioma cells. miR-588 inhibited the invasive, migratory and VM-forming abilities of glioma cells in vitro and in vivo. Mechanistically, roundabout guidance receptor 1 (ROBO1) is a direct, functionally relevant target of miR-588 in glioma. ROBO1 knockdown suppressed the expression of matrix metallopeptidase 2 (MMP2) and matrix metallopeptidase 9 (MMP9), thereby inhibiting the invasive, migratory and VM-forming abilities of glioma. Conclusions: MiR-588 regulated the behaviors of hypoxic glioma cells by targeting ROBO1. miR-588 can be used as a prognostic marker for glioma and has potential implications in glioma gene therapy. In this study, an efficient nano-zero-valent iron supported by alkali-modified sludge biochar (nZVI@NaOH-SBC) composites were synthesized by a liquid-phase reduction method for Cu
2+ removal. The preparation conditions (sodium hydroxide concentration, pyrolysis temperature, solid–liquid ratio, and modification time) of sludge biochar (SBC) were optimized by orthogonal experiments. The results showed that the best SBC was prepared under the conditions of sodium hydroxide concentration of 3 mol/L, pyrolysis temperature of 500 °C, a solid–liquid ratio of 1:20, and modification time of 24 h, and a maximum adsorption capacity was 183.22 mg/g, which is 2.43 times that of the pristine sludge biochar. The adsorption behavior was in accordance with a pseudo-second-order kinetic model, indicating that the removal of Cu2+ was mainly affected by chemisorption. The adsorption isotherm conformed to the Langmuir model, and the maximum adsorption capacity of Cu2+ was 358.42 mg/g at pH = 5, and 25 °C. Reusability tests confirmed the good reusability and stability of the material. Batch experiments and characterization reveal that the mechanism of Cu removal by nZVI@NaOH-SBC composites involves multiple processes: adsorption, precipitation, electrostatic attraction, and reduction. This study proposes a new biochemical adsorbent, which provides a good choice for the treatment of copper-containing wastewater and the resource utilization of sludge. 25. CT-based clinico-radiological nomograms for prognosis prediction in patients with intrahepatic mass-forming cholangiocarcinoma: a multi-institutional study.

Objectives: To establish prognostic nomograms based on CT imaging features for predicting the prognosis in patients with intrahepatic mass-forming cholangiocarcinoma (IMCC) before and after surgery. Methods: Two models were established for overall survival (OS) prediction in a training set (179 IMCC patients underwent surgery at institution 1 from 2009 to 2019): imaging-based nomogram included imaging features and clinical characteristics acquired before surgery; postoperative nomogram included imaging-based score, equal to the linear predictor of the imaging-based nomogram, and pathological parameters. Both prognostic nomograms were validated in an independent external dataset (103 IMCC patients received surgical treatment at two independent institutions from 2009 to 2019). Predictive performance and discrimination were evaluated and compared with the common prognostic models. Results: The imaging-based nomogram was developed according to preoperative serum carbohydrate antigen 19-9 and four imaging features including multiple nodules, arterial enhancement pattern, CT-reported lymph node (LN) metastasis, and capsular retraction; the postoperative nomogram was built based on the imaging-based score and three pathological parameters including tumor differentiation grade, capsular invasion, and LN status. Both nomograms presented improved prognostic performance and discrimination (concordance index, 0.770–0.812; integrated Brier score, 0.120–0.138) compared with the common prognostic models in the training and external validation datasets. Besides, the nomograms stratified IMCC patients into two risk strata for OS. Conclusions: Nomograms based on CT imaging features can provide accurate individual survival prediction for IMCC patients before and after surgery, which may help to improve personalized treatment. Purpose: Spiritual well-being had a protective effect on quality of life in cancer, due to the cultural, regional, and custom differences; it was rarely been discussed between cancer and chronic diseases in Chongqing, China. We aimed at comparing the level of spirituality in two groups and discussing its factors of subjects with cancer at county regions. Methods: A cross-sectional questionnaire survey was distributed to 630 inpatients who received treatment between January and December 2020 in Chongqing University Three Gorges Hospital. In addition to basic demographic data, spirituality was measured using the Chinese version of Functional Assessment of Chronic Illness Therapy-Spiritual Well-Being (FACIT-Sp-12). The mean, standard deviation, independent t-tests, ANOVA, and multiple regression were used for statistical description and analysis. Results: Significant differences were found between cancer and chronic diseases in total scores of FACIT-Sp-12 and each domain (P < 0.05). The meaning, peace, faith, and total scores in cancer were 11.21 ± 3.38, 10.66 ± 4.46, 11.43 ± 3.54, and 33.3 ± 10.35, respectively, which were lower than chronic diseases (13.00 ± 3.21, 12.95 ± 4.76, 12.66 ± 3.64, 38.61 ± 10.88, respectively). The spiritual well-being had significant differences in gender, character, and emotional with spouse for cancer (P < 0.05). The male and extravert character were significantly associated with a greater spiritual well-being. Conclusion: The study shows a medium level of spiritual well-being in cancer, which stands the population with lower economic and education in county regions. It suggests that under the current nursing mode, we should provide specifically spiritual care to the female, introvert, and those with poor relationship with spouses and create a harmonious doctor-patient environment to improve the spiritual well-being. 27. Real-World super-resolution under the guidance of optimal transport.

- Abstract
In the real world, lacking paired training data makes image super-resolution (SR) be a tricky unsupervised task. Existing methods are mainly train models on synthetic datasets and achieve the tradeoff between detail restoration and noise artifact suppression based on a priori knowledge, which indicate it cannot be optimal in both aspects. To solve this problem, we propose OTSR, a single image super-resolution method based on optimal transport theory. OTSR aims to find the optimal solution to the ill-posed SR problem, so that the model can restore high-frequency detail accurately and also suppress noise and artifacts well. Our method consists of three stages: real-world images degradation estimation, LR images generation and model optimization based on quadratic Wasserstein distance. Through the first two stages, the problem of no paired image is solved. In the third stage, under the guidance of optimal transport theory, the optimal mapping from LR to HR image space is learned. Extensive experiments show that our method outperforms the state-of-the-art methods in terms of both detail repair and noise artifact suppression. The source code is available at . Objectives: Acute respiratory failure (ARF) is a common medical complication in patients with cervical traumatic spinal cord injury (TSCI). To identify independent predictors for ARF onset in patients who underwent cervical TSCI without premorbid respiratory diseases and to apply appropriate medical supports based on accurate prediction, a nomogram relating admission clinical information was developed for predicting ARF during acute care period. Methods: We retrospectively reviewed clinical profiles of patients who suffered cervical TSCI and were emergently admitted to Qingdao Municipal Hospital from 2014 to 2020 as the training cohort. Univariate analysis was performed using admission clinical variables to estimate associated factors and a nomogram for predicting ARF occurrence was generated based on the independent predictors from multivariate logistic regression analysis. This nomogram was assessed by concordance index for discrimination and calibration curve with internal-validated bootstrap strategy. Receiver operating characteristic curve was conducted to compare the predictive accuracy between the nomogram and the traditional gold standard, which combines neuroimaging and neurological measurements by using area under the receiver operating characteristic curve (AUC). An additional 56-patient cohort from another medical center was retrospectively reviewed as the test cohort for external validation of the nomogram. Results: 162 patients were eligible for this study and were included in the training cohort, among which 25 individuals developed ARF and were recorded to endure more complications. Despite the aggressive treatments and prolonged intensive care unit cares, 14 patients insulted with ARF died. Injury level, American Spinal Injury Association Impairment Scale (AIS) grade, admission hemoglobin (Hb), platelet to lymphocyte ratio, and neutrophil percentage to albumin ratio (NPAR) were independently associated with ARF onset. The concordance index of the nomogram incorporating these predictors was 0.933 in the training cohort and 0.955 in the test cohort, although both calibrations were good. The AUC of the nomogram was equal to concordance index, which presented better predictive accuracy compared with previous measurements using neuroimaging and AIS grade (AUC 0.933 versus 0.821, Delong's test p < 0.001). Similar significant results were also found in the test cohort (AUC 0.955 versus 0.765, Delong's test p = 0.034). In addition, this nomogram was translated to a Web-based calculator that could generate individual probability for ARF in a visualized form. Conclusions: The nomogram incorporating the injury level, AIS grade, admission Hb, platelet to lymphocyte ratio, and NPAR is a promising model to predict ARF in patients with cervical TSCI who are absent from previous respiratory dysfunction. This nomogram can be offered to clinicians to stratify patients, strengthen evidence-based decision-making, and apply appropriate individualized treatment in the field of acute clinical care. Objective: A systematic review of animal and human studies was conducted on genetically modified (GM) food consumption to assess its safety in terms of adverse effects/events to inform public concerns and future research. Methods: Seven electronic databases were searched from January 1st 1983 till July 11th 2020 for in vivo, animal and human studies on the incidence of adverse effects/events of GM products consumption. Two authors independently identified eligible studies, assessed the study quality, and extracted data on the name of the periodical, author and affiliation, literature type, the theme of the study, publication year, funding, sample size, target population characteristics, type of the intervention/exposure, outcomes and outcome measures, and details of adverse effects/events. We used the Chi-square test to compare the adverse event reporting rates in articles funded by industry funding, government funding or unfunded articles. Results: One crossover trial in humans and 203 animal studies from 179 articles met the inclusion criteria. The study quality was all assessed as being unclear or having a high risk of bias. Minor illnesses were reported in the human trial. Among the 204 studies, 59.46% of adverse events (22 of 37) were serious adverse events from 16 animal studies (7.84%). No significant differences were found in the adverse event reporting rates either between industry and government funding (χ
2 = 2.286, P = 0.131), industry and non-industry funding (χ2 = 1.761, P = 0.185) or funded and non-funded articles (χ2 = 0.491, P = 0.483). We finally identified 21 GM food-related adverse events involving 7 GM events (NK603 × MON810 maize, GTS 40-3-2 soybean, NK603 maize, MON863 maize, MON810 maize, MON863 × MON810 × NK603 maize and GM Shanyou 63 rice), which had all been on regulatory approval in some countries/regions. Conclusion: Serious adverse events of GM consumption include mortality, tumour or cancer, significant low fertility, decreased learning and reaction abilities, and some organ abnormalities. Further clinical trials and long-term cohort studies in human populations, especially on GM food-related adverse events and the corresponding GM events, are still warranted. Objectives: Uric acid(UA) is related with cardiovascular disease, but the association of UA variability with all-cause mortality is rarely known. This study aimed to investigate the relationship between UA variability and all-cause mortality in Kailuan cohort study in northern China. Design: Cohort study. Setting: Kailuan community hospitals in Tangshan, Hebei province, Northern China. Participants: A total of 55717 participants from Kailuan Study were enrolled, and our study followed up biennially from 2006 to 2010. Measurements: Clinical records of the participants enrolled were analyzed. UA variation independent of mean (UAVIM) values were calculated and all the participants were quartile grouped into four groups as: Q1(UAVIM<0.68), Q2(0.68≤UAVIM<1.10), Q3(1.10≤UAVIM<1.67) and Q4(UAVIM≥1.67). The endpoint event was all-cause death. Cox regression model was performed to evaluate the hazard ratios(HRs) of all-cause mortality based on UAVIM groups. Results: During a median follow-up of 6.83 years, 2926 deaths occurred. The accumulated mortality rates were 4.6%, 4.8%, 5.4% and 6.1% in group Q1, Q2, Q3 and Q4 respectively. When adjusted potential confounders, the highest risk for all-cause mortality was in group Q4 and the adjusted HRs and 95% confidence intervals(CIs) of group Q2–Q4 for all-cause death were 1.044(0.937, 1.164), 1.182(1.064, 1.314) and 1.353(1.220, 1.501) compared with group Q1, respectively. Further analysis showed that the risk for all-cause death increased as UAVIM value increased. Sensitive analysis still showed the similar results when excluding participants with hyperuricemia or severe chronic kidney diseases. Sub-group analysis by age, gender, BMI or hypertension history also indicated analogous results. Conclusion: Elevated UAVIM was related with increased all-cause mortality and UAVIM was an independent risk factor for all-cause mortality in the community cohort study. [ABSTRACT FROM AUTHOR]

- Abstract
We recently identified oncologic miR-182 as a new regulator of pulmonary artery hypertension (PAH) that targets myeloid-associated differentiation marker (Myadm), which is expressed in bone marrow stem cells and multipotent progenitors. Both miR-182 and Myadm are expressed in the cardiopulmonary system and correlated with the balance between the bone morphogenetic protein (BMP) and the transforming growth factor (TGF)-β signalling pathways, which are disturbed in PAH. We hypothesize that miR-182/Myadm are involved in BMP-TGF-β-signalling way in PAH. Hypoxia triggered pathological progression in cardiopulmonary PAH in vivo and in vitro; these changes were accompanied by strongly dowregulated BMP/SMAD1/5/8 expression and enhanced TGF-β/SMAD2/3 signalling pathway, favouring SMAD4/SMAD2 transcript formation and inhibiting the PAH negative regulator Id1 expression. miR-182 gain-of-function significantly inhibited the pathological progression in hypoxia-induced PAH (HPH) in vivo and in vitro, with a restoration of the balance in BMP-TGF-β signalling pathway. This recovery was abrogated by overexpression of Myadm. Conversely, loss-of-function of miR-182 increased the pathological progression of HPH followed by severe disturbance of BMP and TGF-β signal transduction and reduced Id1 expression, which was restored by Myadm knockdown. We also showed that the miR-182/Myadm relate BMP-TGF-β pathway is associated with NOS3/NO/cGMP via the crosstalk between endothelial cells and smooth muscle cells. Our findings further support the therapeutic significance of miR-182/Myadm in PAH via the balance of BMP- and TGF-β-associated mechanisms. [ABSTRACT FROM AUTHOR]

This study was to develop a radiomics nomogram mainly using wavelet features for identifying malignant and benign early-stage lung nodules for high-risk screening. A total of 116 patients with early-stage solitary pulmonary nodules (SPNs) (≤ 3 cm) were divided into a training set (N = 70) and a validation set (N = 46). Radiomics features were extracted from plain LDCT images of each patient. A radiomics signature was then constructed with the LASSO with the training set. Combined with independent risk factors, a radiomics nomogram was built with a multivariate logistic regression model. This radiomics signature, consisting of one original and nine wavelet features, achieved favorable predictive efficacy than Mayo Clinic Model. The radiomics nomogram with radiomics signature and age also showed good calibration and discrimination in the training set (AUC 0.9406; 95% CI 0.8831–0.9982) and the validation set (AUC 0.8454; 95% CI 0.7196–0.9712). The decision curve indicated the clinical usefulness of our nomogram. The presented radiomics nomogram shows favorable predictive accuracy for identifying malignant and benign lung nodules in early-stage patients and is much better than the Mayo Clinic Model. 37. Integrating Machine Learning and Tumor Immune Signature to Predict Oncologic Outcomes in Resected Biliary Tract Cancer.

Background: Improved methods are needed to predict outcomes in biliary tract cancers (BTCs). We aimed to build an immune-related signature and establish holistic models using machine learning. Methods: Samples were from 305 BTC patients treated with curative-intent resection, divided into derivation and validation cohorts in a two-to-one ratio. Spatial resolution of T cell infiltration and PD-1/PD-L1 expression was assessed by immunohistochemistry. An immune signature was constructed using classification and regression tree. Machine learning was applied to develop prediction models for disease-specific survival (DSS) and recurrence-free survival (RFS). Results: The immune signature composed of CD3
+ , CD8+ , and PD-1+ cell densities and PD-L1 expression within tumor epithelium significantly stratified patients into three clusters, with median DSS varying from 11.7 to 80.8 months and median RFS varying from 6.2 to 62.0 months. Gradient boosting machines (GBM) outperformed rival machine-learning algorithms and selected the same 11 covariates for DSS and RFS prediction: immune signature, tumor site, age, bilirubin, albumin, carcinoembryonic antigen, cancer antigen 19-9, tumor size, tumor differentiation, resection margin, and nodal metastasis. The clinical-immune GBM models accurately predicted DSS and RFS, with respective concordance index of 0.776–0.816 and 0.741–0.781. GBM models showed significantly improved performance compared with tumor-node-metastasis staging system. Conclusions: The immune signature promises to stratify prognosis and allocate treatment in resected BTC. Background: Hyperuricemia is associated with cardiovascular mortality, but the association of the age at hyperuricemia onset with cardiovascular disease (CVD) and mortality is still unclear. Objective: The purpose of this study was to examine the associations of hyperuricemia onset age with CVD and all-cause mortality. Methods: A total of 82,219 participants free of hyperuricemia and CVD from 2006 to 2015 in the Kailuan study were included. The analysis cohort comprised 18,311 new-onset hyperuricemia patients and controls matched for age and sex from the general population. Adjusted associations were estimated using Cox models for CVD and all-cause mortality across a range of ages. Results: There were 1,021 incident cases of CVD (including 215 myocardial infarctions, 814 strokes) and 1459 deaths during an average of 5.2 years of follow-up. Patients with hyperuricemia onset at an age < 45 years had the highest hazard ratios (HRs) (1.78 (1.14–2.78) for CVD and 1.64 (1.04–2.61) for all-cause mortality relative to controls). The HRs of CVD and all-cause mortality were 1.32 (1.05–1.65) and 1.40 (1.08–1.81) for the 45–54 years age group, 1.23 (0.97–1.56) and 1.37 (1.11 to 1.72) for the 55–64 years age group, and 1.10 (0.88–1.39) and 0.88 (0.76–1.01) for the ≥ 65 years age group, respectively. Conclusions: The age at hyperuricemia onset was identified as an important predictor of CVD and all-cause mortality risk, and the prediction was more powerful in those with a younger age of hyperuricemia onset. Objectives: The aim of this study was to develop and validate a radiomics signature for predicting survival and chemotherapeutic benefits of patients with lower-grade gliomas (LGG). Methods: Radiomics features were extracted from precontrast axial fluid-attenuated inversion recovery (FLAIR) and contrast-enhanced axial T-1 weighted (CE-T1-w) sequence. Lasso Cox regression model was used for feature selection and radiomics signature building. The radiomics signature was developed in a primary cohort that consisted of 149 LGG patients and was then validated on an entirely new validation cohort that contained 66 LGG patients. A radiomics nomogram for the prediction of OS was established by adding the radiomics to clinicopathologic nomogram which developed with clinical data. Results: A radiomics signature derived from joint CE-T1-w and FLAIR images showed better prognostic performance (C-index, 0.798) than signatures derived from CE-T1-w (C-index, 0.744) or FLAIR (C-index, 0.736) sequences alone. Multivariable Cox regression revealed that the radiomics signature was an independent prognostic factor. One radiomics nomogram integrated the radiomics signature from joint CE-T1-w and FLAIR sequences with the clinicopathologic nomogram outperformed the clinicopathologic nomogram based on clinicopathologic data alone in predicting OS of LGG (C-index, 0.821 vs. 0.692; p < 0.001). Further analysis revealed that patients with higher radiomics signature were prone to benefit from chemotherapy. Conclusions: The radiomics signature was independent with clinicopathologic data and was a noninvasive pretreatment predictor for LGG patients' survival. Moreover, it could predict which patients with LGG benefit from chemotherapy. 40. Oxidation behavior of Ni-based superalloy GH738 in static air between 800 and 1000 °C.

- Abstract
The oxidation behavior of a nickel-based superalloy GH738 was studied by isothermal oxidation tests in still air at different temperatures, with exposure time up to 100 h. Oxidation-kinetic curves were plotted using the mass gain method. The surface and cross-sectional morphologies were observed by scanning electron microscopy (SEM). A composition analysis and an oxidation-product identification were conducted using energy-dispersive X-ray spectroscopy (EDS) and an X-ray diffraction (XRD), respectively. The results showed that GH738 exhibited parabolic oxidation-kinetic curves, with stable parabolic-rate constants at each temperatures. The activation energy of oxide growth was calculated to be 329.6 kJ·mol
−1 . Cr2 O3 (chromia) was the external oxidation product at 800 °C. A TiO2 –Cr2 O3 double-layer structure was formed at 900 °C. The position of TiO2 changed from the oxide–metal interface to the air–oxide interface by the diffusion of Ti atoms in chromia during the oxidation time at this temperature. Spallation was observed in the Cr0.12 Ti0.78 O1.74 –Cr2 O3 multi-layer oxide of at 1000 °C, which increased the oxidation rate. For all the tests, the main internal oxide was always Al2 O3 . The entire GH738 oxidation process was interpreted by the competitive diffusion of elements (Cr, Ti, Ni, etc.) in metal matrix and chromia, while a schematic diagram of oxidation process was proposed. Key message: MdHAL3 has PPCDC activity and is involved in the salt tolerance of autotetraploid apple. Apple (Malus × domestica) is the most widely planted fruit tree species worldwide. However, the growth and development of apple have been increasingly affected by abiotic stress, such as high salinity. In our previous study, RNA sequencing (RNA-seq) analysis revealed that the expression level of the MdHAL3 gene was significantly upregulated in the autotetraploid apple cultivar Hanfu. In the present study, we first isolated HAL3, whose product was shown to exert 4′-phosphopantothenoylcysteine decarboxylase (PPCDC) activity, from apple. MdHAL3 was expressed in all organs of apple, and its expression was rapidly induced by salt stress. The MdHAL3 protein was localized to the cytomembrane and cytoplasm. Five MdHAL3 overexpression (OE) lines and five MdHAL3-RNAi apple lines were obtained. We found that MdHAL3 enhanced the salt stress tolerance of apple and that the OE plants rooted more easily than the wild-type (WT) plants. The coenzyme A (CoA) content in the leaves of the OE plants was greater than that in the leaves of the WT plants, and the CoA content in the MdHAL3-RNAi plants was lower than that in the WT plants. This article reports microstructural characterization of as-cast and as-extruded Zn-Fe alloys and their mechanical and corrosion properties. Specifically, 1 wt.%, 2 wt.% and 3 wt.% Fe was added into the Zn matrix to form respective Zn-Fe alloys using casting and indirect hot extrusion processes. As the Fe content increased within the range of 1–3 wt.%, the mechanical properties of as-cast and as-extruded Zn-Fe alloys improved. The as-extruded Zn-Fe alloys showed superior mechanical properties of hardness, ultimate tensile strength (UTS) and elongation to failure (EL) to the as-cast Zn-Fe alloys of the same composition. Specifically, the UTS for as-extruded Zn-Fe alloys was 138–170 MPa, whereas the UTS for as-cast Zn-Fe alloys was 89–125 MPa. The corrosion properties of the as-extruded Zn-Fe alloys were investigated using the electrochemical and immersion methods. 43. Surgical Strategies for the Treatment of Bismuth Type I and II Hilar Cholangiocarcinoma: Bile Duct Resection with or Without Hepatectomy?

- Abstract
Background: The role of hepatic resection in the treatment of type I and II hilar cholangiocarcinoma (HCCA) remains controversial. In the present study, we aimed to identify whether hepatic resection was necessary for type I and II HCCA. Methods: A total of 23 patients classified as type I and II HCCA undergoing surgical resection were included in this study. The patients were divided into two groups: bile duct resection (BDR) group (n = 15) and hepatic resection (HR) group (n = 8). Systematic review and meta-analysis were performed to compare the R0 resection and long-term survival between BDR and HR for Bismuth type I and II HCCA. A total of 7 studies with 260 cases were included in this meta-analysis. Results: In our cohort, the R0 resection rate was 73.3% in BDR group and 87.5% in HR group. The HR group had a higher number of postoperative complications than the BDR group (P = 0.002). There was no difference in long-term survival (P = 0.544) and recurrence (P = 0.846) between BDR and HR in Bismuth type I and II HCCA. The meta-analysis showed that HR was associated with better R0 resection rate (RR 4.45, 95% CI 2.34–8.48) and overall survival (HR 2.15, 95% CI 1.34–3.44) compared with BDR group. There was no publication bias and undue influence of any single study. Conclusions: The meta-analysis showed that HR was associated with better R0 resection rate and overall survival compared with BDR for type I and II HCCA patients. More aggressive surgical strategies should be increasingly considered for the treatment of type I and II HCCA patients. [ABSTRACT FROM AUTHOR]

Contact-electrification is a universal effect for all existing materials, but it still lacks a quantitative materials database to systematically understand its scientific mechanisms. Using an established measurement method, this study quantifies the triboelectric charge densities of nearly 30 inorganic nonmetallic materials. From the matrix of their triboelectric charge densities and band structures, it is found that the triboelectric output is strongly related to the work functions of the materials. Our study verifies that contact-electrification is an electronic quantum transition effect under ambient conditions. The basic driving force for contact-electrification is that electrons seek to fill the lowest available states once two materials are forced to reach atomically close distance so that electron transitions are possible through strongly overlapping electron wave functions. We hope that the quantified series could serve as a textbook standard and a fundamental database for scientific research, practical manufacturing, and engineering. The mechanism of contact electrification remains a topic of debate. Here, the authors present a quantitative database of the triboelectric charge density and band structure of many inorganic materials, verifying that contact electrification between solids is an electron quantum transition effect. The results show that the roadway excavation can be canceled and that the L-type face end structure near the working face end can be formed using low-pressure mining technology without pre-driving roadway, which fundamentally eliminates the impact of advance stress on the deformation of the surrounding rock near the face end and substantially improves the stability of the surrounding rock in front of the face end. Meanwhile, on the rear side of the face end, the stress transfer path of the part roof is cut off by means of roof cutting, and a wider corner effect area is formed. Therefore, the impact of roof caving behind the working face on the deformation of surrounding rock near the L-type face end is weakened, thus significantly improving the stability of the surrounding rock. According to the engineering test, there is a certain range of the low-pressure zone of the L-type face end along the direction of the working face and the axial direction of the roadway. In the former low-pressure zone, the periodic weighting of the working face is not obvious, and the dynamic factor of the support is small; in the latter low-pressure zone, the roof-to-floor deformation of the roadway and support structure stress are both very small as well. The test results are basically consistent with the results of the theoretical analysis. The above results prove that the idea and method of forming the L-type face end using low-pressure mining technology without pre-driving roadway, thus controlling the formation of the surrounding rock near the working face end, is completely effective. Background: PLEKHG5, a Rho-specific guanine-nucleotide exchange factor, is involved in tumor cell migration, invasion and angiogenic potential. In this study, the expression pattern, prognostic value and function of PLEKHG5 in gliomas were investigated. Methods: Immunohistochemistry was used to determine the expression pattern of PLEKHG5 in 61 glioma patients after curative resection. Statistical analysis was performed to evaluate the diagnostic and prognostic significance of PLEKHG5. Gene ontology (GO) analysis, Kyoto encyclopedia of genes and genomes (KEGG) pathway analysis and Gene set enrichment analysis (GSEA) were used to predict potential functions of PLEKHG5. Migration assay and western blot analysis determined PLEKHG5 function in glioma migration and invasion. Results: Increased PLEKHG5 expression levels were associated with higher glioma grades (P < 0.05). In addition, glioblastomas multiforme have higher ratio and stronger intensity of PLEKHG5 expression compared with low-grade gliomas. High expression level of PLEKHG5 indicated poorer prognosis and shorter survival time in all glioma patients (P < 0.001). GO analysis, KEGG pathway analysis and GSEA analysis suggested that PLEKHG5 was involved in glioma migration, invasion and epithelial–mesenchymal transition. Migration assay and western blot analysis revealed PLEKHG5 promoted glioma migration and invasion. Conclusion: Our results demonstrated PLEKHG5 could be used as a novel prognostic biomarker and anti-tumor target for glioma patients. [ABSTRACT FROM AUTHOR]
